A numerical study to evaluate the reliability of 3D steel buildings with moment-resisting frames (MRFs) at the perimeter and gravity frames (GFs) at the interior under the action of seismic loading, is carried out. To this aim, the median values (MV) of the maximum drifts (MD) and fragility curves are obtained. The reliability, expressed as the mean annual rate of exceeding (υ) particular values of MD, is calculated by using the probabilistic seismic hazard analysis format. Three models, representing steel buildings of low-, mid- and high-rise are considered. The interior connections are assumed to be, first perfectly pinned (PP) as considered in practical design, and then as semi-rigid (SR). The Beam-Line Theory together with The Model of Richard are used to represent the SR connections. Results indicate that the MV demands and the seismic fragility can be significantly reduced when the stiffness and dissipation of energy at interior connections are considered; reductions larger than 40% and 60%, respectively, are observed in many of the cases. The structural reliability is also significantly improved; reductions close to 20% in the expected maximum drift are observed in many cases. It is concluded that the effects of the stiffness and energy dissipation of the connections of GFs on the seismic response (in terms of MV), on the fragility curves, and on the mean annual rate of exceedance (υ), are significant so they should not be ignored. Even for shear connections whose contribution in practical design is totally ignored, the effect is considerable.

4. Enabling Fast and Accurate Record Linkage of Large-Scale Health-Related Administrative Databases Through a DNA-Encoded Approach

ObjectivePublic health research frequently requires the integration of information from different data sources. However, errors in the records and the high computational costs involved make linking large administrative databases using record linkage (RL) methodologies a major challenge. We present Tucuxi-BLAST, a versatile tool for probabilistic RL that utilizes a DNA-encoded approach to encrypt, analyze and link massive administrative databases. Materials and MethodsTucuxi-BLAST encodes the identification records into DNA. BLASTn algorithm is then used to align the sequences between databases. We tested and benchmarked on a simulated database containing records for 300 million individuals and also on four large administrative databases containing real data on Brazilian patients. ResultsOur method was able to overcome misspellings and typographical errors in administrative databases. In processing the RL of the largest simulated dataset (200k records), the state-of-the art method took 5 days and 7 hours to perform the RL, while Tucuxi-BLAST only took 23 hours. When compared with five existing RL tools applied to a gold-standard dataset from real health-related databases, Tucuxi-BLAST had the highest accuracy and speed. DiscussionBy repurposing genomic tools, researchers are able to perform subject tracing across multiple large epidemiological databases using a regular laptop. ConclusionTucuxi-BLAST can improve data-driven medical research and provide a fast and accurate way to link individual information across several administrative databases.

The goals of this study were to investigate (a) the localisation of Na+, K+-ATPase in ram sperm; (b) cryopreservation like-changes on median fluorescence intensity (MFI) and the expression of Na+, K+-ATPase on the sperm surface (labeled sperm); and (c) the influence of Na+, K+-ATPase inhibition on sperm kinematics. Exp. 1: pooled semen samples (n = 7) from four rams were diluted and frozen. The following sperm features were evaluated in fresh-diluted and post-thawed samples: MFI and expression of Na+, K+-ATPase (Bodipy® FL-Ouabain), plasma and acrosomal membrane integrity (IP; FITC-PNA), sperm viability and plasma membrane stability (Yo-Pro and Merocyanine), as well as sperm kinematics (CASA). Exp. 2: pooled semen samples (n = 5) from four rams were diluted and incubated in medium treated with or without Na+, K+-ATPase inhibitor (ouabain, 10−4 M). We investigated whether the recombinant leptin (1, 10, 100 ng/mL) influences the meiotic maturation of goat oocytes, whether the MAPK and JAK2/STAT3 pathways mediate the effects of leptin during in-vitro maturation, and whether leptin differently affects the abundance of mRNAs relevant to leptin signal transduction and apoptosis in oocytes and cumulus cells. The addition of leptin to the maturation medium positively affected the number of oocytes that completed nuclear maturation. Nuclear oocyte maturation stimulated by leptin was significantly impaired when we added the specific inhibitors of MAPK (U0126) and JAK2/STAT3 (AG490) to the maturation medium. The addition of leptin (10 ng/mL) during maturation did not affect the expression of AMPKα1, PPARα, Caspase 3, and BCL2 genes in oocytes or cumulus cells. The PPARγ and BAX mRNA abundances were significantly reduced in cumulus cells in the leptin group compared to the control group. Our results demonstrate that supplementation of the in-vitro maturation medium with leptin significantly improves nuclear maturation and reveal the important role of the MAPK and JAK2/STAT3 signaling pathways in establishing the leptin-mediated nuclear maturation of goat oocytes. Moreover, leptin treatment affects PPARγ and BAX gene expression in cumulus cells.

The corrosion process due to the erosion of chloride ions on reinforced concrete (RC) buildings is analyzed in this paper. The main objective of this study is to determine the influence of the corrosion on structural reliability of reinforced concrete buildings under earthquakes. It is assumed that the buildings are located in the city of Acapulco Guerrero in Mexico, and a comparison of the structural reliability when the resistance of the structural members of the buildings under earthquakes is affected by corrosion and when not is computed. In addition, the influence of concrete cover is considered. For the objective of this study, the buildings are modeled as 3D RC frames and they are subjected to several earthquake ground motion records. Furthermore, the analyses are performed by the Monte Carlo simulation technique and stochastic modelling of corrosion initiation, corrosion propagation and corrosion cracking to estimate the steel corrosion. It is shown that omitting the effect of corrosion in reinforced concrete structures can lead to erroneous estimations of the structural reliability of buildings under earthquakes.

In this study, a simplified procedure to predict the maximum inter-story drift demands of mid-rise framed buildings in terms of a spectral-shape-based ground motion intensity measure is proposed. For this aim, the maximum inter-story drift of steel and reinforced concrete (R/C) frames with 4, 6, 8 and 10 stories subjected to several narrow-band ground motions is estimated as a function of the spectral acceleration at first mode of vibration of the structure Sa(T1), which is commonly used in earthquake engineering and seismology, and with the recently developed parameter related to the structural response known as INp. It is observed that the spectral-shape-based intensity measure INp is the parameter best related with the structural response of the selected frames under narrow-band motions. Therefore, a new approach is used to propose equations to compute maximum inter-story drift demands and seismic uncertainty of mid-rise frames as a function of INp. The equations can be used for the rapid seismic assessment and to obtain structural fragility curves of the steel and R/C frame models. The fragility curves obtained with the simplified approach are compared to those obtained with traditional procedure to compute structural fragility. It is observed that the proposed equations provide good accuracy to calculate fragility curves of steel and R/C structures under narrow-band earthquake ground motions in terms of the spectral-shape-based intensity measure INp.

In this study, a new generalized ground motion intensity measure is proposed. The new intensity measure named I B is able to increase the efficiency in the prediction of nonlinear behavior and higher modes effects of structures subjected to earthquake ground motions with different characteristics. The intensity measure is based on a recently proposed proxy of the spectral shape named N p , which has been successfully used in ground motion record selection for nonlinear dynamic analysis. Although the N p parameter was proposed by using the spectral shape in term of pseudo-acceleration, it will be observed that N p can be defined through other types of spectral shapes such as those obtained with velocity, displacement, input energy, inelastic parameters and so on. In addition, it is illustrated that several ground motion intensity measures are particular cases of the new generalized ground motion intensity measure here proposed, which is able to predict both nonlinear structural demands and higher modes effects on buildings under earthquake ground motions.

An evaluation of some structural parameters affecting the amplitude and height-wise distribution of peak and residual drift demands of reinforced concrete frame buildings modeled as 3D complex MDOF systems is presented. In addition, the influence of different ground motion selection methods on the uncertainty in the estimation of both peak and residual drifts is assessed. For this purpose, buildings with different characteristics were subjected to record sets selected: a) randomly, b) considering the magnitude and epicentral distance, and c) to match an objective spectral shape. Results indicate that parameters like the post-yielding stiffness ratio and the structural overstrength importantly affect the magnitude and height-wise distribution of residual drift demands on buildings. The estimation of residual drifts involves a higher level of uncertainty than that of peak drift demands. Selecting records based on a target spectral shape is very effective in reducing the uncertainty on the estimation of both peak and residual inter-story drift demands.
