Background Toxic effects of digoxin may occur with normal therapeutic serum level. However, the underlying mechanisms are not fully understood. Nuclear factor kappa-B (NF-kB) is an important transcription factor in most organ systems and is often implicated in the harmful effects of cardiac injury. NF-kB promotes inflammatory responses, mediates adverse cardiac remodeling and has a function correlation with calcium. The voltage-gated L-type calcium channel CaV1.2 mediates the influx of Ca+2 into the cell in response to membrane depolarization. Our aim was to characterize the role of NF-kB during digoxin toxicity and to assess its correlation with Cav 1.2 in healthy mice in vivo. Methods To address these questions, digoxin was administered in doses of 0.1, 1 or 5 mg/kg orally daily for seven days to the animals. Serum digoxin, serum calcium, atrial and ventricular calcium levels were measured. We, also, looked for NF-kB and CaV1.2 channel expression in cardiac muscle of mice. Results Digoxin at a dose of 0.1 mg/kg did not enhance serum, atrial, and ventricular Ca+2 levels, but were increased when digoxin dose of 1 and 5 mg/kg were administered. Histologically, myocardial necrosis and cellular infiltration on day 7 were significantly more severe in the 5 mg/kg/day digoxin group. Immunohistochemical studies showed more expression of both NF-kB and CaV1.2 in 1 and 5 mg/kg/day digoxin groups. Conclusions These data suggest that NF-kB may be responsible for digoxin toxicity, at least partially via modulation of CaV1.2 and intracellular calcium homeostasis in the myocardium.

The effective parameters for the Al-water reaction are studied. Salts such as sodium chloride (NaCl), potassium chloride (KCl) and barium chloride (BaCl2) were added to Al particles and the mixture were alloyed via high-energy ball milling. It was observed that the reaction of Al/water is evolved and the reaction induction time declined significantly by the application of BaCl2 as new modifier in comparison with NaCl and KCl. The X-ray diffraction patterns revealed that the other valuable reaction byproduct is AlOOH, which can be calcinated to gamma or alpha alumina. Microstructure of alloys were studied via FESEM and it was observed that as the time of ball milling increased the size of particles decreased. Increasing of the salt to Al powder ratio lead to increasing of hydrogen yield as well as hydrogen production rate. Effect of NaOH alkaline solution was also investigated and according to the results, solutions with higher concentration of NaOH generate higher amount of hydrogen.

Background The choice of appropriate antiplatelet therapy before the Pipeline Embolization Device (PED) placement is usually guided by platelet function testing such as light transmission aggregometry (LTA). In this study, we aimed to define the optimal threshold LTA value for clopidogrel responsiveness to predict the risk of postprocedural thromboembolic complications and to help guide appropriate antiplatelet regimen. Methods A prospectively maintained database at an academic neurosurgical center in the United States was retrospectively analyzed from 2014 to 2017 to identify patients with unruptured intracranial aneurysms treated with the PED. Clinical and radiographic data were analyzed to identify thromboembolic complications in the context of platelet function testing performed by LTA. Results A total of 95 procedures were performed for PED placement to treat 110 unruptured intracranial aneurysms. Thromboembolic complications were encountered in 4 (4.2%) of these patients. After stratifying the complication rate based on the maximal extent of platelet aggregation after administration of an exogenous platelet agonist, a marked increase in thromboembolic events was observed in patients with LTA values greater than 50%. When LTA was dichotomized based on this value, patients with an LTA value less than 50% had a thromboembolic complication rate of 1.3% (1/80), compared with 20% (3/15) for those with LTA values ≥50% (P = 0.001). Conclusions We observed the greatest increase in the rate of thromboembolic complications with LTA values of ≥50%. This can serve as an appropriate cut-off value for determining the clopidogrel response in patients undergoing endovascular treatment with the PED.

Here we report experiments dealing with the development of a fully serum-free culture system for AML cell proliferation. A number of compounds were tested for the requirements of AML colony formation and DNA synthesis in vitro in an attempt to replace serum with an artificial mixture. The results indicate that bovine serum albumin (BSA, 15 mg/ml), cholesterol (7.8 micrograms/ml), transferrin (7.7 X 10(-6) M) and insulin (1 microgram/ml) were essential for AML cell proliferation. Linoleic acid, 2-mercaptoethanol and selenite contributed only moderately beneficial effects. This mixture obviated the need for adding exogenous fetal calf or horse serum to the cultures and permitted equivalent and frequently superior AML colony formation and DNA synthesis. This is the first serum-free culture method for human AML that may allow for in-vitro studies of the growth regulation by recombinant growth factors under exactly defined and standardized conditions.
