Abstract

Increasing air conditioner use for cooling indoor spaces has the potential to be a primary driver of global greenhouse gas emissions. Moving indoor air with residential fans can raise the temperature threshold at which air conditioning needs to be turned on to maintain the thermal comfort of building occupants. We investigate whether fans can be used to reduce air conditioner use and associated greenhouse gas emissions.We developed an integrated framework, featuring a dynamic adaptive thermal comfort model with a geographical information system-based spatially gridded map of Australia, further complemented with census data. We assessed the change in energy use and associated greenhouse gas emissions for five scenarios of air conditioner and fan use: an air conditioner-only scenario (no fans); and four fan-first scenarios with fans operating at speeds of 0·1 m/s, 0·3 m/s, 0·8 m/s, and 1·2 m/s, with air conditioning used only once the upper temperature threshold for thermal discomfort is exceeded. For each day of the selected case study year, we estimated the upper temperature limit for thermal comfort and the number of hours in which air conditioning would be switched on.The thermal comfort threshold was increased by the use of fans compared with air conditioner use alone. We found that widespread indoor fan use had the potential to reduce energy demand and greenhouse gas emissions attributable to air conditioner use, without compromising thermal comfort. Taking an annual perspective, the use of fans with air speeds of 1·2 m/s compared with air conditioner use alone resulted in a 76% reduction in energy use (from 5592 GWh to 1344 GWh) and associated greenhouse gas emissions (5091 kilotonnes to 1208 kilotonnes).A common strategy to cope with hot weather is the use of air conditioners, which feed a cycle of high electricity consumption, often delivered by fossil fuel power stations that in turn contribute to further increases in emissions. Abstract

Refractory epilepsy can develop as a late complication for survivors of pediatric cancer with central nervous system involvement who undergo chemoradiation therapy. This case series reports three such patients who presented with atypical non-convulsive seizures up to 14 years after cancer diagnosis. We hypothesize that the development of refractory epilepsy may be a late effect of radiation treatment, as each patient underwent whole brain radiation. Given the high morbidity and mortality associated with epilepsy, early identification is crucial to improve outcomes and quality of life for this vulnerable population.

Abstract

Aquaporin-8 (AQP8) is a peroxiporin, a transmembrane water and hydrogen peroxide (H2O2) transport protein expressed in the mitochondrial and plasma membranes of pancreatic β-cells. AQP8 protein expression is low under physiological conditions, but it increases after cytokine exposure both, in vitro and in vivo, possibly related to a NF-κB consensus sequence in the promoter. AQP8 knockdown (KD) insulin-producing RINm5F cells are particularly susceptible to cytokine-mediated oxidative stress. Cytokine (a mixture of IL-1β, TNF-α, and IFN-γ) treated AQP8 KD cells exhibited pronounced sensitivity to reactive oxygen and nitrogen species (ROS and RNS), resulting in a significant loss of β-cell viability due to enhanced toxicity of the increased concentrations of H2O2 and hydroxyl radicals (●OH) in mitochondria of AQP8 KD cells. This viability loss went along with increased caspase activities, reduced nitrite concentration (representative of nitric oxide (NO●) accumulation) and increased lipid peroxidation. The explanation for the increased toxicity of the proinflammatory cytokines in AQP8 KD cells resides in the fact that efflux of the H2O2 generated during oxidative stress in the β-cell mitochondria is hampered through the loss of the peroxiporin channels in the mitochondrial membranes of the AQP8 KD cells. The increased proinflammatory cytokine toxicity due to loss of AQP8 expression in the KD β-cell mitochondria is thus the result of increased rates of apoptosis. This decreased cell viability is caused by increased levels of oxidative stress along with a ferroptosis-mediated cell death component due to decreased NO● generation.

Abstract

For a semisimple complex Lie algebra g , the BGG category O is of particular interest in representation theory. It is known that Irving's shuffling functors Sh w , indexed by elements w ∈ W of the Weyl group, induce an action of the braid group B W associated to W on the derived categories D b ( O λ ) of blocks of O . We show that for maximal parabolic subalgebras p of sl n corresponding to the parabolic subgroup W p = S n − 1 × S 1 of S n , the derived shuffling functors L Sh s i are instances of Seidel and Thomas' spherical twist functors. Namely, we show that certain parabolic indecomposable projectives P p ( w ) are spherical objects, and the associated twist functors are naturally isomorphic to L Sh w [ 1 ] as auto-equivalences of D b ( O p ) . We give an overview of the main properties of the BGG category O , the construction of shuffling and spherical twist functors, and give some examples how to determine images of both. To this end, we employ the equivalence of blocks of O and the module categories of certain path algebras.

Abstract

Ultrasound Core Laboratories (UCL) are used in multicenter trials to assess imaging biomarkers to define robust phenotypes, to reduce imaging variability and to allow blinded independent review with the purpose of optimizing endpoint measurement precision. The Household Air Pollution Intervention Network, a multicountry randomized controlled trial (Guatemala, Peru, India and Rwanda), evaluates the effects of reducing household air pollution on health outcomes. Field studies using portable ultrasound evaluate fetal, lung and vascular imaging endpoints. The objective of this report is to describe administrative methods and training of a centralized clinical research UCL. A comprehensive administrative protocol and training curriculum included standard operating procedures, didactics, practical scanning and written/practical assessments of general ultrasound principles and specific imaging protocols. After initial online training, 18 sonographers (three or four per country and five from the UCL) participated in a 2 wk on-site training program. Written and practical testing evaluated ultrasound topic knowledge and scanning skills, and surveys evaluated the overall course. The UCL developed comprehensive standard operating procedures for image acquisition with a portable ultrasound system, digital image upload to cloud-based storage, off-line analysis and quality control. Pre- and post-training tests showed significant improvements (fetal ultrasound: 71% ± 13% vs. 93% ± 7%, p < 0.0001; vascular lung ultrasound: 60% ± 8% vs. 84% ± 10%, p < 0.0001). Qualitative and quantitative feedback showed high satisfaction with training (mean, 4.9 ± 0.1; scale: 1 = worst, 5 = best). The UCL oversees all stages: training, standardization, performance monitoring, image quality control and consistency of measurements. Sonographers who failed to meet minimum allowable performance were identified for retraining. In conclusion, a UCL was established to ensure accurate and reproducible ultrasound measurements in clinical research. Standardized operating procedures and training are aimed at reducing variability and enhancing measurement precision from study sites, representing a model for use of portable digital ultrasound for multicenter field studies.
