Abstract

We describe a unique, web-based data visualization portal developed for use by researchers and public transit agencies investigating future shared-taxi fleet scenarios. Augmenting or even replacing fixed-route transit lines with automated, connected, shared taxi fleets may be a desirable alternative in less-densely developed areas. The MATSim agent-based transport microsimulation model is used to study scenarios including the status quo, dynamically-dispatched fleets with drivers, and fully autonomous fleets. This paper focuses on a data visualization portal which includes many interactive views such as agent (taxi) movements color-coded by number of passengers and trip request origins and destinations, changes in roadway and passenger volumes compared to a base case, and more. The agent-based simulation covers a 24 hour simulation period; analysts can hone in on specific times of day to examine, e.g. school pickup/drop-offs or commute trips connecting to rail stations. The tool is in operation for several small cities and rural regions in Germany and was successfully used as an outreach tool in public meetings. In addition, developers of the MATSim DRT extension found the visualizations particularly useful for debugging both the algorithms and the scenario definitions. The code is entirely open source and, while this specific study has a rather esoteric use case, the visualization platform has an extensible design that could be modified for other purposes.

Abstract

This work investigates the densification process of nanocrystalline NdFeB powder by electro-discharge sintering (EDS) and the associated magnetic properties. The EDS technique is used as a fast and energy-saving compaction process for metal powders. A large current is discharged from capacitors into a pre-compacted loose powder, thus resulting in complete compaction. In this study, the microstructure, magnetic, and mechanical properties of the compacted, hard magnetic NdFeB specimens were investigated under variation of the energy EEDS and compression load pEDS. For all specimens, the intrinsic coercivity HcJ decreases on increasing the discharge energy. However, the compaction load has apparently no influence on the coercivity HcJ, whereas the residual induction Br decreases only with increasing discharge energy. An increase in the compression load pEDS causes an increase in the specimens’ density and thus promotes residual induction Br. The applied EDS parameters led to the formation of three different microstructures (insufficiently densified zone, fully densified zone, and remelted zone) along the cross-section of the EDS-densified specimens. Volume fractions of the three different microstructures that form during the EDS process determine the resulting mechanical and magnetic properties of the specimens.

Abstract

The High Energy Stereoscopic System (H.E.S.S.) is one of the three arrays of imaging atmospheric Cherenkov telescopes (IACTs) currently in operation. It is composed of four 12-meter telescopes and a 28-meter one, and is sensitive to gamma rays in the energy range ~30 GeV - 100 TeV. The cameras of the 12-m telescopes recently underwent a substantial upgrade, with the goal of improving their performance and robustness. The upgrade involved replacing all camera components except for the photomultiplier tubes (PMTs). This meant developing new hardware for the trigger, readout, power, cooling and mechanical systems, and new software for camera control and data acquisition. Several novel technologies were employed in the cameras: the readout is built around the new NECTAr digitizer chip, developed for the next generation of IACTs; the camera electronics is fully controlled and read out via Ethernet using a combination of FPGA and embedded ARM computers; the software uses modern libraries such as Apache Thrift, ZMQ and Protocol buffers. Abstract

Inhibition of the glucagon receptor (GCGR) has been identified as a potential therapeutic approach for the treatment of type 2 diabetes. However, a small molecule drug candidate antagonizing GCGR (BAY16) failed during preclinical drug development, in part due to drug induced hepatotoxicity in animals. Since there is evidence to suggest that endogenous GCGR signaling might be important for hepatocyte survival, we hypothesized that on-target effects, i.e., modulation of GCGR activity by BAY16, may contribute to BAY16 hepatotoxicity and associated gene expression changes in rats. To understand the role of GCGR inhibition in BAY16 toxicity, we analyzed cell viability and gene expression profiles in non-silenced and GCGR-targeting siRNA transfected primary rat hepatocytes with and without exposure to BAY16 to discriminate between on- and off-target effects of BAY16. siRNA-mediated silencing of the GCGR did not affect cell viability in primary rat hepatocytes, indicating that cytotoxicity of BAY16 occurs independent of its pharmacological effects. In support of this, gene expression analysis of GCGR silenced hepatocytes revealed no transcriptional alterations relevant to toxicity. In contrast, BAY16 caused a concentration-dependent decrease in cell viability, along with changes in the expression of genes associated with altered xenobiotic metabolism, oxidative stress, increased fatty acid synthesis, and alterations in cholesterol and bile acid metabolic processes. Based on gene expression data, it appears that hepatocytes inhibit cholesterol synthesis and increase detoxifying and eliminating processes in order to protect themselves from accumulation of bile acids, cholesterol or drug intermediates. Importantly, comparison of transcriptional changes in the absence and presence of GCGR revealed that the same pathways were affected in both silenced and non-silenced hepatocytes, indicating that BAY16 toxicity occurs independent of the GCGR receptor.

Abstract

The absolute muon flux between 20 and 3000 GeV is measured with the L3 magnetic muon spectrometer for zenith angles ranging from 0°to 58°. Due to the large exposure of about 150 m2sr d, and the excellent momentum resolution of the L3 muon chambers, a precision of 2.3% at 150 GeV in the vertical direction is achieved.The ratio of positive to negative muons is studied between 20 and 500 GeV, and the average vertical muon charge ratio is found to be 1.285±0.003(stat.)±0.019(syst.)., Physics Letters B, 598 (1-2), ISSN:0370-2693, ISSN:0031-9163, ISSN:1873-2445

Abstract

FeatureIDE is an open-source framework for feature-oriented software development (FOSD) based on Eclipse. FOSD is a paradigm for the construction, customization, and synthesis of software systems. Code artifacts are mapped to features, and a customized software system can be generated given a selection of features. The set of software systems that can be generated is called a software product line (SPL). FeatureIDE supports several FOSD implementation techniques such as feature-oriented programming, aspect-oriented programming, delta-oriented programming, and preprocessors. All phases of FOSD are supported in FeatureIDE, namely domain analysis, requirements analysis, domain implementation, and software generation.

Abstract

We report on the in-core fiber temperature measurements in Yb 2 O 3 -doped aluminum silicate fibers during optical pumping using UV femtosecond-induced fiber Bragg gratings (FBGs). The FBG containing fiber samples show a different temperature behavior during pumping before and after thermal annealing, caused by absorption centers generated during grating inscription. These absorption centers can be removed completely by thermal annealing. In this way, remaining temperature rise during pumping due to the quantum defect of the Yb 3+ ions is determined depending on the Yb concentration in the range 0.14–0.46 mol% and the pump wavelength.

Abstract

In feature-oriented programming (FOP) a programmer decomposes a program in terms of features. Ideally, features are implemented modularly so that they can be developed in isolation. Access control mechanisms in the form of access or visibility modifiers are an important ingredient to attain feature modularity as they allow programmers to hide and expose internal details of a module’s implementation. But developers of contemporary feature-oriented languages have not considered access control mechanisms so far. The absence of a well-defined access control model for FOP breaks encapsulation of feature code and leads to unexpected program behaviors and inadvertent type errors. We raise awareness of this problem, propose three feature-oriented access modifiers, and present a corresponding access modifier model. We offer an implementation of the model on the basis of a fully-fledged feature-oriented compiler. Finally, by analyzing ten feature-oriented programs, we explore the potential of feature-oriented modifiers in FOP.

Abstract

Fiber Bragg grating inscription with a deep ultraviolet (DUV) femtosecond laser and two-beam interferometry allows grating inscription in germanium-free fibers without classical UV photosensitivity. Therefore such a writing technology is a promising solution for the fabrication of fiber Bragg gratings in the active fiber material to realize a fully integrated all-fiber solution. In this paper we report on the use of the DUV femtosecond laser fiber Bragg grating technology to build a grating-stabilized fiber laser with high spectral purity. Output powers up to 158 W at 1083 nm and a slope efficiency of 76% have been demonstrated.

Abstract

Follicular lymphoma (FL) is a germinal centre-derived indolent B-cell lymphoma representing the second most common Non Hodgkin lymphoma in the Western world. This chapter focuses on the pathology of FL and summarizes the current knowledge about genetic and molecular features that are relevant for the pathogenesis of this neoplasm. The translocation t(14;18) is present in approximately 90% of FL leading to the upregulation of the anti-apoptotic protein BCL2, that may constitute a promising molecular target for therapeutic approaches. FL lacking the t(14;18) also exist, and B-cells carrying the t(14;18) can be detected in a subset of healthy individuals. In addition to the t(14;18), secondary genetic alterations are present in most FL and, more recently, deeper insights into the methylation and microRNA expression patterns in the tumour cells have been gained. The tumour microenvironment appears to be particularly important for the biology and the clinical course of FL.

Abstract

p53 limits the proliferation of precancerous cells by inducing cell-cycle arrest or apoptosis. How the decision between survival and death is made at the level of p53 binding to target promoters remains unclear. Using cancer cell lines, we show that the cooperative nature of DNA binding extends the binding spectrum of p53 to degenerate response elements in proapoptotic genes. Mutational inactivation of cooperativity therefore does not compromise the cell-cycle arrest response but strongly reduces binding of p53 to multiple proapoptotic gene promoters (BAX, PUMA, NOXA, CASP1). Vice versa, engineered mutants with increased cooperativity show enhanced binding to proapoptotic genes, which shifts the cellular response to cell death. Furthermore, the cooperativity of DNA binding determines the extent of apoptosis in response to DNA damage. Because mutations, which impair cooperativity, are genetically linked to cancer susceptibility in patients, DNA binding cooperativity contributes to p53's tumor suppressor activity.

Abstract

Alternative splicing of transcripts from many cancer-associated genes is believed to play a major role in carcinogenesis as well as in tumor progression. Alternative splicing of one such gene, the neural cell adhesion molecule CD56 (NCAM), impacts the progression, inadequate therapeutic response, and reduced total survival of patients who suffer from numerous malignant neoplasms. Although previous investigations have determined that CD56 exists in three major isoforms (CD56(120kD), CD56(140kD), and CD56(180kD)) with individual structural and functional properties, neither the expression profiles nor the functional relevance of these isoforms in malignant tumors have been consistently investigated. Using new quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR) strategies and novel CD56 isoform-specific antibodies, CD56(140kD) was shown to be exclusively expressed in a number of highly malignant CD56(+) neoplasms and was associated with the progression of CD56(+) precursor lesions of unclear malignant potential. Moreover, only CD56(140kD) induced antiapoptotic/proliferative pathways and specifically phosphorylated calcium-dependent kinases that are relevant for tumorigenesis. We conclude, therefore, that the specific detection of CD56 isoforms will help to elucidate their individual functions in the pathogenesis and progression of malignant neoplasms and may have a positive impact on the development of CD56-based immunotherapeutic strategies.

Abstract

Due to their ability to degrade RNA, selected members of the bovine pancreatic ribonuclease A (RNase A) superfamily are potent cytotoxins. These cytotoxic ribonucleases enter the cytosol of target cells, where they degrade cellular RNA and cause cell death. The cytotoxic activity of most RNases, however, is abolished by the cytosolic ribonuclease inhibitor (RI). Consequently, the development of RNase derivatives with the ability to evade RI binding is a desirable goal. In this study, tandem enzymes consisting of two RNase A units that are bound covalently via a peptide linker were generated by gene duplication. As deduced from the crystal structure of the RNase A.RI complex, one RNase A unit of the tandem enzyme can still be bound by RI. The other unit, however, should remain unbound because of steric hindrance. This free RNase A unit is expected to maintain its activity and to act as a cytotoxic agent. The study of the influence of the linker sequence on the conformation and stability of these constructs revealed that tandemization has only minor effects on the activity and stability of the constructs in comparison to monomeric RNase A. Relative activity was decreased by 10-50% and the melting temperature was decreased by less than 2.5 K. Furthermore, the cytotoxic potency of the RNase A tandem enzymes was investigated. Despite an in vitro inhibition by RI, tandemization was found to endow RNase A with remarkable cytotoxic activity. While monomeric RNase A is not cytotoxic, IC(50) values of the RNase A tandem variants decreased to 70.3-12.9 microM. These findings might establish the development of a new class of chemotherapeutic agents based on pancreatic ribonucleases.

Abstract

Events with a large rapidity gap in deep inelastic scattering with Q2 greater-than-or-equal-to 10 GeV2 have been studied in the ZEUS detector. The properties of these events with W > 140 GeV are consistent with a leading twist diffractive production mechanism. In the laboratory frame, with E(T)jet greater-than-or-equal-to 4 GeV, 15% of the events are of the 1-jet type with negligible 2-jet production. The single jet is back-to-back in azimuth with the scattered electron. No energy flow is observed between the jet and the proton direction. With a lower jet transverse energy cut 2-jet production is observed both in the laboratory and the gamma*p centre-of-mass systems, demonstrating the presence of hard scattering in the virtual photon proton interactions that give rise to large rapidity gap events.

The Amanda neutrino telescope at the South Pole has been taking data since 1996. Stepwise upgraded, it reached its final stage in January 2000. We present results from the search for extraterrestrial neutrinos, neutrinos from dark matter annihilation and magnetic monopoles.

Abstract

The mass composition of high-energy cosmic rays at energies above 1015 eV can provide crucial information for the understanding of their origin. Air showers were measured simultaneously with the SPASE-2 air shower array and the AMANDA-B10 Cherenkov telescope at the South Pole. This combination has the advantage to sample almost all high-energy shower muons and is thus a new approach to the determination of the cosmic ray composition. The change in the cosmic ray mass composition was measured versus existing data from direct measurements at low energies. Our data show an increase of the mean log atomic mass 〈lnA〉 by about 0.8 between 500 TeV and 5 PeV. This trend of an increasing mass through the "knee" region is robust against a variety of systematic effects. © 2004 Elsevier B.V. All rights reserved.

Abstract

The Antarctic Muon And Neutrino Detector Array (AMANDA) is a high-energy neutrino telescope operating at the geographic South Pole. It is a lattice of photo-multiplier tubes buried deep in the polar ice between 1500m and 2000m. The primary goal of this detector is to discover astrophysical sources of high energy neutrinos. A high-energy muon neutrino coming through the earth from the Northern Hemisphere can be identified by the secondary muon moving upward through the detector. The muon tracks are reconstructed with a maximum likelihood method. It models the arrival times and amplitudes of Cherenkov photons registered by the photo-multipliers. This paper describes the different methods of reconstruction, which have been successfully implemented within AMANDA. Strategies for optimizing the reconstruction performance and rejecting background are presented. For a typical analysis procedure the direction of tracks are reconstructed with about 2 degree accuracy., 40 pages, 16 Postscript figures, uses elsart.sty

Abstract

We report on the analysis of air showers observed in coincidence by the Antarctic Muon and Neutrino detector array (AMANDA-B10) and the South Pole Air Shower Experiment (SPASE-1 and SPASE-2). We discuss the use of coincident events for calibration and survey of the deep AMANDA detector as well as the response of AMANDA to muon bundles. This analysis uses data taken during 1997 when both SPASE-1 and SPASE-2 were in operation to provide a stereo view of AMANDA. © 2003 Elsevier B.V. All rights reserved.

Abstract

The IceCube neutrino telescope, to be constructed near the Antarctic South Pole, represents the next generation of neutrino telescope. Its large 1 km3 size will make it uniquely sensitive to the detection of neutrinos from astrophysical sources. The current design of the detector is presented. The basic performance of the detector and its ability to search for neutrinos from various astrophysical sources has been studied using detailed simulations and is discussed.

We present results from the AMANDA high energy neutrino telescope located at the South Pole. They include measurements of the atmospheric neutrino flux, search for UHE point sources, and diffuse sources producing electromagnetic/hadronic showers at the detector or close to it.

Abstract

We show new results from both the older and newer incarnations of AMANDA (AMANDA-B10 and AMANDA-II, respectively). These results demonstrate that AMANDA is a functioning, multipurpose detector with significant physics and astrophysics reach. They include a new higher-statistics measurement of the atmospheric muon neutrino flux and preliminary results from searches for a variety of sources of ultrahigh energy neutrinos: generic point sources, gamma-ray bursters and diffuse sources producing muons in the detector, and diffuse sources producing electromagnetic or hadronic showers in or near the detector.
