Previous experience is thought to facilitate our ability to extract spatial and temporal regularities from cluttered scenes. However, little is known about how we may use this knowledge to predict future events. Here we test whether exposure to temporal sequences facilitates the visual recognition of upcoming stimuli. We presented observers with a sequence of leftwards and rightwards oriented gratings that was interrupted by a test stimulus. Observers were asked to indicate whether the orientation of the test stimulus matched their expectation based on the preceding sequence. Our results demonstrate that exposure to temporal sequences without feedback facilitates our ability to predict an upcoming stimulus. In particular, observers' performance improved following exposure to structured but not random sequences. Improved performance lasted for a prolonged period and generalized to untrained stimulus orientations rather than sequences of different global structure, suggesting that observers acquire knowledge of the sequence structure rather than its items. Further, this learning was compromised when observers performed a dual task resulting in increased attentional load. These findings suggest that exposure to temporal regularities in a scene allows us to accumulate knowledge about its global structure and predict future events.

Learning and experience are critical for translating ambiguous sensory information from our environments to perceptual decisions. Yet evidence on how training molds the adult human brain remains controversial, as fMRI at standard resolution does not allow us to discern the finer scale mechanisms that underlie sensory plasticity. Here, we combine ultra-high-field (7T) functional imaging at sub-millimeter resolution with orientation discrimination training to interrogate experience-dependent plasticity across cortical depths that are known to support dissociable brain computations. We demonstrate that learning alters orientation-specific representations in superficial rather than middle or deeper V1 layers, consistent with recurrent plasticity mechanisms via horizontal connections. Further, learning increases feedforward rather than feedback layer-to-layer connectivity in occipito-parietal regions, suggesting that sensory plasticity gates perceptual decisions. Our findings reveal finer scale plasticity mechanisms that re-weight sensory signals to inform improved decisions, bridging the gap between micro- and macro-circuits of experience-dependent plasticity.

Extracting the structure of complex environments is at the core of our ability to interpret the present and predict the future. This skill is important for a range of behaviours from navigating a new city to learning music and language. Classical approaches that investigate our ability to extract the principles of organisation that govern complex environments focus on reward-based learning. Yet, the human brain is shown to be expert at learning generative structure based on mere exposure and without explicit reward. Individuals are shown to adapt to-unbeknownst to them-changes in the environment's temporal statistics and predict future events. Further, we present evidence for a common brain architecture for unsupervised structure learning and reward-based learning, suggesting that the brain is built on the premise that 'learning is its own reward' to support adaptive behaviour.

Although language comprehension usually requires multimodal information, no study to date has investigated how comprehenders deal with the revision of a text's interpretation when different modalities are involved. Twenty-four young adults listened to a story prompting an inference (e.g., polar bear), and then saw a picture that was either consistent (polar bear) or inconsistent but still plausible (penguin). Larger negativity (N400) in the inconsistent picture indicated successful inferential monitoring. Subsequently, a sentence carried the disambiguating word which was either expected (“bear”) or unexpected (“penguin”) in relation to the auditory-verbal information. Larger negativity in the unexpected word coming from the consistent picture suggested that comprehenders had difficulties selecting the unexpected concept when previous information was contradictory. More importantly, this effect was modulated by inhibitory control, where a higher resistance to distractor interference (flanker task) was associated with a better ability to suppress pictorial information, therefore preventing semantic competition. Similarly, accuracy measured in a final comprehension question demonstrated that higher inhibitory control was related to a more efficient ability to revise the situation model across modalities. Our findings speak to a relationship between story comprehension and mental flexibility during multimodal processing.

Training is known to improve performance in a variety of perceptual and cognitive skills. However, there is accumulating evidence that mere exposure (i.e. without supervised training) to regularities (i.e. patterns that co-occur in the environment) facilitates our ability to learn contingencies that allow us to interpret the current scene and make predictions about future events. Recent neuroimaging studies have implicated fronto-striatal and medial temporal lobe brain regions in the learning of spatial and temporal statistics. Here, we ask whether patients with mild cognitive impairment due to Alzheimer′s disease (MCI-AD) that are characterized by hippocampal dysfunction are able to learn temporal regularities and predict upcoming events. We tested the ability of MCI-AD patients and age-matched controls to predict the orientation of a test stimulus following exposure to sequences of leftwards or rightwards orientated gratings. Our results demonstrate that exposure to temporal sequences without feedback facilitates the ability to predict an upcoming stimulus in both MCI-AD patients and controls. However, our fMRI results demonstrate that MCI-AD patients recruit an alternate circuit to hippocampus to succeed in learning of predictive structures. In particular, we observed stronger learning-dependent activations for structured sequences in frontal, subcortical and cerebellar regions for patients compared to age-matched controls. Thus, our findings suggest a cortico-striatal–cerebellar network that may mediate the ability for predictive learning despite hippocampal dysfunction in MCI-AD.
