16. Copy number assessment of SMN1 based on real-time PCR with high-resolution melting: fast and highly reliable testing

Abstract

Background: Spinal muscular atrophy (SMA) is a common neuromuscular disorder, caused by absence of both copies of the survival motor neuron 1 (SMN1) gene. Population-wide SMA screening to quantify copy number of SMN1 is recommended by multiple regions. SMN1 diagnostic assay with simplified procedure, high sensitivity and throughput is still needed.Methods: Real-Time PCR with High-Resolution Melting for the quantification of the SMN1 gene exon 7 copies and SMN1 gene exon 8 copies was established and confirmed by multiplex ligation-dependent probe amplification (MLPA). The diagnosis of 2563 individuals including SMA patients, suspected cases and the general population were analyzed by the real-time PCR. The results were compared with the gold standard test MPLA. Results: In this study, the homozygous deletions, heterozygous deletions were identified by Real-Time PCR with High-Resolution Melting method with an incidence of 10.18% and 2.42%, respectively. In addition, the R value distribution (P>0.05) among the 8 replicates and the coefficient of variation (CVConclusions: The qPCR based on High-Resolution Melting provides a sensitive and high-throughput approach to large-scale SMA carrier screening with low cost and labor.

26. Iridoid glucosides from the leaves of Vitex negundo var. cannabifolia

Abstract

Five new iridoid glucosides, cannabifolisides A–E (1 – 5), together with nine analogues (6 – 14), were isolated from the leaves of Vitex negundo var. cannabifolia. Their structures were elucidated by spectroscopic data (NMR, UV, IR, and MS) analyses and comparison of their spectroscopic and physical data with the literature values. The protective effect of isolated compounds on human gastric epithelial cells were evaluated by measuring the enhanced cell viability of GES-1 cell insulted by TCA. Among these, compounds 4, 6, 8, 10, 12, and 14 remarkably increased the cell viability in a concentration-depend manner.

Abstract

MXenes have been long sought-after for their exceptional electrical conductivity, large family members, mechanical stability, tunable surface groups, and ease of processability. In particular, they have become the leading materials for electromagnetic (EM) wave absorption. Herein, MXenes’ crystal and electronic structures, as well as EM wave absorption related to those properties, are discussed. The structures and EM functions of MXene hybrids are systematically arranged. The crystal and electronic structure, EM characteristics, and EM response of MXene-based hybrids, as well as their relationship are revealed. Moreover, we propose the main challenges, and predict future directions in this rapidly broadening field.

38. Fischer-Tropsch synthesis to lower α-olefins over cobalt-based catalysts: Dependence of the promotional effect of promoter on supports

Abstract

Promoters, such as Na and Mn, play an important role in governing the active phase and catalytic performance of cobalt-based FTS catalyst. In this work, the dependence of the promotional effect of promoters on supports has been investigated by using the carbon and silica supported cobalt-based catalysts in the presence of Na, Mn, and both Na and Mn promoters, respectively. The characterization results of H2-TPR, CO-TPD and XRD profiles together with the FTS performances demonstrate the strong dependence of the promotional effect of promoter on the type of support used. The carbon support, which shows weak cobalt-support interaction, facilitates the manifestation of the effect of promoter. In detail, Mn can promote the formation of Co2C sites at the conditions of 260 °C, 0.5 MPa and lower H2/CO ratio, resulting from the promoted CO adsorption and subsequent dissociation. Thus, the suppressed adsorption of H2 and the promoted surface dissociative C* concentration enhances the selectivities to lower olefins (C2=-C4=) and C5+ with a decrease in methane selectivity. Interestingly, the further addition of Na promoter shows synergistic effect of Na and Mn promoter on remarkably increasing selectivity to lower olefins due to the obviously suppressed chain growth. This synergistic effect could be observed only at the weak cobalt-carbon interaction with a more obvious effect at lower H2/CO ratio in feed gas that favors the increase of Co2C sites with mainly exposed facets of (111) and (020). The presence of (020) facet might be more crucial to enhance the formation of lower olefins. However, over silica support with strong cobalt-silica interaction, the promotional effect of Mn is obviously suppressed, which leads to only CoO and metallic Co sites in the FTS reaction and much weaker impact on product selectivity. The further addition of Na into Mn-promoted cobalt catalysts shows similar effect as Na promoter alone in the product selectivity where the chain growth is highly promoted to remarkably suppress methane selectivity and enhance selectivity to C5+ rather than desired lower olefins. In contrast to carbon support, the strong cobalt-silica interaction leads to less promotional formation of CO2 via the Boudouard reaction (2CO*→C* + CO2) due to the increased non-dissociative adsorption of CO.

Abstract

As a post-translational modification, protein glycosylation not only reflects on body health, but also serves as the target of drug treatment. Therefore, it is of great significance to study protein glycosylation for the diagnosis and treatment of diseases. However, many glycoproteins of biological importance exhibit relatively low abundance, high complexity, and wide dynamic range in biological samples. It is indispensable to explore highly efficient sample pretreatment methods. In recent years, monolithic columns have attracted extensive attention in the field of separation and enrichment of glycoproteins due to their rapid separation, high efficiency, and good biological compatibility. In this paper, recent progress of monolithic columns in the field of separation and enrichment of glycoproteins is surveyed, and the future development is prospected.

Abstract

Seven undescribed sesquiterpenoid dimers, commiphomyrones A - G, together with three known analogs, were isolated from the resin of Commiphora myrrha Engl.. The structures of the undescribed compounds were elucidated based on a comprehensive analysis of spectroscopic data (NMR, UV, IR, and MS), and the absolute configurations were defined by comparing the experimental and calculated ECD spectra as well as by performing X-ray crystallographic analysis. All the isolated dimeric sesquiterpenoids feature a 7-oxabicyclo [2.2.1] hept-2-ene moiety formed by the [4 + 2] cycloaddition of two sesquiterpenoids.
