No. of bitstreams: 0 Previous issue date: 2021-12-15 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Hepatic steatosis is directly associated with hepatic inflammation and insulin resistance, which is correlated with hyperglycemia and type 2 diabetes mellitus (T2DM). Aerobic and strength training have been pointed out as efficient strategies against hepatic steatosis. However, little is known about the effects of the combination of those two protocols on hepatic steatosis. Therefore, this study aimed to evaluate the impact of short-term combined training (STCT) on glucose homeostasis and in the synthesis and oxidation of fat in the liver of obesity-induced mice with hepatic steatosis. Swiss mice were distributed into three groups: control lean (CTL), sedentary obese (OB), and combined training obese (CTO). The CTO group performed the STCT protocol, which consisted of strength and aerobic exercises in the same session. The protocol lasted seven days. The CTO group reduced the glucose levels and fatty liver when compared to the OB group. Interestingly, these results were observed even without reductions in body adiposity. CTO group also showed increased hepatic insulin sensitivity, with lower hepatic glucose production (HGP). STCT reduced the expression of the lipogenic genes Fasn and Scd1 and hepatic inflammation, as well as increased the ACC phosphorylation and the oxidative genes Cpt1a and Ppara, reverting the complications caused by obesity. Since this protocol increased lipid oxidation and reduced hepatic lipogenesis, regardless of body fat mass decrease, it can be considered an effective non-pharmacological strategy for the treatment of hepatic steatosis. Laser transmission welding is an established single-stage plastic joining process, which enables hermetically sealed joints under the workpiece surface. The process requires joining partners with proper degrees of transmission and absorption to the processing wavelength. For reaching a stable process an in-process quality assurance is very valuable. Current monitoring systems have a limited usage, as no quantitative information of the weld itself is obtained without its destruction. In medical and pharmaceutical applications a weld with leakage is e.g. unacceptable. The main objective of this paper is the presentation of the optical coherence tomography as a tool for the quality assurance in laser transmission welding. This approach enables the measurement of any residual gap, weld geometry, internal pores and leaks. The presented results show that this technique allows even the characterization of welds using joining partners with thicknesses of 2 mm or with glass fiber reinforcement levels of 30% per weight.

Laser surface structuring is a rapidly developing manufacturing technology. This technology offers a high level of flexibility regarding the geometrical design and a wide range of machinable materials. However, current systems used for laser structuring have a crucial deficit. The existing process control systems have no sufficient feedback infor- mation of the machined surface topology before, during and after the process, not being able to detect errors/devia- tions and so to adapt the structuring strategy while processing. Therefore a novel scanner based inline metrology system has been developed to control the laser surface structuring process by a process monitoring software.

The quality inspection of products poses a challenge towards existing production metrology techniques in terms of accuracy and speed, especially when it comes to inline or in-process measurements. In this terms, optical metrology proves its capabilities, in particular for sensible and fragile work pieces. For the inspection inside small spaces and cavities or for complex geometries, many optical metrology system are to inflexible or cannot be miniaturized enough. Within this work a set-up for a fiber-optic low-coherence interferometer system is described. Due to the high degree of miniaturization of the sensing probe, the system can be used for demanding measuring tasks like roundness of small drilling holes or the run-out of shafts. Different evaluation measurement are presented, which underline the potential of the described system for highly accurate, non-contact inspection of rotational parts.

Repetitive sequences constitute landmarks for genome regulation, evolution, and chromatin architecture. Patterns of specific and non-specific repetitive sequences change in many types and stages of tumor cells, characterized by band loss, gain, and (de) increased staining of pre-existing bands. In this work, repetitive DNA was studied in search of genome instability of skin cancers: basal and squamous cell carcinomas (BCC and SCC), malignant melanoma (MM), melanocytic nevus (MN), and actinic keratosis (AK) lesions. DNAs were extracted from blood and tumor samples from 21 BCC, 7 SCC, 11 MM and 7 lesions. Banding patterns were obtained by random amplification of polymorphic DNA (RAPD), and specific D9S50 and D9S52 microsatellites (9p21). D9S50 patterns revealed microsatellite instability (MSI) and/or loss of heterozygosity (LOH) in 36% BCC, 25% SCC, and 57% MM tumors. D9S52 microsatellite showed 28.5%; 42.8%; and 71.4% altered tumors, respectively. No microsatellite alterations were found in MN and AK. On the other hand, genomic rearrangements detected by RAPD were present in 100% tumors. In BCC, the mean number of tumor DNA alterations showed predominant gain of bands. On the contrary, MM samples presented loss, or decreased intensity signal of RAPD bands. Genome alterations in skin cancers would result from chromosomal rearrangements, aneuploidy and/or polysomies. The low-cost and quick RAPD technique may reveal unknown genes or DNA sequences associated with tumor development and progression, and may be easily implemented in clinical diagnosis.
