Abstract

In this study, we investigated the graphene oxide (GO) nanosheets of specific interaction with metabolome in breast cancer cells. The edge-oxidized form of GO nanosheets have multiple applications in biomedical research. At present, the actual metabolic mechanisms of GO nanosheets is not yet clearly understood. Here, we describe the effects of GO exposure on MCF-7 breast cancer cells using 1H nuclear magnetic resonance (1H-NMR) profiling. We treated MCF-7 cells with different GO concentrations and examined the metabolic distributions. Interestingly, we found that the treatment affected arginine metabolism, proline metabolism, and aminoacyl-tRNA biosynthesis, including anabolism and catabolism. GO dose-dependently increased the number of metabolic disturbances in cancer steroids. Therefore, these data suggest that GO can significantly potentiate the metabolic effect in MCF-7 cells. These results demonstrate that using targeted metabolomics profiling and pattern recognition analysis display the potential metabolic changes and underlying metabolic impacts in human breast cancer cells.

Abstract

Nuclear magnetic resonance (NMR) spectroscopy acts as the best tool that can be used in tissue engineering scaffolds to investigate unknown metabolites. Moreover, metabolomics is a systems approach for examining in vivo and in vitro metabolic profiles, which promises to provide data on cancer metabolic alterations. However, metabolomic profiling allows for the activity of small molecules and metabolic alterations to be measured. Furthermore, metabolic profiling also provides high-spectral resolution, which can then be linked to potential metabolic relationships. An altered metabolism is a hallmark of cancer that can control many malignant properties to drive tumorigenesis. Metabolite targeting and metabolic engineering contribute to carcinogenesis by proliferation, and metabolic differentiation. The resulting the metabolic differences are examined with traditional chemometric methods such as principal component analysis (PCA), and partial least squares-discriminate analysis (PLS-DA). In this review, we examine NMR-based activity metabolomic platforms that can be used to analyze various fluxomics and for multivariant statistical analysis in cancer. We also aim to provide the reader with a basic understanding of NMR spectroscopy, cancer metabolomics, target profiling, chemometrics, and multifunctional tools for metabolomics discrimination, with a focus on metabolic phenotypic diversity for cancer therapeutics.

Abstract

Clinical biomarkers of building construction workers (BCW)-related genetic risk and metabolome fluctuations are still lacking, and profiling and validating them is challenging. Metabolomics is well suited to address this need, and serum is a valuable and accessible biofluid. This study aimed to characterize the 1H nuclear magnetic resonance (NMR)-based serum metabolomics and discrimination of patients with different clinical features of BCW (n = 30) and, healthy control (HC: n = 30). Karyotypes and chemical profiling were analyzed in all 60 subjects. The deproteinized serum samples was discriminated using orthogonal projections to latent structures discriminant analysis (OPLS-DA). With good quality metaphases, we performed in situ karyotyping techniques in blood cells, and identified Trisomy 21 (47, XY, +21), reciprocal translocation (45, XY, der (15;21)), and 7q11 microdeletion syndrome. Target profiling and quantification was performed for around 50 metabolites. With metabolic profiling technology, the chemical reaction between metabolites was measured. Moreover, a cytokinesis block micronucleus cytome assay considerable significant differences between micronuclei, nucleoplasmic bridges, and apoptotic and necrotic cell frequency with healthy voluntaries. Diversified chromosomal aberrations and chromatid breaks were observed in the BCW. The present investigation could contribute substantially to genetic risk and metabolites, with regard to the genotoxic effects in construction workers exposed to various genotoxic compounds.

Abstract

Titanium dioxide (TiO(2)) nanoparticles (NPs), first developed in the 1990s, have been applied in numerous biomedical fields such as tissue engineering and therapeutic drug development. In recent years, TiO(2)-based drug delivery systems have demonstrated the ability to decrease the risk of tumorigenesis and improve cancer therapy. There is increasing research on the origin and effects of pristine and doped TiO(2)-based nanotherapeutic drugs. However, the detailed molecular mechanisms by which drug delivery to cancer cells alters sensing of gene mutations, protein degradation, and metabolite changes as well as its associated cumulative effects that determine the microenvironmental mechanosensitive metabolism have not yet been clearly elucidated. This review focuses on the microenvironmental influence of TiO(2)-NPs induced various mechanical stimuli on tumor cells. The differential expression of genome, proteome, and metabolome after treatment with TiO(2)-NPs is summarized and discussed. In the tumor microenvironment, mechanosensitive DNA mutations, gene delivery, protein degradation, inflammatory responses, and cell viability affected by the mechanical stimuli of TiO(2)-NPs are also examined.
