The Co–Ge binary system was reassessed by CALPHAD method in view of new phase diagram data and drawbacks of the previous modeling. A three-sublattice model (Co,Va)1(Co)1(Ge)1 was used to describe the B82-type βCo5Ge3-phase based on its crystal structure. In order to describe the transformation between ordered L12-type phase (Co3Ge) and the disordered fcc_A1 phase (αCo), one single Gibbs energy function was used for both ordered and disordered phases. In almost all the previous thermodynamic calculations, the ordered phase with a negligible homogeneity range, such as Co3Ge, is treated as a stoichiometric compound. Such a treatment is not physically sound. For (αCo) and (eCo), the magnetic contribution to Gibbs energy is taken into account. Both substitutional solution model and associated model were applied to describe the liquid phase, and thus two sets of self-consistent thermodynamic parameters for this system were obtained. It was found that the associated model can account for the experimental data more satisfactorily than the substitutional solution model, especially for the partial enthalpy of mixing data for the liquid phase. Five representative as-cast Co-Ge alloys were prepared to compare the solidified microstructure with that predicted according to thermodynamic calculations. According to the calculated Scheil solidification curves of two key alloys, the solidified microstructure for the alloys was analyzed. Also, the calculated amounts of the solidified phases in five as-cast alloys are compared with the experimental data resulting from automatic image analysis of the BSE images, showing a good agreement between the calculation and experiment, in particular for the case in which the associated model is used to describe the properties of the liquid phase. It is demonstrated that the combined use of the thermodynamic calculation and decisive experiment is an efficient strategy to obtain the desired microstructure.

Ni and Zn are important alloying elements in Mg-based structural materials and hydrogen storage alloys. The isothermal section of the Mg-Ni-Zn system at 400 °C was investigated by means of X-ray diffraction (XRD) analysis and electron probe microanalysis (EPMA). The NiZn3 phase was observed to be stable in the as-cast and annealed Ni25Zn75 and Ni23Zn77 alloys. The experimental results confirmed the existence of the ternary τ compound at 400 °C. NiZn3, Mg2Zn3, Mg2Ni, MgZn2, MgNi2, and τ show significant homogeneity ranges in the ternary system. The Mg2Zn11 phase is being stabilized in the ternary system at temperatures higher than the melting point of this compound in the Mg-Zn binary system. In the frameworks of the CALPHAD approach, the Ni-Zn system was re-optimized and the thermodynamic description for the Mg-Ni-Zn system was thermodynamically assessed for the first time. The obtained thermodynamic models reasonably reproduce the experimental data on the phase equilibria and thermodynamic properties of the phases. The solidification paths of the representative Mg-Ni-Zn alloys were predicted within the Scheil-Gulliver model using the thermodynamic assessment performed in the present work. The results of the Scheil-Gulliver modeling are in agreement with the EPMA results.

The increasing number of man-made chemicals in the environment that may pose a carcinogenic risk highlights the need for developing reliable time- and cost-effective approaches for carcinogen detection and identification. To address this issue, we investigated the utility of high-throughput microarray gene expression and next-generation genome-wide DNA methylation sequencing for the in vitro identification of genotoxic and non-genotoxic carcinogens. Terminally differentiated and metabolically competent human liver HepaRG cells were treated at minimally cytotoxic concentrations of (i) the genotoxic human liver carcinogen aflatoxin B1 (AFB1) and its structural non-carcinogenic analog aflatoxin B2 (AFB2); (ii) the genotoxic human lung carcinogen benzo[a]pyrene (B[a]P) and its non-carcinogenic isomer benzo[e]pyrene (B[e]P); and (iii) the non-genotoxic liver carcinogen methapyrilene for 72 h and transcriptomic and DNA methylation profiles were examined. Treatment of HepaRG cells with the liver carcinogens AFB1 and methapyrilene generated distinct gene-expression profiles, whereas B[a]P had only a slight effect on gene expression. In contrast to transcriptomic alterations, treatment of HepaRG cells with the carcinogenic and non-carcinogenic chemicals resulted in profound changes in the DNA methylation footprint; however, the correlation between gene-specific DNA methylation and gene expression changes was minimal. Among the carcinogen-altered genes, transferrin (TF) emerged as sensitive marker for an initial screening of chemicals for their potential liver carcinogenicity. Potential liver carcinogens (i.e., chemicals causing altered TF gene expression) could then be subjected to gene-expression analyses to differentiate genotoxic from non-genotoxic liver carcinogens. This approach may substantially enhance the identification and assessment of potential liver carcinogens.

Cockayne syndrome protein B (CSB) is an ATP-dependent chromatin remodeler that relieves oxidative stress by regulating DNA repair and transcription. CSB is proposed to participate in base-excision repair (BER), the primary pathway for repairing oxidative DNA damage, but exactly how CSB participates in this process is unknown. It is also unclear whether CSB contributes to other repair pathways during oxidative stress. Here, using a patient-derived CS1AN-sv cell line, we examined how CSB is targeted to chromatin in response to menadione-induced oxidative stress, both globally and locus-specifically. We found that menadione-induced, global CSB–chromatin association does not require CSB's ATPase activity and is, therefore, mechanistically distinct from UV-induced CSB–chromatin association. Importantly, poly(ADP-ribose) polymerase 1 (PARP1) enhanced the kinetics of global menadione-induced CSB–chromatin association. We found that the major BER enzymes, 8-oxoguanine DNA glycosylase (OGG1) and apurinic/apyrimidinic endodeoxyribonuclease 1 (APE1), do not influence this association. Additionally, the level of γ-H2A histone family member X (γ-H2AX), a marker for dsDNA breaks, was not increased in menadione-treated cells. Therefore, our results support a model whereby PARP1 localizes to ssDNA breaks and recruits CSB to participate in DNA repair. Furthermore, this global CSB–chromatin association occurred independently of RNA polymerase II–mediated transcription elongation. However, unlike global CSB–chromatin association, both PARP1 knockdown and inhibition of transcription elongation interfered with menadione-induced CSB recruitment to specific genomic regions. This observation supports the hypothesis that CSB is also targeted to specific genomic loci to participate in transcriptional regulation in response to oxidative stress.

A strong 8–9 MHz noise from the RF plasma heating generators forms additional difficulties in the diagnostic signal measurements in the U-3M and U-2M torsatrons compared with other stellarators and tokamaks. Set of new optical diagnostics (20-channels SXR and bolometers, 12- channels H α ) with a temporal resolution of up to 4 microseconds has been designed and is successfully used in the noisy RF plasma conditions of U-3M and U-2M torsatrons as well as in the QSPA Kh-50 plasma accelerator. In the original electronics design all the diagnostics components, including the STM-32-microporcessor-based data acquisition system, are assembled in a single unit (without analog signals or digital interface wires). This design allows one to prevent the ground loop interference, to remove the RF load on diagnostics components and to increase substantially the system protection from different electromagnetic noises.

The liver, a central detoxification organ and main regulator of systemic iron homeostasis, is prone to damage by xenobiotics. In the present study, we investigated the effect of the hepatotoxicant and hepatocarcinogen methapyrilene hydrochloride on iron metabolism in rat liver in a repeat-dose in vivo toxicity study and in human HepaRG cells in vitro. Treatment of male Fischer 344 (F344) rats with methapyrilene at doses 40 and 80mg/kg body weight (bw)/day by gavage for 6 weeks resulted in changes in the expression of classic hepatotoxicity-related marker genes and iron homeostasis-related genes, especially a prominent, dose-dependent down-regulation of the transferrin (Tf) gene and an up-regulation of the ferritin, light chain (Ftl) gene. A decrease in the level of TF and an increase in the level of FTL also occurred in methapyrilene-treated differentiated HepaRG cells, indicating the existence of interspecies and in vitro-in vivo similarities in the disturbance of cellular iron homeostasis upon liver injury. In contrast, there was minimal overlap in the expression of liver toxicity-marker genes in the livers of rats and in HepaRG cells treated with methapyrilene. Importantly, the decrease of transferrin at mRNA and protein levels occurred after the treatment with a low dose of methapyrilene that exhibited minimal cytotoxicity. These results demonstrate the significance of the dysregulation of hepatic iron metabolism in the pathogenesis and mechanism of chemical-induced liver toxicity and suggest that these changes may be sensitive and useful indicators of potentially hepatotoxic chemicals.

Excessive alcohol consumption has a significant impact on human health and is a major public health problem worldwide. One of the consequences of long-term excessive alcohol consumption is cellular injury in almost all organs and tissues, with acute kidney injury (AKI) being one of the most common pathological manifestations. In the present study, using a mouse model of alcoholic liver fibrosis-associated AKI induced by a combined treatment with carbon tetrachloride (CCl4) and ethanol and resembling pathological features of AKI in human alcoholic liver fibrosis, we demonstrate alterations in histone modifications in the kidneys and, importantly, in the promoter region of the over-expressed SRY (sex determining region Y)-box 9 (Sox9) gene. The level of SOX9 protein in the kidneys of AKI-mice is reduced and correlates inversely with increased expression of microRNA miR-1247. Mechanistically, the over-expression of miR-1247 is associated with a markedly increase in histone H3 lysine 4 trimethylation in the upstream region of the Mir1247 gene. The results of the present study demonstrate a functional role of epigenetic mechanisms in AKI and indicate the importance of correcting the epigenetic dysregulation for proper renal tubule maintenance and repair.

The enthalpies of mixing of liquid Ni–Ti–Zr alloys were determined at 1873 K by means of a high-temperature isoperibolic calorimeter. The investigations were performed along sections x Ni / x Ti = 4/1 and x Ni / x Zr = 4/1 in the corresponding composition ranges x Zr = 0–0.41 and x Ti = 0–0.48. The limiting partial enthalpy of mixing of undercooled liquid zirconium in liquid Ni–Ti alloy is (−192 ± 8) kJ mol −1 . The limiting partial enthalpy of mixing of undercooled liquid titanium in liquid Ni–Zr alloy is (−130 ± 12) kJ mol −1 . The integral mixing enthalpies are negative over the investigated composition range. An analytical expression for the representation of the integral mixing enthalpy was obtained and used for the calculation of the function in the whole composition range.

A thermodynamic analysis of the phase relations in the Na Sb Sn system was undertaken by applying the CALPHAD method and first principle calculations. The CALPHAD method was used for the optimization of the thermodynamic parameters of the binary and ternary phases, as well as for the calculation and prediction of the phase relations in the systems. The electronic structure calculations were performed using density functional theory to get a measure of formation enthalpies of binary and ternary compounds. New thermodynamic assessments are provided for the Na Sb and Na Sn systems. The formation of new compounds, NaSbSn, Na 3 Sb 3 Sn, and NaSb 2 Sn 3 , in the Na Sb Sn system was proposed.

Thermodynamic optimization of ternary phase diagrams via Calphad approach is a complex procedure. The success and quality of such a Calphad optimization depend on the reliability of the experimental data and on the scrutiny of the critical evaluation of the experimental datasets. With this in mind, we provide a set of recommendations that might facilitate the critical evaluation procedure and improve the quality of the thermodynamic datasets for the calculations of the phase diagrams. The particulars regarding the consistency between binary and ternary phase diagrams as well as the internal agreement between the different ternary datasets, are discussed.

The thermodynamic description of the Al 2 O 3 –Fe 2 O 3 –FeO system has been developed considering newly available experimental data, as well as future compatibility of the present modeling with available descriptions of the Al–Fe, Al–O, and Fe–O systems. All of the experimental data available in the literature have been critically reviewed, and the inconsistent information has been excluded from the optimization. The sublattice model was applied to spinel phase to describe the degree of inversion in FeAl 2 O 4 –Fe 3 O 4 solid solutions as well as an extension of homogeneity range in spinel by the dissolution of Al 2 O 3 and Fe 2 O 3 . The thermodynamic parameters have been evaluated to describe the experimental data in the oxide part of the system.

The enthalpies of mixing of liquid (Co + Cu + Zr) alloys have been determined using the high-temperature isoperibolic calorimeter. The measurements have been performed along three sections (xCo/xCu = 3/1, 1/1, 1/3) with xZr = 0 to 0.55 at T = 1873 K. Over the investigated composition range, the partial mixing enthalpies of zirconium are negative. The limiting partial enthalpies of mixing of undercooled liquid zirconium in liquid (Co + Cu) alloys are (−138 ± 18) kJ · mol−1 (the section xCo/xCu = 3/1), (−155 ± 10) kJ · mol−1 (the section xCo/xCu = 1/1), and (−130 ± 22) kJ · mol−1 (the section xCo/xCu = 1/3). The integral mixing enthalpies are sign-changing. The isenthalpic curves have been plotted on the Gibbs triangle. The main features of the composition dependence of the integral mixing enthalpy of liquid ternary alloys are defined by the pair (Co + Zr) and (Cu + Zr) interactions.

A novel circuit for compact torus (CT) injector operated at high repetition rates has been developed. The core technology adopted in the present work is to charge a large storage capacitor bank and quickly charge the CT capacitor bank through a stack of insulated-gate bipolar transistors (IGBTs). A system consisting of IGBTs and slow banks for the repetitive operation has been developed and installed for each discharge circuit of the University of Saskatchewan Compact Torus Injector (USCTI). A repetition rate up to 1.7 Hz and a burst of 8 CTs have been achieved.

The thermodynamic description of the Cu–Fe–Ni system has been updated considering the newly available experimental data, as well as compatibility of the present modeling with those used for the Cu and Fe systems. All of the experimental data available in the literature have been critically reviewed, and the inconsistent information has been excluded. The thermodynamic parameters have been evaluated in order to properly describe the thermodynamic properties of the liquid phase and miscibility gap in the solid state. A significant improvement in comparison with the previous thermodynamic descriptions has been achieved. Additionally, for the ordered L1 2 phase the four-sublattice model has been adopted to predict the ternary phase equilibria involving this phase. A set of thermodynamic parameters for the phases is given.

The partial enthalpy of mixing of hafnium in liquid Fe–Hf alloys has been investigated at 1873 K in the composition range x Hf = 0−0.06. The limiting partial enthalpy of mixing of undercooled liquid hafnium in liquid iron reaches Δ mix H ‾ Hf ∞ = −128 ± 3 kJ/mol. Composition dependence of the partial mixing enthalpy of hafnium has been represented by the following equation (in kJ/mol): Δ mix H ‾ Hf = - 128 ( 1 - x Hf ) 2 at 0 ⩽ x Hf ⩽ 0.10 . Considerable negative enthalpies of mixing of hafnium with iron indicate preferable interactions of dissimilar atoms in the liquid alloys.

Resume Introduction Le syndrome des vestibulites vulvaires (VVS) est une maladie frequente qui, caracterisee par une dyspareunie superficielle, touche la femme jeune et entraine un fort retentissement psychologique. Les descriptions anatomopathologiques de la litterature sont controversees. Malades et methodes Nous avons etudie d’un point de vue anatomoclinique 14 femmes jeunes souffrant de VVS. Nous avons realise chez ces femmes des biopsies vestibulaires au pourtour de l’orifice d’abouchement de la glande de Bartholin et les avons etudiees selon 25 plans de coupe paralleles apres inclusion et coloration standard et par le PAS. Resultats Les femmes etaient âgees de 26 ans en moyenne et le VVS evoluait depuis 30 mois en moyenne. Nous avons constate un important infiltrat inflammatoire du chorion de la muqueuse vestibulaire constitue majoritairement de cellules mononucleees. Il existait une disposition remarquable de cet infiltrat inflammatoire, periglandulaire, avec parfois des images d’exocytose au sein de l’epithelium glandulaire et des canaux excreteurs. Discussion La majorite des etudes rapportent, chez les femmes porteuses d’un VVS, l’existence d’un infiltrat inflammatoire chronique de la muqueuse vestibulaire vulvaire, predominant autour des glandes vestibulaires mineures. Nous avons pu constater le meme type d’image dans notre etude. L’existence d’une exocytose des cellules inflammatoires dans l’epithelium des glandes vestibulaires mineures et/ou dans leurs canaux excreteurs n’a jamais ete decrite et apporte un argument supplementaire pour l’individualisation de l’entite VVS.

New applications of microarray technology requires new approaches to microarray reader development. Miniaturization of the biochips, necessity of massive parallel microarray-based analysis of various biological samples as well as increasing application of different real-time on-chip reactions for research and diagnostics demand improvement of microarray reader instrumentation. We represent the optical scanning array (OSA) reader system, allowing fast capture of high-resolution multifluorescence images and equipped with thermocontrolled hybridization chamber which is required for registration of a wide range of on-chip real-time processes. We demonstrate the application of OSA microarray reader for optimisation of hybridization and washing conditions for microarray-based detection of Q493X mutation of CFTR gene. It was shown that detailed analysis of on-chip thermodynamic and kinetic behaviour of oligonucleotide/oligonucleotide and ssPCR/oligonucleotide duplexes is very helpful for optimisation of experimental procedure.

An advanced heavy ion beam probing diagnostic (HIBP) based on simultaneous utilization of two different systems for the secondary ions detection (a multiple cell array detector and 30° Proca-green electrostatic energy analyzer) has been developed for the TJ-II flexible heliac. This innovative design aims at enlarging the HIBP capabilities in order to allow the instantaneous measurements of electron density and plasma potential profiles together with their respective fluctuations. This paper presents the detailed description of the main parts of the HIBP equipment and their characteristics obtained during the first operation on TJ-II. Special attention is employed to the control and data acquisition system built on two VME controllers. The results of the diagnostic beam carrying through the magnetic structure of TJ-II into secondary beam line are presented and compared with the trajectory calculations. The problems of plasma loading of the detection system and HX-ray excitement by the probing beam are considered together with the ways of their solution.

This work presents the results of T-10 SOL investigations with multipin Langmuir probes and correlation reflectometry. A special regime with a rail limiter deeply introduced from the bottom of the machine at SOL temperatures of up to 50 eV has been studied. Five types of plasma small scale turbulence were observed in the SOL, including the three previously found types of core turbulence. It was shown that the main SOL parameters are poloidally uniform. A high poloidal asymmetry of the fluctuations and the turbulent flux of particles, was found.

However, this method has its strengths and weaknesses. One of strengths is the ability to model almost any managerial situations. This article covers the methodology and specificity of organization and carrying out the simulation game, 'The formation of the indicators to measure the quality of municipal management of social sphere.' This topic is of particular relevance due to the difficulties of the practical assessment and measurement of the social effectiveness of public and municipal administration.
