- Abstract
An initial concept for the plasma diagnostic and control (D&C) system has been developed as part of European studies towards the development of a demonstration tokamak fusion reactor (DEMO). The main objective is to develop a feasible, integrated concept design of the DEMO D&C system that can provide reliable plasma control and high performance (electricity output) over extended periods of operation. While the fusion power is maximized when operating near to the operational limits of the tokamak, the reliability of operation typically improves when choosing parameters significantly distant from these limits. In addition to these conflicting requirements, the D&C development has to cope with strong adverse effects acting on all in vessel components on DEMO (harsh neutron environment, particle fluxes, temperatures, electromagnetic forces, etc.). Moreover, space allocation and plasma access are constrained by the needs for first wall integrity and optimization of tritium breeding. Taking into account these boundary conditions, the main DEMO plasma control issues have been formulated, and a list of diagnostic systems and channels needed for plasma control has been developed, which were selected for their robustness and the required coverage of control issues. For a validation and refinement of this concept, simulation tools are being refined and applied for equilibrium, kinetic and mode control studies.

To assess the prevalence of age-related macular degeneration (AMD) in a Russian population.Population-based prevalence assessment.The Ural Eye and Medical Study was conducted in a rural and urban area in the Russian republic of Bashkortostan. The study included 5,899 participants aged 40+ years old. AMD, defined according to the Beckman Initiative for Macular Research, was assessed by fundus photographs and optical coherence tomographic images of 4,932 (83.6%) participants.The prevalence of any AMD, early AMD, intermediate AMD, or late AMD, geographic atrophy, and neovascular AMD were 18.2% (95% confidence interval [CI], 16.8-19.6), 11.6% (95% CI, 10.4-12.8), 5.0% (95% CI, 4.2-5.8), 1.6% (95% CI, 1.1-2.0), 0.7% (95% CI, 0.4-1.0) and 0.9% (95% CI, 0.6-1.3), respectively, for individuals55 years old. Applying an age limit of 40+ years for the AMD definition, prevalence of any AMD, early AMD, intermediate AMD, late AMD, geographic atrophy and neovascular AMD were 14.1% (95% CI, 13.1-15.1), 9.4% (95% CI, 8.6-10.2), 3.8% (95% CI, 3.2-4.3), 1.0% (95% CI, 0.7-1.2), 0.4% (95% CI, 0.2-0.6) and 0.5% (95% CI, 0.3-0.7), respectively, for individuals aged 40+ years. Higher AMD prevalence was correlated with older age (odds ratio [OR], 1.15; 95% CI, 1.13-1.16; P0.001), rural region (OR, 1.69; 95% CI, 1.32-2.17; P0.001), lower diabetes prevalence (OR, 0.56; 95% CI, 0.38-0.82; P = 0.003), and shorter axial length (OR, 0.89; 95% CI, 0.79-0.99; P = 0.04). AMD prevalence was not significantly (all P ≥ 0.20) correlated with any systemic parameter examined, except for lower prevalence of diabetes.In this typical, ethnically mixed, urban and rural population from Russia, a higher prevalence for AMD was associated mainly with older age, rural region of habitation, shorter axial length, and lower prevalence of diabetes mellitus. The AMD prevalence was lower than in Europeans and higher than in East Asians.

Block-structured mesh generation techniques have been well addressed in the CFD community for automobile and aerospace studies, and their applicability to magnetic fusion is highly relevant, due to the complexity of the plasma-facing wall structures inside a tokamak device. Typically applied to non-linear simulations of MHD instabilities relevant to magnetically confined fusion, the JOREK code was originally developed with a 2D grid composed of isoparametric bi-cubic Bezier finite elements, that are aligned to the magnetic equilibrium of tokamak plasmas (the third dimension being represented by Fourier harmonics). To improve the applicability of these simulations, the grid-generator has been generalised to provide a robust extension method, using a block-structured mesh approach, which allows the simulations of arbitrary domains of tokamak vacuum vessels. Such boundary-aligned grids require the adaptation of boundary conditions along the edge of the new domain. Demonstrative non-linear simulations of plasma edge instabilities are presented to validate the robustness of the new grid, and future potential physics applications for tokamak plasmas are discussed. The methods presented here may be of interest to the wider community, beyond tokamak physics, wherever imposing arbitrary boundaries to quadrilateral finite elements is required. - Abstract
Neutron activation technique has been widely used for the monitoring of neutron fluence at the Joint European Torus (JET) whereas it is foreseen to be employed at future fusion plants, such as ITER and DEMO. Neutron activation provides a robust tool for the measurement of neutron fluence in the complex environment encountered in a tokamak. However, activation experiments previously performed at JET showed that the activation foils used need to be calibrated in a real fusion environment in order to provide accurate neutron fluence data. Triggered by this challenge, an improved neutron activation method for the evaluation of neutron fluence at fusion devices has been developed. Activation assemblies similar to those used at JET were irradiated under 14 MeV neutrons at the Frascati Neutron Generator (FNG) reference neutron field. The data obtained from the calibration experiment were applied for the analysis of activation foil measurements performed during the implemented JET Deuterium-Deuterium (D-D) campaign. The activation results were compared against thermoluminescence measurements and a satisfactory agreement was observed. The proposed method provides confidence on the use of activation technique for the precise estimation of neutron fluence at fusion devices and enables its successful implementation in the forthcoming JET Deuterium-Tritium (D–T) campaign.

Recently, we identified the yeast red pigment (RP), a polymer of 1-(5'-Phosphoribosyl)-5-aminoimidazole, as a novel potential anti-amyloid agent for the therapy of neurodegenerative diseases. The purpose of this study was to further validate RP for treatment of Parkinson's disease (PD) and to clarify molecular mechanisms involved in the reduction of amyloid cytotoxicity. We investigated RP effects in vivo using Saccharomyces cerevisiae and Drosophila melanogaster PD models. Western blot analysis revealed reduction in the levels of insoluble α-synuclein in both models, while soluble α-synuclein decreased only in Drosophila. In both models RP significantly reduced α-synuclein cytotoxicity, as was revealed by immunohistochemistry in Drosophila (p 0.001, n = 27 flies per genotype/assay) and by flow cytometry in yeast (p 0.05). Data obtained from the yeast PD model suggests that RP antitoxic effects are associated with a drop in ROS accumulation, and slower cellular transition from the early to late apoptotic stage. Using Drosophila brain tissue sections, we have demonstrated that RP helps to compensate for an α-synuclein-mediated reduction in the number of dopaminergic neurons and leads to better performance in animal climbing tests (p 0.001, n = 120-150 flies per genotype/assay). Taken together, these results demonstrate the potential of RP for the treatment of PD, at least in model systems.

International audience; Rare copy-number variation (CNV) is an important source of risk for autism spectrum disorders (ASDs). We analyzed 2,446 ASD-affected families and confirmed an excess of genic deletions and duplications in affected versus control groups (1.41-fold, p = 1.0 × 10(-5)) and an increase in affected subjects carrying exonic pathogenic CNVs overlapping known loci associated with dominant or X-linked ASD and intellectual disability (odds ratio = 12.62, p = 2.7 × 10(-15), ∼3% of ASD subjects). Pathogenic CNVs, often showing variable expressivity, included rare de novo and inherited events at 36 loci, implicating ASD-associated genes (CHD2, HDAC4, and GDI1) previously linked to other neurodevelopmental disorders, as well as other genes such as SETD5, MIR137, and HDAC9. Consistent with hypothesized gender-specific modulators, females with ASD were more likely to have highly penetrant CNVs (p = 0.017) and were also overrepresented among subjects with fragile X syndrome protein targets (p = 0.02). Genes affected by de novo CNVs and/or loss-of-function single-nucleotide variants converged on networks related to neuronal signaling and development, synapse function, and chromatin regulation.

Sigma receptors are usually classified as a separate class of intracellular receptors. Among them the sigma-1 receptor has been the most studied regarding its pharmacological applications. This receptor with average or high affinity binds a wide range of chemical compounds of very different structural classes and a variety of therapeutic and pharmacological properties. The sigma-1 receptor is a trans-membrane protein placed in the endoplasmic reticulum (ER), which regulates the function of inositol-3-phosphate receptor, stabilizing the calcium signaling between ER and mitochondria. There are studies that the sigma-1 receptor is involved in the formation of many neurological and psychiatric conditions. It is assumed that the sigma-1 receptor acts as a sensor of normal calcium operation. The studies over the recent years have shown the role of the violation in calcium signaling in the pathogenesis of Alzheimer's and Huntington's diseases. In particular, changes in calcium homeostasis of the endoplasmic reticulum lead to the break of synaptic connections in the neurons. Thus, the sigma-1 receptor holds promise in application as a potential therapeutic target for the treatment of neuropathological diseases.

A generic time-invariant polling system is modeled as a deterministic hybrid dynamical system in the form of a fluid fabrication facility. The system consists of finitely many infinite size buffers receiving constant-rate inflows from outside the system and a single source of service (finite capacity server). The server can withdraw the buffer’s content but is able to serve at most one buffer at a time, and has to switch among them from time to time, with any switch consuming a nonzero switch-over period. With respect to the long-run maximal scaled wip (work in progress) performance metric, sufficiency of periodic control protocols is established: It is shown that the deepest optimum (that is over all processes in the system, irrespective of their genesis, initial state, and features) is furnished by such a protocol with as high precision as desired. Moreover, sufficiency of special periodic control protocols is established. These protocols prescribe to serve any buffer at the maximal rate until its size reduces to a pre-specified percent of its initial size at the beginning of the visit. This percent is set to be zero for any buffer such that its service at the maximal rate implies reduction of the scaled wip. This is related to another result of the paper that shows optimality of the exhaustive policy for such buffers.

Transverse and longitudinal magnetoresistance in InSb whiskers with different impurity concentration (4.4×1016–7.16×1017) сm−3 were studied at temperature range (4.2–77) K and in magnetic fields (0–14) T. Shubnikov-de Haas oscillations in magnetoresistance for n-type InSb whiskers with impurity concentration in the vicinity to the metal–insulator transition (MIT) at low temperature were revealed. Every peak of longitudinal and transverse magnetoresistance (MR) split as the doublet for concentration 3.26×1017 сm−3. The giant g-factor was defined, that rises from 46 to 60 with increasing of the magnetic field up to 14 T. The oscillation period differs in the range (0.095–0.11) T−1 at various doping concentration in InSb whiskers. The effective mass of electrons mс=0.03m0 and Dingle temperature with range (3–12) K were found in InSb whiskers with impurity concentration in the vicinity to MIT. The observed giant magnetoresistance (increasing in order of magnitude in magnetic field induction 14 T) was used to design the sensitive sensor of magnetic field induction operated at the range of cryogenic temperatures.

Derivatives of phaeosphaeride A (PPA) were synthesised and characterised; then anti-cancer studies were carried out on the A549 cancer cell line. It was found that the acetyl derivative (compound 3) displayed comparable in vitro cytotoxicity to that of PPA (EC50=49±7 μM and EC50=46±5 μM, respectively), while chloroacetyl derivative 6 (EC50=33±7 μM) was found to have better efficacy towards the A549 cancer cell line.

The herein synthesized 7-(4-methylphenyl)thiomethyl and 7-morpholylmethyl derivatives of natural phaeosphaeride A have lower cytotoxic activity towards the human lung adenocarcinoma A549 cell line compared to phaeosphaeride A. The presence of exocyclic C=C bond and N–OMe group in such compounds is suggested to be a factor in their antitumor activity.

A new method was developed to measure the unsteady force measurement by means of six-component strain gauge balance which finds use in technological processes, e.g. in wind tunnel experiments. A mathematical model in the form of damping spring was used as a core of the method. It was verified by results of experiments involving applying harmonic and step force. Applicability of the balance matrix defined in static conditions was validated by an experiment with harmonic steady-state oscillations of the balance. The developed methodology provides the measurement of arbitrary non-stationary load in inertial or non-inertial system using a strain gauge balance with relative deviation at the level of 0.3–3% in the whole balance’ operating frequency range, including the natural frequency. A dynamic calibration method was developed for the axial force of a six-component strain gauge balance by applying a harmonic force to the ground part of the balance.
