Hematopoietic stem/progenitor cell gene therapy (HSPC-GT) has shown clear neurological benefit in rare diseases, which is achieved through the engraftment of genetically modified microglia-like cells (MLCs) in the brain. Still, the engraftment dynamics and the nature of engineered MLCs, as well as their potential use in common neurogenerative diseases, have remained largely unexplored. Here, we comprehensively characterized how different routes of administration affect the biodistribution of genetically engineered MLCs and other HSPC derivatives in mice. We generated a high-resolution single-cell transcriptional map of MLCs and discovered that they could clearly be distinguished from macrophages as well as from resident microglia by the expression of a specific gene signature that is reflective of their HSPC ontogeny and irrespective of their long-term engraftment history. Lastly, using murine models of Parkinson's disease and frontotemporal dementia, we demonstrated that MLCs can deliver therapeutically relevant levels of transgenic protein to the brain, thereby opening avenues for the clinical translation of HSPC-GT to the treatment of major neurological diseases.

Background RCTs that have shown improvement in coefficient of fat absorption with pancreatic enzyme replacement therapy (PERT) have seldom evaluated the impact on overall nutritional status. Objective In this study we evaluated factors responsible for persistence of malnutrition after PERT. Methods In this cross-sectional observational study, patients were enrolled based on predefined enrolment criteria. Patients were divided into those taking PERT regularly (Group A), irregularly (Group B) and not taking (Group C) for at least 3 months. Comprehensive evaluation of anthropometric measurements, nutritional assessment and dietary intake was performed. Malnutrition was measured using the Subjective Global Assessment (SGA) tool. Relationship between PERT status, dietary intake and nutritional status were evaluated using standard statistical methods. Logistic regression was performed to identify factors associated with persistence of malnutrition after PERT. Results 377 patients with CP and 50 controls were included. 95 (25.2%) patients with CP were in Group A, 106 (28.1%) in Group B and 176 (46.7%) in Group C. 130 (34.5%) patients were malnourished, of which 76 (58.5%) were continuing PERT. There were no differences in clinical and biochemical nutritional markers between Groups A, B, and C. Calorie deficit and daily intake of calorie, protein, carbohydrates and fats were not different between those with and without PERT, but was significantly less in those with malnutrition. Logistic regression demonstrated inadequate dietary intake as independent risk factor for persistence of malnutrition. Conclusion Even though PERT is effective in PEI, comprehensive nutritional assessment, personalized nutritional counselling and therapy along with PERT is mandatory.

Cytokines are proteins that play a critical role in the development, maturation, and functional activities of immune cells. For the first time, we have investigated the potential role of Terminalia arjuna as IL-4 and IL-13 modulators for preventing T1DM, i.e., autoimmune diabetes. It has been well documented that the stimulation of IL-4 and IL-13 can regulate the level of type 2 cytokines which can be maintained with the level of type 1 cytokines. In the present study, gallic acid, arjunolic acid, luteolin, ellagic acid, and arjunone were investigated for their potential modulating activity of IL-4 and IL-13. The active amino acid residues identified for IL-4 are VAL51, HIS58, ASP87, THR30, GLN54, THR63, ARG64, LYS84, and GLU60. The active amino acid residues identified for IL-13 are H: GLU46, H: TRP47, H: GLN61, L: PHE98, L: VAL97, L: GLU162, L: THR163, H: ARG105, L: GLN38, L: ASP85, H: GLY42, L: GLY41, H: PRO41, H: TRP47, and L: PHE98. The phytoconstituents demonstrated better modulating activity towards IL-13 than IL-4. Luteolin displayed better potential for both IL-4 and IL-13, and therefore we concluded that it could be used to modulate the activity of IL-4 and IL-13 for the prevention of autoimmune diabetes.

Two polyesters namely poly (glycerol adipate) (PGA) and 4-(1-(4-methoxyphenyl) cyclohexyl) phenyl 6-oxoheptanoate (MPOHP) were synthesised in a facile manner and characterised using Fourier transform infrared spectroscopy (FT-IR) and nuclear magnetic spectroscopy (NMR). The synthesised polyesters were subjected to evaluate the mild steel protecting ability at various concentrations of 10, 50, 100, 500, 1000 ppm in 0.5 M H2SO4 medium. A precise comparison of metal protecting ability rendered by both aliphatic and aromatic moiety was analysed. As an initiation, gravimetric measurements carried out at room temperature resulted with increased inhibition efficiency with increased concentration. Higher temperature studies suggested physisorption mechanism resulting with decreased inhibition efficiency. Experimental results were best fitted with Langmuir adsorption isotherm model. Electrochemical impedance measurements carried out for the selected concentrations of the inhibitors (10, 100, 1000 ppm) revealed increased charge transfer resistance (Rct) with a maximum inhibition efficiency of 54.07% (PGA) and 69.92% (MPOHP) at 1000 ppm indicating the formation of protective barrier. Potentiodynamic polarisation method suggested mixed type of inhibition of both PGA and MPOHP on the metal surface resulting in decreased metal dissolution that could be evidenced from lowered corrosion current density (Icorr).

As a motto of framing the strategies to minimise the deterioration in steel reinforced concrete structures as well as the fact that the work carried out with polymers are rare and scanty, polyesters namely Poly (Glycerol azealate) (PGAZ) and 4-(1-(4-methoxy phenyl) cyclo hexyl)phenyl 9-oxodecanoate (MPOD) were synthesised and evaluated for rebar corrosion in simulated concrete pore solution (SCP) for the first time at selected concentrations of 10, 100, 1000 ppm levels with an aid of electrochemical impedance and potentiodynamic polarisation techniques. Nyquist plots favoured increased Rct values suggesting the formation of barrier. Tafel plots represented cathodic inhibition. The experimental datas were best fitted with Langmuir adsorption isotherm facilitating monolayer adsorption. Both the methods employed were in good agreement with each other favouring maximum inhibition efficiency of 71.81% for MPOD and minimum of 57.64% for PGAZ primarily due the difference in aromatic sites as revealed in mechanism.

Current review survey encompasses the development circumstances of four major recycling industries in Iran and over the world. Thus, evaluation of the individual techniques exploited in the four recycling industries indicated that employed processes had the lowest economic outlay and the highest revenue in comparison to other technologies. Additives can be utilized as raw materials for production of economically valuable bituminous products. The recycling and converting processes are not only cost-effective, but they also descent the environmental risk to a large level, using new techniques to zero. Economic indices, business excellence score and the quality of the obtained products represented that these processes support sustainable development of industries.

Saffron (Crocus sativus L.) is an important plant in medicine. The Kashmir Valley (J&K, India) is one of the world's largest and finest saffron producing regions. However, over the past decade, there has been a strong declining trend in saffron production in this area. Plant Growth Promoting Rhizobacteria (PGPR) are free living soil bacteria that have ability to colonize the surfaces of the roots and ability to boost plant growth and development either directly or indirectly. Using the efficient PGPR as a bio-inoculant is another sustainable agricultural practice to improve soil health, grain yield quality, and biodiversity conservation. In the present study, a total of 13 bacterial strains were isolated from rhizospheric soil of saffron during the flowering stage of the tubers and were evaluated for various plant growth promoting characteristics under in vitro conditions such as the solubilization of phosphate, production of indole acetic acid, siderophore, hydrocyanic acid, and ammonia production and antagonism by dual culture test against Sclerotium rolfsii and Fusarium oxysporum. All the isolates were further tested for the production of hydrolytic enzymes such as protease, lipase, amylase, cellulase, and chitinase. The maximum proportions of bacterial isolates were gram-negative bacilli. About 77% of the bacterial isolates showed IAA production, 46% exhibited phosphate solubilization, 46% siderophore, 61% HCN, 100% ammonia production, 69% isolates showed protease activity, 62% lipase, 46% amylase, 85% cellulase, and 39% showed chitinase activity. Three isolates viz., AIS-3, AIS-8 and AIS-10 were found to have the most plant growth properties and effectively control the growth of Sclerotium rolfsii and Fusarium oxysporum. The bacterial isolates were identified as Brevibacterium frigoritolerans (AIS-3), Alcaligenes faecalis subsp. Phenolicus (AIS-8) and Bacillus aryabhattai (AIS-10) respectively by 16S rRNA sequence analysis. Therefore, these isolated rhizobacterial strains could be a promising source of plant growth stimulants to increase cormlets growth and increase saffron production.

This research propose a VLSI based temperature to digital converter (TDC) which is implemented with the aid of CMOS technology whose precise functionality is to sense the temperature-dependent source-gate voltage (VSG) of PMOS Transistor. It measures an inaccuracy of ±40 mK on 20 samples from one batch ranging between −60 °C to +60 °C temperature. This result is mainly due to the generation of pmos transistors drain currents which minimizes the spread in their gate to source voltages in addition to a PTAT digital circuit in support of the proper stretch in the pmos transistors emitter currents. The entire design has been implemented in 0.045 µm technology and draws a current of 0.3 µA which shows that the proposed TDC can be considered as ultra-low power device as the power consumed by it is 18.09E-12 W between the room temperatures of −60 °C to +60 °C. As per the accuracy of the proposed TDC the calibration cost can be reduced which would avoid the difficulty in the batch calibration. The supply voltage given to the device is 0.45 volts and a temperature inaccuracy of ±18 mk is achieved.

Bisbenzimidazo quinoline receptor 1 bearing diethylene oxide spacer linkage was synthesized and characterized by spectral and analytical techniques. Metal binding properties of the receptor 1 in aqueous ethanol medium were studied through absorption, emission and electrochemical studies. Emission analyses were performed at higher excitation wavelength ( λ ex 750 nm). Binding studies exhibited specific “turn-on-type” fluorescent enhancement character of the receptor 1 for the Fe 2+ ion (K a = 6.67 × 10 5 M –1 ). The appreciable metal binding property of the receptor 1 was theoretically studied by the quantum mechanical calculation. In the illustration of metal binding mechanism three binding modes A , B and C for the receptor 1 with Fe 2+ /Fe 3+ ions were proposed and the active molecular sites were analyzed using DFT descriptors, fukui functions and molecular electron density map (MEP). Theoretically results were correlated to the experimentally observed results.
