Water use traits in lichens are important attributes that determine the duration of hydration and metabolically active periods. In this study, the water holding capacity (WHC) and specific thallus mass (STM) were measured for seven macrolichen species (Parmelia sulcata , Parmelina tiliacea , Evernia prunastri , Ramalina farinacea , Lobaria pulmonaria , Lobarina scrobiculata and Nephroma resupinatum) from a Temperate-Mediterranean fragmented landscape. Twenty Quercus forests with different environmental conditions were selected to gather therein five lichen samples of each species in order to analyse their interspecific and intraspecific variation in WHC and STM. The type of photobiont was mainly responsible for differences in the WHC and the water content per biomass among species. Lichens with cyanobacteria as the main or secondary photobiont showed the highest value for both parameters. However, particular features of species were more important in modulating STM, while growth form had a minor explanatory importance. At the intraspecific level, variation in WHC relied on climatic, microclimatic and forest structure factors, while STM variability was only dependent on the two last predictors. Future research should be focused on other drivers at the microscale to unveil the environmental conditions that shape WHC and STM in lichens. • The type of photobiont primarily shapes water holding capacity in macrolichens. • Specific thallus mass in macrolichens depends on particular features of species. • Growth forms are not so relevant to explain interspecific variability in macrolichens. • The water content per biomass relies on the type of photobiont. • Intraspecifically, water use traits primarily respond to microscale factors. Secondary metabolites of lichens can be involved in production of chelates with heavy metals. We hypothesized that parietin plays important role in protection of photobiont cells in Xanthoria parietina from an excess of cadmium ions. Two types of X. parietina lichen thalli, natural with presence of secondary metabolite parietin (p+) as well as without parietin (p−) were exposed to different doses of cadmium (up to 300 μmol g −1 dw). Based on determination of the total and intracellular Cd-accumulation, ergosterol and thiobarbituric acid reactive substances (TBARS) content did not show statistically significant differences in the response of both types of thalli (p+ and p−). However, a stronger toxic effect of the highest Cd-dose on photosynthetic pigment content and chlorophyll a fluorescence was observed in the parietin-depleted thalli. The protective role of parietin against Cd excess was better supported and concluded from the differences observed in the production of non-protein thiol compounds (cysteine, glutathione and phytochelatins) involved in Cd detoxification. In the p+ thalli Cys content was stable but GSH content slightly decreased in the studied Cd range, while in the p− thalli these compounds were completely absent at high Cd doses. At Cd doses higher than 37.5 μmol Cd g −1 dw, toxic to both types of X. parietina thalli, Cys and GSH contents were significantly higher in p+ than in p− thalli. Also, the photobiont partner in the p+ thalli was better protected of the metal exposition, and able to produce phytochelatins (PCs) over the whole range of metal, while in the p− thalli the production was completely inhibited at 75 μmol Cd g −1 dw and higher concentrations, together with the inhibition of cysteine (Cys) and reduced glutathione (GSH) production. The obtained results indicate that the parietin layer is a natural barrier decreasing Cd access to algal cells in X. parietina . Laser removal of biodeteriogen layers warrants detailed studies due to the advantages it brings with respect to mechanical elimination or the use of biocides. We have investigated elimination of biological crusts on dolomite stones from heritage sites in central Spain. The samples were colonized by epilithic crustose lichens of different species, such as Caloplaca sp. and Verrucaria nigrescens . A comparative study was carried out by applying infrared (1064 nm) and ultraviolet (355 nm) nanosecond laser pulses and sequences pulses of the two wavelengths using a Q-switched Nd:YAG system. To detect anatomical and ultrastructural damage to the lichens, and to assess possible morphological and chemical changes on the underlying stone induced by laser irradiation, we used stereomicroscopy, scanning electron microscopy with backscattered electron imaging and Fourier transform Raman spectroscopy. The optimal conditions for removal of the colonization crust, while ensuring preservation of the lithic substrate, were obtained for dual infrared-ultraviolet sequential irradiation. Abstract: We measured the water holding capacity per area (WHCA; mg H2O cm−2), and thus the saturating rainfall (mm), for sympatric cyano- (Lobaria scrobiculata and Pseudocyphellaria crocata) and cephalolichens (Lobaria pulmonaria) along canopy height gradients in boreal rainforests, to quantify the importance of specimen size, photobiont type and branch height as WHCA drivers. Size increased WHCA by a factor of four. Cyanolichens had ≈1.5 times higher WHCA than cephalolichens. Finally, branch height significantly increased WHCA for the Lobaria species. Reported responses are consistent with higher optimal rainfall requirements for reproducing versus juvenile specimens, and for cyanolichens versus those with green algal photobionts. Increased WHCA with height in the canopy is consistent with acclimation to stronger evaporative demands. The results highlight the link between rainfall patterns and maximal sizes realized for a given lichen species and habitat, and extend our understanding of why the cyanolichen/green algal lichen-ratio increases in forests with increasing rainfall. Functional traits have become important tools for evaluating the response of epiphytic lichens to environmental changes. In this study, we evaluated which predictors related to fragmentation, habitat quality and climate were driving the richness and cover of lichen growth form, type of photobiont and reproduction traits, at both fragment and plot levels in a Temperate-Mediterranean area dominated by Quercus forests. At fragment level, patch size and summer rainfall positively contributed to richness in most of the traits, while tree diameter and slope were the most important drivers, especially for the type of reproduction and growth form at plot scale. High coverage of growth forms especially sensitive to fragmentation were indicative of high values of total species richness, while early-colonizers indicated the opposite. These results provide important information on how lichen traits respond to environmental conditions in an ecotone area where a shift towards a drier climate is more likely to occur. Abstract: To facilitate marker selection in sequence-based studies on genetic diversity and symbiont selectivity in lichens we conducted a comparison of eight molecular markers in the lichen-forming fungus Lasallia pustulata and its trebouxioid photobiont. We compared mtSSU rDNA, mtLSU rDNA, MCM7, TSR1 (mycobiont) and nrITS rDNA, COX2, psbJ-L intergenic spacer, rbcL (photobiont) of 45 individuals from European populations of L. pustulata. Mycobiont and photobiont loci had congruent phylogenetic signals. Based on the results of this study we recommend the use of MCM7 and TSR1 (mycobiont), and nrITS rDNA and COX2 (photobiont). In this specific study system we found no sequence variability in the mycobiont loci EF1, nrITS rDNA, RPB1, and RPB2, which we sequenced for a subset of individuals. We had limited success amplifying GPD (mycobiont), actin and chloroplast LSU rDNA (photobiont), however, we do not rule out that these loci could be valuable markers in other species. Abstract: Little is known about the nature of the association between mycobionts and photobionts in isolated lichen communities. Here we studied the photobiont diversity of different Caloplaca species in a fog-induced community in the Atacama Desert. We compared nrDNA ITS sequences of both symbionts, photobionts and mycobionts, along with morphological characters of the different lichen thalli, to investigate the diversity and to assess the degree of selectivity and specificity of photobiont species in a community of Caloplaca species. Specimens of six fungal species (C. orthoclada, C. fernandeziana, and four undescribed species) were sampled along an altitudinal gradient on a coastal bluff with a strong fog presence, 60km south of Iquique, Chile. The photobiont species in this community belong to three species of the genus Trebouxia in the strict sense: T. arboricola, T. decolorans, and T. gigantea. Most of the fungal species were lichenized with photobionts belonging to different haplotypes of T. arboricola and T. decolorans, although the algae of three specimens, associated with two fungal species (C. orthoclada and C. sp1), were related to representatives of T. gigantea. These results indicate that members of the genus Caloplaca in northern Chile have moderate photobiont selectivity and appear to be selective to members of the T. arboricola group. Also, at high altitudes, changes in the photobiontal haplotype composition were observed in comparison to lower altitudes, probably generated by a higher water availability given higher fog condensation and precipitation in the upper areas of the bluff. This may suggest that ecological factors, such as altitude and water availability could result in a local shift of the associated photobiont and specialization as a product of local adaptation. Abstract: The relative impact of lichen photobiont and mycobiont was evaluated by submitting nine lichen species with: (i) different photobiont types; (ii) different lichen growth forms; and (iii) different nutrients, pH, humidity preferences; to a range of Cu concentrations (μM) supplied in repeated cycles to simulate the natural process of uptake under field conditions. The physiological performance of the photosystem II photochemical reactions was measured using F v/F m and the metabolic activity of the mycobiont was evaluated using ergosterol and intracellular K-loss as indicators. Lichens with higher cation exchange capacity showed higher intracellular Cu uptake and their ecology seemed to be associated with low-nutrient environments. Thus the wall and external matrix, mainly characteristic of the mycobiont partner, cannot be ignored as the first site of interaction of metals with lichens. No common intracellular Cu concentration threshold was found for the physiological impacts observed in the different species. Most physiological effects of Cu uptake in sensitive lichens occurred for intracellular Cu below 200μg/g dw whereas more tolerant species were able to cope with intracellular Cu at least 3 times higher. Cyanobacterial lichens showed to be more sensitive to Cu uptake than green-algal lichens. Within the Trebouxia lichens, different species showed different sensitivities to Cu uptake, suggesting that the mycobiont may change the microenvironment close to the photobiont partner providing different degrees of protection. Despite the fact that the photobiont is the productive partner, the metabolic activity of the mycobiont of lichen species adapted to environments rich in nutrients, showed to be more sensitive to Cu uptake than the photochemical performance of the photobiont. Abstract: The availability of highly variable markers for the partners of a fungal symbiosis enables the integrated investigation of ecological and evolutionary processes at the symbiotic level. In this article we analyze the specificity of the first and to date only microsatellite markers that had been developed for an epiphytic lichen (Lobaria pulmonaria). We used DNA extracts from cultures of the fungal and of the green algal symbionts of L. pulmonaria as well as total DNA extracts from related Lobaria species associated with the same algal partner, and got evidence that five of the previously described microsatellite markers, proposed to be fungus-specific, are indeed alga-specific. Hence, highly variable microsatellite primer sets available for both, the algal and the fungal symbionts of L. pulmonaria are now at our hands, which allow us to investigate so far unexplored biological processes of lichen symbionts, such as codispersal and coevolution. In a broader sense, our work evaluates and discusses the challenges in developing biont-specific molecular markers for fungi forming close associations with other organisms. [Copyright &y& Elsevier]

Abstract: A structural study of the carbohydrates from the aposymbiotically cultured Asterochloris sp., the algal symbiont of the lichen Cladina confusa was carried out for the first time. A xylorhamnogalactofuranan was purified and was predominated by (1→3)-linked galactofuranosyl units with sidechains in position 6 on approximately 6.4% of the units. The sidechains have galactofuranosyl units 5-O and 6-O-substituted, as well rhamnopyranosyl units 2-O, 3-O and 2,3-di-O-substituted. Xylose was detected only as nonreducing end units, together with galactofuranosyl units. Amylose and a β-(1→4)-xylan were also present. These polysaccharides have not been found in the symbiotic thallus of C. confusa, which contained only glucans, galactomannoglucan and galactoglucomannan. A potential role of these carbohydrates in lichen recognition proccess is also discussed. [Copyright &y& Elsevier]

Abstract: An unusual dip (compared to higher plant behaviour under comparable light conditions) in chlorophyll fluorescence induction (FI) at about 0.2–2 s was observed for thalli of several lichen species having Trebouxia species (the most common symbiotic green algae) as their native photobionts and for Trebouxia species cultured separately in nutrient solution. This dip appears after the usual O(J)IP transient at a wide range of excitation light intensities (100–1800 μmol photons m−2 s−1). Simultaneous measurements of FI and 820-nm transmission kinetics (I820) with lichen thalli showed that the decreasing part of the fluorescence dip (0.2–0.4 s) is accompanied by a decrease of I820, i.e., by a reoxidation of electron carriers at photosystem I (PSI), while the subsequent increasing part (0.4–2 s) of the dip is not paralleled by the change in I820. These results were compared with that measured with pea leaves—representatives of higher plants. In pea, PSI started to reoxidize after 2-s excitation. The simultaneous measurements performed with thalli treated with methylviologen (MV), an efficient electron acceptor from PSI, revealed that the narrow P peak in FI of Trebouxia-possessing lichens (i.e., the I–P-dip phase) gradually disappeared with prolonged MV treatment. Thus, the P peak behaves in a similar way as in higher plants where it reflects a traffic jam of electrons induced by a transient block at the acceptor side of PSI. The increasing part of the dip in FI remained unaffected by the addition of MV. We have found that the fluorescence dip is insensitive to antimycin A, rotenone (inhibitors of cyclic electron flow around PSI), and propyl gallate (an inhibitor of plastid terminal oxidase). The 2-h treatment with 5 μM nigericin, an ionophore effectively dissipating the pH-gradient across the thylakoid membrane, did not lead to significant changes either in FI nor I820 kinetics. On the basis of the presented results, we suggest that the decreasing part of the fluorescence dip in FI of Trebouxia-lichens reflects the activation of ferredoxin–NADP+–oxidoreductase or Mehler–peroxidase reaction leading to the fast reoxidation of electron carriers in thylakoid membranes. The increasing part of the dip probably reflects a transient reduction of plastoquinone (PQ) pool that is not associated with cyclic electron flow around PSI.
