Deoxynivalenol (DON) and zearalenone (ZEA) are the two most common mycotoxins in animal feed. In this study, we examined oxidative injury and apoptosis of porcine splenic lymphocytes induced by DON or ZEA and their combination in vitro . Based on IC 50 values, porcine splenic lymphocytes were treated with 0.06, 0.3, 1.5, and 7.5 μg/mL DON, 0.08, 0.4, 2, and 10 μg/mL ZEA, or both DON and ZEA at 0.06 and 0.08 μg/mL, 0.3 and 0.4 μg/mL, and 1.5 and 2 μg/mL, respectively. After 48 h of DON and/or ZEA exposure, the cells were analyzed for antioxidant functions, apoptosis, and mRNA and protein expression of apoptosis-related genes p53, Bcl-2, Bax, caspase-3, and caspase-8 to determine their apoptosis and oxidative damage effects and mechanisms. The results showed that, compared with the control group, SOD, CAT, GPx, GSH, and Bcl-2 mRNA and protein expression levels were significantly reduced in exposed groups ( P < 0.05 or P < 0.01). Furthermore, MDA contents, apoptosis rates, and p53, Bax, caspase-3 and caspase-8 protein and mRNA expression levels were increased significantly ( P < 0.01). The effects of DON and ZEA were dose dependent and synergistic in combination. These data suggest that DON and ZEA induce oxidative damage and apoptosis of porcine splenic lymphocytes. In this study, the in vitro effects of the treatment of concanavalin A (Con A)—stimulated splenic lymphocytes with DON were examined. Splenic lymphocytes isolated from chickens were stimulated with 12.5 μg/mL Con A and exposed to deoxynivalenol (DON) (0–50 μg/mL) for 48 h. The intracellular calcium concentration ([Ca 2+ ]i), pH, calmodulin (CaM) mRNA levels, and Na + ,K + -ATPase and Ca 2+ -ATPase activities were detected. With the DON exposure concentrations increased, the [Ca 2+ ]i and CaM mRNA levels gradually increased in a dose-dependent manner, and all the evaluated conconcentrations affected ATPase activity to the same extent. There were significant differences ( P < 0.05 or P < 0.01) between the treatment groups and the control group. These results indicate that an imbalance in calcium homeostasis and intracellular acidification are components of DON cytotoxicity in chicken lymphocytes. [ABSTRACT FROM AUTHOR]

Nickel (Ni) is an environmental toxicant that can cause toxic damage to humans and animals. Although the hepatotoxicity of Ni has been confirmed, its precise mechanism is still unclear. In this study, the results showed that nickel chloride (NiCl 2)-treatment could induce mice hepatotoxicity including hepatic histopathological alterations and up-regulation of serum AST and ALT. According to the results, NiCl 2 increased malondialdehyde (MDA) production while reducing total antioxidant capacity (T-AOC) activity and glutathione (GSH) content. Additionally, NiCl 2 induced mitochondrial damage which was featured by increase in mitochondrial ROS (mt-ROS) and mitochondrial membrane potential (MMP) depolarization. The mitochondrial respiratory chain complexes I-IV and ATP content were decreased in the liver of NiCl 2 -treated mice. Meanwhile, NiCl 2 caused hepatic ferroptosis accompanied by increased iron content in the liver and up-regulation of cyclooxygenase 2 (COX-2) protein and mRNA expression levels, down-regulation of glutathione eroxidase 4 (GPX4), ferritin heavy chain 1 (FTH1) and nuclear receptor coactivator 4 (NCOA4) protein and mRNA expression levels. Amputation dehorning (AD) is a common practice performed on calves, causing harmful effects such as pain, distress, anxiety, and fear. These effects extend to behavioral, physiological, and hematological responses, prompting serious ethical concerns regarding animal welfare, even when performed with local anesthesia. Meloxicam, a nonsteroidal anti-inflammatory drug, has been widely used to mitigate the side effects of dehorning and disbudding in calves. However, there is a notable gap in research regarding the effects of meloxicam on calves aged 6 wk to 6 mo undergoing AD procedures. This study was designed to assess the effectiveness of co-administering meloxicam with lidocaine, a cornual nerve anesthetic, in alleviating the adverse effects caused by the AD procedure in calves within this age range, compared with the use of lidocaine alone. Thirty Holstein calves were enrolled and randomly divided into 2 groups. The first group received a subcutaneous injection of 5 mL of lidocaine in the horn area and a subcutaneous injection of 0.9% saline at a dose of 0.025 mL/kg in the neck, administered 10 min before the AD procedure. The second group received a combination of lidocaine and meloxicam: a subcutaneous injection of 5 mL of lidocaine in the horn area and a subcutaneous injection of 20 mg/mL meloxicam at a dose of 0.025 mL/kg in the neck, also administered 10 min before the AD procedure. To avoid subjective bias, the researchers were blinded to the treatment groups. Pain-related behaviors, including tail flicking, head shaking, ear flicking, head rubbing, head crossing bar, and kicking, were observed, and physiological parameters, including heart rate, rectal temperature, respiration rate, mechanical nociceptive threshold (MNT), daily active steps, and food intake were monitored. Hematological conditions were determined using enzyme-linked immunosorbent assays and routine blood tests. The data were processed using a generalized linear mixed model. The outcomes demonstrated that the AD procedure increased the frequencies of ear flicking and resulted in rises in the respiration rate, heart rate, rectal temperature, and daily active steps. It also led to decreases in total food intake, forage intake, hay intake, MNT, and increased concentrations of prostaglandin E2 (PgE2), IL-1β, tumor necrosis factor-α (TNF-α), nitric oxide (NO), and malondialdehyde, as well as glutathione peroxidase activity. However, calves that received meloxicam treatment showed significant improvements in response to the AD procedure, including lower respiration rates, heart rates, and rectal temperatures; higher MNT; and lower intermediate cell ratio. They also had higher red blood counts, hemoglobin levels, hematocrit values; larger mean platelet volumes; and lower concentrations of PgE2, IL-1β, TNF-α, and NO. These results suggest that co-administration of lidocaine and meloxicam may aid in mitigating the adverse effects induced by the AD procedure on these calves, thereby supporting the use of meloxicam in conjunction with a local anesthetic in AD procedures for calves aged 6 wk to 6 mo. Trichothecenes are a class of toxic secondary metabolites produced by filamentous fungi such as Fusarium , which pose a threat to human and animal health. Among them, T-2 toxin and deoxynivalenol (DON) have attracted the most attention because of their extensive pollution and strong toxicity. Accumulated studies have found that an increase of reactive oxygen species (ROS) during oxidative stress produces a wide range of secondary consequences by modifying cell signaling proteins, such as DNA RNA damage, inflammatory damage, cell cycle arrest, and apoptosis. Although there has been a comprehensive review of antioxidant agents against trichothecenes, there is no systematic summary of the mechanism of trichothecenes inducing ROS production and then regulating downstream signaling molecules of cells to produce the corresponding effects. Therefore, from the perspective of ROS regulation of downstream signaling pathways, this review mainly explores the mechanisms by which ROS, as trichothecenes' handy weapon, affect an organism. Umbilical cord mesenchymal stem cells (UCMSCs) retain their intrinsic stem cell potential while at the same time displaying high proliferation rates, powerful differentiation capacity, and low immunogenicity. They can also secrete multiple bioactive molecules that exert specific physiological functions. Thus, UCMSCs represent excellent candidates for cell therapy in regenerative medicine and tissue engineering. Abundant preclinical research on different disease models has shown that UCMSCs can accelerate wound or nerve damage recovery and suppress tumor progression. In fact, UCMSCs are thought to possess a higher therapeutic potential than MSCs derived from other tissues. Increasing evidence suggests that the mechanism underlying UCSMCs efficacy depends mostly on cell secretions, in contrast to the early paradigm of cell replacement and differentiation. The present study analyzed the prevalence and antimicrobial resistance of Salmonella along an integrated broiler chicken supply chain. A total of 172 Salmonella isolates were recovered from 1148 samples collected from four sample sources (breeder farms, broiler farms, abattoir, and retail markets), representing nine production stages. These Salmonella isolates were examined for antimicrobial susceptibility to 12 different antimicrobial agents using a disk diffusion assay. Among them, 168 were identified as six different serotypes of Salmonella enterica. The predominant serotype was S. Enteritidis (n = 116), followed by S. Infantis (n = 18), S. Gueuletapee (n = 16), S. Derby (n = 12), S. Meleagridis (n = 4), and S. London (n = 2). The remaining four isolates were serogroup-untypeable. A majority of the 172 isolates (96.51%) was resistant to one or more antibiotics and 61.05% of the Salmonella isolates showed a multidrug resistance phenotype. Statistical analysis indicated the one risk product stage for Salmonella contamination occurred in the sample source at the abattoir, specifically the stage of Carcasses after chilling. The majority of S. Enteritidis isolates shared the same pulsed-field gel electrophoresis (PFGE) cluster, suggesting that the S. Enteritidis strain might spread along the broiler chicken supply chain. The prevalence and antimicrobial resistance of Salmonella in different production stages suggest the importance of controlling Salmonella in the broiler chicken supply chain for public health, underlying the need for improved measures of reducing carcass contamination in abattoirs and the appropriate use of antimicrobials in broiler flocks. The aims of this study were to investigate the pathways which dietary nickel chloride (NiCl 2 ) affects small intestine apoptosis in broiler chickens by observing the ultrastructure, and bcl-2, bax, and caspase-3 protein expression and mRNA expression, and cytochrome C, bak and caspase-9 mRNA expression of the small intestine. A total of 240 one-day-old avian broilers were divided into four groups and fed a corn-soybean basal diet as the control diet or three experimental diets supplemented with 300, 600, and 900 mg/kg of NiCl 2 for 42 days. Ultrastructurally, the microvilli were apparently exfoliated, and the mitochondria were swollen and the number of lysosomes increased in the intestinal cells of three experimental groups. As measured by TUNEL and flow cytometry (FCM), the percentage of apoptotic cells in the small intestine and the lymphocytes in the ileum were significantly increased in three experimental groups when compared with those of the control group. Meanwhile, immunohistochemistry, quantitative real-time polymerase chain reaction (qRT-PCR) and enzyme-linked immuno-sorbent assay (ELISA) tests showed that the protein expression, mRNA expression levels were decreased in the bcl-2, whereas those of bax and caspase-3, and the cytochrome C, bak and caspase-9 mRNA expression levels were increased in three experimental groups. The abovementioned results show that pathway of dietary NiCl 2 -induced small intestine apoptosis is related to the mitochondrial damage and promotion of the cytochrome C release from mitochondria, which activates the mitochondrion-mediated apoptosis pathway. Deoxynivalenol (DON) immunotoxicity and its induction of cytokines and related genes in the splenic lymphocytes of chickens have not been completely elucidated. In the present study, we aimed to evaluate the effects of 48 h of different DON treatments (0 μg/mL, 0.2 μg/mL, 0.8 μg/mL, 3.2 μg/mL, 12.5 μg/mL, and 50 μg/mL) on the secretion and the mRNA expressions of some cytokine genes, such as interleukin-1beta (IL-1β), IL-1RI, IL-2, IL-4, IL-6, IL-10, IL-12β, and IFN-γ in chicken splenic lymphocytes. The concentrations of IL-1RI, IL-6, IL-10, and IFN-γ were increased with the DON concentrations increasing ( P < 0.05 or P < 0.01). However, the concentrations of IL-1β, IL-2, IL-4, and IL-12β were decreased with the DON concentrations increasing ( P < 0.05 or P < 0.01). Except IL-1β, the mRNA expressions of the other cytokines were up-regulated by DON. The highest mRNA expressions values of IL-1RI, IL-4, IL-10, IL-12β, and IFN-γ were at 50 μg/mL DON treatment groups ( P < 0.05 or P < 0.01), while the highest mRNA expressions values of IL-2 and IL-6 were at 12.5 μg/mL DON treatment groups ( P < 0.05 or P < 0.01). To investigate the effects of sodium selenite against aflatoxin B1 (AFB 1), 200 male Avian broilers, divided into five groups, were fed with basal diet (control group), 0.3 mg/kg AFB1 (AFB1 group), 0.3 mg/kg AFB1 + 0.2 mg/kg Se (+Se group I), 0.3 mg/kg AFB1 + 0.4 mg/kg Se (+Se group II) and 0.3 mg/kg AFB1 + 0.6 mg/kg Se (+Se group III), respectively. Compared with the control group, decreased relative weight of bursa of Fabricius and contents of serum immunoglobulin, more vacuoles and debris in the bursal lymphoid follicle, and increased percentage of apoptotic bursal cells were observed in the AFB1 group. Sodium selenite, however, could increase the relative weight of bursa of Fabricius and contents of serum immunoglobulin, and ameliorate histopathological lesions. The percentages of apoptotic bursal cells, through flow cytometry and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling method, in the three +Se groups were lower than those in the AFB 1 group. Compared with the AFB 1 group, moreover, the mRNA expressions of Bax and Caspase-3 by qRT-PCR in the three +Se groups were decreased, while the expression of Bcl-2 was increased. The results indicate that sodium selenite in diet can protect chicken from AFB 1-induced impairment of humoral immune function by reducing bursal histopathological lesions and percentages of apoptotic bursal cells. Deoxynivalenol (DON) is a type of cytotoxic mycotoxin that targets the mitochondria in cells. However, effects of DON on mitochondrial dynamics have not yet been reported. In this study, we investigated the effects of DON on mitochondrial dynamics and mitochondrial autophagy. Spleen lymphocytes of primary pigs were treated with different concentrations of DON. 24 h later, the cells were collected and the indexes were measured as follow: contents of ROS and MDA; activities of SDH and SOD; total antioxidant capacity; expression of MFN1, MFN2 and OPA1; the mRNA expression of the genes encoding mitochondrial autophagy proteins LC3 and P62, as well as mitochondrial respiratory chain complex activities. The results showed that, compared with the control group, the contents of ROS and MDA in the DON groups increased, while the activities of T-AOC, SDH and SOD decreased in a dose-dependent manner. With increasing DON concentrations, the expression levels of MFN1/2, OPA1 and mitochondrial respiratory chain complex activities decreased, while LC3, P62 increased. We suspect that DON causes oxidative damage, which in turn leads to down-regulation of MFN1, MFN2, OPA1 and up-regulation of LC3 and P62 mRNA, thereby inhibiting mitochondrial fusion and promoting mitochondrial autophagy. • The toxicological study of DON was advances to the level of mitochondrial dynamics. • It was found that the effect of DON on mitochondrial dynamics was dose-dependent. • Providing new ideas or strategies for the development of DON antagonists. Fluoride is known to affect the inflammatory process and autoregulation of immune responses, but the molecular mechanism by which fluoride causes innate immune injury remain largely unknown. Also, studies on sodium fluoride (NaF)-caused alteration of TLR signaling are still lacking. In the present study, we examined the effects of NaF on the mRNA and protein expression levels of TLR2/MyD88 signaling pathway molecules in the mouse spleen by using the methods of qRT-PCR and Western blotting. Consequently, we elucidated the mechanism underlying the effects of NaF on innate immunity. Two hundred and forty ICR mice were randomly divided into 4 groups with intragastric administration of distilled water in the control group and 12, 24, 48 mg/kg of NaF treatment in the experiment groups for 42 days. The findings revealed that NaF impaired splenic innate immunity in mice via inactivation of TLR2/MyD88 signaling pathway. NaF-inactivated TLR2/MyD88 signaling pathway was identified by prominently downregulated mRNA and protein expression levels of TLR2/MyD88, IRAK4, IRAK1, TRAF6, TAK1, MKK4/MKK7 and c-Jun, which ultimately altered the expression levels of IL-1β, IL-4, IL-6 and IL-8 to attenuate innate immunity. • NaF exposure can impair splenic innate immunity. • NaF inactivated the TLR2/MyD88 signaling pathway. • NaF induced a causal cascade response to impact immune cytokines production.
