Decreased oocyte quality is a significant contributor to the decline in female fertility that accompanies aging in mammals. Oocytes rely on mRNA stores to support their survival and integrity during the protracted period of transcriptional dormancy as they await ovulation. However, the changes in mRNA levels and interactions that occur during porcine oocyte maturation and aging remain unclear. In this study, the mRNA expression profiles of porcine oocytes during the GV, MII, and aging (24 h after the MII stage) stages were explored by transcriptome sequencing to identify the key genes and pathways that affect oocyte maturation and postovulatory aging. The results showed that 10,929 genes were coexpressed in porcine oocytes during the GV stage, MII stage, and aging stage. In addition, 3037 genes were expressed only in the GV stage, 535 genes were expressed only in the MII stage, and 120 genes were expressed only in the aging stage. The correlation index between the GV and MII stages (0.535) was markedly lower than that between the MII and aging stages (0.942). A total of 3237 genes, which included 1408 upregulated and 1829 downregulated genes, were differentially expressed during porcine oocyte postovulatory aging (aging stage vs. MII stage). Key functional genes, including ATP2A1 , ATP2A3 , ATP2B2 , NDUFS1 , NDUFA2 , NDUFAF3 , SREBF1 , CYP11A1 , CYP3A29 , GPx4 , CCP110 , STMN1 , SPC25 , Sirt2 , SYCP3 , Fascin1/2 , PFN1 , Cofilin , Tmod3 , FLNA , LRKK2 , CHEK1/2 , DDB1/2 , DDIT4L , and TONSL , and key molecular pathways, such as the calcium signaling pathway, MAPK signaling pathway, TGF-β signaling pathway, PI3K/Akt signaling pathway, FoxO signaling pathway, gap junctions, and thermogenesis, were found in abundance during porcine postovulatory aging. These genes are mainly involved in the regulation of many biological processes, such as oxidative stress, calcium homeostasis, mitochondrial function, and lipid peroxidation, during porcine oocyte postovulatory aging. Fertilization capacity and embryo survival rate are decreased in postovulatory aging oocytes, which results in a reduced reproductive rate in female animals. However, the key regulatory genes and related regulatory mechanisms involved in the process of postovulatory aging in oocytes remain unclear. In this study, RNA-Seq revealed that 3237 genes were differentially expressed in porcine oocytes between the MII and aging stages (MII + 24 h). The expression level of FOXM1 was increased at the aging stage, and FOXM1 was also observed to be enriched in many key biological processes, such as cell senescence, response to oxidative stress, and transcription, during porcine oocyte aging. Previous studies have shown that FOXM1 is involved in the regulation of various biological processes, such as oxidative stress, DNA damage repair, mitochondrial function, and cellular senescence, which suggests that FOXM1 may play a crucial role in the process of postovulatory aging. Therefore, in this study, we investigated the effects and mechanisms of FOXM1 on oxidative stress, mitochondrial function, DNA damage, and apoptosis during oocyte aging. Our study revealed that aging oocytes exhibited significantly increased ROS levels and significantly decreased GSH, SOD, T-AOC, and CAT levels than did oocytes at the MII stage and that FOXM1 inhibition exacerbated the changes in these levels in aging oocytes. In addition, FOXM1 inhibition increased the levels of DNA damage, apoptosis, and cell senescence in aging oocytes. A p21 inhibitor alleviated the effects of FOXM1 inhibition on oxidative stress, mitochondrial function, and DNA damage and thus alleviated the degree of senescence in aging oocytes. These results indicate that FOXM1 plays a crucial role in porcine oocyte aging. The conventional finite control set-model predictive control (FCS-MPC) has not been completely embraced by the power industry because of large tracking errors and the high required sampling frequency. Therefore, the double-vector-based model predictive control (DVB-MPC), which enfolds the deadbeat control (DBC) theory, has gradually crept into researchers' horizons in recent years. In the universal DVB-MPC (UDVB-MPC) scheme, selecting two operation vectors in a single sector is a major obstacle to achieving satisfactory tracking performance. There is still a biggish error between the synthesized output voltage and the reference value. To alleviate this issue, this paper proposes an improved double-vector-based model predictive control (IDVB-MPC) scheme. The tracking errors are reduced dramatically by adding two extra vectors in adjacent sectors to the candidate set and dividing a sector into eight zones. Then the power fluctuation is reduced and the grid-connected current quality is improved. To verify the effectiveness of the proposed scheme, the simulation and experiment comparisons between UDVB-MPC and IDVB-MPC are carried out. High-content sugar in honey frequently results in severe matrix effects and requires complex pretreatment prior to analysis, posing significant challenges for the rapid analysis of honey. In this study, the reversal polarity nano-electrospray ionization mass spectrometry (RP-Nano-ESI-MS) analysis was developed for the direct evaluation of honey samples. The results indicated that RP-Nano-ESI-MS significantly mitigated the matrix effects induced by high-content sugar through the implementation of online desalting. Furthermore, RP-Nano-ESI-MS has been proven capable of not only differentiating acacia honey adulterated with 10% rape honey, but also effectively distinguishing six types of honey and exhibiting remarkable proficiency in detecting honey adulteration and botanical traceability. Additionally, RP-Nano-ESI-MS exhibited strong quantitative abilities, effectively characterizing variations in amino acid composition among six types of honey with high stability and reproducibility. Our studies underscore the significant potential of RP-Nano-ESI-MS for its rapid in situ analysis of sugar-rich foods like honey, especially in their authenticity verification. The prevalence of metabolic dysfunction-associated steatotic liver disease (MASLD) is increasing year by year and has become one of the leading causes of end-stage liver disease worldwide. Triggering Receptor Expressed on Myeloid Cells 2 (Trem2) has been confirmed to play an essential role in the progression of MASLD, but its specific mechanism still needs to be clarified. This study aims to explore the role and mechanism of Trem2 in MASLD. Human liver tissues were obtained from patients with MASLD and controls. Myeloid-specific knockout mice (Trem2mKO) and myeloid-specific overexpression mice (Trem2TdT) were fed a high-fat diet, either AMLN or CDAHFD, to establish the MASLD model. Relevant signaling molecules were assessed through lipidomics and RNA-seq analyses after that. Trem2 is upregulated in human MASLD/MASH-associated macrophages and is associated with hepatic steatosis and inflammation progression. Hepatic steatosis and inflammatory responses are exacerbated with the knockout of myeloid Trem2 in MASLD mice, while mice overexpressing Trem2 exhibit the opposite phenomenon. Mechanistically, Trem2mKO can aggravate macrophage pyroptosis through the PI3K/AKT signaling pathway and amplify the resulting inflammatory response. At the same time, Trem2 promotes the inflammation resolution phenotype transformation of macrophages through TGFβ1, thereby promoting tissue repair. Myeloid Trem2 ameliorates the progression of Metabolic dysfunction-associated steatotic liver disease by regulating macrophage pyroptosis and inflammation resolution. We believe targeting myeloid Trem2 could represent a potential avenue for treating MASLD. Streptococcus pyogenes (GAS) is one of the most virulent and infectious bacteria, severely threatening health and lives of people worldwide. Honey has been proven to have effective capability against GAS, but the underlying metabolites and mechanisms are still unclear. In this study, the Castanopsis honey (CH) showed significant antibacterial ability compared to other seven kinds of honey and artificial honey. Furthermore, the antibacterial metabolites and their targets in CH were screened by combined method of metabolomics, network pharmacology, and molecular docking. The results suggested that the activities of two antioxidant enzymes, glutathione peroxidase and tyrosyl tRNA synthetase identified as the primary targets, were significantly inhibited by CH, which significantly increased the level of oxidative stress in GAS. The results revealed a possibly novel mechanism regulating the oxidative stress and inhibits the growth in bacteria, providing strong experimental evidence to support the further development of CH as a novel antibacterial agent. • Amylosucrase and pullulanase treatments of amylopectin produced linear α-glucans. • Linear α-glucans were prone to form V-type complexes with unsaturated fatty acid. • Digestibility of the complexes was controlled by their crystalline structure. • The formation of V-type structure improved the stability of unsaturated fatty acid. In this study, waxy corn starch (WCS) was modified by amylosucrase and pullulanase, producing linear starch chains with elongated length that favored the complexation with unsaturated fatty acids (uFAs). Compared to native WCS, the amylosucrase-modified WCS with an average chain length of 47.8 was easier to form V-type complexes with oleic acid, while increasing the degree of unsaturation impeded the formation of V-type complexes. The pullulanase treatment hydrolyzed the branching points of amylosucrase-modified WCS and the linear starch chains could forme V-type complexes with oleic acid, linoleic acid, and linolenic acid, with V-type crystallinity decreasing from 38.2 % to 20.1 %. V-type complexes had a lower thermal stability than the B-type starch crystallites, and their peak melting temperature ranged from 67.2 to 79.0 °C. The content of resistant starch in the complexes was in the range of 21.8 %-40.9 % and the formation of V-type complexes decreased the susceptibility of uFAs to oxygen. [ABSTRACT FROM AUTHOR]

Although laser-induced graphene (LIG) has great advantages in the cost and manufacturing process, it has been seldom reported in the field of electromagnetic interference (EMI) shielding due to its unobtrusive conductivity and the limitation of inherent substrates. In this work, polybenzoxazine is chosen as precursor to fabricate the LIG via a one-step defocused lasing process. As a result of favorable porous structures and high conductivity, the as-produced LIG exhibits the EMI shielding effectiveness up to 24.8 dB in X-band at the thickness of 68 μm. And the EMI shielding effectiveness of LIG/Fe 3 O 4 composite is increased to 32.7 dB at a smaller thickness of 53 μm which is obtained by a solvent-free approach. When considering the thickness and lightweight properties, the absolute shielding effectiveness is also surpassing most of the other carbon-based shielding materials. More importantly, taking advantage of the difference in the thermal expansion coefficient of carbon material and polymeric substrate, we develop a rapid quench-peeling (RQP) strategy for the separation of LIG from polymeric substrate to obtain the free-standing LIG film. Moreover, the structures and properties of LIG are well preserved. The free-standing LIG films after peeling have wider adaptability in practical EMI shielding applications, and also opened up possibilities for use in other fields, such as Joule heating device. Targeting delivery of photosensitizers to mitochondria as the most sensitive cellular organelles to reactive oxygen species (ROS) by positively charged polymeric nanocarriers (NCs) is one of the useful methods for efficient photodynamic therapy (PDT). However, the NCs with positively charged mitochondria-targeting moieties are easily cleaned during circulation, restricting their in vivo applications. Herein, to address this issue and enhance in vivo PDT efficacy, we developed a sequential-targeting delivery system consisting of mitochondria-targeting micelles as the core prepared from the cationic amphiphilic copolymer for loading chlorin e6 (Ce6) and a tumor-targeting pH-dependent charge transformational layer as the shell obtained from 2,3-dimethylmaleic anhydride modified Biotin-PEG 4000 -NH 2 (BioPEGDMA) via electrostatic interaction. Concealed by the anionic shell, the as-prepared NCs showed longer retention within the first stage of tumor-targeting. Then, the accumulated NCs conversed to positive charge in tumor extracellular microenvironment (pH ∼ 6.5), which could be more effectively internalized by tumor cells, and the re-exposed triphenylphosphonium (TPP) groups endowed their second-stage targetability to the mitochondria. In vivo experiments revealed that the Ce6-loaded NCs exhibited remarkable tumor inhibition rates of 84.1% and 93.2% on BALB/c nude mice and Kunming mice, respectively, under 660 nm NIR irradiation, and stimulated immune responses with upregulated expression of IFN-γ, TNF-α and CD3+ in tumor tissues, and enhanced activation of CD3+/CD4+, CD3+/CD8+ T lymphocytes and DCs in both tumor tissues and lymph glands. This work provided a new pathway for the development of smart drug delivery system with advanced PDT efficacy. Although the existing targeting delivery of photosensitizers to mitochondria by positively charged nanocarriers (NCs) have efficiently enhanced photodynamic therapy (PDT), their positive charges caused rapid clearance during circulation, which has restricted their in vivo applications. Therefore, we fabricated a novel sequential-targeting NC to solve the problem. The tumor accumulated NCs conversed to positive charge in tumor extracellular microenvironment, and the re-exposed triphenylphosphonium groups initiated second-stage targetability to mitochondria. This system exhibited remarkable tumor inhibition efficiency both in vitro and in vivo. Moreover, as we hypothesized, mitochondria-located PDT could promote immune response, resulting in improvement of PDT. The strategy of sequential targeting-based PDT in combination with augmented immune response showed a novel pathway for the development of smart drug delivery system with advanced PDT. Effect of pigskin-originated gelatin on dough properties and bread characteristics, especially on bread staling was investigated in terms of starch retrogradation and water migration in the present study. Results showed that gelatin significantly improved bread qualities during storage. Compared with the control, 7-day staling rate of bread containing 0.5% and 1.0% gelatin decreased by 6.56% and 20.83% respectively, indicating gelatin retarded the staling process of bread. This retardation effect was related to starch retrogradation and water migration. The relative crystallinity decreased from 15.12%, 11.60% to 10.75%, which demonstrated the alleviation effect of amylopectin crystallization. Bread samples containing 1.0% gelatin had the lowest retrogradation enthalpy of 2.93 ± 0.24 J/g followed by 0.5% gelatin bread and the control group, further indicating that gelatin may induce less water available to form crystal lattice and restrict the migration of starch molecules. The 5-day moisture loss rate for the control group was highest with the values of 12.39%, which was 1.43% and 1.83% higher than that of bread with 0.5% gelatin and 1.0% gelatin, respectively. Water activity and T 2 relaxation results provided the information that gelatin inhibited water migration upon storage and gelatin interacted with bread components. However, the effect of gelatin on improving bread quality and retarding staling could not be reflected by the dough properties measured through farinograph, extensograph and rheometer. This study suggested gelatin from pigskin had great potential to be served as an effective bread improver. [Display omitted] • A novel mass spectrometry method is developed to analyze of honey. • Four botanical origins of honey are distinguished with multivariate analysis. • Potential markers for three different types of honey are identified and quantified. • The results derived from the new method are verified by UPLC-Q/TOF-MS. The botanical origins of honey are important for the quality control and commercialization of honey. In this research, we established a nanoliter electrospray ionization mass spectrometry (Nano-ESI-MS) method to identify Castanopsis honey (CH), Eurya honey (EH), Dendropanax dentiger honey (DH), and Triadica cochinchinensis honey (TH). In total, 38 compounds were identified based on the collision-induced dissociation experiments by Nano-ESI-MS with 16 differential compounds and 7 quantified as potential differential markers. These four types of honey were distinguished from each other by their mass spectrometry data combined with multivariate analysis with three out of the 7 differential markers, i.e., phenethylamine, tricoumaroyl spermidine, and (+/–)-abscisic acid, identified as potential markers for CH, EH, and DH, respectively. Both the qualitative and quantitative results derived from Nano-ESI-MS were further verified by UPLC-Q/TOF-MS. Laser-ablated beryllium atoms have been reacted with silane molecules during condensation in excess neon and argon at 4 K. Absorptions due to HBeSiH 3 and HBe(μ-H) 3 Si were observed and identified on the basis of isotopic IR spectroscopy, deuterium substitution with SiD 4 , and quantum chemical frequency calculations. The observed results show excited Be atom ( 1 P 1 :2s 1 2p 1 ) can insert into Si H bond spontaneously and the insertion product rearranges to HBe(μ-H) 3 Si upon photolysis. The electron localization function (ELF) analysis suggests that 3c-2e hydrogen bridge bond (Be H Si) was formed by the donation of electrons for Si H σ bond to the empty p orbital of Be atom for HBe(μ-H) 3 Si molecule, which shows much difference from C H bond complexes.
