The coatings application onto medical devices has experienced a continuous growth in the last few years. Medical device coating market is expected to grow at a CAGR of 5.16% to reach USD 10 million by 2023 due to the increasing geriatric population and the growing demand for continuous innovation. Layer-by-Layer (LbL) assembly represents a versatile method to modify the surface properties, in order to control cell interaction and thus enhance biological functions. Furthermore, LbL is environmentally friendly, able to coat all types of surfaces with the creation of homogenous film and to include and control the release of biomolecules/drugs. This feature review provides a critical overview on recent progresses in functionalizing materials by LbL assembly for bone regeneration and disorder treatment. An overview of emerging and visionary opportunities on LbL technologies and further combination with other existing methods used in biomedical field, is also discussed to evidence the new challenges and potential developments in bone regenerative medicine. Chitosan (CS) membranes obtained by electrospinning are potentially ideal substrates for soft tissue engineering as they combine the excellent biological properties of CS with the extracellular matrix (ECM)-like structure of nanofibrous mats. However, the high amount of acid solvents required to spun CS solutions interferes with the biocompatibility of CS fibres. To overcome this limitation, novel CS based solutions were investigated in this work. Low amount of acidic acid (0.5 M) was used and dibasic sodium phosphate (DSP) was introduced as ionic crosslinker to improve nanofibres water stability and to neutralize the acidic pH of electrospun membranes after fibres soaking in biological fluids. Randomly oriented and aligned nanofibres (128 ± 19 nm and 140 ± 41 nm, respectively) were obtained through electrospinning process (voltage of 30 kV, 30 μL/min flow rate and temperature of 39 °C) showing mechanical properties similar to those of soft tissues (Young Modulus lower than 40 MPa in dry condition) and water stability until 7 days. C2C12 myoblast cell line was cultured on CS fibres showing that the aligned architecture of substrate induces cell orientation that can enhance skeletal muscle regeneration. [ABSTRACT FROM AUTHOR]

The current trend of peripheral nerve tissue engineering is the design of advanced nerve guidance channels (NGCs) acting as physical guidance for regeneration of nerves across lesions. NGCs should present multifunctional properties aiming to direct the sprouting of axons from the proximal nerve end, to concentrate growth factors secreted by the injured nerve ends, and to reduce the ingrowth of scar tissue into the injury site. A critical aspect in the design of NGCs is conferring them the ability to provide topographic, chemotactic and haptotactic cues that lead to functional nerve regeneration thus increasing the axon growth rate and avoiding or minimizing end-organ (e.g. muscle) atrophy. The present work reviews the recent state of the art in NGCs engineering and defines the external guide and internal fillers structural and compositional requirements that should be satisfied to improve nerve regeneration, especially in the case of large gaps (>2 cm). Techniques for NGCs fabrication were described highlighting the innovative approaches direct to enhance the regeneration of axon stumps compared to current clinical treatments. Furthermore, the possibility to apply stem cells as internal cues to the NGCs was discussed focusing on scaffold properties necessary to ensure cell survival. Abstract: Bone regeneration can be accelerated by localized delivery of appropriate growth factors/biomolecules. Localized delivery can be achieved by a 2-level system: (i) incorporation of biomolecules within biodegradable particulate carriers (nanoparticles), and (ii) inclusion of such particulate carriers (nanoparticles) into suitable porous scaffolds. In this study, freeze-dried porous chitosan–gelatin scaffolds (CH–G: 1:2 ratio by weight) were embedded with various amounts of poly(lactide-co-glycolide) (PLGA) nanoparticles, precisely 16.6%, 33.3% and 66.6% (respect to CH–G weight). Scaffolds loaded with PLGA nanoparticles were subjected to physico-mechanical and biological characterizations including morphological analysis, swelling and dissolution tests, mechanical compression tests and cell viability tests. Results showed that incorporation of PLGA nanoparticles into porous crosslinked CH–G scaffolds: (i) changed the micro-architecture of the scaffolds in terms of mean pore diameter and pore size distribution, (ii) reduced the dissolution degree of the scaffolds, and (iii) increased the compressive modulus. On the other hand, the water uptake behavior of CH–G scaffolds containing PLGA nanoparticles significantly decreased. The incorporation of PLGA nanoparticles did not affect the biocompatibility of CH–G scaffolds. [Copyright &y& Elsevier]

Posttraumatic peripheral nerve repair is one of the major challenges in restorative medicine and microsurgery. Despite the recent progresses in the field of tissue engineering, functional recovery after severe nerve lesions is generally partial and unsatisfactory. Autograft is still the best method to treat peripheral nerve lesions, although it has several drawbacks and does not allow complete functional recovery. Full recovery of nerve functionality could ideally be achieved by proper guiding axon regeneration toward the original target tissues, through the use of purposely engineered artificial nerve guidance channels (NGCs). In the last decade, artificial NGCs have been produced using a variety of both natural and synthetic, biodegradable and nonbiodegradable polymers. Several techniques have been developed to obtain porous and nonporous NGCs and to realize and incorporate bioactive fillers for NGCs. Some of the developed products have been approved for clinical applications. Many other NGC typologies have been object of interest and are currently under investigation. The current trend of nerve tissue engineering is the realization of biomimetic NGCs, providing chemotactic, topological, and haptotactic signalling to cells, respectively by surface functionalization with cell binding domains, the use of internal‐oriented matrices/fibres and the sustained release of neurotrophic factors. The present contribution provides a balanced integration of the most recent achievements of tissue engineering in the field of peripheral nerve repair. By an accurate evaluation of the status of research, the review delineates the most promising directions to which research should address for consistent progress in the field of peripheral nerve repair. Composites reinforced with micro-particles are widely used in engineering fields. Unfortunately, the traditional manufacturing processes do not allow customization and shape complexity achievable with additive manufacturing technologies. In particular, VAT polymerization enables the highest printing resolution. Here, we demonstrate the fabrication of fully-biobased composites consisting of a matrix of acrylated soybean oil (ASO) and a lignocellulose waste derived from the macadamia nut industry (macadamia nut-shell, MAC) as reinforcing agent. Different formulations were prepared by varying the content of MAC, and their reactivity was evaluated by means of Fourier transform infrared spectroscopy and photorheology. The suitability of the formulated novel biobased inks for digital light processing (DLP) 3D-printing was illustrated by successful printing of complex 3D-structures with a high resolution. The thermo-mechanical and mechanical properties of the 3D-printed composites were tested by DSC, DMTA and tensile tests, which revealed that. increasing MAC contents reflected into higher glass transition temperatures, ranging from 25 to 46 °C, and higher Young's modulus, ranging from 15 to 100 MPa. The morphology of the samples and the dispersion of the MAC in the ASO network was investigated by field emission scanning electron microscopy, while the surface roughness of the printed composites was analysed with profilometry. Cytotoxicity, cell adhesion and cell proliferation tests with human fibroblast cells were carried out in view of the fabrication of scaffolds for bioengineering and the results envisage good potential for these fully bio-based composites in the biomedical field.
