34. Cardiovascular risk and the COVID-19 pandemic: A retrospective observational study in a population of healthcare professionals.

Abstract

To contain the spread of COVID-19, many countries imposed several restrictive measures, leading to radical changes in daily life behaviors. Healthcare workers experienced additional stress due to the increased risk of contagion, possibly causing an increase in unhealthy habits. We investigated changes in cardiovascular (CV) risk assessed by the SCORE-2 in a healthy population of healthcare workers during the COVID-19 pandemic; an analysis by subgroups was also conducted (sportspeople vs sedentary subjects). We compared medical examination and blood tests in a population of 264 workers aged over 40, performed yearly before (T0) and during the pandemic (T1, T2). We found a significant increase in the average CV risk, according to SCORE-2, during the follow-up in our healthy population, with a shift from a mean low-moderate risk profile at T0 (2.35%) to a mean high-risk profile at T2 (2.80%). Furthermore, in sedentary subjects was observed a greater and early increase in SCORE-2 compared to sportspeople. Since 2019, we observed an increase in CV risk profile in a healthy population of healthcare workers, particularly in sedentary subjects, highlighting the need to reassess SCORE-2 every year to promptly treat high-risk subjects, according to the latest Guidelines. • The spread of COVID-19 forced billions of people to remain at home, leading to radical changes in daily life behaviors. • Moreover, healthcare workers experienced additional stress due to the increased risk of contagion. • We found a significant increase in cardiovascular risk profile during the pandemic in healthy healthcare workers. • The increase in systolic blood pressure was independent predictor of the reported increasing in cardiovascular risk. Abstract

Although the epigenetic regulatory protein histone deacetylase 6 (HDAC6) has been recently implicated in the etiology of Alzheimer's disease (AD), little is known about the role of HDAC6 in the etiopathogenesis of AD and whether HDAC6 can be a potential therapeutic target for AD. Here, we performed positron emission tomography (PET) imaging in combination with histopathological analysis to better understand the underlying pathomechanisms of HDAC6 in AD. We first developed [18F]PB118 which was demonstrated as a valid HDAC6 radioligand with excellent brain penetration and high specificity to HDAC6. PET studies of [18F]PB118 in 5xFAD mice showed significantly increased radioactivity in the brain compared to WT animals, with more pronounced changes identified in the cortex and hippocampus. The translatability of this radiotracer for AD in a potential human use was supported by additional studies, including similar uptake profiles in non-human primates, an increase of HDAC6 in AD-related human postmortem hippocampal tissues by Western blotting protein analysis, and our ex vivo histopathological analysis of HDAC6 in postmortem brain tissues of our animals. Collectively, our findings show that HDAC6 may lead to AD by mechanisms that tend to affect brain regions particularly susceptible to AD through an association with amyloid pathology. A novel PET imaging probe [18F]PB118 was demonstrated as a valid HDAC6 radioligand that can quantify and measure the change of HDAC6 in health and Alzheimer's disease (AD). Abstract

The realization and characterization of a high quality factor resonator composed of two hollow-dielectric cylinders with its pseudo-TM030 mode resonating at 10.9 GHz frequency is discussed. The quality factor was measured at the temperatures 300 K and 4 K obtaining Q 300 K = ( 150 , 000 ± 2 , 000 ) and Q 4 K = ( 720 , 000 ± 10 , 000 ) respectively, the latter corresponding to a gain of one order of magnitude with respect to a traditional copper cylindrical-cavity with the corresponding TM010 mode resonating at the same frequency. The implications to dark-matter axion-searches with cavity experiments are discussed showing that the gain in quality factor is not spoiled by a reduced geometrical coupling C 030 of the cavity mode to the axion field. This reduction effect is estimated to be at most 20%. Numerical simulations show that frequency tuning of several hundreds MHz is feasible.
