Bacillus cereus is a typical spore-forming strain that can transform into dormant spores to resist unfavorable growth conditions. Among the microorganisms that cause spoilage of dairy products, B. cereus accounts for a large proportion. In this study, we evaluated the inhibitory activity of monolauroyl-galactosylglycerol (MLGG) on B. cereus spores and explored its inhibitory mechanism. The minimum inhibitory spore concentration (MISC) and the minimum sporicidal concentration (MSC) of MLGG against B. cereus spores were 0.313 mg/mL and 2.5 mg/mL, respectively. Scanning electron microscopy was used to observe the changes in spore morphology after MLGG treatment. It was found that low concentrations of MLGG caused irregular spore shape, with a few spores slightly ruptured and high concentrations of MLGG increased the degree of spore rupture, resulting in significant leakage of spore contents. By measuring the effect of MLGG on spore structure, it could be determined that MLGG damaged the coat of the spores and weakened the resistance of the spores, but did not affect the activity of cortical lyases. Therefore, spores could not resume growth even with the addition of exogenous lytic enzymes. After propidium iodide (PI) staining of the spores, flow cytometry analysis revealed that the positive area gradually increased, indicating that the permeability of the inner membrane of the spores was disrupted, leading to dipicolinic acid (DPA) release. The amount of DPA release was proportional to the MLGG concentration, and the growth rate of the spores decreased under hypertonic conditions. In April 2023, the World Health Organization (WHO) reported that 17.5% of the global adult population experience infertility. What may be the contribution of per-and polyfluoroalkyl (PFAS) to this global public health problem? This study explored the associations between in vitro fertilization (IVF) outcomes and plasma concentrations of individual PFAS and PFAS mixtures in women undergoing in vitro fertilization and embryo transfer (IVF-ET) and how these exposures might affect IVF outcomes. We analyzed 8 PFASs in plasma samples from women (N = 259) who underwent IVF treatment. In multivariable generalized linear mixed models, there were statistically significant associations of higher plasma concentrations of PFNA with reduced numbers of total retrieved oocytes [12.486 (95%CI: 0.446,25.418), p trend = 0.017], 2 PN zygotes [6.467(95%CI: 2.034,14.968), p trend = 0.007], and cleavage embryos [6.039(95%CI: 2.162,14.240), p trend = 0.008]. Similarly, there was a continuous decline in the numbers of retrieved 2 PN zygotes and cleavage embryos with increasing concentration of PFOS [6.467(95%CI: 2.034,14.968), p trend = 0.009 and 6.039(95%CI: 2.162,14.240), p trend = 0.031,respectively] and a negative association between PFHxS concentrations and clinical pregnancy during the initial cycles of frozen ET [0.525(95%CI:0.410,0.640), p trend = 0.021]. To investigate the joint effect of PFAS mixtures, a confounder-adjusted BKMR model analysis showed inverse relationship between PFAS mixtures and the number of high-quality embryos, 2 PN zygotes and cleavage embryos, to which the greatest contributors to the mixture effect are PFDeA and PFBS, respectively. It demonstrated that PFAS exposure might exert negative effects on oocyte yield, fertilization and high-quality embryo in women undergoing IVF. These findings suggest that exposure to PFAS may increase the risk of female infertility and further studies are needed to uncover the potential mechanisms underlying the reproductive effects associated with PFAS. Objective: This study aimed to analyze the application value of intracavitary electrocardiogram (ECG) classification in peripherally inserted central catheter (PICC) tip localization in patients with cancer.Methods: Using a self-control study method, 325 patients with cancer underwent intracavitary ECGs to position the tip of a PICC catheter. The P wave, QRS wave amplitude, and waveform changes of each intracavitary ECG were recorded. Chest X-ray examination was performed after the catheterization to compare the results of different intracavity ECG maps with the results of the chest X-ray.Results: The intracavitary ECG positioning maps of the 325 patients were divided into four categories: (1) increased P wave (293 cases), accounting for 90.15% (293/325) of all cases; compared with the positioning results of the chest X-rays, the placement rate was 98.98% (290/293); (2) negative deepening of the P wave (1 case), accounting for 0.31% (1/325) of all cases and with a placement rate of 100% (1/1); (3) no change in P wave (19 cases), accounting for 5.85% (19/325) of all cases and with a placement rate of 42.11% (8/19); (4) atrial fibrillation/atrial flutter (12 cases), accounting for 3.69% (12/325) of all cases and with a placement rate of 58.33% (7/12). The four types of intracavitary ECG positioning maps had statistically significant differences (χ2 = 133.924, P = 0.000).Conclusion: There are four types of intracavitary ECG localization maps: increased P wave, negative deepening of the P wave, no change in P wave, and atrial fibrillation/atrial flutter. The increased P wave pattern had the highest occurrence probability and high positioning accuracy. [Display omitted] • Soluble dietary fiber (SDF) from millet bran prevented obesity in mice. • Millet bran SDF improved gut microecology in mice fed with high-fat diet. • Fermented soluble dietary fiber (FSDF) is more effective in preventing obesity. • Unfermented soluble dietary fiber prevented obesity by regulating lipid metabolism. • FSDF reduced appetite in mice through the gut-brain axis, thereby preventing obesity. Obesity-related diabetes, cardiovascular disease, and hypertension pose many risks to human health. Thus, mice on a high-fat diet were gavaged with millet bran (unfermented/fermented) soluble dietary fiber (RSDF/FSDF, 500 mg·kg−1) for 10 weeks in current research, and then evaluated the various biological indicators. These findings revealed that RSDF and FSDF supplements could prevent fat synthesis by inhibiting sterol regulatory element-binding protein-1c gene expression. The RSDF supplements can also accelerate fat catabolism through enhanced the mRNA expression levels of adipose triglyceride lipase and peroxisome proliferator-activated receptor α. FSDF supplements can prevent obesity by decreasing 3-hydroxy-3-methyl-glutaryl-CoA reductase expression and increasing cholesterol 7α-hydroxylase expression. Moreover, FSDF also controls obesity development by lowering total cholesterol and low-density lipoprotein cholesterol levels in the blood, triglyceride, total cholesterol, and bile acid levels in the liver. Notably, FSDF supplements can promote Bacteroides and Prevotella propagation; excretive propionic acid binds to free fatty acid receptor 2/3 and then stimulates intestinal epithelial cells to generate glucagon-like-peptide-1 and peptide YY, which can reduce food and energy intake and ultimately prevent obesity. As an effective anticancer drug, the clinical limitation of doxorubicin (Dox) is the time- and dose-dependent cardiotoxicity. Yes-associated protein 1 (YAP1) interacts with transcription factor TEA domain 1 (TEAD1) and plays an important role in cell proliferation and survival. However, the role of YAP1 in Dox-induced cardiomyopathy has not been reported. In this study, the expression of YAP1 was reduced in clinical human failing hearts with dilated cardiomyopathy and Dox-induced in vivo and in vitro cardiotoxic model. Ectopic expression of Yap1 significantly blocked Dox-induced cardiomyocytes apoptosis in TEAD1 dependent manner. Isorhapontigenin (Isor) is a new derivative of stilbene and responsible for a wide range of biological processes. Here, we found that Isor effectively relieved Dox-induced cardiomyocytes apoptosis in a dose-dependent manner in vitro. Administration with Isor (30 mg/kg/day, intraperitoneally, 3 weeks) significantly protected against Dox-induced cardiotoxicity in mice. Interestingly, Isor increased Dox-caused repression in YAP1 and the expression of its target genes in vivo and in vitro. Knockout or inhibition of Yap1 blocked the protective effects of Isor on Dox-induced cardiotoxicity. In conclusion, YAP1 may be a novel target for Dox-induced cardiotoxicity and Isor might be a new compound to fight against Dox-induced cardiotoxicity by increasing YAP1 expression. Kansui, the root of Euphorbia kansui T.N. Liou ex T.P. Wang (Euphorbiaceae), is a well-known poisonous traditional Chinese medicine (TCM). However, many monographs of TCM indicated that it cannot be co-used with licorice, as kansui-licorice is a typical “eighteen incompatible” medicaments. Our previous studies have indicated that kansui was effective in treating malignant pleural effusion (MPE), and the efficacy could be weakened by the co-use of licorice, even causing serious toxicity at the given ratio. Nevertheless, the actual mechanisms of their dosage-toxicity-efficacy relationship need to be well clarified. The present study aimed to investigate the effect of individual and combined use of kansui and licorice on MPE rats, and explain the underlying mechanisms from a metabolomic perspective. Urine samples were analyzed by ultra-high-performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry (UHPLC-QTOF/MS). Partial least-squares discriminate analysis (PLS-DA) models were built to evaluate the interaction between kansui and licorice. Seven potential biomarkers contribute to the separation of model group and control group were tentatively identified. And selenoamino acid metabolism and nicotinate and nicotinamide metabolism with the impact-value 0.31 and 0.24, respectively, were filtered out as the most important metabolic pathways. Kansui and kansui-licorice at a ratio of 4:1 can treat MPE rats by adjusting abnormal metabolic pathways to the normal state, while it may have opposite result with kansui-licorice 1:4. The different influences to the two metabolic pathways may partially explain the dosage-toxicity-efficacy relationship of kansui-licorice with different ratios. Three strains, including L. fermentum , L. plantarum and S. thermophilus, were combined to ferment blueberry juice. Through the sequential simplex lattice mixture design, regression modeling and genetic algorithm optimization, it was found that the combination of S. thermophilus with either L. fermentum or L. plantarum weakened the capacity of Lactobacillus strains to enrich phenolics, and the combinations of these strains had no synergistic effect of synthesizing lactic acid. The resulting optimal inoculation proportion to enrich phenolics was the mixed L. fermentum and L. plantarum at 0.5:0.5. After fermentation for 48 h, total phenolic, ferulic acid, rutin, and quercetin-3-rhamnoside of mixed fermented samples were 82.19 %, 15.22 %, 79.08 % and 98.59 % higher than the unfermented juice, and their contents were all highest among the fermented samples. Moreover, the samples fermented by mixed strains possessed higher amounts of 3-methyl-1-butanol, 2-methyl-1-propanol, 2-heptanone and 2-pentanone than samples fermented by L. fermentum , S. thermophilus and unfermented samples. In this study, black chokeberries were dehydrated by sequential calcium pretreatment and ultrasonic/microwave drying. The investigation focused on exploring the mechanisms of water transport and metabolic pathways of polyphenols. A diffusional model was modified by considering the factors such as shrinkage, porosity and temperature-dependent water diffusivity to simulate the moisture transport. Accordingly, the Spatio-temporal evolutions of cytoplasm water, extracellular water and total water content in black chokeberry throughout drying were successfully revealed. On the other hand, the quality attributes involved in the metabolic pathways of polyphenols, i.e., free/bound phenolics, cell wall pectin, polyphenol oxidase (PPO) or peroxidase (POD), and viscoelastic properties under these drying treatments were intensively analysed. Through correlation analysis, it was found that CaCl 2 pretreatment strengthened the oxidation of free procyanidin B 2 by PPO. In contrast, both ultrasound and microwave drying weakened the negative influence of PPO on the stability of free rutin. Besides, temperature rise promoted the loss of free polyphenols and the degradation of cell wall pectin, weakening the attachment of bounded-caffeic acid on the cell wall.
