Population-based sewage surveillance has emerged as a promising approach for studying the prevalence of antibiotic resistance in pathogens. To determine the temporal prevalence of cefotaxime-resistant Escherichia coli in sewage from five sewage treatment plants located in Bergen city, to determine whether ESBL- and carbapenemase-producing E. coli are consistently disseminated in the receiving environment through sewage. A total of 569 cefotaxime-resistant E. coli were isolated over a period of 19 months (August 2020 to February 2022) using ECC CHROMagar™ plates from 82 samples, antibiotic sensitivity profiles were determined, using Sensititre™ plates. The draft genome sequences were determined, using Illumina MiSeq-based sequencing. Complete genome sequences were determined, using Oxford Nanopore-based sequencing. All 569 strains obtained from influent (N= 461) and effluent (N= 108) were multi-drug resistant. Most of the sequenced strains (52 of 61) carried bla CTX-M-15 (38.5%) and bla CTX-M-27 (34.6%). The most prevalent sequence types (STs) for ESBL-carrying strains were ST131 (32.8%) and ST38 (21.3%). All CTX-M-27-carrying ST131 strains belonged to clade A or C1, while CTX-M-15-harbouring strains were present in all the clades. Five OXA-244-producing ST38 strains, genetically similar to epidemic-causing strains from Western Norway, France and the Netherlands, were isolated only from raw and treated sewage of the treatment plant receiving hospital sewage. This is the first study showing persistent dissemination of OXA-244-producing ST38 clones through sewage in Norway, demonstrating that hospital sewage is the likely source of OXA-244-producing ST38 clones reaching the receiving environment. Urban environments present new challenges to the integration of photovoltaic systems onto the building envelope. This study describes a multi-objective genetic algorithm developed for the maximization of the PV electrical production and minimization of relevant system costs, such as wiring, modules and inverter costs. Two case studies are analyzed, featuring a partially shaded rooftop and a vertical facade. The PV layouts suggested by the genetic algorithm outperform the conventional configurations by reducing the cost of produced electricity between 6% and 18%. The optimization results also show that layouts with more but shorter PV strings achieve higher energy yields. However, when compared with longer clustered strings in the most sunlit areas of the surfaces, this results in the doubling of costs. [ABSTRACT FROM AUTHOR]

The present work addresses the topic of automated calibration of numerical models starting from the experimental characterization of the structure's dynamic behaviour. The importance of the topic is well known in the literature, especially in cases where it is necessary to have at disposal validated numerical models, necessary for the correct evaluation of the safety of existing buildings. Generally, the calibration problem is developed with a manual approach (manual tuning), with a positive outcome whenever there is a good knowledge of the boundary and internal constraint conditions and the elastic mechanical properties of the construction's constituent materials. Conversely, the positive outcome is particularly difficult to achieve manually when there are non-homogeneous and/or complex structures, as in the cases of historic masonry structures, which are often the result of constructions carried out at different times, organized in aggregates whose interaction between the portions is not simple to understand. For this purpose, the present work, using commercial software and specially prepared routines, illustrates a semi-automatic procedure, which employs genetic algorithms, suitable for the optimized identification of the numerical model that best represents the structure's experimental dynamic behaviour. The procedure is presented with reference to two case studies: the Gabbia Tower historic masonry aggregate in Mantua and the bell tower of the Monastery of the Ursuline nuns in Capriolo, Brescia. In the first case, in addition to the experimental dynamic characterization, a good instrumental characterization of the tower's masonry mechanical properties is available. In the second case, alongside a good experimental dynamic characterization, only a qualitative estimate of the masonry mechanical properties, based on visual inspections, is available. The two case studies allow for testing the validity of the numerical models' calibration procedure, necessary for their application in the field of safety checks. Finally, for the case studies analysed the work presents an assessment of seismic vulnerability starting from the models identified with the semi-automatic procedure. Abstract: The aim of this study was to assess the effects of dietary leucine supplementation in lactating dams, particularly on energy homeostasis through signaling mechanisms in the central nervous system. Dams were fed ad libitum with standard diet during pregnancy (control dams) or supplemented with 2% leucine (leucine-supplemented dams) from delivery onwards. Food intake, body weight and composition were periodically recorded. Hypothalamus was collected at the end of lactation, and the expression of neuropeptide Y (NPY), agouti-related protein (AgRP) pro-opiomelanocortin (POMC), cocaine and amphetamine regulated transcript (CART), insulin receptor (InsR), ghrelin receptor (GSHR), melanocortin receptor (MCR4), leptin receptor (Ob-Rb) and suppressor of cytokine signaling 3 (SOCS3) were analyzed. Dietary leucine supplementation to lactating rats increased plasma leucine by 56%, modulated body composition and contributed to a tendency of higher ratio of lean/fat mass content of dams during lactation, without affecting food intake, thermogenesis capacity or body or tissue/organs weights. No differences in body weight of offspring from control and leucine-supplemented dams were found. The expression of orexigenic peptides (NPY and AgRP) decreased in leucine-dams, whereas the expression of anorexigenic peptides (POMC and CART), the hypothalamic receptors of insulin, ghrelin, melanocortin and leptin and SOCS3 did not change by leucine supplementation. In conclusion, increased leucine intake during lactation may contribute to a healthier profile of body composition in dams, without compromising the growth and development of the progeny by a mechanism associated with lower expression of orexigenic neuropeptides in hypothalamus. • Roller-crimping cover crops reduced cabbage yield compared to full incorporation. • Legume/rye mixtures reduced available N and cabbage root growth compared to legumes. • Roller-crimping reduced soil microbial activity compared to full incorporation. • Roller-crimping suggested reduced N leaching risk during winter. • Roller-crimping suppressed weed emergence by 63% compared to full incorporation. Roller-crimping of cover crops grown during winter may facilitate reduced tillage efforts in organic vegetable production. A one-year field experiment was conducted twice (autumn 2015–2017) in sandy loam soil in Denmark testing two cover crop termination systems, roller-crimping (RC) without tillage before crop planting, and full incorporation by tillage (FI) of mowed cover crops in organic white cabbage (Brassica oleracea L. convar. capitata var. capitata f. alba) production. Three legume species were investigated: winter faba bean (Vicia faba L.), winter pea (Pisum sativum L.), hairy vetch (Vicia villosa Roth), in pure stands or in a mixture with winter rye (Secale cereale L.). Roller-crimping reduced total white cabbage biomass by 31% (2016) and 19% (2017). Marketable white cabbage yield was 100% (2016) or 24% (2017) lower under RC than FI, likely caused by delayed N release from cover crops and reduced soil dehydrogenase activity. White cabbage root growth was reduced by RC following pea/rye, where N availability was low. Despite reduced root growth and yield, RC still had the advantage of reducing weed growth by 63% compared with FI three weeks after cover crop termination in 2017 and of decreasing N leaching risk, indicated by reduced soil mineral N content in 0.5–1.5 m depth in autumn. Marketable yield was 35% higher following legumes compared with legume/rye mixtures in both termination systems in 2017, due to 105 kg N ha−1 higher mineral soil N content in 0–2.5 m depth in the spring and faster N mineralisation from plant material with a lower C/N ratio. This yield increase corresponded with increased root growth following RC legumes. In contrast, legume/rye mixtures had the advantage of decreasing weed growth by 50–68% and N leaching risk by 22 kg ha−1 in 0.5−2 m depth. Although RC demonstrated ecological benefits such as weed suppression and indicated reduced N leaching risk, the trade-off with yield losses could create a barrier to adoption.
