28. Unveiling the human nitroproteome: Protein tyrosine nitration in cell signaling and cancer.

Abstract

Covalent amino acid modification significantly expands protein functional capability in regulating biological processes. Tyrosine residues can undergo phosphorylation, sulfation, adenylation, halogenation, and nitration. These posttranslational modifications (PTMs) result from the actions of specific enzymes: tyrosine kinases, tyrosyl-protein sulfotransferase( s), adenylate transferase(s), oxidoreductases, peroxidases, and metal-heme containing proteins. Whereas phosphorylation, sulfation, and adenylation modify the hydroxyl group of tyrosine, tyrosine halogenation and nitration target the adjacent carbon residues. Because aberrant tyrosine nitration has been associated with human disorders and with animal models of disease, we have created an updated and curated database of 908 human nitrated proteins. We have also analyzed this new resource to provide insight into the role of tyrosine nitration in cancer biology, an area that has not previously been considered in detail. Unexpectedly, we have found that 879 of the 1971 known sites of tyrosine nitration are also sites of phosphorylation suggesting an extensive role for nitration in cell signaling. Overall, the review offers several forward-looking opportunities for future research and new perspectives for understanding the role of tyrosine nitration in cancer biology. [ABSTRACT FROM AUTHOR]

Prenner (a), Eric Schenkel (b) Keywords: Allergic rhinitis; Desloratadine; Perennial allergic rhinitis; Seasonal allergic rhinitis; Treatment Abstract: Allergic rhinitis is a common medical condition characterized by nasal, throat, and ocular itching; rhinorrhea; sneezing; nasal congestion; and, less frequently, cough. The treatment of allergic rhinitis should control these symptoms without adversely affecting daily activities or cognitive performance and should prevent sequelae such as asthma exacerbation or sinusitis. This review describes a stepwise approach to treatment of allergic rhinitis derived from a synthesis of clinical trial results, patient preferences, and real-world tolerability data. Key clinical considerations include frequency and intensity of symptoms, patient age, comorbidities, compliance with treatment regimens (influenced by formulation, route and frequency of administration), and effects on quality of life. Oral second-generation antihistamines, versus first-generation agents and inhaled corticosteroids, should be considered first-line treatment because they provide rapid relief of most allergic rhinitis symptoms without safety and tolerability issues. Additional therapeutic agents can then be added or substituted based on individual symptom response. Abstract

Highlights • Thyme extract KMTv24497 containing authorised medicinal product (Aspecton® Oral Drops) showed antiviral activity. • Aspecton® Oral Drops was shown to have Antiviral activity against human rhinovirus and against influenza A virus. • Concentrations showing an antiviral effect are non-toxic. Abstract The common cold is a respiratory disease often caused by viral pathogens, including a major group of human rhinoviruses (HRV). Currently, there is no specific antiviral approach available against these viruses. Another viral pathogen that causes respiratory diseases is the influenza virus (IV). Even though a few approved drugs are available for antiviral treatment against IV, these are prone to lose their effectiveness due to the rapid emergence of resistant virus variants. Therefore, new effective approaches characterised by a broad antiviral activity and wide availability would be very advantageous. In this study the authors aimed to investigate the antiviral potential of an authorised medicinal product (Aspecton® Oral Drops) containing a specific thyme extract (KMTv24497) against human rhinovirus serotype 1a (HRV1a), as well as against an influenza A virus isolate of the 2009 pandemic (H1N1pdm09) at non-toxic concentrations. To the best of the authors knowledge, they were able to demonstrate, for the first time, the in vitro antiviral activity of a thyme extract containing an authorised medicinal product in non-toxic concentrations against an HRV, as well as against an influenza A virus strain in cell culture. This study represents a first approach to prove a causal therapeutic effect in addition to the already described symptomatic effects for a thyme preparation. Patients with common cold infections may profit from the additional causal therapeutic effects of an expectorant. [ABSTRACT FROM AUTHOR]
