No previous controlled studies have been specifically designed or adequately powered to show the efficacy of topical oxybutynin for palmar hyperhidrosis by using quantitative measures. To evaluate efficacy of 20% oxybutynin hydrochloride lotion (20% OL) in reducing palmar sweat volume in patients with primary palmar hyperhidrosis (PPHH). In a randomized controlled trial, Japanese patients with PPHH aged 12 years and older received either 20% OL (n = 144) or placebo (n = 140) on both palms once daily for 4 weeks. Palmar sweat volume was measured by the ventilated capsule method. For the primary outcome, response was defined as a reduction of sweat volume of at least 50% from baseline. At week 4, the responder rate for sweat volume was significantly higher in the 20% OL arm than in the placebo arm (52.8% vs 24.3%, respectively; treatment difference, 28.5% [95% CI, 17.7% to 39.3%]; P <.001). No serious adverse events occurred, and no adverse events led to treatment discontinuation. The treatment period was only 4 weeks. Abstract: The mutual interaction of oxybutynin hydrochloride (OB) with bovine serum albumin (BSA) was investigated by fluorescence, UV–vis absorption, circular dichroism (CD), and Fourier transform infrared (FT-IR) spectroscopies under simulative physiological conditions. The results of fluorescence titration revealed that OB could quench the intrinsic fluorescence of BSA by static quenching and there was a single class of binding sites on BSA for this drug. The thermodynamic parameters ΔH, ΔS, and ΔG calculated at different temperatures indicated that hydrogen bonds and van der Waals interactions were the dominant intermolecular forces in stabilizing the OB–BSA complexes. According to the theory of Förster’s non-radiation energy transfer, the binding distance r between OB and BSA was evaluated to be 3.27nm. The displacement experiments confirmed that OB could bind to site I of BSA. The FT-IR and CD spectra showed that the binding of OB to BSA induced conformational changes in BSA. Abstract: Simple, accurate, sensitive and validated UV spectrophotometric, chemometric and HPTLC-densitometric methods were developed for determination of oxybutynin hydrochloride (OX) in presence of its degradation product and additives in its pharmaceutical formulations. Method A is the first derivative of ratio spectra (DD1) which allows the determination of OX in presence of its degradate in pure form and tablets by measuring the peaks amplitude at 216nm. Method B and C are principal component regression (PCR) and partial least-squares (PLS) for determination of OX in presence of its degradate in pure form, tablets and syrup. While, the developed high performance thin layer chromatography HPTLC-densitometric method was based on the separation of OX from its degradation product, methylparaben and propylparaben followed by densitometric measurement at 220nm which allows the determination of OX in pure form, tablets and syrup. The separation was achieved using HPTLC silica gel F254 plates and chloroform:methanol:ammonia solution:triethylamine (100:3:0.5:0.2, v/v/v/v) as the developing system. The accuracy, precision and linearity ranges of the developed methods were determined. The results obtained were statistically compared with each other and to that of a reported HPLC method, and there was no significant difference between the proposed methods and the reported method regarding both accuracy and precision. Abstract: Two high performance liquid chromatographic (HPLC) methods are presented for the determination of oxeladin citrate (OL) and oxybutynin hydrochloride (OB) and their degradation products. The first method was based on HPLC separation of OL from its degradation product using a Nucleosil C18 column with a mobile phase consisting of acetonitrile –0.1% phosphoric acid (60:40 v/v). The second method was based on HPLC separation of OB from its degradation product using a VP-ODS C18 column with a mobile phase consisting of acetonitrile/0.01 M potassium dihydrogen phosphate/diethylamine (60:40:0.2). Quantitation was achieved with UV detection at 220 nm based on peak area. The two HPLC methods were applied for the determination of OL or OB, their degradation products, methylparaben and propylparaben in pharmaceutical preparations. The proposed methods were used to investigate the kinetics of acidic and alkaline degradation processes of OL and OB at different temperatures and the apparent pseudofirst-order rate constant, half-life and activation energy were calculated. The pH-rate profiles of degradation of OL and OB in Britton–Robinson buffer solutions within the pH range 2–12 were studied. [Copyright &y& Elsevier]

Abstract: The purpose of this investigation is to understand the influence of gastrointestinal (GI) pH on the gel layer formation and its dynamics for various hydrophilic/swellable matrices, in the process of developing a pH-independent controlled release system for a basic drug, oxybutynin hydrochloride (OXB). Cylindrical matrices (8-mm diameter) without and with fumaric acid, were readily prepared by direct compression. Formulations were evaluated for in vitro drug release, and gel layer dynamics was studied by viscosity measurements and texture profiling analysis. In the in vitro drug release study, OXB, which shows pH-dependent solubility, showed faster release from all the matrices in pH 1.2 medium. Release rates enhanced to a lesser extent with change of medium from pH 6.8 to pH 1.2, for HPMC polymer matrices. Anionic polymer matrices showed drastic differences in the release rates when medium was changed from pH 6.8 to pH 1.2. Addition of fumaric acid to matrices demonstrated pH-independent drug release, which was attributed to the micro-environmental pH manipulation within the hydrated gel layer. Viscosity and texture profiling studies revealed that saturation solubility of drug at swelling front play a major role in the pH-dependent drug release from HPMC matrices, while both saturation solubility and the altered gel consistency as a function of pH are involved with anionic polymer matrices. Presence of fumaric acid in HPMC matrices showed efficient retardation and pH-independent drug release. In conclusion, understanding the influence of GI physiological pH on the gel layer dynamics and manipulating the micro-environmental pH provides efficient and predictable in vivo performance from these swellable cylindrical matrices. A new UV spectrophotometric method and a reversed-phase HPLC method were developed for quantitative evaluation of oxybutynin hydrochloride (OXB) formulations. Determination of OXB by UV spectroscopic method was based on complexation of OXB with picric acid to form picrate, which was extracted to chloroform. The picrate complex showed quantifiable absorbance at 344nm. Chromatography was carried out at 25°C on a 4.6mm × 250mm 5μm cyano column that contained USP packing L10 with water:methanol:acetonitrile::48:12:40 (v/v), as mobile phase. UV detector was set at 203nm. Both methods were found to be selective, linear, accurate and precise in the specified ranges. The LOD and LOQ of HPLC method were 0.5 and 1.65μg/ml, respectively. Intra-day and inter-day variability for both methods were <2% RSD. These methods were successfully used for quantification of OXB in drug-release studies from immediate-release tablets and controlled-release (CR) formulations. [Copyright &y& Elsevier]
