Abstract

There is an association between body composition and lung function, assessed by spirometry, but the effects of body compartments on static lung volumes and its changes during lung growth remain to be explored. We aimed to investigate the association of appendicular lean mass, reflecting skeletal muscle mass, and fat mass on forced and static lung function measures in childhood and adolescence. In total, 1489 children and adolescents (6–18 years) of the observational, longitudinal (first and second visit within 4 years), general population-based LEAD study have been investigated. The association of appendicular lean mass and fat mass indices (ALMI and FMI; assessed by dual-energy X-ray absorptiometry) on lung function by spirometry (FEV 1 , FVC) and body plethysmography (TLC, RV, FRC) was investigated cross-sectionally. Longitudinal associations between lung function and body compartment changes between the two visits were analyzed. The ALMI is positively associated with FEV 1 , FVC, and TLC. Contrary, FMI is inversely associated with lung function measures including FRC and RV. During the phase of lung growth, higher gain in muscle mass is associated with higher increases of FVC and TLC. This study demonstrates the different effects of muscle and fat mass on forced expiratory and static lung volumes. Achieving and maintaining muscle mass in childhood and adolescence might become an important preventive strategy for lung health in adulthood. [ABSTRACT FROM AUTHOR]

Abstract

The transcriptional regulator BRD4 has been shown to be important for the expression of several oncogenes including MYC. Inhibiting of BRD4 has broad antiproliferative activity in different cancer cell types. The small molecule JQ1 blocks the interaction of BRD4 with acetylated histones leading to transcriptional modulation. Depleting BRD4 via engineered bifunctional small molecules named PROTACs (proteolysis targeting chimeras) represents the next-generation approach to JQ1-mediated BRD4 inhibition. PROTACs trigger BRD4 for proteasomale degradation by recruiting E3 ligases. The aim of this study was therefore to validate the importance of BRD4 as a relevant target in colorectal cancer (CRC) cells and to compare the efficacy of BRD4 inhibition with BRD4 degradation on downregulating MYC expression. JQ1 induced a downregulation of both MYC mRNA and MYC protein associated with an antiproliferative phenotype in CRC cells. dBET1 and MZ1 induced degradation of BRD4 followed by a reduction in MYC expression and CRC cell proliferation. In SW480 cells, where dBET1 failed, we found significantly lower levels of the E3 ligase cereblon, which is essential for dBET1-induced BRD4 degradation. To gain mechanistic insight into the unresponsiveness to dBET1, we generated dBET1-resistant LS174t cells and found a strong downregulation of cereblon protein. These findings suggest that inhibition of BRD4 by JQ1 and degradation of BRD4 by dBET1 and MZ1 are powerful tools for reducing MYC expression and CRC cell proliferation. In addition, downregulation of cereblon may be an important mechanism for developing dBET1 resistance, which can be evaded by incubating dBET1-resistant cells with JQ1 or MZ1. [ABSTRACT FROM AUTHOR]

Abstract

The cosmopolitan dream of a borderless world has little to do with reality. Today's borders bear witness to regulatory intervention in the circulation of goods, information, capital and people. These interventions, naturally, have an impact at border regions. For analyzing these impacts, we map, quantify and relate border typologies, development dynamics near borders, and economic and political indicators of neighboring nation-states. We do so on global scale for all current 315 land borders. We rely on data from a mix of border dossiers, in-depth literature review, censuses and multi-temporal mapping products from satellite imagery. Our analysis strategy is two-fold: First, in a descriptive analysis, we map the various border typologies. And, we also compute development dynamics over a 15-year period from 2000 to 2015. Since there are few consistent, appropriately spatially resolved, and globally available datasets, we measure development by the proxies 'settlements' and 'population' instead of the usual economic characteristics. We use an ensemble of metrics that show not only the developments in the border region but also the dynamics in the border region relative to the respective nation-state. By means of a global ranking, we show the variability of development dynamics at borders across the globe. Second, we relate these dynamics to the different border typologies, and to economic and political differences of neighboring nation-states. We find the following trends: higher political or economic differences of neighboring nation-states relate to stronger border fortification, greater economic or political disparities relate to stronger population or settlement accumulation at the poorer or less free side of the border, and stronger fortification hinders settlement and population development to a certain degree. These empirically measured trends, however, are only partially statistically significant and not as strong or unambiguous as assumed. In a critical discussion, we reflect on the capabilities and limitations of such an empirical global approach. • Global classification of five border typologies for all 615 land borders. • Ranking of development dynamics in terms of settlements and population in all 615 border regions across the globe. • Relationships are found between the dynamics in border regions and different border typologies. • Greater political or economic disparities of neighboring countries are found to relate to stronger border fortification. • These disparities relate to stronger population or settlement accumulation at the poorer or less free side of the border. Abstract

The cleanability of food contact surfaces is determined by surface characteristics and the properties of the soiling material. Effects of selected physicochemical parameters of biopolymer soils on the removal from stainless steel were studied using a laboratory flow cell system with continuous removal measurement and, additionally, gravimetric detection. The results show the potential for improving cleaning processes on the basis of structural properties of the biopolymers. A higher cleaning efficiency was achieved when surface energy and cationic charge of the polymers was lower, indicating that adhesive interactions (van der Waals forces of attraction and electrostatic forces) between surface and polymer play a dominant role. The effects of pre-heating treatments of the soils, inducing molecular changes, on removal behavior were however not significant. The results indicate that a holistic view of product and process design is useful to develop efficient cleaning protocols. [ABSTRACT FROM AUTHOR]

Abstract

Diabetic dyslipoproteinemia characterized by hypertriglyceridemia, low high-density lipoprotein (HDL) cholesterol, and often elevated low-density lipoprotein (LDL) cholesterol with predominance of small, dense LDL is a strong risk factor for atherosclerosis. It is unclear whether fibrate or statin therapy is more effective in these patients. We compared atorvastatin (10 mg/day) with fenofibrate (200 mg/day), each for 6 weeks separated by a 6-week washout period in 13 patients (5 men and 8 women; mean age 60.0+/-6.8 years; body mass index 30.0+/-3.0 kg/m2) with type 2 diabetes mellitus (hemoglobin A1c 7.3+/-1.1%) and mixed hyperlipoproteinemia (LDL cholesterol 164.0+/-37.8 mg/dl, triglycerides 259.7+/-107 mg/dl, HDL cholesterol 48.7+/-11.0 mg/dl) using a randomized, crossover design. Lipid profiles, LDL subfraction distribution, fasting plasma viscosity, red cell aggregation, and fibrinogen concentrations were determined before and after each drug. Atorvastatin decreased all LDL subfractions (LDL cholesterol, -29%; p <0.01) including small, dense LDL. Fenofibrate predominantly decreased triglyceride concentrations (triglycerides, -39%; p <0.005) and induced a shift in LDL subtype distribution from small, dense LDL (-31%) to intermediate-dense LDL (+36%). The concentration of small, dense LDL was comparable during therapy to both drugs (atorvastatin 62.8+/-19.5 mg/dl, fenofibrate 63.0+/-18.1 mg/dl). Both drugs induced an increase in HDL cholesterol (atorvastatin +10%, p <0.05; fenofibrate +11%, p = 0.06). In addition, fenofibrate decreased fibrinogen concentration (-15%, p <0.01) associated with a decrease in plasma viscosity by 3% (p <0.01) and improved red cell aggregation by 15% (p <0.05), whereas atorvastatin did not affect any hemorheologic parameter. We conclude that atorvastatin and fenofibrate can improve lipoprotein metabolism in type 2 diabetes. However, the medications affect different aspects of lipoprotein metabolism. Abstract

Electric vehicles (EVs) have high energy efficiency and low pollutant and greenhouse gas (GHG) emissions compared with conventional internal combustion engine vehicles (ICEVs). This study examines the economic competitiveness of battery electric vehicles (BEVs) in the Chinese market. BEVs are compared with ICEVs using benefit-cost analyses from the perspectives of consumers, society and GHG emissions. A life-cycle cost model is developed to evaluate the lifetime cost of a vehicle. The results show that, with central government subsidies, the BEV life-cycle private cost (LCPC) is about 1.4 times higher than comparable ICEVs. Central government subsidies on BEVs will not be cost effective and efficient unless the annual external cost reduction from using BEV reaches $2500 for a compact vehicle or $3600 for a multi-purpose vehicle. That total cost level would imply a carbon cost of more than $2100 per ton. The current life-cycle external cost reductions from using BEV are around $2000–$2300, which are smaller than government subsidies or LCPC differences between BEV and ICEV.
