15. Dual immunoplatform to assess senescence biomarkers TIMP-1 and GDF-15: Advancing in the understanding of colorectal cancer.

Abstract

In this work, we report the development of electrochemical bioplatforms for both the single determination of TIMP-1 and the simultaneous determination of TIMP-1 and GDF-15, two biomarkers of cellular senescence and prognosis in colorectal cancer (CRC). The designed immunoplatforms rely on a sandwich-type configuration labeled with horseradish peroxidase (HRP) built on magnetic microsupports and involve a pair of specific antibodies to capture and detect each target protein. Amperometric readout was performed upon trapping the magnetic bioconjugates on the surface of either single or dual disposable carbon electrodes, using the hydroquinone/hydrogen peroxide (HQ/H 2 O 2) system. The attractive analytical performance of the TIMP-1 bioplatform, achieving a LOD of 13.0 pg mL‒1 and a dynamic range between 43.4 and 2500 pg mL‒1, led us to exploit it for TIMP-1 determination in cell extracts and tissues of CRC samples. The results showed the possibility of discriminating the metastatic capabilities of cells and detecting CRC patients, after just a simple dilution and in only 60 min. The multiplexing feasibility of the developed immunoplatform allowed the implementation of a dual assay for the simultaneous determination of TIMP-1 (LOD 19.2 pg mL‒1) and GDF-15 (LOD 16.6 pg mL‒1). The dual platform was used for the analysis of CRC samples (plasma and cell secretomes) in just 75 min and using a very simple protocol. These characteristics make the developed immunoplatform a very attractive tool to face particularly challenging samples, such as secretomes or plasma from CRC patients, due to the required sensitivity, thus providing a better snapshot of CRC. Abstract

Ethylene oxidation by TiO 2 photocatalysis was studied in a mili-photoreactor (NETmix), as a potential clean postharvest technology to scavenge ethylene from fruit. The NETmix photoreactor comprises a stainless steel slab imprinted with a network of cylindrical chambers interconnected by prismatic channels, sealed by a UVA transparent borosilicate slab (BS), and a UVA LED plate paced above the reactor window. A thin film of the photocatalyst was immobilized in the NETmix network or on the side of the borosilicate slab in contact with the ethylene gas stream. Ethylene oxidation in single-pass mode (continuous operation) was evaluated as a function of TiO 2 crystallinity, TiO 2 loading, radiant power, illumination mechanism, feed stream composition (ethylene and water concentration), and flow rate. The catalyst loading and deposition surface, and relative humidity (RH) of the feed stream strongly affected the ethylene conversion, mineralization, and reaction rate. Even under severe conditions (RH = 25% and Q feed = 571 cm3 min−1 for the back side illumination), the NETmix removed at least 0.11 μmol ethylene min−1. The high efficiency of the photocatalytic NETmix reactor allows the removal of ethylene from the storage system at the beginning of fruit ripening, the period of the highest gas production, enabling it to be used as a promising technology in controlling fruit ripening. Abstract

The adverse effects of excessive and problematic social networking sites (SNSs) use are well described in academic and practitioner literature. However, it remains unclear how these problematic uses arise. By exploring the phenomenon of SNS addiction, this paper identifies how the problematic use of an SNS emerges and develops. We hypothesize that the flow state induced by using SNSs and the sense of belonging are two mediators of the influence of the user's motivations on the development of an SNS addiction. We used variance-based structural equation modeling (PLS-SEM) to analyze data collected from a sample of TikTok SNS users and test the research hypotheses. The results suggest that different addiction mechanisms are associated with different motivations to continue using the social network. This research brings new insights to the social media addiction literature by identifying one new alternative addiction mechanism – via the sense of belonging – by which addiction to social networks can develop. Such knowledge may help inform social networking providers and organizations of social media users' engagement triggers. It also helps inform public health officials on how to help users overcome addictive behaviors on social media. • SNS addiction occurs through two different mechanisms depending on the user's motivations fulfilled by the SNS. • The need for escapism can lead to addiction through the application-induced flow state and/or the sense of belonging state. • Social interaction needs can lead to addiction exclusively through a feeling of belonging to the TikTok community. • The gratification sought by the user is paramount in determining the source of addiction and respective solutions. Abstract

Abstract: A new class of bis-di-organotin (IV) compounds were synthesized in good yields (ca. 80%) by a one-step five molecules reaction in which intervene two molecular equivalents of leucine, one of 5,5′-methylene-bis-salicylaldehyde and either two of di-n-butyltin (IV) or diphenyltin (IV) oxides, that independently provided two bis-di-organotin derivatives. The structures of these two compounds were established by 1H, 13C and 119Sn NMR, as well as by IR and elemental analysis. In both cases the 119Sn chemical shifts are characteristic of pentacoordinated atoms in solution of non-coordinating solvent (CDCl3). Once characterized, the novel compounds were tested as corrosion inhibitors at 25, 50 and 100mgL−1 on mild steel surfaces in sour brine by linear polarization resistance and potentiodynamic curves. These experiments prove that these derivatives present top efficiencies of 54% and 68% for butyl and phenyl moieties, respectively. In addition, these two compounds were tested in vitro against Bacillus subtilis (Gram-positive, strain ATCC 6633), Escherichia coli (Gram-negative, strain DH5) and Pseudomonas fluorescens strains to assess their bactericidal activities, being more effective for the E. coli strain, at concentrations as low as 25mgL−1. Moreover, in order to correlate different activities and ascertain homologous action mechanisms, the two derivatives were tested for their asphaltene inhibition/dispersion activity. The results suggest that the presence of the four n-butyl moieties provide the needed amphiphillic balance to obtain better efficiencies in the tested activities. [Copyright &y& Elsevier]

Abstract: PCR and Southern blot hybridization were used to determine the distribution of Trypanosoma cruzi clones in 37 chronic chagasic cardiopathic and non-cardiopathic patients. Parasite DNA amplified from peripheral blood or dejections of Triatoma infestans fed on patient blood was hybridized with probes containing hypervariable minicircle nucleotide sequences capable of detecting three sublineages of T. cruzi. Probes Z-I and Z-IIb detect unique sequences in lineages TcI and TcIIb, respectively. Probe Z-hybrid detects sequences of lineages TcIId and TcIIe. T. cruzi clones of the Z-I sublineage were detected in 62.2% of T. infestans dejections and 5.4% of peripheral blood samples. Clones of Z-IIb and Z-hybrid sublineages had similar distribution in blood and dejection samples. Interestingly, clones of the Z-IIb sublineage were significantly lower in cardiopathic than in non-cardiopathic patients (23.5% versus 75%; P =0.0006). Clones of the Z-hybrid sublineage were found in 29.4% of cardiopathic and 75% of non-cardiopathic patients, respectively (P =0.0051). By contrast, clones of sublineage Z-I were similarly distributed in both groups of patients. The low frequency of Z-IIb and Z-hybrid sublineage clones detected in cardiopathic patients suggests that the immunological mechanisms involved in controlling and eliminating these T. cruzi parasites may be detrimental to the host, leading to the development of chagasic cardiomyopathy. Abstract: The crystal structure of the elicitin β-cinnamomin (β-CIN) was determined in complex with ergosterol at 1.1 Å resolution. β-CIN/ergosterol complex crystallized in the monoclinic space group P21, with unit cell parameters of a =31.0, b =62.8, c =50.0 Å and β =93.4° and two molecules in the asymmetric unit. Ligand extraction with chloroform followed by crystallographic analysis yielded a 1.35 Å structure of β-CIN (P43212 space group) where the characteristic elicitin fold was kept. After incubation with cholesterol, a new complex structure was obtained, showing that the protein retains, after the extraction procedure, its ability to complex sterols. The necrotic effect of β-CIN on tobacco was also shown to remain unchanged. Theoretical docking studies of the triterpene lupeol to β-CIN provided an explanation for the apparent inability of β-CIN to bind this ligand, as observed experimentally. [Copyright &y& Elsevier]

Abstract

This work presents a disruptive approach to promote highly-efficient photo-Fenton process at neutral pH under continuous mode operation. The system consists of a tube-in-tube membrane reactor designed for continuous-flow titration of low iron doses to the annular reaction zone (ARZ). A concentrated acidic ferrous ion (Fe2+) solution is fed by the lumen-side of the membrane, permeating through the membrane pores (inside-out mode), being dosed and uniformly delivered to the membrane shell-side. Polluted water, containing amoxicillin (AMX) and oxidant (H 2 O 2), flows continuously in the reactor annulus (space between the membrane shell-side and an outer quartz tube). The catalyst radial dispersion is enhanced by the helicoidal movement of water around the membrane shell-side, efficiently promoting its contact with H 2 O 2 and UV light. The efficiency of photochemical and photocatalytic oxidation was evaluated as a function of catalyst dose, catalyst injection mode (radial permeation vs injection upstream from the reactor inlet), light source (UVA vs UVC) and aqueous solution matrix (synthetic vs real wastewater). At steady-state, photo-Fenton reaction with Fe2+ radial addition, driven by UVC light, showed the highest AMX removal for synthetic (∼65%, removal rate of 44 μM AMX /min, using [Fe2+] ARZ = 2 mg/L and [H 2 O 2 ] inlet = 10 mg/L) and real municipal wastewaters (∼45%, removal rate of 31 μM AMX /min, with [Fe2+] ARZ = 5 mg/L and [H 2 O 2 ] inlet = 40 mg/L), with a residence time of only 4.6 s. Highlights from the article: To describe frequency of elementary lesion on protocol and for cause transbronchial biopsy (TBB) based on a "real life" systematic prospective analysis in lung transplanted patients. 346 patients and 2182 TBB (1767 protocol and 415 for cause, median (IQR 25-75) number of biopsy/patient: 6.3 (4-8)) were included. These preliminary analysis demonstrate a relatively higher frequency of lesion free biopsies in protocol biopsy, with rare isolated cellular rejection A. Capillary lesion, intra alveolar fibrin and pneumocyte hyperplasia were more frequent in for cause biopsies than protocol.
