The identification of anticancer active ingredients and their molecular targets in traditional Chinese medicine is a great challenge in modern pharmacology research. Evodiamine is one of the bioactive components isolated from the fruit of Wu-Zhu-Yu and displays significant pharmacological activities, especially anti-inflammatory, antimicrobial, regulate metabolic syndrome and neuroprotective activity. Evodiamine has attracted great interest recently for its potential anticancer activities, and has also been found to inhibit the proliferation of various cancer cells and arrest cell cycle and cell migration. In recent years, evodiamine has been found to have potential toxic effects, mainly manifested as hepatotoxicity and cardiotoxicity. However, the pharmacological and toxicological mechanism of evodiamine is not clear. In this review, we summarized the anticancer effects of evodiamine and its target molecules in vitro and in vivo, focusing on key molecules such as HIF-1α, NF-κB and STAT3, and proposed that epigenetic modifications (DNA methylation, histone acetylation and microRNA) mediate the regulation of key molecules, which are still being explored and excavate into clinical practice. It is known that the male hypogonadism plays an important role in regulating adipose metabolism. In the present study, fifteen pairs of full male sibs were divided into a castrated group and an intact group with a paired experiment design. The pigs were slaughtered at an age of 175 days. The carcass characteristics and fat deposit of the studied animal were measured, and the hormone and serum lipid levels of the peripheral blood samples were determined, and the differentially expressed genes of the back fat between the two groups were screened with porcine genome array. Our results showed that the absence of male gonadal steroids attributed to castration significantly raised the serum lipid levels and increased fat accumulation in the pigs. A total of 225 differentially expressed genes were identified between the boars and barrows and 135 of them were upregulated. The analysis of Gene Ontology categories and KEGG pathway indicated that these differentially expressed genes were mainly involved in metabolism of lipid, carbohydrate, amino acid, xenobiotics biodegradation, and immune diseases pathways. Our results indicated that there were higher capacity of fatty acid of synthesis, enhanced uptaking capacity of fatty acids and cholesterol, inhibited lipolysis, and enhanced carbohydrate metabolism in the adipose tissue of barrows compared to boars. In this study, the pectin of waste seed melon peel in Xinjiang was used as raw material to prepare seed melon pectin oligosaccharide (Sm-POS). The separation and purification of Sm-POS were carried out by HPD-750 macroporous resin chromatography column and gel chromatography to further study the molecular structure of its active component with antibacterial effect by FT-IR, NMR, and LC/MS. The antibacterial activity and mechanism of the Sm-POS active component were investigated by inhibition zone experiment, minimum inhibitory concentration determination, cell growth curve determination, scanning electron microscopy, and flow cytometry analysis. Results showed that Sm-POS-2 was an oligosaccharide composed of two galacturonic acids, one glucose, one arabinose, and one xylose. It showed good antibacterial activity and suppressed the growth of E. coli and S. aureus at minimum inhibitory concentrations (MIC) of 25.0 and 50.0 g/L, respectively. The inhibitory effect of Sm-POS on E. coli and S. aureus was mainly due to cell membrane contraction or rupture, leading to the apoptosis or necrosis of bacterial cells. [ABSTRACT FROM AUTHOR]

In the realm of contemporary medical research, the extraction of pivotal attributes associated with Parkinson's disease (PD) from intricate Magnetic Resonance Imaging (MRI) data characterized by multifaceted dimensions and non-linearity stands as a formidable challenge in the pursuit of realizing automated diagnostic support. To address this issue, an approach is introduced, denoted as the Deep Frequency-Channel Attention Factorization (Deep FCAF), designed to address the complexities inherent to the neurodegenerative ailment of Parkinson's. The Deep FCAF amalgamates three key elements: (1) the inherent capacity for non-linear processing exhibited by neural networks, (2) the inherent capability of the attention mechanism to capture global interdependencies, and (3) the underpinning principles of tensor decomposition within the order of multi-dimensional data structures. The experiments conducted on the identification of PD underscore the efficacy of Deep FCAF in assimilating structural and non-linear factor matrices, consequently leading to an improved classification performance. Notably, the elucidation of functional connectivity based on the acquired factor matrices align with previous scholarly investigations.
