Summary: Administration of Penicillium polonicum extract to male Sprague-Dawley rats (200 g), either mixed in feed or given daily by gavage, for 5 days, had no clinical effects. However, at necropsy on day 6 marked histopathological changes occurred in renal tubule epithelia, including mitotic figures, karyomegalic nuclei, and frequent apoptosis identified specifically by TUNEL methodology and confocal microscopy. Ochratoxin A given similarly to rats (daily, 1mg or 0.2 mg) was also clinically asymptomatic except for the 1mg dose given by gavage; rats in this group lost weight. Marked renal tubular necrosis, though even without any significant accompanying apoptosis, was evident only at this higher dose by gavage; it was associated also with the highest incidence of renal DNA adducts and a disproportionately high concentration of ochratoxin A in plasma on day 6. Significantly fewer renal DNA adducts were detected in rats given 1mg ochratoxin A in feed. The study demonstrates the potential for exaggerated toxicological responses to ochratoxin A administered by gavage through predicted consequential surges in the circulating concentration of the mycotoxin. A methodology was developed to quantify the efficiency of yeast-based products for adsorption of three mycotoxins: zearalenone (ZEA), aflatoxin B1 (AFB1), and ochratoxin A (OTA). Eight products were tested (yeast cell wall or inactivated yeast). The described experimental protocol based on in vitro tests provided reliable isotherms for each mycotoxin. The most suitable models were the Hill model for ZEA, the Langmuir model for AFB1, and the Freundlich model for OTA. From these models, original mathematical affinity criteria were defined to quantify the product adsorption performances for each mycotoxin. The best yeast product, a yeast cell wall from baker's yeast, can adsorb up to 68% of ZEA, 29% of AFB1, and 62% of OTA, depending on the mycotoxin concentrations. The adsorption capacity largely depended both on yeast composition and mycotoxin, but no direct correlation between yeast composition and adsorption capacity was found, confirming that adsorption of mycotoxin on yeast-based products involves complex phenomena. Zearalenone (ZEA) contamination in food samples plays a critical role in food safety, since it causes serious health problems. Usage of microorganisms, such as lactic acid bacteria (LAB), is a promising new approach for detoxification. Eight Lactobacillus pentosus strains were evaluated for their ability to remove ZEA from a sodium acetate buffer solution with initial ZEA concentrations of 5.51–74.70 μg/mL. The adsorption capacity increased with increasing ZEA concentrations. The strain JM0812 showed the highest adsorption capability, at 83.17%, in solution containing 74.70 μg/mL ZEA, followed by UM054 (82.78%) and UM055 (81.69%), respectively. Three adsorption isotherms were applied to predict the removal efficiency of ZEA and the Freundlich isotherm appeared to have the best-fit for ZEA sorption onto bacterial cells. Abstract: The application of membrane bioreactor (MBR) technology was investigated with the aim of evaluating its potential for cytostatic drug and cytotoxicity bioremoval. The toxicity removal was assessed from biomarker test. CP removal of up to 80% was achieved under the operating conditions studied (HRT of 48h and a SRT of 50days). The increase of TMP was associated with an increase of supernatant toxicity as if fouling led to retention of the toxicity. Peaks of supernatant cytotoxicity were correlated with peaks in supernatant humic acid contents. It may suggest that molecules with a toxic effect may be adsorbed or entrapped in humic acids substances. Our study then points out that advances in wastewater treatment using an MBR can provide a suitable process for lowering CP concentrations before discharge into the aqueous environment. However, a tertiary treatment is necessary if complete elimination of toxicity is targeted. [Copyright &y& Elsevier]

Abstract: The influence of cyclophosphamide and its principal metabolites (CPs) on the physicochemical properties of the mixed liquor of a cross-flow membrane bioreactor and the consequences for membrane fouling were investigated. The influence of CPs was determined by comparing the performance of two bioreactors running in parallel, MBR-CPs (with CPs) and MBR-control (without CPs). The physicochemical properties of the mixed liquor were characterized by soluble extracellular polymeric substances (soluble EPS content), particle size distribution and specific cake resistance. Results suggested that the CP toxicity altered the characteristics of the biological matrix of the activated sludge. Micro-organisms exposed to CPs showed higher endogenous respiration rates than MBR-control micro-organisms. The accumulation of soluble EPS and the formation of small particles (in MBR-CPs after cross-flow velocity was raised) increased the resistance to filtration. The fouling potential of the supernatant seemed to be linked more closely to the concentration of polysaccharides than of proteins and humic substances. Modifications of the membrane performance were observed. Under operating conditions, membrane fouling was faster in MBR-CPs than MBR-control. Moreover, the membrane played an important role in the permeate quality and the overall performance of the process, making possible the biological treatment of such an effluent. Abstract: The possible coexistence of three mycotoxins in rice, including aflatoxin B1 (AFB1), citrinin (CIT) and ochratoxin A (OTA), was investigated. The samples of rice were collected in large markets in five provinces of the central region of Vietnam. These toxins were extracted, purified and finally quantified by HPLC with fluorimetry detection. Contamination of AFB1 was found to be the most, followed by OTA, while contamination of CIT was insignificant. The coexistence of CIT with AFB1/OTA in rice was found in high percentage. Some samples overpassed the authorized limit by Europe in OTA and/or AFB1. [Copyright &y& Elsevier]

Abstract: Crops may be contaminated by mycotoxins which can persist in the final products. Forty-five breakfast cereals were collected in French supermarkets. Ochratoxin A (OTA) and citrinin (CIT) were simultaneously extracted by a new method based on solvent partition validated in-house. The recoveries were over 80% for CIT and OTA. Fumonisin B1 (FB1) was analysed by an IUPAC method. The recoveries for FB1 ranged from 50% to 70%, depending on the matrix. The losses were located at the step of immunoaffinity clean-up. OTA was detected in 69% of the samples; 20% of them were above the EU limit of 3 μg/kg. Twenty percent contained CIT (1.5–42 μg/kg). FBs were detected, not only in cornflakes, but also in products containing oats or rice, in the range 1–1110 μg/kg. Some samples were contaminated by all three mycotoxins. [Copyright &y& Elsevier]

Ochratoxin A (OTA) exposure can result in chronic renal diseases and cancer. The incidence of kidney, renal pelvis, and ureter malignant neoplasms in the Czech Republic is approximately 29.5 renal tumours per 100,000 inhabitants. The question arises whether mycotoxins are also involved in kidney disease and cancer. A sensitive validated analytical methodology, based on an immunoaffinity clean-up followed by HPLC with fluorescence detection, was developed to explore whether OTA accumulates in clear renal cell carcinoma-adenocarcinoma in Czech patients. Simultaneously, DNA-adducts and OTA metabolites were qualitatively analysed in tissues and urine. OTA was analysed in 33 kidney and tumour samples from 26 men and 7 women collected during nephrectomy from patients of the East Bohemian region from 2015 to 2017. OTA was found in 76% of the analysed samples. Its concentrations ranged from not detectable to 390 ng/kg with a median of 167 ng/kg in kidney samples and from not detectable to 430 ng/kg with a median of 122 ng/kg in tumour samples. Urinary OTA metabolites and DNA adducts were qualitatively analysed for the corresponding 20 patients. The presence of some OTA metabolites such as ochratoxin A hydroquinone and/or decarboxylated ochratoxin A hydroquinone correlate with the presence of OTA-DNA adducts. • HPLC with fluorescence detection method was developed for ochratoxin A (OTA) determination in tumour kidney samples. • OTA determination directly in samples of clear cell renal carcinoma was used for the first time. • OTA metabolites and DNA adducts were qualitatively analysed in tissues and urine. • Presence of some OTA metabolites correlate with the OTA-DNA adducts. • OTB-GSH, OTHQ-GSH, and relevant NAC-conjugates can serve as useful OTA biomarkers.
