Odours emitted by biological wastewater treatment plants (WWTPs) may cause a nuisance and negatively impact people’s health. It is possible to tackle this issue using known deodorization technologies such as adsorption, absorption, combustion, catalytic oxidation, and biofiltration. However, applying some of these may lead to secondary pollution, high operating and investment costs, periodic replacing, utilising or regenerating reactor or filter bed, or using expensive catalysts. It is possible to avoid problems of this kind using the compact trickle-bed bioreactor (CTBB) technology to biodegrade odours emitted from WWTPs. A pilot-scale CTBB reactor, with a total volume of 1.07 m³, diameter of 0.8 m and height of 2.13 m, was installed on the premises of a municipal WWTP. At variable parameters of the biodegradation process, odour reduction was investigated using mobile measuring devices to detect hydrogen sulfide (H2 S) and volatile organic compounds (VOCs). The factor of H2 S conversion was 71%–97%, and that of VOC conversion was 82%–94% when the gaseous-phase flow rate ranged from 7–30 m³ h–1, at pH = 7 in the liquid-phase. The very-low calorie ketogenic diet (VLCKD) is recommended as an effective dietary approach for the management of obesity. This study investigated changes in circulating biomarkers of redox homeostasis induced by a multiphase VLCKD in obese individuals. A total of 40 obese subjects were prescribed a multiphasic VLCKD for eleven weeks. Anthropometric measurements, body composition parameters, calorimetric measures, and standard laboratory markers of glucose and lipid metabolism were evaluated at baseline (T 0) and at the end of the dietary intervention (T 1). Additionally, circulating markers of oxidative damage and antioxidant status were analyzed in serum and erythrocytes. Compared to T 0 , at T 1 the multiphase VLCKD induced significant weight loss and reduction of waist circumference, with beneficial effects on body composition parameters and the glucose/lipid biochemical profile. Moreover, a decrease in serum markers of oxidative damage was reported at T 1 , while no changes in serum markers of antioxidant status and in erythrocyte redox markers were observed. In addition, a significant association was found between variations in anthropometric measurements, body composition, glucose metabolism parameters, and changes in circulating markers of oxidative damage. Regression models showed that variation in lipofuscin was significant predictor of changes in body mass index, fat mass, visceral adiposity, and insulin sensitivity. In conclusion, this study demonstrated that the multiphase VLCKD improves serum redox balance by reducing markers of oxidative damage in obese individuals, highlighting the interplay between adiposity, glucose metabolism, and redox homeostasis in the pathogenesis of obesity. PTMC-PEG-PTMC triblock copolymers were prepared by ring-opening polymerization of trimethylene carbonate (TMC) in the presence of dihydroxylated poly(ethylene glycol) (PEG) with Mn of 6000 and 10,000 as macro-initiator. The copolymers with different PTMC block Lengths and the two PEGs were end functionalized with acryloyl chloride. The resulting diacrylated PEG-PTMC-DA and PEG-DA were characterized by using NMR, GPC and DSC. The degree of substitution of end groups varied from 50.0 to 65.1%. Hydrogels were prepared by photo-crosslinking PEG-PTMC-DA and PEG-DA in aqueous solution using a water soluble photo-initiator under visible light irradiation. The effects of PTMC and PEG block lengths and degree of substitution on the swelling and weight loss of hydrogels were determined. Higher degree of substitution leads to higher crosslinking density, and thus to lower degree of swelling and weight loss. Similarly, higher PTMC block length also leads to lower degree of swelling and weight loss. Freeze dried hydrogels exhibit a highly porous structure with pore sizes from 20 to 100 µm. The biocompatibility of hydrogels was evaluated by MTT assay, hemolysis test, and dynamic clotting time measurements. Results show that the various hydrogels present outstanding cyto- and hemo-compatibility. Doxorubicin was taken as a model drug to evaluate the potential of PEG-PTMC-DA and PEG-DA hydrogels as drug carrier. An initial burst release was observed in all cases, followed by slower release up to more than 90%. The release rate is strongly dependent on the degree of swelling. The higher the degree of swelling, the faster the release rate. Finally, the effect of drug loaded hydrogels on SKBR-3 tumor cells was evaluated in comparison with free drug. Similar cyto-toxicity was obtained for drug loaded hydrogels and free drug at comparable drug concentrations. Therefore, injectable PEG-PTMC-DA hydrogels with outstanding biocompatibility and drug release properties could be most promising as bioresorbable carrier of hydrophilic drugs. Graphical abstract Abstract Filomicelles (worm-like micelles) possess high drug loading capacity and long circulation time in the bloodstream. A novel approach can be filomicelles with folic acid (FA) as a targeting moiety. Folate-drug delivery systems can target FA receptors (FAR) that are overexpressed in several human carcinomas, which can potentially maximize therapeutic efficacy while minimizing side effects. The aim of this study was to develop filomicelles from combination of poly(L-lactide)-Jeffamine-folic acid and poly(L-lactide)-poly(ethylene glycol) for delivery of betulin derivative. Phosphate derivative of betulin reveals high cytotoxicity against cancer cells, however its application is restricted due to poor solubility in water. Incorporation into hydrophobic core of micelles can effectively solubilize the drug. Three kinds of micelles were obtained with high drug loading capacity. Based on TEM analysis, the copolymers formed exclusively filomicelles or mixture of filomicelles and spherical micelles. All kinds of micelles provided release of betulin derivative for over 9 days and apart the very initial phase displayed similar release profile. The influence of PLA block on initial burst effect was revealed. The in vitro cytotoxicity of betulin derivative loaded micelles against FAR-positive HeLa cells was confirmed, which proves their usefulness for targeted delivery of cytostatic drug. • A machine-learned surrogate model is developed to predict ground surface tilts induced by pressurized fractures. • The model has been trained for 7 dip angles: 0, 15, 30, 45, 60, 75 and 90°, but can predict for any given dip angles. • The model trained using 11,000 samples of single fractures, however, it satisfactorily predicts the tilts from multiple interacting fractures. • The model trained for a given set of model parameters and a reference depth, but is capable of predicting the tilts for different parameters and depths. Fast and reliable estimation of engineered fracture geometries is a key factor in controlling undesirable fractures and enhancing stimulation design. Measuring the surface deformation gradient (tilt) for engineered fractures in shallow depths (<1000 m) has been proven a reliable source of data to infer fracture geometry, thanks to the impressive resolution of tiltmeter units (in the order of nano-radians). However, solving the inverse problem requires reliable and fast forward models. In this study, we present a fast and reliable machine-learned surrogate model to estimate the ground surface tilt induced by pressurised fractures. The proposed surrogate model, based on Conditional Generative Adversarial Networks (cGAN), receives a fracture aperture map in XY and XZ planes as input and predicts the corresponding surface tilts (in X and Y directions). The surrogate model with Wasserstein loss and gradient penalty has been trained using 11,000 samples and tested for a range of input parameters such as depth, dip angles, elastic properties, fluid pressures and fracture shapes. The testing results show excellent performance of the surrogate model compared with the forward finite element model for both single and multiple pressurised fractures, while running hundreds to potentially thousands of times faster. The aim of the study was to investigate whether α-tocopherol supplementation for workers who are chronically exposed to lead would normalize/improve the values of parameters that are associated with the lead-induced oxidative stress. Study population included chronically lead exposed males who were divided into two groups. Workers in the first group (reference group) were not given any antioxidants, while workers in the second group (αT group) received supplementation with α-tocopherol. After treatment, the blood lead and leukocyte malondialdehyde levels decreased significantly in the αT group compared to the baseline levels and reference group. However, the erythrocyte malondialdehyde, conjugated dienes, and lipofuscin levels significantly increased compared to the baseline levels. The glutathione level significantly increased compared with the baseline. Effects of supplementation with α-tocopherol on oxidative damage were not satisfactory. Therefore, there is no reason to administer α-tocopherol to workers chronically exposed to lead as a prophylaxis of lead poisoning. [ABSTRACT FROM AUTHOR]

The main aim of this study was to optimize and maximize the impacts of odor and volatile organic compounds (VOCs) biodegradation in a wastewater treatment plant utilizing a pilot-scale compact trickle bed bioreactor (CTBB). A CTBB was built and tested for its long-term performance during which gases were supplied from the tank containing semi-liquid fats, oils, and fat waste. The concentrations of pollutants ranged from 0 to 140.75 mg/m3 H 2 S, 0 to 2500 mg/m3 VOCs, and 0 to 21.5 mg/m3 NH 3. The CTBB was tested at different gas flow rates and at two pH values for the liquid phase: pH = 7.0 and 5.0. In the liquid phase, the pollutant removal efficiency was higher at pH = 7.0 than at pH = 5.0. Overall, the removal efficiency was between 81.5 % and 99.5 % for the VOCs and 87.5 % and 98.9 % for H 2 S, while NH 3 removals were >99 %. [ABSTRACT FROM AUTHOR]

Abstract: In this study, we sought to understand the influence of occupational lead-exposure on the gene expression (Sod1) and activity (SOD) of superoxide dismutase, catalase and glutathione peroxidase (GPx, Gpx1) in leukocytes and erythrocytes. The study group consisted of 45 healthy male employees of a lead-zinc works and was divided into two subgroups: those with low exposure to lead (LE) and those with high exposure to lead (HE). In addition, 17 healthy male administrative workers participated in the study as the control group. The gene expression levels of both Sod1 and Gpx1 were significantly increased in the LE group as compared to the control group. By contrast, we noted only an insignificant tendency for increased gene expression of both Sod1 and Gpx1 in the HE group. The expression and activity of catalase were unchanged. Nevertheless, SOD and GPx activities in erythrocytes was significantly elevated in both examined subgroups, whereas SOD activity in leukocytes was raised only in the LE group. The results of this study led us to conclude that lead has a significant influence not only on the activities of antioxidant enzymes but also on the dose-dependent expression in their genes. The emission of lead (Pb) occurring during the extraction, processing and industrial applications of this element remains a significant environmental risk factor. The absorbability of lead in humans is strongly associated with the general health status of exposed individuals. Existing mineral deficiencies are considered being a predisposition to an increased Pb uptake. Both, iron deficiency and lead poisoning are the major causative factors responsible for the prevalence of anemia within the vulnerable population, especially in children. Although some of the intervention programs of counteracting lead poisoning by iron supplementation proved to be effective in the Pb-exposed population, the exact mechanisms of this interaction still require further studies. The objective of the presented study was to examine the association of iron level on oxidative stress measures and its effects on the severity of lead toxicity in the exposed population. The analyzed population consisted of 270 male workers from the lead-zinc smelter. The studied population was divided into two sub-groups based on the serum iron concentration: low iron level group (L-Fe; Fe < median value) and high iron level group (H-Fe; Fe > median value). Measured traits comprised of blood lead (PbB), serum Fe and zinc protoporphyrin (ZPP) levels as well as a blood count and oxidative stress markers. No significant correlation between serum iron concentration and PbB in the tested cohort was found. On the contrary, the analysis of ZPP levels (long-term marker related to a hematologic toxic effect of Pb) within the subgroups differing in serum Fe level shown that ZPP was 12.3 % lower (p = 0.043) in subjects classified within the H-Fe group. A positive correlation of serum Fe and total antioxidant capacity (TAC) was found (R = 0.1999). The conducted 3-D PCA analysis showed that individuals classified within the H-Fe group were characterized by the co-occurrence of higher Fe levels, lower ZPP, and higher TAC value. These results support the existing evidence providing that maintaining the optimal status of Fe may play a significant role in preventing the lead poisoning and alleviating harmful effects of Pb on the oxidative balance in humans. Objective: Osteoarthritis of the knee (KOA) is a common, age-related, joint disorder associated with loss of articular cartilage, osteophyte formation, sub-chodral bone change and synovitis. Recent studies have shown that reactive oxygen species (ROS) may participate in the initiation and progression of KOA. This study examines potential changes in the activities of antioxidant enzymes (superoxide dismutase, both isoenzymes zinc-copper superoxide dismutase and manganese superoxide dismutase) and glutathione transformation enzymes (glutathione peroxidase, glutathione reductase and glutathione-S-transferase) in synovial fluid of KOA patients, and estimates their relationship to the degree of lipid peroxidation in synovial fluid evaluated by malondialdehyde concentration, synovial fluid viscosity, type and duration of KOA.Design: Synovial fluid samples obtained by transdermal arthrocentesis from 41 patients with KOA (23 had primary KOA and 18 had secondary KOA) and 22 control subjects were analyzed. Activities of antioxidant enzymes were analysed with the use of kinetic method, MDA concentration was measured fluorometrically by the Ohkawa method, and synovial fluid viscosity was measured using a cone-late viscometer Brookfield DV-II+ and a test by Ropes.Results: Patients with KOA had significantly increased activities of all enzymes when compared to the control subjects for both KOA subgroups. The synovial fluid viscosity was significantly decreased and the synovial fluid test by Ropes was abnormal in KOA patients, mainly in the secondary KOA subgroup. The activities of all antioxidant enzymes were significantly negatively correlated with synovial fluid viscosity and duration of KOA.Conclusions: Patients with KOA display abnormal antioxidant status of synovial fluid with increased activities of antioxidant enzymes and decreased synovial fluid viscosity. The aim of this study was to examine the association between combined concentrations of macro and trace elements and markers of oxidative stress and antioxidative defense system function together with selected cytokine levels. Based on the combined medians of the seminal plasma levels of calcium, magnesium, zinc, copper, iron, and selenium, the study subjects (88 fertile male volunteers) were divided into the following two subgroups: the Me-L group (low level of metals) and the Me-H group (high level of metals). There was a tendency toward reduced motility in the Me-H group compared to that in the Me-L group. The total protein, albumin, and total oxidation status (TOS) levels were significantly higher in the Me-H group than in the Me-L group. The total superoxide dismutase (SOD), Mn-SOD, and CuZn-SOD, activity in spermatozoa were significantly lower in the Me-H group than in the Me-L group. In seminal plasma, the Mn-SOD activity was significantly higher in the Me-H group, whereas the CuZn-SOD activity was significantly lower. Additionally, the activity levels of glutathione peroxidase (GPx) and glutathione-S-transferase (GST) were lower in the Me-H group. The medians of IL-1β, IL-10, and IL-12 were significantly higher in the Me-H group than in the Me-L group, whereas the medians of IL-2, IL-5, and IL-13 were significantly lower. Higher levels of macro and trace elements in the seminal plasma of fertile males may be associated with decreased motility. Higher levels of the examined metals are associated with elevated oxidative stress accompanied by decreased activities of some of the antioxidant enzymes and increased pro-inflammatory cytokine levels. Abstract: The aim of this experiment was to establish an efficient method for isolation and further culture in vitro of the normal chicken oviduct epithelial cells (COEC) for cell-based research models. Different factors were tested to optimize COEC primary culture for repeatable results: the origin of isolated cells (oviduct Infundibulum or Magnum section); the oviduct tissue dissociation procedure (mechanical scrapping or mincing), tissue digestion times (15, 30 and 45 min), the culture plates coating (colagene I, polystyrene surface or 3T3 feeder layer), the growth media (classic DMEM/Ham''s F12 and defined serum-free medium, Lonza Switzerland), incubation temperature (37 °C vs 41°C) and different cell seeding numbers: 0.2M, 0.5M and 1.0M cells/well. The COEC isolated by mincing the Infundibular neck and digestion of tissue for 30 min formed cell aggregates of bright colour and gave proliferating colonies of epithelial-like character which was the best result obtained from all applied procedures in our studies. The fibroblast-like cells considered as contaminants occurred only sporadically up to day 7 of culture. Seeding about 1M cells in 1 mL of serum-free medium onto 12-well dishes gave the optimal growth of colonies resulting in 5 to 7 confluent culture wells from a single oviduct sample. Feeder layer and collagen I did not improve adhesion of the COEC to the culture vessel. Adoption of 37 °C and 41 °C did not reveal apparent differences to the condition of cultured COEC. Cell differentiation and proliferation potential depends on number and replicative capacity of isolated progenitors. The progenitors are responsible for holoclones formation and good culture growth. The percentage of colonies developed from the cells isolated from Infundibulum was greater than that of other samples in our studies. We conclude that the model of COEC primary cultures from different segments of oviduct, in particular infundibulum, should be incorporated to the range of avian cells research as this work generates questions about undocumented sources of oviduct progenitor cells. Abstract: The Agri-Environment Footprint Index (AFI) has been developed as a generic methodology to assess changes in the overall environmental impacts from agriculture at the farm level and to assist in the evaluation of European agri-environmental schemes (AES). The methodology is based on multi-criteria analysis (MCA) and involves stakeholder participation to provide a locally customised evaluation based on weighted environmental indicators. The methodology was subjected to a feasibility assessment in a series of case studies across the EU. The AFI approach was able to measure significant differences in environmental status between farms that participated in an AES and non-participants. Wider environmental concerns, beyond the scheme objectives, were also considered in some case studies and the benefits for identification of unintentional (and often beneficial) impacts of AESs are presented. The participatory approach to AES evaluation proved efficient in different environments and administrative contexts. The approach proved to be appropriate for environmental evaluation of complex agri-environment systems and can complement any evaluation conducted under the Common Monitoring and Evaluation Framework. The applicability of the AFI in routine monitoring of AES impacts and in providing feedback to improve policy design is discussed. [Copyright &y& Elsevier]

Abstract: The effect of poly(l-lactide-co-TMC) chain microstructure (and its changes during degradation) on immunosuppressive drugs’ release process was analyzed. Three kinds of poly(l-lactide-co-TMC) (PLATMC) – two semiblock and one random were used to prepare matrices containing cyclosporine A or rapamycine and drug free matrices. All of them degraded slowly enough to provide long term delivery of immunosuppressive agents. Moreover, copolymer chain microstructure determined the effect of drug loading on the degradation process. It was observed that matrices without drug obtained from semiblock copolymer degraded differently than matrices containing cyclosporine A or rapamycine, whereas all kinds of matrices obtained from random PLATMC degraded in similar way. This is the evidence that the only in case of semiblock copolymer factors concerning the presence of drug and the kind of drug influenced degradation process. Based on the obtained results, correlations between copolymer degradation and drug release process are proposed. According to our outcomes, regular drug release process may be obtained from highly randomized copolymers (R ≈1) that remain amorphous during degradation process. Determination of this factor may help in development of biodegradable systems, in which drug release rate and profile can be tailored by synthesis of polymer with appropriate chain microstructure. Abstract: Terpolymers of lactide, glycolide and trimethylene carbonate initiated on zirconium acetylacetonate (IV) exhibit shape-memory behaviors in the range of body temperature when appropriate monomer composition and characterizing conditions are applied. In this paper, the procedure of characterizing these terpolymers by means of nuclear magnetic resonance spectroscopy is presented. The 1H and 13C NMR spectra allow to characterize the chain microstructure of these polyester-carbonates in spite of overlapping resonance signals due to the possibility of forming different co-monomeric sequences. Detailed assignment was given for spectral lines in proton and carbon spectra recorded in polar and non-polar deuterated solvents. For quantitative analysis, equations for calculations of average block lengths, long and mixed glycolidyl, lactidyl and carbonate blocks as well as formulas allowing to investigate intramolecular transestrification processes were elaborated. Abstract: The influence of the chain microstructure on release process of doxorubicin from polymeric matrices was analyzed. Aliphatic polyester copolymers with optimal chain microstructure, i.e. poly(glycolide-co-l-lactide, 15/85) (PGLA) and poly(glycolide-co-ɛ-caprolactone, 10/90) (PGCA) were synthesized for long-term doxorubicin delivery systems. Various release profiles from PGLA and PGCA matrices were obtained. The investigations revealed the most steadily doxorubicin release from PGCA matrices with 5% (w/w) of drug content. Degradation of matrices with and without drug was monitored by means of NMR spectroscopy and confirmed stability of degradation process. From PGCA matrices the increase of released doxorubicin amount was observed during first 60 days. On the contrary in case of matrices obtained from PGLA the delay of doxorubicin release was observed during first 50 days, what was caused by interaction of drug molecules with polylactide chain of polymer matrix. The interaction between doxorubicin molecules and polylactide chains was confirmed by IR spectroscopy. This fact can be used for designing of delivery systems consisting of combination of matrices with different microstructure of copolymer chains in order to adjust concentration of released doxorubicin and stabilization of drug release process. Abstract: An aggregated farm-level index, the Agri-environmental Footprint Index (AFI), based on multiple criteria methods and representing a harmonised approach to evaluation of EU agri-environmental schemes is described. The Index uses a common framework for the design and evaluation of policy that can be customised to locally relevant agri-environmental issues and circumstances. Evaluation can be strictly policy-focused, or broader and more holistic in that context-relevant assessment criteria that are not necessarily considered in the evaluated policy can nevertheless be incorporated. The Index structure is flexible, and can respond to diverse local needs. The process of Index construction is interactive, engaging farmers and other relevant stakeholders in a transparent decision-making process that can ensure acceptance of the outcome, help to forge an improved understanding of local agri-environmental priorities and potentially increase awareness of the critical role of farmers in environmental management. The structure of the AFI facilitates post-evaluation analysis of relative performance in different dimensions of the agri-environment, permitting identification of current strengths and weaknesses, and enabling future improvement in policy design. Quantification of the environmental impact of agriculture beyond the stated aims of policy using an ‘unweighted’ form of the AFI has potential as the basis of an ongoing system of environmental audit within a specified agricultural context. Abstract: Electrospray ionization-mass spectrometry (ESI-MS) and proton nuclear magnetic resonance (1H NMR) have been used to investigate the hydrolytic degradation of copolymers obtained by bulk ring-opening copolymerization of glycolide and ɛ-caprolactone with monomer ratios ranging from 70/30 to 30/70. NMR allows changes of the average sequence distribution and composition of the components to be followed. In contrast, ESI-MS is able to reveal the detailed chemical structures of various sequences despite the molecular weight limit of 2000Da. Combination of ESI-MS with NMR can thus provide information to describe microstructure changes during degradation. The distribution of various oligomers shown in the form of planar projections is of great interest for the design of biodegradable system aimed at medical applications. [Copyright &y& Elsevier]

The automotive painting industry is a source of environmental pollution caused by Volatile Organic Compounds (VOCs) present in the discharged ventilation air. To contribute to the mitigation of this type of air pollution, Ekoinwentyka Ltd. developed – from pilot scale to full scale – and adapted the technology of Compact Trickle Bed Bioreactor (CTBB) whose operating principle builds upon co-current downflow of the gas phase (polluted air) and liquid phase (solution of mineral salts) through a packed bed where active microorganisms are immobilized in the biofilm on the surfaces of packing elements. Pilot-scale bioreactor 0.32 m in diameter and 1.50 m total height had its packed bed inoculated with a consortium of microorganisms dominated by Pseudomonas fluorescens bacteria. During the experimental programme that lasted several months, the flow rate of air drawn from the ventilation system of the painting shop was changing between 1.0 and 10.0 m3/h and the inlet concentration of VOCs ranged from 10 to 200 ppm. By measuring VOC concentration in the purified air, the factor of VOC biodegradation was found to range between 85 and 99%. Based on pilot-scale experiments, full-scale CTBB has been developed 2.8 m in diameter and 10 m total height and installed as an add-on component of the ventilation system of the painting shop. Test operation at gas flow rates up to 6000 m3/h, confirmed VOC biodegradation factor at the level of 85–99% thus proving a positive result of CTBB technology adaptation to the conditions of the automotive painting industry. The present study investigated associations between environmental exposure to cobalt (Co) and the levels of oxidative stress parameters and the antioxidant defense system in the seminal plasma of fertile males. The study population consisted of 117 healthy, non-smoking, fertile men from the southern region of Poland. The study was carried out in 2021–22. Based on the median cobalt levels in seminal plasma, subjects were divided into two groups: those with low (Co-L) and high (Co-H) cobalt concentrations. Semen parameters assessed according to WHO 2021 recommendations. After the analysis of spermiograms, observed reduction in progressive motility after 1 h was found in the Co-H group. Moreover, superoxide dismutase (SOD), glutathione S-transferase (GST) in the Co-H group had lower activity and GR higher activity. The OSI (Oxidative stress index) were higher in the group with high cobalt concentration in semen. The concentrations of redox balance parameters: TOS, TAC and OSI significantly were higher in the Co-high group as well as GR activity. Environmental exposure to cobalt decreases sperm motility in both normal and abnormal semen.
