This technical report is based on the study for the evaluation of the processes of reuse and recycling of reverse osmosis components and membranes in the Canary Islands and Macaronesia, within the DESAL+ project and in the framework of the DESAL+ LIVING LAB platform, coordinated by the Canary Islands Technological Institute (ITC) and the Canary Islands Agency for Research, Innovation and Information Society (ACIISI), with the support of the Interreg-MAC Programme. Reverse osmosis membranes could be reused in the same or another desalination plant by replacing the membranes in the first, dirtier positions with those in the last, less damaged positions. Also, by changing the best first-stage membranes to the second and vice versa, the useful life of these membranes could be extended through chemical cleaning and a second life could be given in tertiary treatment plants, reuse in industrial processes where they use special reverse osmosis membranes and degrade rapidly, in processes with leachate from landfill waste and also an interesting option is the oxidation of reverse osmosis elements to obtain nanofiltration, ultrafiltration or microfiltration membranes for the removal of physical dirt. The main categories of thermal processing recycling commonly used in industry include incineration and pyrolysis to produce energy, gas and fuel. These processes can be applied to mixed plastic waste, such as the combination of materials used in the manufacture of reverse osmosis membranes. The recycling of reverse osmosis elements from desalination plants is shown as an opportunity, nowadays existing pioneering initiatives in Europe. Energy recovery, via incineration, is feasible but is not considered in accordance with the environmental, social and political problems that this may generate. However, the recycling of the reverse osmosis elements via pyrolytic industry for fuel production can be centralized in a new industry already planned in the Canary Islands and all the osmosis membranes that are obsolete can be sent there. Abstract Marine pollution, overrepresented by plastic, is a growing concern worldwide. However, there is little knowledge on occurrence and detrimental impacts of marine debris in cetaceans. To partially fill in this gap of knowledge, we aimed to investigate the occurrence and pathologies associated with foreign bodies (FBs) in a large cohort of cetaceans (n = 465) stranded in the Canary Islands. The Canary Islands shelter the greatest cetacean biodiversity in Europe, with up to 30 different species, of which nine are regularly present year around. We found at least one ingested FB in 36 out of 465 (7.74%) studied cetaceans, involving 15 different species, including eight out of the nine (80%) cetacean species present year-round in the Canary Islands. Risso's dolphin was the species most affected, followed by sperm whale, beaked whale and mysticetes. Plastic FB were the most common item found (80.56%). FB was directly associated with death in 13/36 (36.11%) animals. Poor body condition and deep diving behavior were found to be risk factors for FB ingestion, whereas the adult age was a protective factor. To the authors knowledge this is the first study that use statistical analysis to investigate risk and protective factors for FB ingestion. This study also provides insights of the potential impact caused by ingested FBs on the animal's health and mortality. This knowledge is critical to better understand and assess the impact of FB in cetaceans setting the scientific basis for prospective impact monitoring and future conservation policies. Summary Spanish goat encephalitis virus (SGEV) is a recently described member of the genus Flavivirus belonging to the tick-borne encephalitis group of viruses, and is closely related to louping ill virus (LIV). Naturally acquired disease in goats results in severe, acute encephalitis and 100% mortality. Eighteen goats were challenged subcutaneously with SGEV; nine were vaccinated previously against LIV and nine were not. None of the vaccinated goats showed any clinical signs of disease or histological lesions, but all of the non-vaccinated goats developed pyrexia and 5/9 developed neurological clinical signs, primarily tremors in the neck and ataxia. All non-vaccinated animals developed histological lesions restricted to the central nervous system and consistent with a lymphocytic meningomyeloencephalitis. Vaccinated goats had significantly ( P <0.003) greater concentrations of serum IgG and lower levels of IgM ( P <0.0001) compared with unvaccinated animals. SGEV RNA levels were below detectable limits in the vaccinated goats throughout the experiment, but increased rapidly and were significantly ( P <0.0001) greater 2–10 days post challenge in the non-vaccinated group. In conclusion, vaccination of goats against LIV confers highly effective protection against SGEV; this is probably mediated by IgG and prevents an increase in viral RNA load in serum such that vaccinated animals would not be an effective reservoir of the virus. Enhancing the quality of beef meat is an important goal in terms of improving both the nutritional value for the consumer and the commercial value for producers. The aim of this work was to study the effects of different vegetable oil supplements on growth performance, carcass quality and meat quality in beef steers reared under intensive conditions. A total of 240 Blonde D’ Aquitaine steers (average BW=293.7±38.88 kg) were grouped into 24 batches (10 steers/batch) and were randomly assigned to one of the three dietary treatments (eight batches per treatment), each supplemented with either 4% hydrogenated palm oil (PALM) or fatty acids (FAs) from olive oil (OLI) or soybean oil (SOY). No differences in growth performance or carcass quality were observed. For the meat quality analysis, a steer was randomly selected from each batch and the 6th rib on the left half of the carcass was dissected. PALM meat had the highest percentage of 16:0 (P<0.05) and the lowest n-6/n-3 polyunsaturated fatty acids (PUFA) ratio (P<0.05), OLI had the highest content of t11-18:1 (P<0.01) and c9,t11-18:2 (P<0.05) and SOY showed the lowest value of monounsaturated fatty acids (MUFA) (P<0.001), the highest percentage of PUFA (P<0.01) and a lower index of atherogenicity (P=0.07) than PALM. No significant differences in the sensory characteristics of the meat were noted. However, the results of the principal component analysis of meat characteristics enabled meat from those steers that consumed fatty acids from olive oil to be differentiated from that of steers that consumed soybean oil. Assisted reproductive technologies are of great importance for increasing the genetic diversity in captive animals. The use of bovine or murine oocytes in heterologous IVF provides advantages compared to homologous IVF in nondomestic animals, such as the accessibility to oocytes and the availability of well-developed in vitro maturation systems. The aim of this study was to determine the heterologous IVF parameters using cryopreserved dolphin spermatozoa and zona-intact bovine or murine oocytes and to examine the nuclear chromatin status of the dolphin spermatozoa. All the processes involved in the fertilization including embryo cleavage were observed by confocal microscopy and hybrid embryo formation was confirmed by polymerase chain reaction. Heterologous bovine IVF showed no polyspermy, lower percentages of pronuclear formation, and a lower cleavage rate compared to homologous IVF group (34.8% vs. 89.3%). Heterologous murine IVF showed a lower cleavage rate than homologous IVF (9.6% vs. 77.1%). With respect to dolphin sperm chromatin, it was more stable, i.e. more resistant to EDTA-SDS decondensation than the bovine sperm chromatin. This study revealed the stability of the dolphin sperm chromatin and the ability of the dolphin spermatozoa to penetrate zona-intact bovine and murine oocytes, leading to hybrid embryo formation. Abstract: Objective: The aims of this study were to explore the influence of personality disorders (PDs) in Spanish adolescents with Axis I psychiatric disorders on their use of mental health services and to analyze the risk of having a comorbid PD in relation to psychiatric service use. Methods: The Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition (DSM-IV) and International Classification of Diseases, Tenth Revision (ICD-10) modules of the semistructured interview International Personality Disorders Examination were administered to a sample of 112 adolescent psychiatric patients (mean age = 15.8 years; SD, 0.8; range, 15-17; 79% women) at the point of initiating treatment. On the basis of the interview, subjects were divided into two groups: a PD group (PDG) and a non-PD group (NPDG). After 3 years of treatment, clinical records were retrospectively analyzed. Results: The PDG showed a significantly higher number of psychiatric admissions (P < .001), days per psychiatric admission (P < .001), and psychiatric emergencies (P < .010) than the NPDG, although the number of outpatient consultations was not significantly higher. Logistic regression analysis showed that the probability of belonging to the PDG rather than the NPDG increased with each psychiatric admission (odds ratio [OR] = 1.67 for DSM-IV criteria and OR = 1.59 for ICD-10 criteria), after controlling by sex, age, and comorbidity (Axis I disorders). Conclusions: Patients with comorbid PD used more inpatient and emergency psychiatric services than did patients without a PD. To evaluate the effect of stocking density and transport time on physiological responses and meat quality, 72 male suckling lambs were transported by road to the slaughterhouse at three different stocking densities (0.12, 0.20 or 0.25m2/lamb) and two transport times (5 h or 30 min). Blood samples were collected pre-transport at the farm and after unloading in the slaughterhouse to measure levels of cortisol, creatine kinase (CK) and lactate dehydrogenase (LDH). After slaughter, the weights of the hot carcass, liver and spleen were recorded and pH in Musculus longisimus thoracis et lumborum (L), Musculus semitendinosus (ST) and Musculus psoas major (PM) were determined. Colour, water-holding capacity (WHC), texture and thiobarbituric acid-reactive substances (TBARS) values were measured in samples of L, at 24 h post mortem and after 5 days of ageing. Cortisol and LDH were higher in suckling lambs transported for 30 min than those transported for 5 h. Stocking density did not affect blood parameters studied. Transport time significantly affected some carcass quality parameters, but stocking density had no significant effect. Suckling lambs transported for 5 h had lower liver weights and dressing percentages than those transported for 30 min. Transport time influenced pH values, with lambs subjected to the longer journey showing the lowest pH at 0 h in the three muscles studied, with the lowest final pH in L and PM. The PM lambs transported at high density (0.12m2/lamb) had the lowest pH at 24 h. Transport time and stocking density did not greatly affect colour and texture parameters. The meat from lambs transported for 30 min had higher WHC than meat from lambs transported for 5 h. Animals transported for longer journeys showed higher lipid oxidation after 5 days of ageing than those transported for 30 min. Abstract: The aim of this study was to evaluate the fatty acid composition of intramuscular fat and the vitamin E content of beef from different production systems. Four cattle production systems were used: cattle reared under intensive conditions and fed concentrate (INT1) from Spain, cattle raised at pasture and supplemented with concentrate (SUP1) from the United Kingdom, cattle raised at pasture and on corn silage (SUP2) from Germany and cattle reared under extensive conditions slaughtered at two and three years old (EXT2 and EXT3) from Uruguay. The highest proportion and content (mg per 100g of muscle) of linoleic acid (C18:2n-6) and arachidonic acid (C20:4n-6) and the lowest saturated fatty acids (SFA) was found in INT1 beef. In contrast, beef reared under extensive conditions showed the highest proportion and content of linolenic acid (C18:3n-3), docosapentaenoic acid (DPA, C22:5n-3) and eicosapentaenoic acid (EPA, C20:5n-3), and SUP1 and SUP2 beef showed the highest level of palmitic acid (C16:0) and SFA. Beef from intensive system (INT1) had the lowest P/S (PUFA/SFA) ratio, whereas beef from extensive system (EXT2 and EXT3) had the lowest n-6/n-3 ratio. The results of the PCA (principal components analysis) of fatty acid composition showed that beef from intensive system (INT1) was clearly differentiated from the other meats and was located with the C18:2n-6 and C20:4n-6 and the n-6/n-3 ratio. EXT2 and EXT3 beef were located with C18:3n-3 and long chain fatty acids. Beef from extensive systems had the highest concentration of vitamin E (3.91mg α-tocopherol/kg muscle). Abstract: Body composition was evaluated in 49 males and females suckling lambs of the Manchego breed slaughtered at 10, 12 and 14kg. Lambs slaughtered at 14kg had a greater proportion of loin-rib than those slaughtered at 10kg. Male lambs exhibited a larger forequarter (neck, anterior rib and shoulder) while females had larger loin-rib. Slaughter weight affected the distribution of fat, and proportion of fat in loin-rib increased with slaughter weight. Males at 10kg had more muscle in shoulder and neck than females. Significant slaughter weight×gender interaction was observed for total fat proportion in leg, loin-rib, anterior rib and flank. These proportions increased with slaughter weight in male lambs while in female lambs remained constant. The carcass composition of the suckling lamb showed significant correlations with muscle, fat and bone of the individual cuts, mainly leg and loin-rib. As expected, the proportion of muscle in the carcass was highly correlated with muscle in loin-rib, leg and shoulder. Ti–Al intermetallic coatings on titanium have been produced by laser surface alloying. A 5-kW CO2 laser was employed to melt the surface of the titanium specimens and to alloy it with aluminium powder in a nitrogen atmosphere. The combination of titanium, aluminium and nitrogen and the specific characteristics of the high-power laser allow a new coating structure of titanium aluminides and nitrides to be produced. These coatings present a surface Al–TiAl3 composition that gradually evolves to Ti3Al close to the titanium base. A dendrite structure of titanium nitrides (TiN and Ti2N) and titanium aluminium nitrides (Ti2AlN) is formed throughout the laser track. In this paper we present the results corresponding to on-road traffic emissions measurements obtained during two field campaigns developed in the Madrid region (Spain) during 2014 and 2015 in the framework of the CORETRA project. The experimental strategy was based on the use of a RSD 4600 remote sensor in interurban roads. These measurements have produced a global database of > 190,000 vehicles with their associated emission data (NO/CO 2 , HC/CO 2 and CO/CO 2 ), which can be considered representative of the current Spanish circulating fleet. The results of M1 vehicles were analysed according to their distribution by Euro Standard and engine model. One of the relevant findings is that, despite the progressive introduction of increasingly stringent standards, no NO emission reduction is observed for diesel vehicles with time, although this behavior shows significative differences among brands and engine models. We have also investigated the presence of “high emitter” (HE) vehicles in the Spanish M1 circulating fleet and most of the HE detected corresponded to diesel vehicles with very high NO/CO 2 values. With these results at hand, we strongly propose the future incorporation of the “high emitter vehicle” definition into the European environmental legislation, as well as the establishment of specific strategies in each country/region in order to identify these anomalous vehicles. Identification and repair of HE vehicles within the European circulating fleets, although are not easy tasks, should be considered very important for the improvement of air quality in the EU. The use of non-intrusive optical technologies (i.e. RSD) is an excellent option to provide instantaneous real emission data of each individual vehicle without disturbing traffic and for on-road fleet monitoring. In summary, it is a good strategy to obtain valuable information about the long term surveillance of real vehicle emission trends, specially after the introduction of new standard. Performance of metal–organic framework MIL-101 (Cr) to remove, through adsorption, organic nitrogen (N-) compounds from straight run gas oil (SRGO) and straight run gas oil/light cycle oil (SRGO/LCO) mixtures was compared against that for four commercial adsorbents: silica gel, Selexsorb® CD, Selexsorb® CDX and activated carbon in batch and fixed-bed experimental set-ups. For fixed-bed tests, MIL-101 (Cr) was pelletized using carboxymethylcellulose as agglomerant. Room temperature batch data at equilibrium, showed that MIL-101 (Cr) has, by far, the highest adsorption capacity, following both Langmuir and Freundlich models for the adsorption of N-compounds. Kinetic data show that MIL-101 (Cr) has also the fastest adsorption rate treating SRGO under room conditions. Lumped kinetic analysis showed that adsorption is described by a pseudo second-order rate equation. Correspondingly, flow fixed-bed data showed that MIL-101 (Cr) achieved a higher adsorption loading of N-compounds than commercial adsorbents, being the flow rate and Nitrogen content in feed the main variables affecting the cycle length before adsorbent regeneration was needed. Additionally, MIL-101 (Cr) showed a good stability, maintaining its initial adsorption capacity after 300 regeneration procedures (from 90 to 85%). It is expected an useful life in industrial application of at least 1000 cycles. According to the aforementioned, MIL-101 (Cr) confirmed to be a promising material for the adsorption of N-compounds from middle distillates as a pretreatment step toward ultra low sulfur diesel production. Abstract: The effect of the zwitterionic buffers HEPES, TES and MOPS and of PBS used for out-of-incubator procedures during standard in vitro embryo production on bovine oocytes and embryo development, morphology and on the expression patterns of eight selected genes: Fgf-4, Lama1, Ube2a, Gsta4, Il6, Sod1, Prss11 and Hspb1, was evaluated. All buffers were prepared at a concentration of 10mM in TALP medium, with the exception of PBS. The total time of oocyte/embryo exposure to each buffer was ∼41min. The cleavage rates and number of embryos that developed to ≥8 cells at day 4 were no different among the buffers tested, however, more blastocysts developed at day 7, 8 and 9 in HEPES and MOPS treatments than in PBS and TES (P <0.05). No difference between buffers in total and apoptotic cell number was found. Except for Hspb1 and Ube2a genes, the levels of expression of the six remaining transcripts were higher in in vivo than in in vitro embryos irrespective of buffer used (P <0.05). In addition, higher expression of Hspb1 and lower expression of Ube2a and Lama1 were observed in PBS and TES than in MOPS and HEPES treatments (P <0.05). Expression of Fgf-4 and Gsta4 in the in vitro embryos was lower in PBS than in the remaining three buffers (P <0.05) and the level of expression of the Il6 gene was not affected by any buffer tested but was lower in in vitro than in in vivo derived embryos. Expression of both Sod1 and Prss11 genes in MOPS were at the level of the in vivo embryos. These results showed that the choice of buffer and short exposure time of ∼41min, affects mRNA expression of in vitro produced bovine embryos. [Copyright &y& Elsevier]

Abstract: Objective: Diabetes is one of the main causes of end-stage renal disease (ESRD) and admission to hemodialysis, and the demand for kidney transplantation in this population has increased. Our aim was to evaluate the clinical aspects and survival of diabetic patients with kidney transplants by comparing them with the nondiabetic population. Materials and Methods: Patients transplanted during the period from 1994 to 2003 were evaluated for this study. The transplant and demographic characteristics were analyzed by the chi-square test and Student t test according to the type of variable. Kaplan-Meier curves and the log-rank test were used to evaluate the graft and patient survival. Results: From a total of 523 consecutive renal transplants, 35 (6.6%) were diabetics who were older than nondiabetics (47 ± 11 years vs 37 ± 16, P < .002). Patients received immunosuppression with cyclosporine (84.3%), tacrolimus (11.2%), azathioprine (46.6%), mycophenolate mofetil (43.5%), and steroids (all patients). The diabetic patients had a higher percentage of living donors (33.5% vs 17.2%; P = .04). Graft survival rates at 1, 3, and 5 years were 82.7%, 70.9%, and 63.0% in the diabetic patients and 87.6%, 79.0%, and 72.5% (P = .6) in the nondiabetic patients. Patient survival at 5 years was 90.5% in diabetic patients vs 89.0% in nondiabetic patients (P = .9). Conclusions: No differences were found in our series in transplant complications or survival in the diabetic patients compared with the nondiabetic patients. Abstract: The application of biosolids such as sewage sludge is a concern, because of the potential release of toxic metals after decomposition of the organic matter. The effect of application of sewage sludge (Sw) and compost (C) to the soil (S) on the Cu and Cd sorption, distribution and the quality of the dissolved organic matter (DOM) in the soil, was investigated under controlled conditions. Visible spectrophotometry, infrared spectroscopy, sorption isotherms (simple and competitive sorption systems), and sequential extraction methods were used. The E 4/E 6 (λ at 465 and 665nm) ratio and the infrared spectra (IR) of DOM showed an aromatic behaviour in compost–soil (C–S); in contrast sewage sludge–soil (Sw–S) showed an aliphatic behaviour. Application of either Sw or C increased the Cu sorption capacity of soil. The Cd sorption decreased only in soil with a competitive metal system. The availability of Cu was low due to its occurrence in the acid soluble fraction (F3). The Cu concentration varied in accordance with the amounts of Cu added. The highest Cd concentration was found in the exchangeable fraction (F2). The Sw and C applications did not increase the Cd availability in the soil. [Copyright &y& Elsevier]

Abstract: The fatty acid composition of commercial lambs from different production systems of Spain, Germany, United Kingdom and of two types of Uruguayan lambs (heavy and light) was studied. Concentrate fed lambs, as Spanish lambs, displayed the highest proportions of linoleic acid (C18:2), while Uruguayan lambs, reared under extensive grazing conditions, showed the highest proportions of linolenic acid (C18:3), due to the great concentration of this fatty acid in grass. German and British lambs, which were fed grass and concentrate, displayed intermediate proportions of linolenic acid (C18:3). Heavy Uruguayan lambs had higher intramuscular fat content (5.92%) than German (4.25%) and British (4.32%) lambs, and this content was twofold higher than light lambs (Spanish (2.41%) and light Uruguayan (3.05%)). Heavy Uruguayan, German and British lambs had a low polyunsaturated/saturated (P/S) ratio due to their high saturated fatty acid (SFA) content and proportion. Principal component analysis was performed to study the relationship between fatty acids. Spanish lambs were clearly separated from the other types and were situated close to the proportions of short chain and n −6 fatty acids and n −6/n −3 ratio in the data plot for fatty acid proportions. Light Uruguayan lambs were located close to long chain fatty acids, and heavy Uruguayan and British lambs were placed near the antithrombotic potential (ATT), stearic acid (C18:0), SFA and conjugated linoleic acid (CLA) proportions. This is the first work that applies immunohistochemistry in the characterisation of the inflammatory infiltrate of verminous bronchopneumonia associated with lungworm parasites in stranded dolphins. Samples from three different species (Stenella coerulealba, Stenella frontalis and Delphinus delphis) stranded in the Canary Islands were used. The most common findings found in these animals varied from bronchitis to verminous bronchopneumonia with different degree of severity. The immunohistochemical study showed variable expressions of Lysozyme, MHC-II, iNOS and IgG. The main population presenting in the inflammatory infiltrates were CD3+ lymphocytes. However, moderate number of Foxp3+ lymphocytes was found in lymph nodes even though no Foxp3+ cells were found in lung lesions in any of the samples analysed. This study revealed that lung lesions showed a chronic inflammatory infiltrate mainly composed by lymphocytes CD3+. Deeper studies are needed in order to provide a more complete scope about the infiltrates involved in these types of lesions. [ABSTRACT FROM AUTHOR]

Essential oils (EO) and short-chain fatty acids have potential antimicrobial activity in broilers. This study aimed to investigate the effect of a specific blend of EO and a combination of this blend of EO with sodium-butyrate on growth performance and Salmonella colonization in broilers. A total of 480 one-day-old male broilers were distributed into 5 treatments (8 pens per treatment and 12 birds per pen) and reared during 42 d in experimental conditions. Dietary treatments consisted of the addition of different doses of EO (0 mg/kg, control; 50 mg/kg, EO50 and 100 mg/kg, EO100) or a combination of EO with 1 g/kg of sodium-butyrate (B; EO50 + B, EOB50 and EO100 + B, EOB100) to a basal diet. All birds were orally infected with 108 cfu of Salmonella Enteritidis on d 7 of study. Individual BW and feed intake per pen were measured at arrival and on a weekly basis. The prevalence and enumeration of Salmonella in feces was determined per treatment at 72 h postinfection and on d 23 and 37 of study. At slaughter, cecal content and liver samples from 16 birds per treatment were cultured for Salmonella and cecal pH was measured. No differences were observed on growth performance among treatments. All fecal samples analyzed were positive for Salmonella from d 10 to the end of the rearing period. At slaughter, Salmonella contamination (positive samples) in cecum was lower in birds fed EOB50 compared with the other treatments (P < 0.05), whereas birds fed the control diet showed the highest colonization rates. The pH of the cecal content was not different among treatments. Thus, EO or its combination with sodium-butyrate did not affect growth performance. Abstract: The effect of slaughter at arrival (L0) or after 3 (L3), 6 (L6), 12 (L12) h of lairage on carcass and meat quality of suckling lambs has been studied. Lairage time had a slight effect on carcass quality traits, but most of the meat quality parameters at 24h post mortem were affected. Weight losses increased and glycogen content of liver and longissimus muscle decreased as lairage time increased. Longissimus muscle from L3 lambs had the highest pH, shear force and toughness and the lowest b* and C* values, at 24h post mortem. L3 and L6 had the higher proportion of expelled juice, or low water holding capacity (WHC), at 24h post mortem. Texture parameters and WHC were similar among groups after 5days of storage. Despite the fact that the effects of lairage time on meat quality disappears after storage, from the point of view of weight losses the slaughter of suckling lambs at arrival is recommended. Abstract: Haematological and molecular analysis of blood samples was carried out during an outbreak of bovine anaplasmosis in Hungary. Acute disease was observed in five animals, two of which died. Anaplasma-carrier state was diagnosed in 69 (92%) of cattle. Further evaluation of 24 blood samples revealed concurrent infections with Mycoplasma wenyonii and ‘Candidatus M. haemobos’ in 22 and 21 animals, respectively. In addition, two cows were identified with rickettsaemia. Regarding molecular investigation of potential hard tick vectors, Haemaphysalis inermis and Dermacentor marginatus males collected from the animals were PCR-negative. However, in one pool (out of 18) of Ixodes ricinus males, and in six pools (out of 18) of D. reticulatus males the msp4 gene of Anaplasma marginale was detected. In the same I. ricinus pool Anaplasma ovis was also identified. All ticks were negative for haemoplasmas. Anaplasma sequences yielded 97–99% homology to sequences deposited in the Genbank. This is the first report of fatal bovine anaplasmosis associated with divergent A. marginale genotypes and concurrent ‘Candidatus M. haemobos’ infection, as well as of an A. ovis strain in ticks collected from cattle. Abstract: Fatty acid content and sensory characteristics of meat from light lambs fed three diets supplemented with different sources of n−3 fatty acids (fish oil, extruded linseed and extruded linseed plus microalgae) and a control diet during refrigerated storage have been studied. The meat from lambs fed linseed diets had the highest levels of C18:3 n−3, while animals fed fish oil had the highest long-chain n−3 polyunsaturated fatty acids (PUFA). Thus, 100g of meat from lamb fed the fish oil diet provided 183mg of long-chain n−3 PUFA, representing 40% of the daily recommended intake. The levels of n−3, n−6 and long-chain n−3 PUFA decreased during a 7-day storage period. The meat from lambs fed the fish oil diet had high scores of fish odour and flavour and rancid odour and flavour and the lowest overall liking. This study investigated whether a low pathogenicity isolate of Anaplasma marginale with an appendage (UFMG1) could protect calves from infection with a pathogenic A. marginale isolate (UFMG2). Two groups of five Friesian calves were each inoculated with UFMG1 by intravenous injections of either A marginale-infected tick cell cultures (group 1 ) or blood stabilates (group 2); a third (control) group was injected with saline. All animals were inoculated with a blood stabilate containing a high pathogenicity A. marginale isolate (UFMG2) 75 days after the UFMG1 inoculation. After infection with UFMG2, animals in groups 1 and 2 presented low rickettsaemia, but no clinical signs and no reduction in packed cell volume (PCV). Control animals became sick, with high rickettsaemia (16% infected erythrocytes) and a reduction in PCV (71%), resulting in 60% deaths. Up to 2 weeks after the UFMG2 inoculation, msp1α UFMG1 sequences were detected in groups 1 and 2. Four weeks after UFMG2 inoculation, UFMG2 sequences were detected in these animals, along with a new msp1α genotype sequence, closely related to that of the UFMG2 isolate. Control animals had UFMG2 msp1α sequences up to 4 weeks after inoculation with UFMG2 and the new msp1α genotype sequence could be detected on the sixth week. The origin of the new A. maginale genotype was unknown, but may represent the first example of MSP1α antigenic variation in infected cattle. The results confirmed the low pathogenicity of the UFMG1 isolate, which provided clinical protection against the highly pathogenic A. marginale UFMG2. Abstract: The Eurasian wild boar (Sus scrofa) is considered a reservoir for bovine tuberculosis (bTB) caused by Mycobacterium bovis and closely related members of the Mycobacterium tuberculosis complex in south-central Spain. The vaccination of wildlife with BCG offers an alternative to culling and to movement restriction for the control of bTB among wildlife reservoirs. In this study, we hypothesized that oral BCG immunization of wild boar would affect the expression of immunoregulatory genes and confer protection against M. bovis. Three groups were used to describe the infection, pathological findings and gene expression profiles in wild boar: BCG-vaccinated and M. bovis-challenged (vaccinated challenged group; N =6), non-vaccinated and M. bovis-challenged (non-vaccinated challenged group; N =4), and non-vaccinated and mock-infected (control group; N =2) animals. M. bovis was isolated from 50% (3/6) and 75% (3/4) of vaccinated challenged and non-vaccinated challenged animals, respectively. All four wild boar from the non-vaccinated challenged group developed bTB-compatible lesions 114 days after challenge. In contrast, only 50% of vaccinated challenged wild boar developed lesions. The PBMC mRNA levels of IL4, RANTES, C3, IFN-gamma and methylmalonyl-CoA mutase (MUT) were analyzed at several days post-vaccination (dpi). When vaccinated challenged animals were compared to controls, all five genes were significantly upregulated at the time of M. bovis infection at 186dpi but IFN-gamma levels were also upregulated at 11 and 46dpi. The C3 and MUT mRNA levels were higher at 46dpi, and 11 and 186dpi, respectively, in vaccinated protected wild boar when compared to non-vaccinated challenged animals. At the end of the experiment (300dpi), the mRNA levels of selected genes were lower in non-vaccinated challenged animals when compared to control wild boar. Exposing wild boar to a dose of 104 cfu of M. bovis by the oropharyngeal route is an adequate protocol to produce an infection model in this species. Our results suggested that oral BCG immunization of wild boar results in the upregulation of immunoregulatory genes that may be associated with protective response to M. bovis infection in this species. More studies on vaccine efficacy, delivery, and safety will be needed to confirm if oral vaccination with BCG could be used in bTB control programs for reducing M. bovis infection and clinical disease in wild boar. Abstract: Anaplasma marginale is a tick-borne pathogen of cattle responsible for the disease anaplasmosis. Data suggest that Rhipicephalus (Boophilus) microplus and R. annulatus may be the major tick vectors of A. marginale in tropical and subtropical regions of the world. In this work we demonstrated the first infection and propagation of a Brazilian isolate of A. marginale (UFMG1) in the BME26 cell line derived originally from embryos of R. (Boophilus) microplus. The establishment of A. marginale infection in a cell line derived from R. (Boophilus) microplus is relevant for studying the A. marginale/tick interface. [Copyright &y& Elsevier]
