The hypothesis is that dual attention integrated Residual Network (DA-ResNet) together with a Principal Component Analysis (PCA) model accurately maps the measured on-treatment CBCT projections from different phases to each phase's 3D-CBCT real timely. Finally, it achieves real time 4D-CBCT. The DA-ResNet contains a CBAM (Convolutional Block Attention Module) and a SPA (Spatial Pyramid Structure). Hundreds of forward projections from one view of patient 4D-CT were simulated to train DA-ResNet. The extracted motion features were expressed as PCA coefficients. To test DA-ResNet performance, several single measured on-treatment projection is sent into it and the network output several groups of PCA coefficients real timely. Each group of PCA coefficient stands for the corresponding deformation field between the 0% phase and the predicted phase that the PCA label stands for. In labels such as Mean Absolute Error (MAE), R square, Normalized Cross Correlation (NCC), Structural Similarity Index Measure (SSIM) were used to evaluate DA-ResNet performance with other state-of-the-art networks such as CNN/Unet/ResNet. DA-ResNet outperforms other popular networks on all of the quantification labels, with MAE (14.41 ± 8.85), R square (0.95 ± 0.07) in PCA coefficient prediction, and NCC (0.9981 ± 0.0006), SSIM (0.98 ± 0.04) in image reconstruction. Compared with conventional ResNet, DA-ResNet reduced MAE by 79%. Meanwhile R square rises from 0.88 to 0.95, NCC rises from 0.996 to 0.998, SSIM rises from 0.94 to 0.98. DA-ResNet offers a solution for real time on-treatment 4D-CBCT image guidance by using single 2D on-line projections. It offers better motion prediction and image reconstruction accuracy compared with traditional deep learning networks (e.g., CNN/Unet/ResNet). The initial results are promising. An improved rheology model, inspired from explicit experiments is conceived to represent rate-dependent cyclic shear behavior of high damping rubber bearings at subzero and room temperatures. Total stress has been decomposed into nonlinear rate independent elasto-plastic stress, nonlinear elastic stress and nonlinear visco-elasto-plastic overstress branches. To represent nonlinear viscosity behavior, ‘overstress branch’ has been generalized by putting linear elastic spring in parallel to nonlinear elasto-plastic model, placed in series with nonlinear dashpot. Constitutive relations for model elements have been designated for respective fundamental phenomenon observed in constant strain rate experiments. An optimum calculation approach is developed to determine a unique set of overstress parameters capable not only of representing constant strain rate cyclic tests but also sinusoidal tests with variable input strain rates. Essential abilities of the proposed model and adequacy of estimated parameters have been confirmed by comparing numerical simulation results with experiments conducted at −30 °C, −10 °C and 23 °C. Abstract: Erythropoietin (EPO) can induce a series of cytoprotective effects in many non-hematopoietic tissues through interaction with the erythropoietin receptor (EPOR), but whether EPO can prevent the overproduction of reactive oxygen species (ROS) and apoptosis in diabetes remains unclear. Here, we report that renal tubular cells possess EPOR and that EPO reduces high glucose-induced oxidative stress in renal tubular cells. Further, we found that EPO inhibited high glucose-induced renal tubular cell apoptosis and that this protective effect was dependent on reduction of Bax/caspase-3 expression as well as elevation of Bcl-2 expression. Our results suggest that EPO can inhibit high glucose-induced renal tubular cell apoptosis through direct effect on anti-oxidative stress and that EPOR may play a key role in this process. Abstract: An integrated process was developed to hydrolyze the phytates in light steep water (LSW) and to simultaneously isolate inorganic phosphate (Pi) and myo-inositol products. The proposed integrated process will be helpful in resolving the environmental and nutritional concerns in the use of corn gluten feed (CGF) in the animal diets. This process comprised of partial and total hydrolysis of LSW and intermediate anion exchange separation technique. The phytates in LSW were initially degraded to negatively charged myo-inositol phosphates (InsP2–InsP5). The optimized experimental parameters for the partial hydrolysis of LSW were determined to be 2h hydrolysis with 1FTU Aspergillus niger/g substrate at 35°C. The negatively charged species of the partially hydrolyzed substrate were separated on a strong base anion exchange resin. The negatively charged species, retained by the resin, were eluded with 1M NaCl solution and were subjected to complete hydrolysis with the Escherichia coli, A. niger derived phytases and their respective combinations. The maximum amount of myo-inositol released from the anion exchange column was 3.73±0.03mg/NaCl elution which was detected after 48h reactions catalyzed by 100FTU E. coli, 150FTU E. coli, and 150FTU the combination of A. niger and E. coli. The time course of Pi released showed a similar trend to that of myo-inositol and the released Pi reached a maximum amount of 3.30±0.05mg/g NaCl elution after 48h incubation at the enzyme loadings for which the maximum concentration of myo-inositol were reached. L'artérite à cellules géantes (ACG) est une vascularite granulomateuse affectant les gros vaisseaux. Le rôle des monocytes/macrophages et des cellules musculaires lisses (CML) semble prépondérant dans la physiopathologie de la maladie. Le traitement standard de l'ACG repose sur les glucocorticoïdes (GC), qui sont très efficaces, mais n'empêchent pas l'apparition de lésions vasculaires et leurs conséquences. Ainsi, une meilleure compréhension des mécanismes physiopathologiques impliqués dans l'ACG semble nécessaire pour améliorer la prise en charge des patients. La voie de l'EGFR (Epithelial Growth Factor Receptor) pourrait jouer un rôle dans certains maladies inflammatoires, ainsi que dans la migration et la prolifération des CML. L'HBEGF (Heparin-binding epidermal growth factor) et l'amphiréguline (AREG) sont les ligands de l'EGFR les plus connus. De plus, leur rôle dans les maladies inflammatoires émerge dans la littérature. L'objectif de cette étude était donc d'étudier le rôle de la voie de signalisation de l'EGFR dans la physiopathologie de l'ACG. Du matériel humain et des lignées cellulaires ont été utilisés dans cette étude. Les échantillons de sérum et les biopsies d'artère temporale (BAT) ont été obtenus auprès de patients participant à l'étude VASCO (VASculitis COhort), une cohorte prospective de patients atteints de vascularite systémique. Deux lignées cellulaires ont par ailleurs été utilisées : HAoSMC (CML aortique humaine) et THP-1, monocytes humains. Grâce à des techniques d'immunohistochimie (IHC) multiplex et de quantification, nous avons mis en évidence que les BAT de patients atteints d'ACG exprimaient des niveaux significativement plus élevés d'AREG, d'HBEGF, d'EGFR et de p-EGFR par rapport aux BAT de témoins. Les images de colocalisation ont montré qu'AREG, HBEGF et EGFR étaient exprimés par les macrophages et HBEGF, EGFR et p-EGFR par les CML (Figure). En utilisant une technique d'IHC multiplex et des tests ELISA, nous avons constaté que l'AREG, le HBEGF, l'EGFR et le pEGFR étaient significativement plus élevés dans les macrophages que dans les THP-1, en particulier dans les macrophages polarisés pro-inflammatoires (M1). Les niveaux d'AREG et d'HBEGF étaient augmentés dans le surnageant des macrophages polarisés M1 et l'HBEGF dans le surnageant des macrophages M2. Par ailleurs, la stimulation des cellules THP-1 par l'AREG ou le HBEGF induisait la phosphorylation de p38 et l'activation de la voie MAPK. Sur une technique d'imagerie de cellules vivantes (Live cell imaging) AREG et HBEGF augmentaient à la fois la réplication et la migration des CML. À l'inverse, AG1478, un inhibiteur de l'EGFR stoppait à la fois la migration et la prolifération des CML, suggérant un rôle clé de la voie de l'EGFR sur les CML. Ni l'AREG ni l'HBEGF n'augmentaient la production de cytokines pro-inflammatoires par les THP1, les macrophages ou les CML (Figure 1). La voie de l'EGFR et son activation par l'AREG et l'HBEGF pourrait jouer un rôle clé dans la physiopathologie de l'ACG, en particulier dans la phase de remodelage vasculaire, qui est responsable des complications tardives et qui n'est pas bien ciblée actuellement par les thérapies immunosuppressives. Une stratégie thérapeutique ciblant la voie de l'EGFR ou ses ligands pourrait ainsi permettre un effet épargneur de corticoïdes et améliorer le traitement des patients atteints d'ACG. La glomérulonéphrite extra-membraneuse (GEM) lupique représente 5 à 20 % des atteintes rénales du lupus systémique [1]. Si le traitement des classes prolifératives (III et IV de la classification ISN-RPS 2003) est relativement consensuel, celui des GEM lupiques (classe V de la classification ISN-RPS 2003) n'est toutefois pas clairement défini [2]. Notre étude a pour objectif de décrire les caractéristiques cliniques, biologiques et histologiques des patients avec GEM lupique et d'apprécier l'efficacité des différentes thérapeutiques. L'étude menée est rétrospective, observationnelle, multicentrique, conduite sur 18 centres français. Les patients majeurs avec un diagnostic de première poussée de GEM lupique pure entre 2000 et 2020 étaient inclus. Les patients avec une poussée antérieure de GEM lupique étaient exclus. Une rémission complète était définie comme un rapport protéinurie/créatininurie < 0,05 g/mmol et une rémission partielle comme une réduction de > 50 % mais comprise entre 0,05 et 0,3 g/mmol de la protéinurie initiale avec stabilité de la fonction rénale conformément aux recommandations KDIGO 2024 [2]. Deux cent 1Xhuit patients (87,6 % de femmes avec un âge médian [IQR] de 27 [21–35 ans]) ont été inclus. Parmi eux, 97 (44,5 %) étaient d'origine afro-caribéenne. Dans 89 cas (36,7 %) la GEM lupique était inaugurale de la maladie. Soixante-six (30,3 %) patients avaient présenté une poussée rénale antérieure, dont 22 (33,3 %) une classe III ± V et 27 (40,9 %) une classe IV ± V. Au diagnostic de GEM lupique, les médianes de débit de filtration glomérulaire (DFG) étaient de 117 [94–131] mL/min/1,73 m2, d'albuminémie de 29 [21,2–34] g/L et de protéinurie de 0,23 [0,13–0,45] g/mmol ; 88/200 patients (44 %) présentaient une hématurie. Au total, 75/210 (35,7 %) patients avaient un syndrome néphrotique et 77 (35,3 %) présentaient une atteinte rénale isolée. Initialement, 51 patients (23,4 %) bénéficiaient d'un traitement néphroprotecteur seul alors que 30 (13,8 %), 64 (29,3 %), et 73 (33,5 %) patients bénéficiaient d'un traitement par Rituximab (RTX) ± corticothérapie, Mycophénolate mofétil (MMF) ± corticothérapie, ou un autre traitement immunosuppresseur respectivement. Dix (19,6 %) patients sous traitement néphroprotecteur nécessitaient l'introduction d'un traitement immunosuppresseur avant le 6e mois de suivi (19,6 %). Les patients sous traitement néphroprotecteur présentaient un taux d'albumine significativement plus important (p < 0,001) et une protéinurie plus faible par rapport aux 3 autres groupes. Le nombre de patient présentant un syndrome néphrotique était plus grand dans le groupe RTX (n = 16/34, 47,1 %) que dans le groupe néphroprotecteur (n = 9/41, 22 %), MMF (n = 25/67, 37,3 %) ou autre (n = 24/76, 31,6 %), p = 0,08. La réponse globale (partielle + complète) à 12 mois ne différait pas entre les 4 groupes (70,4 % dans le groupe néphroprotecteur, 68,7 % dans le groupe MMF, 75,7 % dans le groupe RTX et 64,3 % dans le groupe « autre », p = 0,7). Le nombre de rechute d'une GEM lupique au décours du traitement n'était pas non plus différent (16,7 %, 18,7 %, 16,7 % et 20,9 % respectivement, p = 0,9), de même que le risque de rechute rénale ou extrarénale tout venant. Le DFG était préservé dans les 4 groupes après un suivi médian de 63,8 [34,7–101] mois. La GEM lupique présente un taux de rémission complète plus élevée que dans les formes prolifératives mais sous réserve de l'emploi d'immunosuppresseurs dans 71 % des cas. Si le type de traitement immunosuppresseur n'est pas consensuel, il ressort que le RTX ± corticothérapie ne semble pas inférieur au MMF ou à d'autres d'immunosuppresseurs [3]. [ABSTRACT FROM AUTHOR]

Abstract: When the prime word is masked and flashed very quickly, its presence cannot be perceived by the subjects, but it can still accelerate the processing of the subsequent relevant target stimulus. This is known as the masked priming effect. Adopting a similar experimental paradigm, in this study we take the gray scale pictures describing ordinary objects as prime stimuli and the environmental sounds as target stimuli. The subjects are then asked to classify the target sounds to investigate whether incongruent target sounds elicit the N400 semantic priming effect. At the same time, the prime-mask withdraw value is made at these time intervals: 16ms, 33ms, 50ms, 66ms and 83ms, so as to reveal whether there exists a correlation between the prime duration and the priming effect. The result shows that, at each time interval, the processing of sounds that are congruent with prime pictures is sped up and sounds that are incongruent with prime pictures trigger the N400 effect. The amplitude of the N400 effect is enlarged proportionally with the time interval and gradually approaches the point of saturation. Given this, it is believed that the N400 effect reflects automatic semantic processing, that consciousness is not a prerequisite of semantic priming, and that early visual information has an accumulative priming effect on target sounds. Abstract: In this paper, we investigated event-related potentials (ERPs) evoked by visual stimuli with different three-dimensional visual depths. Before the experiment, tests for binocular advantage and for sensitivity to three-dimensional depth were carried out, respectively. At the same time, it should be ensured that the distinguishing degree of different depths is big enough. After the qualified participants were selected, pictures with five different three-dimensional depths were presented to the participants who were asked to give a judgment of the depths. The behavioral results showed that under the five different depths, the participants had a very high accuracy of judgment. ERP results showed that in the time windows of 90–130ms and 150–200ms after the onset of visual target stimuli, P1 and N2 components appeared in the area from parietal to occipital regions. It was interesting that there existed a positive correlation between the amplitude of the N2 component and the absolute value of the depth. Meanwhile, time–frequency analysis results showed that, in the time window of 150–200ms after the onset of visual target stimuli, a similar positive correlation between the time–frequency distribution and the absolute value of the depth was also found. Additionally, in the time window of 200–600ms after the onset of visual target stimuli, the alpha waves evoked under the five different depths were almost the same, which reflected that the cognitive process of three-dimensional visual depth might be finished by 200ms after the onset of visual target stimuli, when the brain is in a state of relaxation.
