Abstract

The SARS-CoV-2 Omicron variant emerged in South Africa in November 2021, and has later been identified worldwide, raising serious concerns. A real-time RT-PCR assay was designed for the rapid screening of the Omicron variant, targeting characteristic mutations of the spike gene. The assay was used to test 737 sewage samples collected throughout Italy (19/21 Regions) between 11 November and 25 December 2021, with the aim of assessing the spread of the Omicron variant in the country. Positive samples were also tested with a real-time RT-PCR developed by the European Commission, Joint Research Centre (JRC), and through nested RT-PCR followed by Sanger sequencing. Overall, 115 samples tested positive for Omicron SARS-CoV-2 variant. The first occurrence was detected on 7 December, in Veneto, North Italy. Later on, the variant spread extremely fast in three weeks, with prevalence of positive wastewater samples rising from 1.0% (1/104 samples) in the week 5–11 December, to 17.5% (25/143 samples) in the week 12–18, to 65.9% (89/135 samples) in the week 19–25, in line with the increase in cases of infection with the Omicron variant observed during December in Italy. Similarly, the number of Regions/Autonomous Provinces in which the variant was detected increased from one in the first week, to 11 in the second, and to 17 in the last one. The presence of the Omicron variant was confirmed by the JRC real-time RT-PCR in 79.1% (91/115) of the positive samples, and by Sanger sequencing in 66% (64/97) of PCR amplicons. In conclusion, we designed an RT-qPCR assay capable to detect the Omicron variant, which can be successfully used for the purpose of wastewater-based epidemiology. We also described the history of the introduction and diffusion of the Omicron variant in the Italian population and territory, confirming the effectiveness of sewage monitoring as a powerful surveillance tool.

10. Validation plan of bone marrow collection, processing and distribution using the failure mode and effect analysis methodology: a technical report

Abstract

Background aims: Bone marrow (BM) is commonly used in the pediatric and adult setting as a source of hematopoietic stem cells (HSCs). The standards of the Joint Accreditation Committee of the International Society for Cell & Gene Therapy & European Society for Blood and Marrow Transplantation (JACIE) include specific requirements regarding BM collection, processing and distribution. To run this process, each transplant team develops a series of JACIE-compliant procedures, customizing them with regard to local settings and paths. Moreover, JACIE standards require that transplant teams validate and periodically revise their procedures to keep the entire process under control. In this article, the authors describe the methodology adopted in our center to fulfill the aforementioned JACIE requirements. Methods: The authors developed a validation plan based on the failure mode and effect analysis (FMEA) methodology. According to the FMEA approach, the authors carefully revised activities and procedures connected to BM collection, processing and distribution at our institution. The entire process was initially divided into five main phases (assessment of donor eligibility, perioperative autologous blood donation, preparation of BM collection kit, BM harvesting and BM processing and distribution), comprising 17 subphases and 22 activities. Results: For each activity, one or more failure modes were identified, for a total of 28 failure modes, and a risk priority number (RPN) was then assigned to each failure mode. Although many procedures were validated, others were subjected to substantial changes according to the RPN rating. Moreover, specific indicators were identified for subsequent monitoring to contain the risk of failure of steps emerging as critical at FMEA. Conclusions: This is the first study describing use of the FMEA methodology within an HSC transplant program. Abstract

Background: The coronavirus disease 2019 (COVID-19) pandemic has deeply modified the complex logistical process underlying allogeneic hematopoietic stem cell transplant practices. Aim: In light of these changes, the authors compared data relative to allogeneic transplants carried out from 2018 at their center before (n = 167) and during the pandemic (n = 45). Methods: The authors examined patient characteristics, donor and graft types, cell doses and main transplant outcomes. Moreover, the authors evaluated the rise of costs attributable to additional COVID-19-related procedures as well as the risk of adverse events these procedures conveyed to grafts or recipients. Results: Overall, the number of transplants did not decrease during the pandemic, whereas patients at high relapse risk were prioritized. Transplants were mainly from matched unrelated donors, with a significant decrease in haploidentical related donors. Moreover, the use of bone marrow as a graft for haploidentical transplant was almost abandoned. Cryopreservation was introduced for all related and unrelated apheresis products, with a median storage time of 20 days. Notably, transplant outcomes (engraftment, acute graft-versus-host disease and non-relapse mortality) with cryopreserved products were comparable to those with fresh products. Conclusions: Considering that the emergency situation may persist for months, cryopreserving allogeneic grafts can offer a lifesaving opportunity for patients whose allogeneic transplant cannot be postponed until after the end of the COVID-19 pandemic.

Abstract

We analyzed data relative to cell content in 88 consecutive patients receiving HLA haploidentical bone marrow (BM) transplants with post-transplantation cyclophosphamide (PT-CY). Median age was 54.5 (range, 17–72); diagnoses were acute leukemia (n = 46), lymphoproliferative disorders (n = 24), myelofibrosis (n = 11) and myelodysplastic syndromes (n = 5). Total nucleated cell (TNC) and CD34+, CD3+, CD4+ and CD8+ cell doses were stratified as higher than first, second and third quartile and the dose effect on various clinical outcomes was assessed. Median time to engraftment was 17 days for neutrophils and 24 days for platelets. To receive a dose of TNC ≥3.2 x 106/kg or CD34+ cells ≥2.7 x 106/kg significantly shortened the time to neutrophil and platelet engraftment and reduced the blood product requirements in the 30-day period after transplantation. Overall, TNC and CD34+ cell doses had no effect on acute graft-versus-host disease (GVHD) incidence, whereas patients receiving higher CD3+ and CD8+ cell doses seemed to have less chronic GVHD. No effect on non-relapse mortality, progression-free survival and overall survival was observed at different cell dose thresholds. These data suggest that in HLA haploidentical BM transplant with PT-CY, appropriate cell doses are relevant to the engraftment. The association between low CD3+/CD8+ cells and chronic GVHD deserves further investigation.

19. Pesticide-induced alterations in zebrafish (Danio rerio) behavior, histology, DNA damage and mRNA expression: An integrated approach.

Abstract

To assess the impact of glyphosate and 2,4-D herbicides, as well as the insecticide imidacloprid, both individually and in combination, the gills of adult zebrafish were used due to their intimate interaction with chemicals diluted in water. Bioassays were performed exposing the animals to the different pesticides and their mixture for 96 h. The behavior of the fish was analyzed, a histological examination of the gills was carried out, and the genotoxic effects were also analyzed by means of the comet assay (CA) and the change in the expression profiles of genes involved in the pathways of the oxidative stress and cellular apoptosis. The length traveled and the average speed of the control fish, compared to those exposed to the pesticides and mainly those exposed to the mixture, were significantly greater. All the groups exposed individually exhibited a decrease in thigmotaxis time, indicating a reduction in the behavior of protecting themselves from predators. Histological analysis revealed significant differences in the structures of the gill tissues. The quantification of the histological lesions showed mild lesions in the fish exposed to imidacloprid, moderate to severe lesions for glyphosate, and severe lesions in the case of 2,4-D and the mixture of pesticides. The CA revealed the sensitivity of gill cells to DNA damage following exposure to glyphosate, 2,4-D, imidacloprid and the mixture. Finally, both genes involved in the oxidative stress pathway and those related to the cell apoptosis pathway were overexpressed, while the ogg1 gene, involved in DNA repair, was downregulated. 21. Betweenness to assess leaders in criminal networks: New evidence using the dual projection approach

Abstract

Brokerage is crucial for dark networks. In analyzing communications among criminals, which naturally induce bipartite networks, previous studies have focused on the classic Freeman's betweenness, conceived for one-mode matrices and possibly biasing the results. We explore different betweenness centrality including three inspired by the dual projection approach recently suggested by Everett and Borgatti 2013. We test these measures in identifying criminal leaders in a meeting participation network. Despite the expected high correlations among them, the measures yield different node rankings, capturing different characteristics of brokerage. Overall, the dual projection approaches show higher success than classic approaches in identifying the criminal leaders.

Abstract

Brokerage is crucial for dark networks. In analyzing communications among criminals, which naturally induce bipartite networks, previous studies have focused on the classic Freeman's betweenness, conceived for one-mode matrices and possibly biasing the results. We explore different betweenness centrality including three inspired by the dual projection approach recently suggested by Everett and Borgatti 2013. We test these measures in identifying criminal leaders in a meeting participation network. Despite the expected high correlations among them, the measures yield different node rankings, capturing different characteristics of brokerage. Overall, the dual projection approaches show higher success than classic approaches in identifying the criminal leaders.

Abstract

Variations in the concentration of salivary alpha amylase (sAA) may indicate a change in the autonomic nervous system functionality. In osteopathic medicine it has long been stated that the osteopathic manipulative treatment (OMT) can modulate the autonomic nervous system. Studies carried out on the compression of the fourth ventricle (CV4) have shown a positive effect in reducing the sympathetic tone. The goal of this pilot study is measuring the physiological response of the sAA levels after CV4 technique. 90 subjects were randomly assigned to a sham, a control or a CV4 group. Randomization accounted for sex and score in the STAI-2 (form Y) questionnaire. Each subject completed the STAI-1 (form Y) questionnaire to evaluate the anxiety of the moment. sAA activity and saliva flow rate were measured. Saliva was collected before, immediately after and 30 min after treatment. Within group analysis revealed that sAA activity increased significantly immediately after the technique application only in the CV4 group (p = 0,05). Between groups analysis show a significant difference of the sAA activity in the CV4 group respect the control group (p < 0,05), but no significant difference between CV4 and sham group (p > 0,05). The effect in the CV4 group after the intervention is highly variable and appeared to be related to the level of stress measured with the STAI-Y1 questionnaire (p = 0,002). This study shows a positive effect of the CV4 procedure on sAA activity even if not significantly different from the sham procedure, probably due to the confounding effect of stress variability between groups. This work describes modeling and performance prediction of a kW-size reciprocating piston expander adopted in micro-Organic Rankine Cycle (ORC) energy systems. Two selected semi-empirical models have been opportunely adapted, calibrated and validated over a full set of experimental data to detect the best method for the simulation of a reciprocating machine. The first modelling approach is based on polynomial correlations of the expander efficiencies and it has been extended to account for the heat losses to ambient. The second one is a lumped parameters model using few key geometrical data and some physical equations to describe the process. Within the calibration range, the considered models show similar performance results of the output variables: the lowest mean relative error value is obtained on the prediction of exhaust temperature (lower than 2%). Maximum relative errors are obtained in the evaluation of rotational speed for the polynomial fitting model (equal to 10%) and in the electric power output for the lumped parameters approach (equal to 8%). The global error function value is close to 5% for both the applied approaches. Conversely, when compared outside of the calibration range, the polynomial fitting functions model proves to be less accurate overestimating the expander rotational speed while underestimating the value of the filling factor at high pressure ratios, and overestimating the isentropic efficiency at low pressure ratios. Abstract Natural gas compressor stations represent a huge potential in terms of waste heat recovery. Typical installations consist of multiple gas turbine units, in mechanical drive arrangement, operated most of the time under part-load conditions. The paper investigates the feasibility of Organic Rankine Cycle application as bottomer recovery technology in natural gas compressor facilities. The aim of the performed analysis is to obtain a detailed techno-economic and environmental scenario of the integrated system on yearly base. Different commercial gas turbine models, in the size range from 3 to 30 MW, have been taken into account as representative of mechanical driver units. Bottomer configurations (with & without intermediate loop) are modelled and compared assuming two different organic fluids. A sensitivity analysis of the bottomer cycle is carried out aimed at maximizing ORC shaft power output for each investigated layout. Off-design part-load operation of the integrated cycles have been simulated with reference to one minute data typical GT operation on a yearly base. The goal of this work is: (i) to assess the actual performance of merging gas turbines and ORC units for efficient power generation under variable operating conditions; (ii) to analyze the real potential of state-of-the art technology in the proposed innovative application. Abstract The aim of this study is to characterize a prototype of reciprocating piston expander integrated into a micro-ORC system test bench (in the kW range of power), installed at the laboratory of the university of Bologna. In order to simulate behavior and performances of the expander in not yet explored operating conditions, two semi-empirical models proposed in the literature have been opportunely adapted to the case of study and calibrated over a full set of available experimental data. One model is based on polynomial correlations on the expander efficiencies, whereas the other one is based on a lumped parameters approach with a more physical sense. Both the models have been evaluated on the error on predict the outputs and compared into performance prediction maps. The preliminary results demonstrate that the polynomial fitting functions model is the most accurate in predicting the outputs in the range of the explored working conditions. However, in order to verify the models extrapolation capability, more experimental points should be collected. Abstract Nowadays cogeneration is recognized as one of the most effective techniques to meet the increasingly stringent requirements regarding energy efficiency increase and energy saving in buildings. In this context, the aim of this study is the definition of reference parameters for the optimal energy systems design in residential applications. To this purpose, a generation scenario with cogeneration units, heat pumps, auxiliary boilers and chillers (both compression and absorption machines) has been set for the fulfillment of residential users' needs (in terms of electrical, thermal and cooling loads). For a given number of involved households, commercial cogeneration units have been selected, sized on the basis of the electrical peak need, and the generation scenario has been optimized by an in-house developed software, obtaining the optimal energy systems design and operation. Then, a parametric analysis has been carried out varying the number of considered households in order to define the optimal range of the energy systems size. In particular, specific values in terms of installed power for household and installed power for unitary peak load have been determined. For completeness, an economic analysis has been finally carried out for the evaluation of the return on investment and of the differential net present value – with respect to a standard generation scenario (only natural gas boilers for thermal needs fulfillment and electricity purchase from the grid for electrical and cooling needs) – considering a time horizon of ten years. Abstract

Making use of a majorization technique for a suitable class of graphs, we derive upper and lower bounds for some topological indices depending on the degree sequence over all vertices, namely the first general Zagreb index and the first multiplicative Zagreb index. Specifically, after characterizing c-cyclic graphs (0≤c≤6) as those whose degree sequence belongs to particular subsets of Rn, we identify the maximal and minimal vectors of these subsets with respect to the majorization order. This technique allows us to determine lower and upper bounds of the above indices recovering some existing results in the literature as well as obtaining new ones.
