Extracellular vesicles (EVs) harvested from conditioned media of human mesenchymal stromal cells (MSCs) suppress acute inflammation in various disease models and promote regeneration of damaged tissues. After successful treatment of a patient with acute steroid-refractory graft-versus-host disease (GVHD) using EVs prepared from conditioned media of human bone marrow–derived MSCs, this study focused on improving the MSC-EV production for clinical application. Independent MSC-EV preparations all produced according to a standardized procedure revealed broad immunomodulatory differences. Only a proportion of the MSC-EV products applied effectively modulated immune responses in a multi-donor mixed lymphocyte reaction (mdMLR) assay. To explore the relevance of such differences in vivo , at first a mouse GVHD model was optimized. The functional testing of selected MSC-EV preparations demonstrated that MSC-EV preparations revealing immunomodulatory capabilities in the mdMLR assay also effectively suppress GVHD symptoms in this model. In contrast, MSC-EV preparations, lacking such in vitro activities, also failed to modulate GVHD symptoms in vivo. Searching for differences of the active and inactive MSC-EV preparations, no concrete proteins or miRNAs were identified that could serve as surrogate markers. Standardized MSC-EV production strategies may not be sufficient to warrant manufacturing of MSC-EV products with reproducible qualities. Consequently, given this functional heterogeneity, every individual MSC-EV preparation considered for the clinical application should be evaluated for its therapeutic potency before administration to patients. Here, upon comparing immunomodulating capabilities of independent MSC-EV preparations in vivo and in vitro , we found that the mdMLR assay was qualified for such analyses. [ABSTRACT FROM AUTHOR]

In performance based fire safety design, ensuring safe egress, e.g. by visibility of safety signs, is a crucial safety goal. Compliance with the building requirements is often demonstrated by simulations of smoke spread. Numerical models like the Fire Dynamics Simulator generally compute visibility as a local quantity using the light extinction coefficient, without the consideration of the actual light path to a safety sign. Here, visibility maps are introduced, providing an approach for post-processing fire simulation data. They indicate safe areas along egress routes, with respect to visibility. At each location, the available visibility is calculated using Jin's empirical relation, as an integrated value of the extinction coefficient along the line of sight to the closest exit sign. The required visibility results from the distance between those points. Additional parameters like view angle or visual obstructions are considered. STATEMENT: The International Society for Cellular and Gene Therapies (ISCT) and the International Society for Extracellular Vesicles (ISEV) recognize the potential of extracellular vesicles (EVs, including exosomes) from mesenchymal stromal cells (MSCs) and possibly other cell sources as treatments for COVID-19. Research and trials in this area are encouraged. However, ISEV and ISCT do not currently endorse the use of EVs or exosomes for any purpose in COVID-19, including but not limited to reducing cytokine storm, exerting regenerative effects or delivering drugs, pending the generation of appropriate manufacturing and quality control provisions, pre-clinical safety and efficacy data, rational clinical trial design and proper regulatory oversight. This study applies Value-Belief-Norm (VBN) theory to investigate fishers' pro-environmental behavioral intention and their support for policies to reduce marine litter. While pro-environmental behavioral intention is often associated with personal environmental intention at the household level, policy support represents their support for government action at the political level. Therefore, we examine whether fishers are willing to engage in both of these dimensions. Data from 369 Vietnamese fishers are analyzed using structural equation modeling with FIMIX-PLS and PLS-POS. The results show that the sequence of values, beliefs, and personal norms can explain the fishers' pro-environmental behavioral intention and their support for policies addressing marine litter. Two segments of the fishers are detected from the sample with different perspectives of engagement intention. Fishers in segment 1 placed great emphasis on the consequences of marine litter on fishing activities as a motivator for their responsible actions. Meanwhile, fishers in segment 2 express pro-environmental engagement intention driven by the concern about responsibility and moral obligation. A binary logistic regression indicates that the risks of marine litter on fishing activity can provide the explanations for the differences between them. Our findings highlight both fishers' pro-environmental behavior and policy support as important strategies for dealing with marine litter in fisheries. If implemented simultaneously, they hold promise to alleviate the environmental and economic impact of marine litter in Vietnam. • Fishers felt a responsibility to behave pro-environmentally. • Combination of values, beliefs, and personal norms could predict pro-environmental behavioral intention and policy support. • Management measures to address marine litter should emphasize both fishers' pro-environmental behavior and policy support. This research takes a holistic approach to considering the consequences of marine plastic pollution. A semi-systematic literature review of 1191 data points provides the basis to determine the global ecological, social and economic impacts. An ecosystem impact analysis demonstrates that there is global evidence of impact with medium to high frequency on all subjects, with a medium to high degree of irreversibility. A novel translation of these ecological impacts into ecosystem service impacts provides evidence that all ecosystem services are impacted to some extent by the presence of marine plastic, with a reduction in provision predicted for all except one. This reduction in ecosystem service provision is evidenced to have implications for human health and wellbeing, linked particularly to fisheries, heritage and charismatic species, and recreation. • Marine plastic has a global impact on all ecological subjects reviewed. • There are negative impacts on the provision of almost all marine ecosystem services. • Negative human wellbeing impacts, particularly fisheries, heritage, recreation. • Economic costs conjectured: $3300–$33,000 per tonne of marine plastic per year. Two photometric measurement methods (Radiance method and LEDSA) were compared against the established MIREX measurement apparatus under controlled laboratory conditions to assess their capability of measuring extinction coefficients in real-scale fires on a temporal and spatial scale. LEDSA is a tomographic technique based on direct measurements of light intensity from individual LEDs using commercially available DSLR cameras. By discretizing the domain into horizontal layers with homogeneous smoke density, values of the extinction coefficient can be computed using an inverse model based on Beer Lambert's law. The Radiance method involves measuring the contrast of light and dark areas in images and/or video footage. It was originally developed to investigate the descent of the smoke layer in high-temperature fire events. In this work, the extinction coefficient was deduced from measurements on a contrast board by a straightforward analytical approach. Both methods were shown to yield similar extinction coefficient results in line with the MIREX for an EN 54-7 TF5 n-heptane fire. The Radiance method is able to generate accurate patterns but not values for a TF2 wood smouldering fire, while LEDSA is generally able to reflect the MIREX measurement values, yet requires higher computational effort. • Real-scale visibility experiments. • High resolution measurement of extinction coefficients in compartment fires. • Verification of two novel photometric approaches against established MIREX apparatus. • Experiments similar to the EN 54-7 TF5 (flaming) and TF2 (non-flaming) test fires. Promoting tolerance towards wildlife that pose a threat to humans or their livelihoods is critical to achieving species conservation goals. While conservation discourses often revolve around retaliation, tolerance and intolerance can vary from passive acceptance to active support, and according to level and scale of loss. We developed an experimental tool with a gamified survey-based field experiment to test multiple intended behaviours along a gradient of livestock loss to predation by jaguars (Panthera onca) in Calakmul, Mexico. Using a within-subject design, participants played a card-choice game against livestock loss scenarios. The choices offered were "No action [taken]", "Protect [domestic animals]", "Kill [the jaguar]". First, our experimental design was validated by increased frequencies of "Kill" responses in reaction to hypothetical jaguar incursion with increasing damage. The greatest difference occurred between zero and minimal damage (one chicken), showing that whether damage occurs at all is at least as important as its extent. Secondly, average frequency of intentional retaliation to jaguar incursion was higher for participants who had already killed a jaguar. Finally, age, education, and ethnicity all correlated with intentional behaviours to implement preventive measure or kill jaguars. Past experience of attack had no effect. Our results showed that survey/game-based experiments can assess tolerance to potentially hazardous wildlife interactions, while allowing more nuanced understandings of people's tolerance on a gradient of loss. This allows us to move away from a dualistic model of "conflict" versus "coexistence" towards a more complex understanding incorporating individual attitudes and experience, cultural significance, and socio-economic context.
