The following investigation was carried out to determine the effects of Selenium nanoparticles (Se NPs) on the growth rates, nutrient digestibility, and hematology of Cirrhinus mrigala fingerlings fed sunflower meal as basal diet. The experiment included seven test diets with varying Se levels (0, 0.5, 1, 1.5, 2, 2.5, and 3 mg/kg) based on Se NPs supplementation. Chromic oxide, an inert maker, was also added. Fingerlings were fed at a rate of 5% of their body weight. The test meal of 1 mg/kg Se NPs resulted in the highest weight gain (12.31 g) and the lowest feed conversion ratio (1.58). Best hematological indices (RBCs 2.84 106 mm−3, WBCs 7.79 103 mm−3, PLT 66, Hb 8.5 g/100 ml, PCV 25% and MCV 190 fl) and maximum nutrient absorption (crude protein 72%, ether extract 73% and gross energy 67%) were also observed in the case of 1 mg/kg supplementation of Se NPs. Hematology studies indicated that when fish were fed 0.5 mg/kg Se NPs, their levels began to rise. Maximum results were achieved with feed containing 1 mg/kg of Se NPs, but when the concentration increased above 1 mg/kg, the values began to decline. Instead, nutrient digestibility began to increase when the concentration of Se NPs increased to 1 mg/kg and abruptly started to decline with a further increase in Se NPs. The results demonstrated that a sunflower meal-based diet supplemented with Se NPs (1 mg/kg) increased the growth performance, nutritional digestibility, and hematology of C. mrigala fingerlings. Perfluorooctane sulfonate (PFOS) is an endocrine disruptor chemical (EDC) with potentially adverse effects on the male reproductive system. Pachypodol (5,4′-dihydroxy-3,7,3′-trimethoxyflavone) is a promising flavonoid isolated from Pogostemon cablin (Blanco) Benth that shows a broad range of pharmacological properties. However, the potential curative effects of pachypodol on testicular toxicity are not available until now. Therefore, this research was proposed to examine the efficiency of pachypodol against PFOS-induced testicular toxicity in adult male rats. The experiments were conducted on Sprague-Dawley rats (n = 48), which were equally distributed into four groups: control, PFOS (20 mg/kg), PFOS + Pachypodol (20 mg/kg + 10 mg/kg respectively), and Pachypodol (10 mg/kg). After 56 days of treatment, testes were excised by slaughtering rats, weighed, and stored till further analysis. The estimated parameters include biochemical markers, spermatogenic indices, hormonal and histopathological profiles. PFOS exposure disturbed the biochemical profile by altering the antioxidant/oxidant balance. For instance, it decreased the activities of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPx), and glutathione reductase (GSR) while increasing the concentration of reactive oxygen species (ROS) and level of thiobarbituric acid reactive substances (TBARS). PFOS intoxication also led to a notable decline in viability, motility, epididymal sperm count, and the number of HOS coiled-tail sperms, whereas the higher level of abnormality in the head, mid-piece, and tail of sperms were observed. Besides, it lowered luteinizing hormone (LH), follicle-stimulating hormone (FSH), and plasma testosterone. In addition, PFOS exposure led to histopathological damages in testicles. However, pachypodol treatment potently alleviated all the illustrated impairments in testes. Bisphenol A (BPA) is an industrial toxicant that can potentially damage the liver. Tangeretin (TGN) is a natural flavonoid that displays various pharmacological activities. This experiment was carried out to evaluate the protective effects of TGN against BPA-induced hepatic impairment in the male albino rat. Twenty-four male albino rats were equally divided into four different groups: control, BPA (100 mg/kg), BPA + TGN (100 mg/kg + 50 mg/kg) and TGN (50 mg/kg). BPA exposure significantly decreased the activities of catalase (CAT), superoxidase dismutase (SOD), peroxidase (POD), glutathione reductase (GSR), glutathione S-transferase (GST), and glutathione (GSH) content while substantially increasing the thiobarbituric acid reactive substances (TBARS) and hydrogen peroxide (H 2 O 2) levels. A substantial increase in the levels of alanine aminotransferase (ALT), alkaline phosphatase (ALP), aspartate aminotransferase (AST) was also observed in BPA treated rats. Moreover, BPA significantly increased the inflammatory markers, including tumor necrosis factor-α (TNF-α), nuclear factor kappa-B (NF-κB), Interleukin-6 (IL-6), Interleukin-1β (IL-1β)levels, cyclooxygenase-2 (COX-2) activity, and histopathological damages. However, co-treatment with TGN efficiently minimized the BPA-induced biochemical, inflammatory, and histopathological impairments in rat liver. Small non-coding RNAs (sRNAs) are about 50–500 nucleotides in length, regulate the gene expression at both transcriptional and posttranslational levels, and are critical players in microbial adaptation to different environmental conditions. Our current understanding of molecular processes underlying adaptation to harsh environments, including cold environments, is far from being complete. To understand the regulation of gene expression in psychrophilic bacteria and the roles of sRNAs in cold adaptation, we scanned the genome of psychrophilic and halotolerant bacterium Psychrobacter aquaticus using a comparative genomics-based approach. In total, 111 putative cis -regulatory RNAs (cis -sRNAs) exhibiting thermodynamic stability of their secondary structure were predicted, 43 of which were predicted to be able to bind to their respective targets and studied further. Six of the 43 cis -sRNAs, including cis2, cis9, cis19, cis28, cis32, and cis40, were predicted as regulators of regulatory proteins, revealing that regulatory proteins themselves are regulated by sRNAs. A major class of cis -sRNAs are considered regulators of the genes related to synthesizing and transporting amino acids, nucleotides, and other metabolites in Psychrobacter aquaticus. Several cis -sRNAs were identified to putatively function in modulating the expression of genes related to stress responses. Particularly, cis38 was identified as a regulator of the rhlB gene crucial for bacterial adaptation to cold environments and, therefore, was regarded as a cold regulator. A putative cis -sRNA homologous to cis38 was also found in Psychrobacter alimentarius , a psychrophilic bacterium isolated from Antarctica and Korean fermented food. Both cis8 and its homolog shared a conserved domain interacting with their respective targets, mRNAs of the rhlB genes. Understanding the mechanisms of cold regulators will provide future opportunities to engineer bacterial species encoding cold-adapted enzymes for industrial applications. In an epoch of escalating number of antibiotic-resistance bacteria, there is a dire need to develop efficient and novel feeding strategies for animal nutrition as alternatives to antibiotics. Here, implicating nutrigenomic approach, phytobiotics and organic acids were used to evaluate ghrelin gene expression levels, gut microflora composition, performance parameters and intestinal histomorphological changes in broiler chickens. One-day-old chicks (n = 315) were reared for 42 days and distributed randomly into five experimental groups; each with three replicates (21 birds per replicate). Experimental groups were control: basal diet only, antimicrobial growth promoter: 40 g/metric ton of basal diet (virginiamycin), organic acids: 4 kg/metric ton of basal diet, phytobiotics: 3 kg/metric ton of basal diet, combination: 7 kg/metric ton of basal diet (organic acids 4 kg and phytobiotics 3 kg metric ton of feed). Growth performance, histological and ghrelin gene expression analysis were executed on 21 and 42 days while, quantitative bacterial analysis of cecum and ileum was performed on day 42. Increased feed intake and body weight (p < 0.05) were noticed in phytobiotics group. Addition of phytobiotics significantly improved (p < 0.05) villus height and ratio of villus height/crypt depth in ileum, jejunum, and duodenum and down-regulated ghrelin gene expression levels. Total coliform and Escherichia coli in cecal and ileal digesta were decreased significantly (p < 0.05) in organic acids group. Correlation analysis revealed Lactobacillus spp. were positively correlated to villus height/crypt depth ration in duodenum. The findings indicated the importance of gene-nutrient-microbiota interactions based on nutrigenomics approach. Reproductive success is attained by various mechanisms in insects. Prolonged post insemination association is one such mechanism to increase the reproductive success. The present study was conducted to assess the role of post insemination association of mating partners on reproductive performance in Chrysomelidae beetle, Zygogramma bicolorata Pallister. The matings were disrupted at different time intervals and fecundity and percent egg viability of the females were recorded. In addition, the mounting attempts, mating attempts, time to commencement of mating and latent period were also recorded. It was hypothesized that: (1) the mounting and mating attempts would not exist, (2) copulation duration, would not affect the reproductive performance, and (3) the beetle would not exhibit the mate guarding behaviour. Interestingly, results revealed that 6.00 ± 1.3 and 6.59 ± 0.93 mounting and mating attempts are needed to establish successful mating. The results revealed that males improved their percent egg viability with a mating duration ranging from nearly 30–50 min. While fecundity increased with a mating duration of above 30 min and up to a duration of 60 min. This result concluded that males of this beetle display post copulatory mate guarding behaviour after 60 min in which male rides on female's back with his aedeagus inserted in the female genital tract. Researchers worldwide with great endeavor searching and repurpose drugs might be potentially useful in fighting newly emerged coronavirus. These drugs show inhibition but also show side effects and complications too. On December 27, 2020, 80,926,235 cases have been reported worldwide. Specifically, in Pakistan, 471,335 has been reported with inconsiderable deaths. Identification of COVID-19 drugs pathway through drug-gene and gene−gene interaction to find out the most important genes involved in the pathway to deal with the actual cause of side effects beyond the beneficent effects of the drugs. The medicines used to treat COVID-19 are retrieved from the Drug Bank. The drug-gene interaction was performed using the Drug Gene Interaction Database to check the relation between the genes and the drugs. The networks of genes are developed by Gene MANIA, while Cytoscape is used to check the active functional association of the targeted gene. The developed systems cross-validated using the EnrichNet tool and identify drug genes' concerned pathways using Reactome and STRING. Five drugs Azithromycin, Bevacizumab, CQ, HCQ, and Lopinavir, are retrieved. The drug-gene interaction shows several genes that are targeted by the drug. Gene MANIA interaction network shows the functional association of the genes like co-expression, physical interaction, predicted, genetic interaction, co-localization, and shared protein domains. Our study suggests the pathways for each drug in which targeted genes and medicines play a crucial role, which will help experts in-vitr o overcome and deal with the side effects of these drugs, as we find out the in-silico gene analysis for the COVID-19 drugs. The present work aims to synthesize zinc oxide (ZnO) nanoparticles via green approaches using leaf extract of Parthenium hysterophorus. UV–vis and FT-IR tests confirmed the existence of biomolecules, active materials, and metal oxides. The X-ray diffraction structural study exposes the ZnO nanoparticles formation with hexagonal phase structures. SEM and TEM analysis reveal surface morphologies of ZnO nanoparticles and most of them are spherical with a size range of 10 nm. ZnO nanoparticles were revealed strong antimicrobial activity against both bacterial and fungal strains. The germination of seeds and vegetative growth of Sesamum indicum has been greatly improved. Pollution is severely threatening the wetland habitats. Heavy metals are one among of the major pollutants in wetland habitats. The cadmium (Cd), copper (Cu), chromium (Cr), cobalt (Co), lead (Pb), mercury (Hg), nickel (Ni) and zinc (Zn), were assessed in the water, sediment, benthic species (polychaetes, mollusc and crustaceans), prawn and fishes. The assessment of heavy metals was done by using double-beam Atomic Absorption Spectrophotometer (AAS). The Hg, Cr and Co were greater in sediment, Ni and Cd were higher in polychaetes and molluscs respectively. However, the Cu and Pb greater in crabs and the Zinc was greater in fishes. The concentration of metals showed significant differences among the various sources examined (P < 0.05) except Cr (P > 0.05). The inter-correlational analysis among the metals assessed from the various sources showed that the Cr and Pb not correlated among the eight metals examined. However, the Cu and Co were correlated with Hg (r = 0.307) and (r = 0.788) respectively. The nickel was correlated with Hg (r = 0.367), Cu (r = 0.362) and Co (r = 0.432). The Zinc was correlated with the Cd (r = 0.331) and Hg (r = 0.737). However, correlation of metals among the different sources shown that the metals of polychaetes correlated with sediment r = 0.637, the metals of crabs correlated with the sediment and polychaetes r = 0.630 and r = 842 respectively, the metals of molluscs was also correlated with sediment (r = 0.636), polychaetes (r = 0.889) and crabs (r = 0.894). In addition to that the metals of prawns was correlated with the polychaetes (r = 839), crabs (r = 0.628) and molluscs (r = 0.634). The metals of fishes correlated with polychaetes (r = 0.529), crabs (r = 0.710), molluscs (r = 0.493) and prawns (r = 0.593). Indeed the multiple regression model explained that the metals of sediments influence the accumulation of metals in biotic species such as polychaetes, molluscs, crustaceans, prawns and fishes with 84% (F = 21.079; p < 0.001).The order of the heavy metals in the water, sediment and biotic species was Hg > Pb > Ni > Cr > Zn > Co > Cu > Cd. The study found that the level of heavy metals at various sources in the sanctuary is showing considerable warning and the sanctuary is required intensive assessment on various aspects of pollution since the Point Calimere Wildlife Sanctuary is supporting several species of migratory and endangered shorebirds seasonally. Chronic diabetes extensively complicates the glucose metabolism to onset and progress the complication. Concurrently, several contemporary medicines, especially organo-metallic formulations, are emerging to treat hyperglycemia. The current study aims to emphasize the gold nanoparticles (GNPs) potential for glucose metabolism regulation in Streptozotocin (STZ) induced diabetes. Quantitative real-time polymerase chain reaction (RT-PCR) was carried out to detect the mRNA expression of Glucose transporters 2 (GLUT2), Glucokinase (GK) and Glucose 6 Phosphatase (G-6-Pase). The study shows remarkable results such as the prognostic effect of GNPs in reinforcing the repression of enzyme complex G-6-Pase about 13.3-fold when compared to diabetes control. Also, molecular docking studies showed significant inhibition of G-6-Pase by the terpenoid ligands with alpha and beta amyrin from leaf extract of Couroupita guianensis. Thus the study explored the novel mechanism of G-6-Pase downregulated by GNPs intervention that majorly contributes to the regulation of circulatory glucose homeostasis during diabetes. The entomofaunal survey and its toxicity of Blumea mollis (Asteraceae) leaf aqueous extract-mediated (Bm -LAE) silver nanoparticles (AgNPs) were assessed against selected human vector mosquitoes (HVMs). A total of 1800 individuals of 29 species belongs to 7 genera were identified. Month-wise and Genus-wise abundance of HVMs larval diversity were calculated and one-way ANOVA statistically analyzed the average physico-chemical characteristics. The relationship between physicochemical characteristics and HVMs larvae in KWS was interpreted. The total larval density and container index were 23530.18 and 1961.85 examined against 10 different containers. Various spectroscopic and microscopic investigation characterized Bm -AgNPs. The Bm- AgNPs tested against HVMs larvae, the predominant LC 50 /LC 90 values of 18.17/39.56, 23.45/42.49 and 21.82/40.43 μg/mL were observed on An. subpictus Cx. vishnui and Ae. vittatus, respectively. The findings of this investigation, improperly maintained drainages, containers and unused things in study sites, are engaged to HVMs development. This will be essential for designing and implementing HVMs control. Thimerosal is ethyl mercury based compound which is being used as a preservative in vaccines since decades. Pharmaceutical products and vaccines that contain thimerosal are among the potential source of mercury exposure. Current research was intended to ascertain the reprotoxic effects of thimerosal on rat testes. Twenty-four adult male albino rats were sorted into four groups (n = 6). The first group was a control group. Rats of experimental Group 2, 3 and 4 were treated with various dosages of thimerosal (0.5, 10, 50 mg/kg) respectively. Rats were decapitated after thirty days of trial and different parameters were analyzed. Thimerosal exposure resulted in a significant decrease in antioxidant enzyme activities including catalase (CAT), peroxidase (POD), superoxide dismutase (SOD), glutathione reductase (GSR) and increased levels of thiobarbituric acid reactive substances (TBARS). Different doses of thimerosal significantly decreased (p < 0.05) the concentration of plasma testosterone, luteinizing hormone (LH) and follicle stimulating hormone (FSH). Additionally, Daily sperm production (DSP) and efficiency of daily sperm production were significantly reduced followed by thimerosal exposure. Moreover, thimerosal significantly (p < 0.05) decreased the primary spermatocytes, secondary spermatocytes, number of spermatogonia along with spermatids. Thimerosal induced adverse histopathological and morphological changes in testicular tissues such as decreased Leydig cells, diameter of seminiferous tubules, tunica albuginea height and epithelial height. On the other hand, the increase in tubular lumen and interstitial spaces was observed due to thimerosal. These outcomes indicated that thimerosal has potential reprotoxic effects in male albino rats. Zygogramma bicolorata Pallister (Coleoptera: Chrysomelidae) is a satisfactory potential candidate for controlling Parthenium hysterophorus L. The study was conducted in order to evaluate the bio-control potential of Z. bicolorata to satisfactory reduced the density of parthenium weed in natural field conditions. Therefore, beetles were released in the twelve study sites @ 12 adults and/or larvae/m2 in their respective sites on 5th, 10th, 15th and 20th day after initial field releases. Observations were made on defoliation by the beetle , perthenium plant height, plant biomass, leaf injury rating and soil seed reservoir. Present study showed that field releases of Z. bicolorata considerably lowering down the parthenium density at all treated sites. At larval release sites by the 60th day of observation a complete defoliation was noted, however at adult release sites it took 80 days for complete defoliation. Moreover, significantly higher number of developmental stages i.e. eggs, larvae and adults were noted in the adults released sites than to larval released sites and the controls. It was also noted that plant height and its density, flowers & leaves production , biomass of parthenium plant and seed reservoir in soil was negatively affected in treated sites as compared to untreated sites. The study is to scrutinize andrographolides with Indoleamine 2,3-dioxygenase (IDO) inhibitory potential, its molecular mechanism against streptozotocin (STZ) diabetic retinopathy (DR) in Wistar rats. Oxidative stress markers such as Kynurenine metabolites, retinal histopathological changes have been studied. Further, IDO gene expression and docking studies have been performed. Andrographolide treated rats have been reducing the level of thiobarbituric acid reactive substances and protein carbonyls Kynurenine metabolites with an improvement in the level of GSH and expression of IDO as revealed by morphological changes in inner and outer nuclear layer of the retina. The current results of this study have been generated information about an activity of the andrographolide in the essential pocket of IDO. Our results explain, involving IDO and andrographolide would constitute an attempt to identify natural products with therapeutic value and further studies in this direction would be of immense significance in the administration of diabetes and its related problems. Fish kidneys are sensitive to chemical changes in the freshwater ecosystem because they are directly and constantly exposed to chemicals dissolved in the water. This study evaluated nephrotoxicity in Wallago attu and Cirrhinus mrigala harvested from the Chenab River in an area of industrial and sewage waste disposal. Induced histological alternation data were correlated to the severity of environmental degradation in order to determine whether this biological system can be used as a tool for environmental monitoring programs. Kidneys from two fish species occupying different niches were collected and stored for 24 h in 10% formalin. Control fish were collected upstream of the polluted river area. Specimens were processed using topical histological methods. The major histological alterations observed in both species were renal tubule myxospora, hyperemia, glomerulonephritis, degeneration of renal tubule cells, dilation of glomerular capillaries, presence of pycnotic nuclei in the hematopoietic tissue, epithelial hypertrophy, vacuolization, reduced lumen of renal tubules, and shrinkage of glomeruli. Renal tubular atrophy, degeneration due to extensive degranulation, necrosis of glomeruli, glomerular expansion, absence of Bowman's space, hypertrophied nucleus, necrosis and hyalinization of the interstitium, clogging of tubules, and regeneration of tubules was also observed. Wallago attu exhibited the maximum incidence of moderate to severe changes and was defined as having the highest "histopathologic alteration index". These severe alterations were found to be related to environmental degradation, indicating the presence of stressors in freshwater. Control groups showed normal tissue morphology in the kidneys. Infectious bursal disease virus (IBDV) is the causative agent of Infectious Bursal Disease (IBD), the disease causes immunosuppression which leads to secondary infections among rearing poultry flocks. Characterization of the virus is important for its control and eradication. The circulating IBDVs are classified on the basis of their antigenic and pathogenic properties. The virus is categorised as classical, variant and very virulent IBDV (vvIBDV). IBDV is a non-envelop, icosahedral double stranded virus. Viral protein 2 (VP2) is the major structural protein of capsid that determines the host-pathogen relationship. The aim of this study was to characterise the IBD virus of Pak-Asian region. IBDV suspected flocks were examined in Punjab, Pakistan from 2014–2018. Two hundred and fifty samples were collected with complete history of the disease. The suspected samples were collected from broiler, layer and rural poultry farms. RNA was extracted and hyper-variable region of VP2 gene was amplified using specific primers. Nucleotide sequence of the VP2 gene was determined and its Amino Acid sequence was deduced. Moreover, phylogenetic analysis was also performed. The current classifications based in a hyper-variable region of the capsid protein VP2 (hvVP2), classification of IBDVs is split into newly proposed geno-groups according to Jackwood group. Among these prevailing, some IBDVs are limited geographically whereas, others are reported cosmopolitan. Genetic alterations are continuously playing role in evolution of new strains of the virus. During this study it was found that isolates of IBDV fall in first three geno-groups. Abstract Introduction Nanoparticles (NPs) have become very important owing to their various uses. In this research, an environmentally friendly biological technique was used to synthesize silver nanoparticles with Coriandrum sativum L. The objective of this research to use the source for the fabrication of silver NPs from C. sativum L., and to check the activity of the fabricated silver NPs was determined versus a couple of gram negative and a couple of gram positive bacteria in the presence of antibiotic viz. gentamicin to judge their impact. Methodology A silver nitrate solution, which served as the reducing and capping agent, was mingled with coriander leaf extract. The solution's temperature and pH were maintained at 75 °C and 8.6, respectively. The observed mean particle size (z-average) and polydispersity index were 390.2 nm and 0.452, respectively. The synthesized Ag NPs were characterized using different techniques, including scanning electron microscopy (SEM), X-ray diffraction, and Fourier transmission infrared (FTIR) analysis. The globular shape of the silver nanoparticles was depicted in SEM illustrations. Results XRD data revealed the mean size of the particles was 11.9 nm. The FTIR analysis showed the existence of various functional groups, including C O and O H. When their antibacterial ability was tested, it was found that the fabricated Ag NPs inhibited Bacillus subtilis , Pasteurella multocida, Enterobacter aerogenes, and Staphylococcus aureus, with a greater effect against B. subtilis and P. multocida compared to E. aerogenes and S. aureus. Conclusion It has been concluded small silver NPs benefited from a higher surface area ratio, as shown by the results of experiments where smaller particles had a better bactericidal proficiency than large silver-based NPs. Silver-based NPs infiltrate bacterial cells, as well as interfere with their exterior membrane. In the present study, Neodymium (Nd), Cadmium (Cd), and the various molar ratios of Nd: Cd doped copper oxide nanocomposites (CuO NCs) were prepared by the co-precipitation method. The as-synthesized Nd, Cd, and Nd:Cd doped CuO NCs [Cu 1-x + y Nd x Cd y O, (x:y = 0.006:0.00, 0.00:0.006, 0.005:0.001, 0.004:0.002, 0.003:0.003, M)] were characterized through various instrumentation techniques such as TGA, UV–Vis–NIR, FTIR, Raman, FL, XRD, ZP, FE-SEM with EDAX elemental mapping, HR-TEM and XPS analyses. Further, the antibacterial activity of doped CuO NCs was tested against Staphylococcus aureus and Escherichia coli. An equal molar ratio of Nd and Cd doped CuO NCs showed excellent antibacterial activity mainly due to the synergistic effect of sufficient Nd3+, Cd2+, and Cu2+ ions releasing ability. Interestingly, the doping effect enhances surface defects and decreases the ability to scavenge free radicals compared to pure CuO nanomaterials. At the same time, the cytotoxicity of NCs was evaluated on the human lung epithelial L132 cell line. Evidently, 75 μg/ml concentration of Nd:Cd doped CuO NCs samples shows 80% viability, which confirms their negligible cytotoxic effect. The Nd:Cd doped CuO NCs (94:3:3) had a high aspect ratio shape, remarkable ion-releasing ability, and biocompatibility while being thermally stable. Because of these qualities, they are well suited for treating bacterial infections in the biomedical area. • Nd and Cd doped CuO NCs was successfully synthesized by co-precipitation method. • The equal molar ratio of Nd and Cd excellently substituted the CuO matrix and extends growth direction [0 1 0] plane. • CuO NCs showed ROS-mediated antibacterial activity in S. aureus and E. coli via releasing Nd3+ and Cd2+ ions. • Cytocompatibility of CuO NCs against L132 cell line opens-up the possibilities for further pre-clinical investigations.
