Environmental factors significantly affected the succession of microbial communities during the fermentation of Huangjiu Raw wheat Qu (RWQ). Influence of environmental factors on microbial community during fermentation of Chinese Huangjiu RWQ. The interaction of environmental factors and microorganisms affects the changes of microbial communities and key enzyme transcript levels in different stages during the production of Huangjiu RWQ. This study combined environmental factor analysis and high-throughput sequencing technology to comprehensively analyze the specific changes of microbial communities and environmental factors at each stage of Huangjiu production. In addition, the influence of the interaction of environmental factors and microorganisms on microbial succession was investigated by correlation analysis, and the enzymes related to saccharification and ethanol fermentation during the production of RWQ were explored. The results showed that Bacillus , Weisseria , Thermophilic actinomycetes and Lactobacillus were the dominant bacteria in RWQ, while Aspergillus , Thermomyces , Fusarium and Alternaria were the dominant fungi. Fungi were active in the early stage of RWQ fermentation, while bacteria were active in the middle and late stage of fermentation. Microbial community succession was closely related to environmental humidity and CO 2. These environmental factors were positively correlated with most fungi, but negatively correlated with Thermophilic actinomycetes, Glycopolyspora and Acinetobacter. In fungi, the enzymes related to saccharification and fermentation mainly came from Thermophilic ascomycetes and thermophilic fungi , while in Bacteria , these two enzymes mainly came from weisseria. Pydiflumetofen (PYD), a highly effective and broad-spectrum fungicide, is commonly employed for the control of fungal diseases. In this study, the uptake, translocation, and biotransformation of PYD by wheat (Triticum aestivum L.) were firstly investigated at a chiral level. The findings revealed that the residue concentration of R -PYD in wheat was higher than that of S -PYD, because of its higher uptake rate (k 1 = 0.0421 h−1) and lower elimination rate (k2 = 0.0459 h−1). Additionally, R -PYD exhibited higher root bioconcentration factors and translocation factors compared with S- enantiomer, indicating R -PYD was more easily accumulating in roots and translocating to shoots. Furthermore, a total of 9 metabolites, including hydroxylated, demethylated, demethoxylated, dechlorinated, hydrolyzed, and glycosylated-conjugated products, were detected qualitatively in wheat roots or shoots. Symplastic pathway-mediated uptake, which predominantly relied on aquaporins and anion channels, was confirmed by root adsorption and inhibition experiments, without displaying any enantioselective effect. Molecular simulations demonstrated that R -PYD exhibited stronger binding affinity with Ta LTP 1.1 with a lower grid score (−6.79 kcal/mol), whereas weaker interaction with the metabolic enzyme (CYP71C6v1) compared to the S- enantiomer. These findings highlight the significance of plant biomacromolecules in the enantioselective bioaccumulation and biotransformation processes. Importantly, a combination of experimental and theoretical evidence provide a comprehensive understanding of the fate of chiral pesticides in plants from an enantioselective perspective. Perfluorooctane sulfonamide (PFOSA) is a precursor of perfluorooctane sulfonic acid (PFOS) and can be broken down to PFOS in environment and biota. In the present work, PFOSA was spiked in soil and its biodegradation in soil, uptake and metabolism in wheat ( Triticum aestivum L.) and earthworms ( Eisenia fetida ) were investigated. The results indicated that PFOSA could be biodegraded to highly stable PFOS, which has the same perfluorinated carbon chain length as PFOSA, by microbes in soil. PFOSA could be taken up by wheat root and earthworm from soil with higher bioaccumulation ability than PFOS. In both wheat and earthworms, PFOSA also degraded to PFOS. However, other shorter-chain perfluoroalkane sulfonates (PFSAs), including perfluorohexane sulfonate (PFHxS) and perfluorobutane sulfonate (PFBS), were observed in wheat, but not in soil and earthworms, suggesting that wheat displayed distinctly different degradation mechanisms to PFOSA from soil microbes and earthworms. Wheat chaff was prepared for the enzymatic conversion by ultrasound, alkali or hydrothermal pretreatment while obtained waste liquid fractions were evaluated for their antioxidant activity. Waste streams were assayed by standard antioxidant methods to evaluate their potential as sources of valuable products. Results showed that all waste streams expressed high antioxidative activities with sample after alkali pretreatment being even superior in comparison to standard antioxidants. The most of the antioxidative activities of assayed waste fractions showed strong correlation with concentration of reducing sugars, which can be attributed to the identified presence of the xylooligosaccharides. Results indicated potential use of waste streams from investigated pretreatments for co-production of valuable products with high antioxidative capacities. [ABSTRACT FROM AUTHOR]

Abstract: On sites where C4-plants have replaced C3-plants, changes in soil δ13C allow the turnover of C3- and C4-derived C to be separated. Studies of decadal scale turnover of soil C following conversion to C4-plants generally lack δ13C values for previous C4-residue inputs and assume that estimates of C4-derived soil C to be based on a fixed δ13C value. Further assumptions are that changes in the initial (time-zero) soil δ13C values are insignificant following conversion to C4-plants. We tested these assumptions by measuring: 1) the δ13C of annual samples of silage maize biomass (C4-plant) and winter wheat grains (C3-plant) grown during 1988 to 2006, and 2) the δ13C of soil kept under bare fallow during 1956 to 1983. The δ13C of plants was related to climate variables, and the impact of maize δ13C was based on estimates of maize-derived soil C using different approaches to establish the δ13C in maize inputs. The δ13C of both maize and wheat decreased with time, but the rate of change and annual variations were considerably larger for wheat than for maize. Maize as well as wheat δ13C was best related to year (probably reflecting a decrease in atmospheric δ13C) and the water balance during the active growth period. Using the smallest (−12.44‰) and the largest (−11.26‰) δ13C measured during 1988 and 2006, estimates of maize-derived C in soil after 18 years ranged from 13.2% to 14.2% of the soil total C. Despite a loss of 31% of the soil C pool under bare fallow, the increase in soil δ13C was significant only at P < 0.10. We conclude that annual variations in maize δ13C values and changes in the δ13C of the soil C fraction derived from the pre-conversion C3-vegetation have only little impact on estimates of maize-derived soil C that cover a few decades. For estimates covering several decades to centuries, the subtle but consistent changes in plant and soil δ13C need to be accounted for. The variability in δ13C in wheat grains suggest that the use of any fixed δ13C value for C3-residues in estimates of C turnover in soils on which C4-plants have been replaced by C3-plants can be associated with considerable uncertainty. Abstract: This study determined hydrogen production, volatile fatty acids (VFAs) generation and cellulose solubilisation from anaerobic dark fermentation of wheat stalk and showed the effect of different mixed microflora. The cumulative hydrogen yields of anaerobic digested activated sludge (AS)-inoculated and anaerobic digested dairy manure (DM)-inoculated system were 23.3 and 37.0mL/g VS at 204h, respectively. A modified Gompertz equation was able to adequately describe the production of hydrogen from the batch fermentation by both mixed microflora. During the process, acetate and butyrate accounted for more than 76.1% of total VFAs for both fermentations. The extent of cellulose solubilisation approached 46.6% and 75.2% for AS- and DM-inoculated fermentation, respectively. The X-ray diffraction (XRD) showed that the crystallinities of both fermented stalks were partly disrupted by the mixed microflora, and DM-inoculated fermentation had more disruption than AS-inoculated one. Abstract: Restructuring the current fermentation and recovery practices employed for the production of polyhydroxyalkanoates is essential for the commercialisation of environmentally benign and cost competitive biodegradable plastics. This study presents the potential of a wheat-based biorefinery for the production of poly(3-hydroxybutyrate) (PHB). Fed-batch bioconversions using Wautersia eutropha growing on wheat-derived media led to the production of 162.8g/l PHB. A high PHB to total dry weight (TDW) yield of 93% (w/w) was achieved due to microbial autolysis at the end of fermentation. Images of bacterial cells taken with a Transmission Electron Microscope (TEM) indicated the potential of bacterial autolysis as a mean to shorten downstream processing for PHB purification. The consumption of amino acids and peptides derived from wheat gluten hydrolysis resulted in a high glucose to PHB conversion yield of 0.47g/g. The respective yield regarding the amount of wheat used for the production of enzymes and PHB was around 0.3gPHB/g wheat, which corresponds to 82.8% of the maximum theoretical conversion yield. The productivity achieved was around 0.9g/lh. Fermentations carried out on wheat-derived media and media formulated with various commercial sources of nutrients (glucose, yeast extract, soy-protein acid hydrolysate, casein hydrolysates, corn steep liquor and various inorganic chemicals) showed that the proposed wheat-based biorefinery strategy enhanced PHB production. Abstract: The fine specialty chemicals production from agro-industrial by-products such as cereal bran is a topic of high interest in wheat-producing countries. In this work, the possibility of obtaining vanillin from bioconversion of ferulic acid derived from enzymatic hydrolysis of wheat bran was explored. Freely available ferulic acid was first obtained through wheat bran hydrolysis by using commercially available, technical-grade enzyme preparations. Then, resting cells of an engineered E. coli strain, JM109(pBB1), containing the genes responsible for conversion of ferulic acid into vanillin, were used to transform ferulic acid available in the hydrolyzed mixture into vanillin. A vanillin molar yield of 50% was obtained, but most of the vanillin produced was reduced to vanillyl alcohol. The purification of the hydrolyzate with a ionic exchange resin to remove most of the carbohydrates allowed to obtain both higher bioconversion yields (up to 70%) and lower vanillin reduction. This is the first work in which vanillin is produced from ferulic acid obtained through wheat bran hydrolysis. The results obtained demonstrate the potential application of the proposed strategy in vanillin biosynthesis from agro-industrial by-products. [Copyright &y& Elsevier]

Hexachlorocyclohexane isomers (HCHs) are persistent organic pollutants being responsible for environmental contamination worldwide. In order to characterize transformation of HCHs in different plant compartments during uptake, a hydroponic experimental setup was designed using wheat as the test plant. The extent of transformation was determined by using compound-specific isotope analysis (CSIA) and enantiomer fraction (EF) analysis. In nutrient solutions, no change of carbon (δ13C) and chlorine isotope ratios (δ37Cl) of α-HCH and β-HCH was detected throughout the experiment indicating no transformation there. In wheat leaves, stems and roots, however, transformation of α-HCH due to a C‒Cl bond cleavage was indicated by increasing δ13C and δ37Cl compared to the nutrient solution. In addition, 1,3,4,5,6-pentachlorocyclohexene (PCCH) was identified as the major metabolite of α-HCH transformation. For β-HCH, in contrast, no transformation was detected. The evaluation of enantiomer fraction analysis revealed no change of the EF(-) in the nutrient solution or on root surface but a decrease in the wheat compartments, providing an evidence for the preferential biological transformation of (-)α-HCH in wheat.
