To investigate the effect of the 8th American Joint Committee on Cancer (AJCC) pathological prognostic staging on chemotherapy decision-making for triple-negative breast cancer (TNBC) patients with T1-2N0M0 disease. Patients diagnosed with T1-2N0M0 TNBC were retrieved from the Surveillance, Epidemiology, and End Results program. Statistical methods including Kaplan-Meier survival curve, receiver operating characteristics curve, and Cox proportional hazard model. We identified 12,156 patients, including 9371 (77.1%) patients who received chemotherapy. Overall, 57.4% of patients (n = 6975) were upstaged after being reassigned by the 8th AJCC staging. However, the 8th staging of AJCC did not have a greater prognostic value compared to the 7th staging (P = 0.064). The receipt of chemotherapy significantly improved the breast cancer-specific survival for stage T1c and T2 tumors (P < 0.001), but not for stage T1a (P = 0.188) and T1b (P = 0.376) tumors. Using AJCC 8th staging, chemotherapy benefit was only found in stage IIA patients (P = 0.002), but not for stage IA (P = 0.653) and IB (P = 0.492) patients. There were 9564 patients with stage T1c and T2 diseases and 4979 patients with 8th AJCC stage IIA disease. Therefore, approximately half of patients (47.9%, n = 4585) may be safe to omit chemotherapy using the AJCC 8th staging compared to the current chemotherapy recommendation for T1-2N0M0 TNBC. The 8th AJCC staging system did not demonstrate the superior discriminatory ability of prognostic stratification than the 7th AJCC staging system in T1-2N0M0 TNBC. The incidence of critical leptospirosis manifested as massive pulmonary hemorrhage has been significantly reduced, which has been rarely reported in recent years, while the mortality rate is extremely high once it occurs. A 54-year-old man with no HIV infection was admitted to the local county hospital due to high-grade continuous fever lasting four days (38.5–40.5C), upper limb and shoulder-back muscle pain, and general fatigue. The chest CT (Aug 26, 2021) showed "multiple patchy, cloudy, and fuzzy shadows in both lungs, mainly under the pleura of the upper and lower lobes of both lungs; some lymph nodes in the mediastinum are enlarged". Despite being diagnosed with "common community-acquired pneumonia" and starting injectable levofloxacin, the symptoms worsened, and massive hemoptysis occurred. However, after being transferred to our hospital, the patient was diagnosed with the "pulmonary hemorrhage type of leptospirosis" through comprehensive dynamic analysis. The patient recovered very well after undergoing "penicillin 3MIU q6h" alone for two weeks to fight the infection. Leptospirosis has a high mortality rate when it becomes critical or severe. Diagnosis typically relies on factors such as epidemiology, clinical symptoms, and pathogenetic testing. Metagenomic next-generation sequencing (mNGS) is more effective in sensitivity and speed than traditional detection methods, making it an excellent option for diagnosing challenging and severe infections in emergencies. • We propose a generalized radial basis function interpolant with difference constraints. • Various types of constraints are derived to satisfy the interpolation conditions. • An adaptive iterative algorithm is used for approximating gradient and tangent constraints. • The numerical results reveal the improved performance of our method. In this work, we present a new algorithm for reconstruction of implicit surfaces from a set of cloud points with normals (Hermite data), based on the generalized radial basis functions interpolant with various types of constraints. Our key contribution is a novel construction of difference constraints in the interpolant to satisfy certain linear functional data, named as the domain constraints, the difference constraints of the gradient and the difference constraints of the tangent. To avoid ambiguous constraints, we apply an iterative algorithm to determine the adaptive distance in the difference constraints. The extended interpolant can not only provide an exact interpolation to the function values, but also approximate the function's derivatives by constructing a difference operator along the gradient or tangent direction. We interpolate the sampling points using our method by constructing a signed distance field in the geometry domain. Compared to the Hermite–Birkhoff interpolation with RBFs, the interpolant we use has faster solution efficiency. Microbe-mediated arsenic (As) metabolism plays a critical role in global As cycle, and As metabolism involves different types of genes encoding proteins facilitating its biotransformation and transportation processes. Here, we used metagenomic analysis based on high-throughput sequencing and constructed As metabolism protein databases to analyze As metabolism genes in five paddy soils with low-As contents. The results showed that highly diverse As metabolism genes were present in these paddy soils, with varied abundances and distribution for different types and subtypes of these genes. Arsenate reduction genes ( ars ) dominated in all soil samples, and significant correlation existed between the abundance of arr (arsenate respiration), aio (arsenite oxidation), and arsM (arsenite methylation) genes, indicating the co-existence and close-relation of different As resistance systems of microbes in wetland environments similar to these paddy soils after long-term evolution. Among all soil parameters, pH was an important factor controlling the distribution of As metabolism gene in five paddy soils ( p = 0.018). To the best of our knowledge, this is the first study using high-throughput sequencing and metagenomics approach in characterizing As metabolism genes in the five paddy soil, showing their great potential in As biotransformation, and therefore in mitigating arsenic risk to humans. Solar cells that convert sunlight into electrical energy are the main component of a solar power system. Quality inspection of solar cells ensures high energy conversion efficiency of the product. The surface of a multi-crystal solar wafer shows multiple crystal grains of random shapes and sizes. It creates an inhomogeneous texture in the surface, and makes the defect inspection task extremely difficult. This paper proposes an automatic defect detection scheme based on Haar-like feature extraction and a new clustering technique. Only defect-free images are used as training samples. In the training process, a binary-tree clustering method is proposed to partition defect-free samples that involve tens of groups. A uniformity measure based on principal component analysis is evaluated for each cluster. In each partition level, the current cluster with the worst uniformity of inter-sample distances is separated into two new clusters using the Fuzzy C-means. In the inspection process, the distance from a test data point to each individual cluster centroid is computed to measure the evidence of a defect. Experimental results have shown that the proposed method is effective and efficient to detect various defects in solar cells. Quercetin and quercetin glucosides are the major flavonols present in onion ( Allium cepa L . ) and are predominantly present as quercetin, quercetin-3,4′-diglucoside and quercetin-4′-glucoside. Effect of different light wavelengths on onion after harvest and storage, with fluorescent, blue, red and ultra violet light influenced the quercetin and quercetin glucosides profile. In a peeled onion, all the light treatments elevated quercetin content in bulb. Among them, particularly fluorescent light effect was more eminent which stimulates the maximum synthesis of quercetin in onion. In case of whole onion bulb, skin and pulp showed different responses to light treatment, respectively. The pulp had the highest quercetin glucosides under blue light, whereas the lowest under fluorescent light. Onion skin showed nearly opposite pattern as compared to the pulp. In particular, light treatment proved to be a better way to increase the level of quercetin content in onions which might be utilized for industrial production of bioactive compounds from onion and onion waste products. Abstract: Experimental and theoretical investigations of water-side fouling have been performed inside four corrugated plate heat exchangers. They have different geometric parameters, such as plate height, plate spacing, and plate angle. Fouling experiments including particulate fouling tests and composite particulate and precipitation fouling tests have been performed. The tests are primarily focused on the effects of concentration and average velocity. Scanning electron microscope (SEM) was used to investigate the microscopic structures of composite fouling. Heat transfer coefficients and friction factors have been obtained in clean tests. The plate heat exchanger with the largest d e and height to pitch ratio shows the best anti-fouling performance. The Von-Karman analogy was used to obtain mass transfer coefficient to develop appropriate semi-theoretical fouling correlations; The wall shear stress is obtained from the pressure drop. Abstract: An experimental investigation was performed for single-phase flow and condensation characteristics inside five micro-fin tubes with the same outer diameter 5mm and helix angle 18°. Data are for mass fluxes ranging from about 200 to 650kg/m2 s. The nominal saturation temperature is 320K, with inlet and outlet qualities of 0.8 and 0.1, respectively. The results suggest that Tube 4 has the highest condensation heat transfer coefficient and also the highest condensation pressure drop penalty, while Tube 5 has the highest enhancement ratio due to its lowest pressure drop penalty and intermediate heat transfer coefficient. Condensation heat transfer coefficient flattens out gradually as G decreases when G <400kg/(m2 s) for Tube 2 and Tube 4. This nonlinear mass-flux effect may be explained by the complex interactions between micro-fins and fluid, including liquid drainage by surface tension and interfacial turbulence. In addition, the experimental data was analyzed using seven existing pressure-drop correlations and four heat-transfer models to verify their respective accuracies. [Copyright &y& Elsevier]

Objective: To explore Effects of marine collagen peptides (MCPs) on markers of metablic nuclear receptors, i.e peroxisome proliferator-activated receptor (PPARs), liver X receptor (LXRs) and farnesoid X receptor (FXRs) in type 2 diabetic patients with/without hypertension. Method: Study population consisted of 200 type 2 diabetic patients with/without hypertension and 50 healthy subjects, all of whom were randomly assigned to MCPs-treated diabetics (n=50), placebo-treated diabetics (n=50), MCPs-treated diabetics with hypertension (n=50), placebo-treated diabetics with hypertension (n=50), and healthy controls (n=50). MCPs or placebo (water-soluble starch) were given daily before breakfast and bedtime over three months. Levels of free fatty acid, cytochrome P450, leptin, resistin, adiponectin, bradykinin, NO, and Prostacyclin were determined before intervention, and 1.5 months, and 3 months after intervention. Hypoglycemia and the endpoint events during the study were recorded and compared among the study groups. Result: At the end of the study period, MCPs-treated patients showed marked improvement compared with patients receiving placebo. The protection exerted by MCPs seemed more profound in diabetics than in diabetics with hypertension. In particular, after MCPs intervention, levels of free fatty acid, hs-CRP, resistin, Prostacyclin decreased significantly in diabetics and tended to decrease in diabetic and hypertensive patients whereas levels of cytochrome P450, leptin, NO tended to decrease in diabetics with/without hypertension. Meanwhile, levels of adiponectin and bradykinin rose markedly in diabetics following MCPs administration. Conclusion: MCPs could offer protection against diabetes and hypertension by affecting levels of molecules involved in diabetic and hypertensive pathogenesis. Regulation on metabolic nuclear receptors by MCPs may be the possible underlying mechanism for its observed effects in the study. Abstract: This study focuses on the thermodynamic analysis of saturated vapor flowing slowly onto and condensing on an elliptical tube with variable wall temperature. An entropy generation minimization (EGM), technique is applied as a unique measure to study the thermodynamic losses caused by heat transfer and film-flow friction for the laminar film condensation on a non-isothermal horizontal elliptical tube. The results provide us how the geometric parameter ellipticity and the amplitude of non-isothermal wall temperature variation affect entropy generation during filmwise condensation heat-transfer process. The optimal design can be achieved by analyzing entropy generation in film condensation on a horizontal elliptical tube with further account for the amplitude of non-isothermal wall temperature variation. Abstract: This paper aims to perform thermodynamic analysis of saturated vapor flowing slowly onto and condensing on an elliptical cylinder. This analysis provides us how the geometric parameter-ellipticity affects entropy generation during film-wise condensation heat transfer process. The results observe that local condensate film thickness decreases with an increase in ellipticity of a cylinder. From the first law point of view, the local heat transfer coefficient enhances as ellipticity increases. Meanwhile, from the second law point of view, entropy generation increases with increasing the value of ellipticity. We derive an expression for entropy generation, which accounts for the combined action of the specified irreversibilities. The result demonstrates that thermal irreversibility dominates over film flow friction irreversibility. Finally, an expression of minimizing entropy generation in laminar film condensation heat transfer is obtained. [Copyright &y& Elsevier]

Muscle tissue engineering is a promising therapeutic strategy for volumetric muscle loss (VML). Among them, decellularized extracellular matrix (dECM) biological scaffolds have shown certain effects in restoring muscle function. However, researchers have inconsistent or even contradictory results on whether dECM biological scaffolds can efficiently regenerate muscle fibers and restore muscle function. This suggests that therapeutic strategies based on dECM biological scaffolds need to be further optimized and developed. In this study, we used a recellularization method of perfusing adipose-derived stem cells (ASCs) and L6 into adipose dECM (adECM) through vascular pedicles. On one hand, this strategy ensures sufficient quantity and uniform distribution of seeded cells inside scaffold. On the other hand, auxiliary L6 cells addresses the issue of low myogenic differentiation efficiency of ASCs. Subsequently, the treatment of VML animal experiments showed that the combined recellularization strategy can improve muscle regeneration and angiogenesis than the single ASCs recellularization strategy, and the TA of former had greater muscle contraction strength. Further single-nucleus RNA sequencing (snRNA-seq) analysis found that L6 cells induced ASCs transform into a new subpopulation of cells highly expressing Mki67 , CD34 and CDK1 genes, which had stronger ability of oriented myogenic differentiation. This study demonstrates that co-seeding ASCs and L6 cells through vascular pedicles is a promising recellularization strategy for adECM biological scaffolds, and the engineered muscle tissue constructed based on this has significant therapeutic effects on VML. Overall, this study provides a new paradigm for optimizing and developing dECM-based therapeutic strategies. Study on the therapeutic effect and mechanism of vascularized adECM biological scaffolds co-recellularized by ASCs and L6 cells (myoblasts) for VML treatment. Seeding ASCs and L6 cells into biological scaffolds via vascular pedicles ensures uniform cellular distribution within the scaffold and promotes functional muscle fibers regeneration. Meanwhile, under the assistance of L6 cells, ASCs in adECM biological scaffolds transform into a new subpopulation with high expression of Mki67 , CD34 and CDK1 genes. P2X receptors are a class of nonselective cation channels widely distributed in the immune and nervous systems, and their dysfunction is a significant cause of tumors, inflammation, leukemia, and immune diseases. P2X7 is a unique member of the P2X receptor family with many properties that differ from other subtypes in terms of primary sequence, the architecture of N- and C-terminals, and channel function. Here, we suggest that the observed lengthened β2- and β3-sheets and their linker (loop β2,3), encoded by redundant sequences, play an indispensable role in the activation of the P2X7 receptor. We show that deletion of this longer structural element leads to the loss of P2X7 function. Furthermore, by combining mutagenesis, chimera construction, surface expression, and protein stability analysis, we found that the deletion of the longer β2,3-loop affects P2X7 surface expression but, more importantly, that this loop affects channel gating of P2X7. We propose that the longer β2,3-sheets may have a negative regulatory effect on a loop on the head domain and on the structural element formed by E171 and its surrounding regions. Abstract: This paper provides a numerical and experimental analysis on precipitation and particulate fouling in corrugated plate heat exchangers with different geometric parameters which are plate height, plate spacing, and plate angle. The Realizable κ–ε model with non-equilibrium wall functions is used in the 3D numerical simulation considering the realistic geometries of the flow channel to obtained Nusselt number and wall shear stress, while Von-Karman analogy is used to obtain mass transfer coefficient. Numerical analysis is verified by experimental study. The predicted influence of fluid velocity in fouling resistance is compatible with experimental data that it can help to optimize the design of plate heat exchangers. This investigation significantly simplifies the fouling analysis of complex flow fields and can be used to assess the fouling potential of corrugated plate heat exchangers. [Copyright &y& Elsevier]

Abstract The mixing of land uses lies at the core of understanding their characteristics in cities. Some existing land use mix measures have been used to quantify the mix degree from different perspectives. However, these existing indices mainly cover only two dimensions of the land use mix concept, namely, distance and quantity, and cannot effectively delineate the picture of land use mix. Thus, we propose a new quantification framework of land use mix by involving a third dimension, that is, the "attribute" level of the land use mix concept. On the basis of this framework, we develop a novel measure called (weighted) vector-based mix degree index (VMDI/WVMDI) to address the compatibility mix degree by incorporating the compatibility relations between different land uses. This novel measure adopts vector land use data and compatibility relations. To some extent, VMDI/WVMDI can overcome some of the deficiencies of existing land use mix measures. We select Guangzhou as a study area in evaluating the proposed measure. In addition, the "head/tail breaks" classification scheme is employed for results with heavy-tailed distribution. By using this scheme, we clearly identify several clustered regions of land use mix in Guangzhou, which might be useful for urban planners and policy makers. Compared with some traditional measures, VMDI/WVMDI can be more efficient in depicting the compatibility relation between land use parcels. Abstract It is widely known that transcriptional diversity contributes greatly to biological regulation in eukaryotes. With the development of next-generation sequencing (NGS) technologies, several studies on RNA sequencing have considerably improved our understanding of transcriptome complexity. However, obtaining full-length (FL) transcripts remains a considerable challenge because of difficulties in short read-based assembly. In the present study, single-molecule real-time (SMRT) sequencing and NGS were combined to generate the complete and FL transcriptome of Manis javanica. The results provide a comprehensive set of reference transcripts and hence contribute to the improved annotation of the M. javanica genome. We obtained 45,530 high-confidence transcripts from 19,109 genic loci, of which 8014 genes have not yet been annotated within the M. javanica genome. Furthermore, we revealed 8824 long-chain noncoding RNAs (lncRNAs). A total of 30,199 alternative splicing (AS) and 11,184 alternative polyadenylation (APA) events were identified in the sequencing data. The structure and expression level of 59 digestive enzyme genes, including 13 carbohydrase genes, 28 lipase genes and 18 protease genes, were analyzed, which might provide original data for further research on M. javanica. Newly assembled dengue viruses (DENV) undergo maturation to become infectious particles. The maturation process involves major rearrangement of virus surface premembrane (prM) and envelope (E) proteins. The prM-E complexes on immature viruses are first assembled as trimeric spikes in the neutral pH environment of the endoplasmic reticulum. When the virus is transported to the low pH environment of the exosomes, these spikes rearrange into dimeric structures, which lie parallel to the virus lipid envelope. The proteins involved in driving this process are unknown. Previous cryoelectron microscopy studies of the mature DENV showed that the prM-stem region (residues 111-131) is membrane-associated and may interact with the E proteins. Here we investigated the prM-stem region in modulating the virus maturation process. The binding of the prM-stem region to the E protein was shown to increase significantly at low pH compared with neutral pH in ELISAs and surface plasmon resonance studies. In addition, the affinity of the prM-stem region for the liposome, as measured by fluorescence correlation spectroscopy, was also increased when pH is lowered. These results suggest that the prM-stem region forms a tight association with the virus membrane and attracts the associated E protein in the low pH environment of exosomes. • Recent understanding of ClpX had been summarized in this review. • A single molecular system had been designed to study the mechanical changes of ClpX. • ClpX mutants could be constructed to mimick their state of ATP-binding and ATP-free. • Experimental results would elucidate the mechanism of power stroke of ClpX. ClpXP in Escherichia coli is a proteasome degrading protein substrates. It consists of one hexamer of ATPase (ClpX) and two heptamers of peptidase (ClpP). The ClpX binds ATP and translocates the substrate protein into the ClpP chamber by binding and hydrolysis of ATP. At single molecular level, ClpX harnesses cycles of power stroke (dwell and burst) to unfold the substrates, then releases the ADP and Pi. Based on the construction and function of ClpXP, especially the recent progress on how ClpX unfold protein substrates, in this mini-review, a currently proposed single ClpX molecular model system detected by optical tweezers, and its prospective for the elucidation of the mechanism of force generation of ClpX in its power stroke and the subunit interaction with each other, were discussed in detail.
