Since the first discovery of grapevine brown leaf spot disease in Turpan, Xinjiang, China, in 2007, it has been a common occurrence in grapevine-growing regions. Grapevine brown leaf spot seriously decreases fruit yield and has become one of the most important leaf diseases in the Turpan region. However, thus far, there have been no reports on the evaluation of grapevine germplasm resources for resistance to brown leaf spot disease. In addition, the response mechanism of grapevine leaves to brown leaf spot infection has not been revealed. To better understand the resistance of grapevine germplasm resources to brown leaf spot disease, we first evaluated resistance in grapevine varieties. On this basis, the most susceptible variety ‘Sultanina’ was selected as the experimental material for this study. Transcriptome analysis and carbohydrate content analysis were performed on ‘Sultanina’ leaves with different levels of disease susceptibility. As the severity of the disease increased, the content of fructose gradually decreased, while the content of trehalose gradually increased. Transcriptome data revealed that differentially expressed genes were enriched in the pentose and glucuronate interchange pathway. These results suggest that the sugar trehalose may play an important role in the response of ‘Sultanina’ leaves to brown leaf spot infection. In addition, the pentose and glucuronate interchange pathway may be involved in the response mechanism of brown leaf spot disease. Our work provides new insights into the mechanism of the ‘Sultanina’ leaf response to brown leaf spot infection.

Insect hormones, such as juvenile hormone (JH), precisely regulate insect life-history traits. The regulation of JH is tightly associated with the tolerance or resistance to Bacillus thuringiensis (Bt). JH esterase (JHE) is a primary JH-specific metabolic enzyme which plays a key role in regulating JH titer. Here, we characterized a JHE gene from Plutella xylostella (PxJHE), and found it was differentially expressed in the Bt Cry1Ac resistant and susceptible strains. Suppression of PxJHE expression with RNAi increased the tolerance of P. xylostella to Cry1Ac protoxin. To investigate the regulatory mechanism of PxJHE, two target site prediction algorithms were applied to predict the putative miRNAs targeting PxJHE, and the resulting putative miRNAs were subsequently verified for their function targeting PxJHE using luciferase reporter assay and RNA immunoprecipitation. MiR-108 or miR-234 agomir delivery dramatically reduced PxJHE expression in vivo, whilst only miR-108 overexpression consequently increased the tolerance of P. xylostella larvae to Cry1Ac protoxin. By contrast, reduction of miR-108 or miR-234 dramatically increased PxJHE expression, accompanied by the decreased tolerance to Cry1Ac protoxin. Furthermore, injection of miR-108 or miR-234 led to developmental defects in P. xylostella, whilst injection of antagomir did not cause any obvious abnormal phenotypes. Our results indicated that miR-108 or miR-234 can be applied as potential molecular targets to combat P. xylostella and perhaps other lepidopteran pests, providing novel insights into miRNA-based integrated pest management.

This work describes the successful design and synthesis of a new fluorescence resonance energy transfer (FRET)-based sensor, denoted as RD1. This sensor incorporates a robust dual-fluorophore design, which combines a rhodamine and a dansyl derivative, functionalized with a thiosemicarbazide group that acts as Hg(II) specific recognition site. A synthetic pathway was developed that allowed the efficient synthesis of RD1 with a remarkable overall yield of 44% over four steps, through microwave-assisted protocols. The influence of ethyl, benzyl and phenyl substituents of isothiocyanate in the preparation of the thiosemicarbazide moiety was studied, revealing a crucial dependence of the nature of the isothiocyanate in the formation of the recognition site. Owing to its characteristic ratiometric detection, RD1 exhibited remarkable robustness to external parameters such as pH and solvent composition. The sensor demonstrated a hybrid two-stage response to Hg(II), with an initial quenching of fluorescence followed by an enhancement of emission through a FRET mechanism, both stages being corroborated by DFT (density functional theory) calculations. In vitro studies demonstrated that RD1 presents excellent cytocompatibility and capacity to permeate cellular membranes and be effectively internalized by L929 cell line. Importantly, RD1 retained its sensory ability in a complex cellular environment, affirming its efficacy as a fluorescent sensor for the in vitro detection of bioaccumulated mercury species. This study investigates hypoxic processes beneath lake ice and explores the interactions between water environmental factors and dissolved oxygen (DO) during winter, aiming to systematically assess the influence of winter climate changes on lake ecosystems through environmental variables (physical, chemical, and biological characteristics of water). A complete hypoxic cycle in a high-altitude shallow lake was recorded using high-frequency monitoring technology, and based on a predetermined hypoxic threshold (DO<2 mg/L), it was divided into three phases: stable and decay phase, hypoxic phase, and recovery phase. Significant trend changes of environmental variables under different levels of DO were analyzed for each phase using the Mann-Kendall non-parametric test. On the basis of evaluating the overall trend of the time series, a GAM multi-factor optimization model integrating interactive effects was constructed to explore the response mechanism of environmental variables to hypoxic processes. The optimized model showed excellent explanatory performance with R 2 values of 0.833 for the stable and decay phase and 0.932 for the recovery phase, with AIC values of 1554.72 and 721.03, respectively. The trend test combined with model analysis indicates that: (1) the decay of DO under the ice is primarily affected by the increase in EC, BGA, and EC*Temp, (2) the phytoplankton biomass in shallow lakes during winter has a definite contribution to the occurrence of oxygen deficiency, and (3) the water body experiences rapid reoxygenation after the ice layer breaks, but the DO level is restricted by the increase in temperature. The Yesso scallop is a large and ancient molluscan group with great economic value; however, it has recently suffered severe cases of Polydora infection. Polydora parasitizes the shells of scallops, badly damaging shell structures and affecting growth and mortality. To investigate the molecular mechanism of Yesso scallops’ response to Polydora infection, proteomic profiling changes in the mantle tissues of Polydora-infected (diseased) and healthy scallops were systematically analysed by tandem mass tags (TMT) labelling technology in this study. A total of 519 differentially expressed proteins (DEPs) were identified. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed most innated immune-related functions and pathways were significantly downregulated in diseased scallops, except the phagocytosis pathway, indicating an important role of phagocytosis in response to Polydora infection. DEPs involved in the phagocytosis pathway were associated with phagocytic receptor recognition, phagosome biogenesis and pathogen degradation, and they were further verified by quantitative real-time PCR. The results elucidate the molecular components of phagocytosis in molluscs for the first time. Polydora can be encapsulated by melanization with an obvious appearance in shells; indeed, melanization-related DEPs were upregulated in diseased scallops. Inhibition of apoptosis and nervous modulation may be also involved in the response mechanism, with some highly associated proteins significantly differentially expressed. Finally, a protein–protein interaction network was constructed to provide a global view of the interaction relationships of the DEPs. The study predicts the molecular response mechanism of Yesso scallops to Polydora infection, and lays a theoretical foundation for Polydora disease control.

Micro/nano-plastics (MNPs) are emerging non-point source pollutants that have garnered increasing attention owing to their threat to ecosystems. Studies on the effects of MNPs on horticultural crops are scarce. Specifically, whether MNPs can be absorbed and transported by grapevines have not been reported. To fill this gap, we added polystyrene nanoplastics (PS-NPs, 100 nm) to a hydroponic environment and observed their distribution in grape seedlings of Thompson Seedless (TS, Vitis vinifera L.). After 15 d of exposure, plastic nanospheres were detected on the cell walls of the roots, stems, and leaves using confocal microscopy and scanning electron microscopy. This indicated that PS-NPs can also be absorbed by the root system through the epidermis-cortex interface in grapevines and transported upward along the xylem conduit. Furthermore, we analyzed the molecular response mechanisms of TS grapes to the PS-NPs. Through the measurement of relevant indicators and combined omics analysis, we found that plant hormone signal transduction, flavonoid and flavonol biosynthesis, phenylpropanoid biosynthesis, and MAPK signaling pathway biosynthesis played crucial roles in its response to PS-NPs. Spirulina platensis can secrete extracellular polymeric substances (EPS) helping to protect damage from stress environment, such as cadmium (Cd 2+ ) exposure. However, the responding mechanism of S. platensis and the secreted EPS to exposure of Cd 2+ is still unclear. This research focuses on the effects of Cd 2+ on the composition and structure of the EPS and the response mechanism of EPS secretion from S. platensis for Cd 2+ exposure. S. platensis can produce 261.37 mg·g -1 EPS when exposing to 20 mg·L -1 CdCl 2 , which was 2.5 times higher than the control group. The S. platensis EPS with and without Cd 2+ treatment presented similar and stable irregularly fibrous structure. The monosaccharides composition of EPS in Cd 2+ treated group are similar with control group but with different monosaccharides molar ratios, especially for Rha, Gal, Glc and Glc-UA. And the Cd 2+ treatment resulted in a remarkable decline of humic acid and fulvic acid content. The antioxidant ability of S. platensis EPS increased significantly when exposed to 20 mg·L -1 CdCl 2 , which could be helpful for S. platensis protecting damage from high concentration of Cd 2+ . Overuse of insecticides has accelerated the evolution of insecticide resistance and created serious environmental concerns worldwide, thus incentivizing development of alternative methods. Bacillus thuringiensis (Bt) is an insecticidal bacterium that has been developed as a biopesticide to successfully control multiple species of pests. It operates by secreting several insect toxins such as Cry1Ac. However, metabolic resistance based on ATP-binding cassette (ABC) transporters may play a crucial role in the development of metabolic resistance to Bt. Here, we characterized an ABCG gene from the agricultural pest Plutella xylostella (PxABCG3) and found that it was highly expressed in a Cry1Ac-resistant strain, up-regulated after Cry1Ac protoxin treatment. Binding miR-8510a-3p to the coding sequence (CDS) of PxABCG3 was then confirmed by a luciferase reporter assay and RNA immunoprecipitation. miR-8510a-3p agomir delivery markedly reduced PxABCG3 expression in vivo and consequently decreased the tolerance of P. xylostella to Cry1Ac, while reduction of miR-8510a-3p significantly increased PxABCG3 expression, accompanied by an increased tolerance to Cry1Ac. The combination of amendments has emerged as a potential strategy to efficiently alleviate salt stress in saline-alkali soil. However, knowledge regarding how to optimize the proportion of different amendment materials, comprehensively assess the contribution of each component, and clarify the response mechanisms of the amendment-saline-alkali soil-plant system is incomplete. Based on this, we conducted a pot experiment to evaluate the improvement effect of the combined application of different amendment materials at varying levels and the contribution of the amendment components to alleviating salt stress. Overall, T6 exhibited the most significant improvement effect on the physicochemical and biological properties of the saline-alkali soil and promoted the growth of oilseed rape, with the levels of 2.0 % phosphogypsum, 2.0 % humic acid, 0.25 % bentonite, and 0.03 % sodium carboxymethyl cellulose. Compared with the control group, the EC decreased by 1.51 % to 33.49 %, the soil salt content dropped by 11.40 % to 35.46 %, and the soil soluble Na + concentration significantly declined by 39.47 % to 63.20 %. Additionally, the soil nutrient content and soil microbial community structure were enhanced in treatment groups. Meanwhile, amendments alleviated salt stress in the oilseed rape plant by activating anti-oxidative enzymes and osmoregulatory substances such as soluble sugar and proline, thus improving their ability to remove reactive oxygen species (ROS). The anti-oxidative enzymes such as superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were significantly increased, with an increase of 10.68 % (SOD, T2) ∼207.31 % (CAT, T6) compared to the control group. The structural equation modeling (SEM) analysis and simulation experiments indicated that the amendment components synergically promoted the amelioration effect on salt stress, and effectively improved soil properties, which affected the response of oilseed rape to soil environment. To date, studies on the effect of sewage disturbances on treatment facilities were based on fixed-length flow variations, which are incapable of imitating the actual dynamic flow characteristics of municipal sewage. Here, an innovative dynamic influent disturbance control system is established in this study and applied in a novel denitrifying phosphorus removal (anaerobic anoxic oxic-biological contact oxidation, AAO-BCO) system to simulate seasonal and diurnal sewage fluctuations in laboratory-scale experiments. The results showed that, under sinusoidal influent flow perturbation, the effluent pollutant content followed a relatively gentle sinusoidal trend and did not always result in desired level of pollutant removal. The ability of the system to cope with sinusoidal flow variations was susceptible to the amplitude of diurnal sewage fluctuation, while stronger tolerance capacity was observed to seasonal and momentary increase in wastewater flowrate. There was also a discrepancy in the system buffering capacity towards various pollutants removal (COD > TIN > PO 4 3- ), which may be attributed to wide fluctuations in PO 4 3- /NO 3 - and different decrease in metabolic activity of denitrifying phosphorus removal (DPR) sludge caused by extreme hydraulic retention times. One of the significant limitations of aquaculture worldwide is the prevalence of divalent copper (Cu). Crayfish (Procambarus clarkii) are economically important freshwater species adapted to a variety of environmental stimuli, including heavy metal stresses; however, large-scale transcriptomic data of the hepatopancreas of crayfish in response to Cu stress are still scarce. Here, integrated comparative transcriptome and weighted gene co-expression network analyses were initially applied to investigate gene expression profiles of the hepatopancreas of crayfish subjected to Cu stress for different periods. As a result, 4662 significant differentially expressed genes (DEGs) were identified following Cu stress. Bioinformatics analyses revealed that the "focal adhesion" pathway was one of the most significantly upregulated response pathways following Cu stress, and seven DEGs mapped to this pathway were identified as hub genes. Furthermore, the seven hub genes were examined by quantitative PCR, and each was found to have a substantial increase in transcript abundance, suggesting a critical role of the "focal adhesion" pathway in the response of crayfish to Cu stress. Triclocarban (TCC) is an emerging environmental contaminant, posing potential ecological risks. Displaying a high accumulation effect and 120-day half-life in the soil environment, the toxic effects of TCC to soil organisms have been widely reported. Previous studies have confirmed that TCC can induce the oxidative stress and changes in superoxide dismutase (SOD) and catalase (CAT) activities in earthworms, but the underlying mechanisms of oxidative stress and disorder in antioxidant enzyme activities induced by TCC have not yet been elucidated. Here, we explored the multiple response mechanisms of SOD and CAT under the regulation of oxidative stress induced by TCC. Results indicated that higher-dose (0-2.0 mg/L) TCC exposure triggered the overproduction of ROS in Eisenia foetida coelomocytes, causing oxidative damage and a decrease in cell viability that was response to ROS accumulation. The TCC-induced inhibition of intracellular SOD/CAT activity was found under the regulation of oxidative stress (SOD: 29.2 %; CAT: 18.5 %), and this effect was blunted by antioxidant melatonin. At the same time, the interaction between antioxidative enzymes and TCC driven by various forces (SOD: electrostatic interactions; CAT: van der Waals forces and hydrogen bonding) led to inhibited SOD activity (9.84 %) and enhanced CAT activity (17.5 %). Then, to elucidate the binding mode of TCC, we explored the changes in SOD and CAT structure (protein backbone and secondary structure), the microenvironment of aromatic amino acids, and aggregation behavior through multispectral techniques. Molecular docking results showed that TCC inhibited SOD activity in a substrate competitive manner and enhanced CAT activity by the stabilizing effects of TCC on the heme groups. The diamondback moth (DBM), Plutella xylostella (L.), has evolved resistance to multiple insecticides including Bacillus thuringiensis (Bt). ATP-binding cassette (ABC) transporters are a class of transmembrane protein families, involved in multiple physiological processes and pesticide resistances in insects. However, the role and regulatory mechanism of ABC transporter in mediating the response to Bt Cry1Ac toxin remain unclear. Here, we characterized a MAPK signaling pathway-enriched ABCG subfamily gene PxABCG20 from DBM, and found it was differentially expressed in the Cry1Ac-resistant and Cry1Ac-susceptible strains. RNAi knockdown of PxABCG20 increased the tolerance of DBM to Cry1Ac protoxin. To explore the regulatory mechanism of PxABCG20 expression, we predicted the potential miRNAs targeting PxABCG20 using two target prediction algorithms. Luciferase reporter assay confirmed that novel-miR-310 was able to down-regulate PxABCG20 expression in HEK293T cells. Furthermore, injection of novel-miR-310 agomir markedly inhibited PxABCG20 expression, resulting in increased tolerance to Cry1Ac protoxin in susceptible strain, while injection of novel-miR-310 antagomir markedly induced the expression of PxABCG20, leading to decreased tolerance to Cry1Ac protoxin. In this work, a toxicity monitoring microbial system (TMMS) with a nitrifying biofilm as a sensing element and cathode oxygen reduction as an electrical signal was successfully constructed for trichloroacetaldehyde (TCAL) detection. The current and nitrification rate showed a linear relationship with TCAL concentration from 0 to 100 μg/L (R 2 current  = 0.9892, R 2 nitrification  = 0.9858), indicating that the target substrate concentration can be directly obtained from an electrical signal without further sample concentration. High-throughput sequencing revealed that the TMMS was composed of autotrophic/heterotrophic nitrifying and denitrifying microorganisms. Further analysis via a symbiotic relationship network demonstrated that unclassified&#95;Comamonadaceae and unclassified&#95;Xanthobacteraceae were the core nodes for maintaining the interaction between autotropic and heterotrophic nitrifying bacteria. Kyoto Encyclopedia of Genes and Genomes analysis showed that the electron transfer process primarily relied on ferredoxin and cytochromes under TCAL stress, and the abundance of functional enzymes involved in the process of nitrification was decreased, resulting in changes in electrical signal output. Trichloroacetic acid (TCA), a toxic substance produced in the disinfection process of wastewater treatment plants, will accumulate in the receiving water. The detection of TCA in the water can achieve the purpose of early warning. However, currently there are few reports on microbial sensors used for TCA detection, and the characteristics of their microbial communities are still unclear. In this work, a toxicity monitoring microbial system (TMMS) with nitrifying biofilm as a sensing element and cathode oxygen reduction as a current signal was successfully constructed for TCA detection. The current and nitrification rate showed a linear relationship with low TCA concentration from 0 to 50 μg/L (R 2 current  = 0.9892, R 2 nitrification  = 0.9860), and high concentration range from 50 to 5000 μg/L (R 2 current  = 0.9883, R 2 nitrification  = 0.9721). High-throughput sequencing revealed that the TMMS was composed of autotrophic/heterotrophic nitrifying and denitrifying microorganisms. Further analysis via symbiotic relationship network demonstrated that Arenimonas and Hyphomicrobium were the core nodes for maintaining interaction between autotropic and heterotrophic nitrifying bacteria. Kyoto Encyclopedia of Genes and Genomes analysis showed that after adding TCA to TMMS, the carbon metabolism and the abundance of the tricarboxylic acid cycle pathway were reduced, and the activity of microorganisms was inhibited. TCA stress caused a low abundance of nitrifying and denitrifying functional enzymes, resulting in low oxygen consumption in the nitrification process, but more oxygen supply for cathode oxygen reduction. Seasonal hypoxia, as an increasingly recognized environmental issue, frequently occurred in marine ranching from northern Yellow Sea, China. Although microorganisms play an important ecological role in marine ecosystems, but little is known on the response mechanism of microbial community to seasonal hypoxia in marine ranching. A total of 132 seawater samples and 47 sediment samples were collected from the marine ranching, both in the death disaster zone of sea cucumbers and the non-disaster zone, and in different months. 16S rRNA gene high-throughput sequencing was used to explore the microbial community and its influencing factors. The results showed that the stratification in community composition and dissolved oxygen content appeared in August. The Alpha diversity in seawater was higher in summer than in winter, and significant differences in Beta diversity appeared between the death disaster zone of sea cucumbers and the non-disaster zone in sediments. In addition, environmental effects explained more of the variation in bacterial community composition in seawater as compared with spatial effects did, whereas, sedimentary bacterial communities were more closely related to spatial effects. Phenol and ammonia in wastewater pose a serious threat to ecosystems and human health. However, the currently limited studies on single bacterium simultaneously removing phenol and nitrogen pollution have not fully elucidated the relevant metabolic mechanisms. The differences in proteomic profile after supplementing with phenol and ammonia for 6 and 24 h, respectively, were evaluated to explore the metabolic characteristics and adaptive mechanism of Cupriavidus oxalaticus T2 during the simultaneous removal process of phenol and nitrogen. Results revealed that a new potential phenol para-degradation pathway appeared in T2. Phenol induced changes in nitrogen metabolism, resulting in increased denitrification and decreased synthesis of glutamate from ammonia at 6 h. In addition, phenol exposure enhanced the expression of cytochrome oxidases with high oxygen affinity and increased ATP synthesis. The increase in chemotaxis and flagellar assembly was conducive to the uptake and utilization of phenol. The synthesis of lipoic acid and biotin was also promoted to resist phenol toxicity. Moreover, phenol triggered cellular stress response, thereby leading to the upregulation of anti-stress proteins, such as universal stress protein, iron‑sulfur cluster protein, and chaperones. This study contributes to revealing the metabolic characteristics and adaptive mechanism of T2 during simultaneous nitrogen and phenol removal. SIGNIFICANCE: Phenol and ammonia often co-exist in wastewater, causing serious environmental problems. The information on the metabolic mechanism of simultaneously removing these two pollutants by bacteria is insufficient at present. Moreover, phenol is toxic to microbial and causes cells damage. Therefore, exploring the response mechanism of bacteria to phenol stress is conducive to understand their tolerance mechanism to aromatic compounds. In this study, the metabolic characteristics and adaptive mechanism of C. oxalaticus T2 during the simultaneous removal of phenol and nitrogen process were evaluated by comparing the proteome profiles at different stages. The results revealed the degradation pathways of phenol and nitrogen by strain T2. A variety of phenol response mechanisms were determined, including enhanced energy production, improved cell motility, increased the synthesis of lipoic acid and biotin, and combined action of multiple anti-stress proteins. Selenium (Se) is an essential mineral nutrient for animals. Se deprivation can lead to many disorders and even death. This study investigated the response of Przewalski's gazelle (P. przewalskii) to Se-deprived environment. We found that Se deprivation in soil and forage not only influenced the mineral contents of the blood and hair in P. przewalskii, but also severely disrupted their blood parameters. We identified significant changes in the abundance of 146 proteins and 25 metabolites (P < 0.05) in serum, including the selenoproteins L8IG93 (glutathione peroxidase) and F4YD09 (Cu/Zn superoxide dismutase). Furthermore, the major known proteins and metabolites associated with the Se stress response in P. przewalskii were Cu/Zn superoxide dismutase, the vitamin K-dependent protein C, the C4b-binding protein alpha chain, complement component C7, lipase linoleic acid, peptidase D, thymidine, pseudo-uridine, L-phenylalanine, L-glutamine, PGA1, and 15-deoxy-delta-12,14-PGJ2. The main signaling pathways involved included complement and coagulation cascades, metabolic pathways, and stress granule formation. Our results indicate that the intake of Se-deficient forage elicited an oxidative stress response in P. przewalskii. These findings provide insights into the response mechanisms of this threatened gazelle to Se stress, and enable the development of conservation strategies to protect populations on the Qinghai-Tibetan Plateau. SIGNIFICANCE: This study is the first to point out the presence of oxidative stress in P. przewalskii in selenium-deficient areas through proteomics and metabolomics studies. Cysteine(Cys) is tightly related to physiological and pathological of human, and the imbalance of concentration of cysteine in the intracellular are associated with many diseases. Here, a novel NIR fluorescent probe TCF-Cys was designed and synthesized, and both the optimal excitation and emission wavelength of them were between 650 and 900 nm, that within the "optical window" of biological tissues. In aqueous solution, TCF-Cys, which with an acrylate extremity as a recognizing unit, exhibited excellent "turn-on" fluorescence response for Cys superior to other amino acids and thiols with a limit of detection of 0.1323 μM. Moreover, as an excellent naked-eye colorimetric indicator, TCF-Cys could effectively distinguishing the Cys, Hcy and GSH in aqueous solution through color change. Then, the response mechanism of TCF-Cys for Cys was revealed by TLC, 1 H NMR, HPLC, HRMS and DFT calculation. Finally, TCF-Cys was successfully employed to fluorescence specifically map of exogenous and endogenous Cys in living cells and zebrafish with low toxicity., (Copyright © 2019 Elsevier B.V. Chlorella vulgaris was cultivated in the gradually increased proportion of toxic sludge extracts for acclimation, which was obtained from SBR treated synthetic wastewater containing mixed chlorophenols (2,4,6-trichlorophenol and 4-chlorophenol). The growth of C. vulgaris was obviously improved after acclimation with the cell number in the 100% sludge group was 22.75±0.85∗10 6 cellmL -1 , which was relatively more than the BG11 control group's (20.80±0.35∗10 6 cellmL -1 ) and apparently over the 100% sludge group (10.78±0.45∗10 6 cellmL -1 ). Compared with the sludge control sludge group, C. vulgaris in the acclimation group gained 24.1% and 18.2% more relative removal rate about TOC and ecotoxicity, respectively. Proteomics analysis showed that protein spots were more clear and centralized and the clarifications of the different protein spots narrowed from 8 to 5 after acclimation. Proteins related to oxidoreducase activity and energy metabolism were over expressed and C. vulgaris could select the metabolic pathways, especially enhanced pyruvate fermentation, TCA cycle, and glycolysis after acclimation, by over accumulating the corresponding vital enzymes. Conclusively, acclimation was a good method to improve the removal ability and growth of C. vulgaris and algae could acclimatize itself to grow upon the toxic sludge extracts by metabolic selection. The gas phase ionization detectors are the most widely used detectors for gas chromatography. The column and makeup gases commonly used in gas chromatography are near perfect insulators. This facilitates the detection of a minute number of charge carriers facilitating the use of ionization mechanisms of low efficiency while providing high sensitivity. The main ionization mechanism discussed in this report are combustion in a hydrogen diffusion flame (flame ionization detector), surface ionization in a plasma (thermionic ionization detector), photon ionization (photoionization detector and pulsed discharge helium ionization detector), attachment of thermal electrons (electron-capture detector), and ionization by collision with metastable helium species (helium ionization detector). The design, response characteristics, response mechanism, and suitability for fast gas chromatography are the main features summarized in this report. The illegal feeding of clenbuterol (CLB) to domestic animals and the potential harm of it to human health lead an urgent requirement for the efficient detection of CLB, especially in the edible meat. In this paper we reported a new fluorescence method for the detection of trace amount of CLB by using the BSA-protected gold nanoclusters (AuNCs@BSA). Under the excitation of either 280 or 500 nm the emission of AuNCs@BSA was quenched obviously by diazotized CLB, supplying a dual-responsive fluorescence method to detect CLB in aqueous solution. In addition, the linear response of the fluorescence intensity of AuNCs@BSA to diazotized CLB allowed the quantitative detection of CLB in a range of 4.0 nM-300 μM upon excitation at two wavelength, and the limit of detection for CLB was 3.0 nM upon 280 nm excitation and 1.6 nM upon 500 nm excitation, respectively. In addition, the dual-responsive mechanism of AuNCs@BSA to CLB was investigated in detail by using several CLB analogues and reference compounds. Particularly, the proposed method was successfully applied to detect CLB in pork mince and the results were validated well by HPLC, illustrating it could be used as a reliable, rapid, and cost-effective technique for the determination of CLB residues in real samples., (Copyright © 2015.
