Summary: The lymphatic fluid is the conduit by which part of the tissue “omics” is transported to the draining lymph node for immunosurveillance. Following cannulation of the pre-nodal cervical and mesenteric afferent lymphatics, herein we investigate the lymph proteomic composition, uncovering that its composition varies according to the tissue of origin. Tissue specificity is also reflected in the dendritic cell-major histocompatibility complex class II-eluted immunopeptidome harvested from the cervical and mesenteric nodes. Following inflammatory disruption of the gut barrier, the lymph antigenic and inflammatory loads are analyzed in both mice and subjects with inflammatory bowel diseases. Gastrointestinal tissue damage reflects the lymph inflammatory and damage-associated molecular pattern signatures, microbiome-derived by-products, and immunomodulatory molecules, including metabolites of the gut-brain axis, mapped in the afferent mesenteric lymph. Our data point to the relevance of the lymphatic fluid to probe the tissue-specific antigenic and inflammatory load transported to the draining lymph node for immunosurveillance.

In an islanded hybrid microgrid, frequency deviation is caused by the intermittent nature of renewable resources and load variations. Maintaining nominal frequency requires the secondary controller to send a signal to the controllable energy sources so that supply and demand are synchronized. Time delays in the system's communication channels, however, can reduce its efficiency. To compensate for the system uncertainties, modeling error and variant nature of renewable resources, an optimized flatness based active disturbance rejection controller have been proposed. Mitigating the effects of communication delay in the system and to synchronize the inputs to the extended state observer, a smith predictor based low pass filter is employed. Furthermore, an improved beluga whale optimization (IBWO) technique is employed to tune the proposed controller gains. The comparative analysis of the proposed IBWO is performed with existing algorithms. Moreover, the proposed controller superiority over the standard PID and LADRC approach has been investigated by perturbation in renewable resources, uncertainty in parameters, variation and mismatch in communication delay. Simulation results provide the testimony for the efficient performance of the designed controller. Lastly, the proposed controller is further evaluated in multi microgrid environment.

Background: Since 2020 SARS-CoV-2 spreads pandemically, infecting more than 119 million people, causing >2·6 million fatalities. Symptoms of SARS-CoV-2 infection vary greatly, ranging from asymptomatic to fatal. Different populations react differently to the disease, making it very hard to track the spread of the infection in a population. Measuring specific anti-SARS-CoV-2 antibodies is an important tool to assess the spread of the infection or successful vaccinations. To achieve sufficient sample numbers, alternatives to venous blood sampling are needed not requiring medical personnel or cold-chains. Dried-blood-spots (DBS) on filter-cards have been used for different studies, but not routinely for serology. Methods: We developed a semi-automated protocol using self-sampled DBS for SARS-CoV-2 serology. It was validated in a cohort of matched DBS and venous-blood samples (n = 1710). Feasibility is demonstrated with two large serosurveys with 10247 company employees and a population cohort of 4465 participants. Findings: Sensitivity and specificity reached 99·20% and 98·65%, respectively. Providing written instructions and video tutorials, 99·87% (4465/4471) of the unsupervised home sampling DBS cards could be analysed. Interpretation: DBS-sampling is a valid and highly reliable tool for large scale serosurveys. We demonstrate feasibility and accuracy with a large validation cohort including unsupervised home sampling. This protocol might be of big importance for surveillance in resource-limited settings, providing low-cost highly accurate serology data.
