Powder Bed Fusion Laser Beam (PBF-LB) is an attractive additive manufacturing technique for fabricating K447A superalloys components. Unfortunately, these as-built parts generally exhibit unsatisfied mechanical properties because they are highly susceptible to internal defects. Herein, we propose different post-treatments, including laser shock peening (LSP), ultrasonic shock peening (USP), laser after ultrasonic shock peening (ULSP), and ultrasonic after laser shock peening (LUSP) for tuning the microstructures and performances of a PBF-LB fabricated K447A superalloys. The microstructural response, residual stress and microhardness redistribution, as well as tensile properties of the K447A superalloys treated by different post-treatments are systematically studied. Results indicate that a gradient heterogeneous structure, which is primarily characterized with gradient variations in grain size, precipitation phase size and dislocation density, is formed on the K447A specimen surface under LUSP treatment. The surface residual stress is completely transformed into compressive state with a maximum amplitude value of −185 MPa, and the peak value of microhardness is improved to 568 HV. The various strengthening mechanisms of gradient heterogeneous structures, as well as dislocation multiplication, grain refinement and precipitated phase strengthening, are responsible for achieving strength-ductility synergy. This work provides promising route and valuable theoretical guidance for optimizing mechanical properties of additively manufactured metastable high γ′ phase fraction superalloys via tuning microstructures.

The stimulator of interferon genes (STING) plays a pivotal role in orchestrating innate immunity, and dysregulated activity of STING has been implicated in the pathogenesis of autoimmune diseases. Recent findings suggest that bacterial infection activates STING, relieving ER stress, and triggers non-canonical autophagy by spatially regulating STX17. Despite these insights, the precise mechanism governing the dynamics of autophagosome fusion elicited by STING remains unclear. In this study, we demonstrate that dynamic STING activation guides the autophagy flux, mirroring the trajectory of canonical autophagy adaptors. STING engages in a physical interaction with STX17, and agonist-induced phosphorylation or degradation alleviates STING's inhibitory effects on the assembly of the STX17-SNAP29-VAMP8 complex. Consistent with these findings, degradation-deficient mutants hinder autophagy flux by impeding STX17-mediated autophagosome-lysosome fusion. Moreover, STING mutants associated with lupus disrupt the assembly of the STX17-SNAP29-VAMP8 complex and autophagy process, which lead to persistent STING activation and elevated IFN-β production. Our results highlight that the intracellular trajectory of STING, coupled with autophagy flux, guides the assembly and membrane fusion of the STX17-SNAP29-VAMP8 complex, ensuring the accurate regulation of innate immunity.

When using current methods to predict electricity theft behavior, there are problems of large sample data errors before training, high line loss rate, and low prediction effect. To this end, a power-stealing behavior prediction method based on multi-dimensional features and BP neural network is proposed. The method first extracts the abnormal power usage characteristics according to the abnormal features of the user’s power usage behavior. Then the genetic algorithm is used to optimize the BP network, and the optimized network is used to train the abnormal power consumption characteristics. Finally, a prediction model is established based on the training results to achieve the final prediction. The experimental results show that the training error test, line loss rate test and prediction effect test of the method before and after training verify the practicability and effectiveness of the method.

Many brittle materials, such as single-crystal materials, amorphous materials, and ceramics, are widely used in many industries such as the energy industry, aerospace industry, and biomedical industry. In recent years, there is an increasing demand for high-precision micro-machining of these brittle materials to produce precision functional parts. Traditional ultra-precision micro-machining can lead to workpiece cracking, low machined surface quality, and reduced tool life. To reduce and further solve these problems, a new micro-machining process is needed. As one of the nontraditional machining processes, rotary ultrasonic machining is an effective method to reduce the issues generated by traditional machining processes of brittle materials. Objective: To determine the efficacy and safety of the Yinmei Kuijie decoction combined with 5-aminosalicylic acid (5-ASA) in treating mildly to moderately active ulcerative colitis (UC) under real-world conditions. Methods: This multicenter, prospective, non-randomized, observational study will be conducted in real-world settings. A total of 204 eligible patients will be consecutively enrolled in the study. Patients in the combination treatment group will receive Yinmei Kuijie decoction in combination with 5-ASA, whereas those in the control group will be treated with 5-ASA alone. The primary endpoint will be a clinical response at week 12, defined as a ≥3 point and ≥30% reduction from baseline in the Mayo total score with ≥1 reduction in rectal bleeding or rectal bleeding score = 0 or 1. Secondary efficacy endpoints at week 12 will include health-related quality of life, mucosal healing, and inflammation indicators. Conclusion: The results of this study may provide evidence of the efficacy and safety of Yinmei Kuijie decoction combined with 5-ASA in treating patients with mildly to moderately active UC under real-world principles. The results will provide a basis for further confirmatory studies on the efficacy of Yinmei Kuijie decoction.

This paper discusses how managers adjust their strategies to allocate relevant resources more effectively and maximize economic benefits when major technological changes are predicted for the future. For a supply chain system consisting of a single manufacturer and two competing retailers as the research object. First, random stop model is applied to portray the impact of technological innovation on the decision-making of supply chain members. On this basis, differential game models for supply chain members are constructed based on different cooperation modes, including centralized, decentralized, and retailers alliance. Second, we solve and compare the optimal decision-making, emissions reduction, low-carbon goodwill, and profit levels before and after technological innovation in different modes. Finally, we design a bilateral cost-sharing contract to achieve coordination. Results demonstrate that: (1) Before the success of technological innovation, when a higher probability of success and uplift rate is predicted can incentivize supply chain members' emissions reduction and low-carbon promotion inputs; (2) In the presuccess period of technological innovation, members' independent decision-making (decentralized decision-making) can optimize the retailer's low-carbon promotional inputs under certain conditions. In contrast, the optimality of decentralized decision-making after technological innovation depends only on the influence of competition intensity. (3) The bilateral cost-sharing contract designed in this paper can optimize supply chain-related inputs and performance levels to achieve perfect coordination within the supply chain system, given that specific preconditions are satisfied.

Inflammation-mediated carcinogenesis develops in the context of chronic inflammation and is a significant cause of cancer within the digestive system. In the chronic inflammation microenvironment, the metabolic activity of tissue cells undergoes extensive changes, which interfere with the normal function of immune cells. Dysregulation of cell metabolism and immune function has been identified as a key factor contributing to inflammation-mediated carcinogenesis within the major digestive organs, such as the stomach, liver, and colorectum. This metabolic–immune imbalance also corresponds to traditional Chinese medicine (TCM) theories of “yin-yang disharmony” and “disharmony between Ying-nutrients and Wei-defense.” The metabolic–immune imbalance has also been regarded as the key factor supporting “treatment of different diseases with the same method,” in which the same approach is adopted in the treatment of different conditions. In the TCM treatment process, it is necessary to first identify TCM patterns and then apply the corresponding TCM to correct the dysregulated metabolic and immune function, thereby blocking the progression from inflammation to malignancy. Our study findings deepen the TCM understanding of metabolic–immune dysregulation and the relationship between metabolic–immune dysregulation, pattern identification, and treatment method. They also provide new insights for the treatment of inflammation-mediated carcinogenesis in major digestive organs and help us further explore the scientific connotation of the TCM strategy of “treating different diseases with the same method.
