The development of tools that can provide a holistic picture of the evolution of the tumor microenvironment in response to intermittent fasting on the prevention of breast cancer is highly desirable. Here, we show, for the first time, the use of label-free Raman spectroscopy to reveal biomolecular alterations induced by intermittent fasting in the tumor microenvironment of breast cancer using a dimethyl-benzanthracene induced rat model. To quantify biomolecular alterations in the tumor microenvironment, chemometric analysis of Raman spectra obtained from untreated and treated tumors was performed using multivariate curve resolution-alternative least squares and support vector machines. Raman measurements revealed remarkable and robust differences in lipid, protein, and glycogen content prior to morphological manifestations in a dynamically changing tumor microenvironment, consistent with the proteomic changes observed by quantitative mass spectrometry. A photo/electrochemical coupling interface of Ru[dcbpy] 3 2+ -AMT/Au (AMT; 5-Amino-1,3,4-thiadiazole-2-thiol) was fabricated using a dehydration condensation sulfhydrating method. For the interface functional properties, a combined dual-signal recording (CDSR) method was applied to characterize the response characteristics, and a scanning electrochemical microscopy-electrochemiluminescence (SECM-ECL) imaging was developed to assess the interface distribution uniformity. The interface biosensing compatibility was validated by constructing a simple DNA sensor. The research results show that the interaction between the two functional parameters follows a synergistic effect mechanism in the coupling conditions and an interference effect mechanism in the detection condition. Under optimized conditions, the saturation dual-signal response values are 156.0 and 86.8 μA, respectively. The statistics and imaging comparison analysis validate good interface distribution uniformity and stability performance. The DNA sensor's dual-signal detection limits to the signal probe (SP) are ∼30 fM and 0.3 pM with linear ranges of 100.0 fM ∼ 1.0 nM and 1.0 pM ∼ 10.0 nM, respectively. The fabricated interface exhibits an effective bi-functional response performance compatible with biosensing. The world's largest "green tide" (Ulva prolifera) has occurred every year since 2007 in the Yellow Sea. The Subei Shoal area is thought to be the origin of the green tide. Based on field data from 2016 to 2023, seasonal and interannual variations of dissolved nutrients and their ecological effects in the Subei Shoal were analyzed. Spatial distribution of dissolved inorganic nitrogen (DIN), dissolved inorganic phosphorus (DIP) and dissolved silicate (DSi) showed clear terrestrial sources, while ammonia (NH 4 -N) and dissolved organic nitrogen (DON) were not solely controlled by terrestrial sources. The seasonal variations of NH 4 -N, DIN, DON, DIP and DSi concentrations were significant, and the interannual variations of DIN, DON, DIP and DSi concentrations showed general decreasing trends from 2016 to 2023. The key factors affecting the seasonal and interannual variations of DIN and DIP concentrations were terrestrial input, aquaculture wastewater discharge, atmospheric deposition, submarine groundwater discharge and macroalgae absorption, while the dominant factor determining the variations of DSi concentrations was terrestrial input. NH 4 -N and DON concentrations were mainly influenced by aquaculture wastewater discharge and the absorption and release of macroalgae. The presence of organic-complexed copper and zinc in anaerobic digestate effluent (ADE) poses persistent ecological toxicity. This study investigated the detoxification performance and biotic responses of indigenous bacteria against ethylene diamine tetraacetic acid (EDTA)-complexed Cu(II) and Zn(II). Heavy metals (HMs) stress induced reactive oxygen species (ROS) generation and enhanced extracellular polymeric substances (EPS) secretion. At a Cu(II) influent concentration of 20.0 mg·L -1 , indigenous bacteria removed 88.2% of Cu(II) within nine days. The majority of copper and zinc sequestered by bacteria were stored in the cell envelope, with over 50% of copper and 60% of zinc being immobilized. Transmission electron microscopy mapping (TEM-mapping) revealed significant mineralization of copper and zinc on the cell wall. Proteins abundant in EPS, alongside humic acid-like substances, effectively adsorbed HMs. Indigenous bacteria exhibited the capacity to reduce cupric to the cuprous state and cupric is preferentially reduced to cuprous before reaching reducing capacity saturation. Sulfur precipitation emerges as a crucial pathway for Zn(II) removal. Seasonal sediment deposition-erosion events are dominant drivers of particle-solute dynamics in large-river delta-front estuaries (LDEs), but their influence on elemental cycles is not yet fully understood. To better constrain the role of deposition-erosion events on elemental cycling in LDEs, benthic fluxes of dissolved inorganic carbon (DIC), oxygen, and pore-water solute profiles were measured over different seasons in the Changjiang LDE. Benthic DIC efflux (23.4 ± 6.0 mmol C m -2 d -1 ) was greater than oxygen influx (7.5 ± 2.0 mmol O 2 m -2 d -1 ) in summer but less in winter (7.7 ± 1.2 mmol C m -2 d -1 and 10.1 ± 1.5 mmol O 2 m -2 d -1 , respectively). The additional oxygen consumption in sediments in winter was likely due to the oxidation of inorganic diagenetic reductive products (IDRP) (e.g., NH 4 + , Fe 2+ , and Mn 2+ ) in deeper sediments exposed by erosion, which resulted in the development of an "oxygen debt". Sedimentary oxygen respiration accounted for at least 48 % of total oxygen consumption (oxygen consumption in both water column and sediment) in winter and was significantly greater than in summer (∼15 %); this highlighted the importance of winter sediment erosion in oxygen depletion. In addition to IDRP oxidation, the remineralization of resuspended sedimentary organic carbon in water column also contributed to the oxygen consumption. Objectives: To assess the efficacy and safety of rifampicin-based triple therapy (rifampicin, isoniazid, and ethambutol) for treating NPM., Methods: This single-center, single-arm, prospective clinical trial was conducted at the Second Hospital of Shandong University (Jinan, China). Patients with pathologically diagnosed granulomatous lobular mastitis and periductal mastitis received triple drugs, i.e., rifampicin (450 mg/day), isoniazid (300 mg/day), and ethambutol (15 mg/kg/day), until complete response or the investigator decided to discontinue treatment. The primary endpoint was the complete response rate (CRR) assessed by the investigator. The secondary endpoints included the overall remission rate (ORR), recurrence rate (RR), and safety., Results: A total of 218 patients were enrolled in the study between January 1, 2013 and October 31, 2020. With a median follow-up time of 48 months, the CRR and the ORR were 78.44% and 94.04%, respectively. While 13 patients (5.96%) demonstrated no response and 19 relapsed (8.72%). Adverse events (AEs) were not common. The most common AEs during treatment were liver dysfunction (1.83%), gastrointestinal reactions (1.83%), fatigue (1.83%), erythema (1.38%), and menstrual disorders (0.92%)., Conclusion: Rifampicin, isoniazid, and ethambutol demonstrated promising response rates with acceptable safety profiles in patients with NPM. Glycerol dialkyl glycerol tetraether core lipids (GDGTs) are microbial biomarkers ubiquitously distributed in terrestrial and marine environments. Dispersal and fate of GDGTs in an estuary largely depends on sediment grain size, however, their size distribution patterns remain poorly understood. Here, surface sediments collected from the Changjiang Estuary were separated into <20, 20-32, 32-63, 63-125 and >125 μm fractions, and analyzed for GDGTs as well as total organic carbon (TOC), stable isotopic composition (δ 13 C) of TOC and lignin phenols, to investigate the size and spatial distributions of GDGTs and the particle size effects on GDGTs proxies in this large river delta-front estuary. The concentrations of isoprenoidal GDGTs (isoGDGTs) were higher in the finest fractions and in off-estuary sites. On the contrary, branched GDGTs (brGDGTs) were high not only in the finest fractions but in coarser fractions (>32 μm fractions), and thus at both near- and off-estuary sites. The branched and isoprenoid tetraether (BIT) index increased with increasing grain size, and decreased sharply from the estuary (~0.52) to the shelf (~0.16). BrGDGTs were positively correlated with crenarcheaol in both high and low BIT regions. The brGDGTIIIa/IIa ratios in all size fractions were <0.59, further indicating that the brGDGTs were mainly derived from terrestrial input with minimum in-situ production. Fractional TOC source assignments derived from the BIT index was significantly positively correlated with the fractions of terrestrial OC from a mixing model based on δ 13 C-TOC and lignin contents, indicating that BIT may track a broader pool of terrestrial OC than just soil OC. This work provides novel, yet preliminary insights into the size fractionated distribution characteristics of GDGTs and the applicability of BIT as a proxy for OC sources in estuarine sediments. Aim: This study aims to identify suitable candidates for axillary sentinel lymph node biopsy (SLNB) or targeted axillary dissection (TAD) among clinical N2 (cN2) triple-negative (TN) or HER2 positive (HER2+）breast cancer patients following neoadjuvant therapy（NAT）., Background: Despite the substantial axillary burden in cN2 breast cancer patients, high pathological response rates can be achieved with NAT in TN or HER2+ subtypes, thus enabling potential downstaging of axillary surgery., Methods: A retrospective analysis was conducted on data from the CSBrS-012 study, screening 709 patients with initial cN2, either HER2+ or TN subtype, from January 1, 2010 to December 31, 2020. The correlation between axillary pathologic complete response (pCR) (yPN0) and breast pCR was examined., Results: Among the 177 cN2 patients who achieved breast pCR through NAT, 138 (78.0 %) also achieved axillary pCR. However, in the 532 initial clinical N2 patients who did not achieve breast pCR, residual axillary lymph node metastasis persisted in 77.4 % (412/532) of cases. Methylmercury is a neurotoxic compound that can enter rice fields through rainfall or irrigation with contaminated wastewater, and then contaminate the human food chain through the consumption of rice. Flooded paddy soil has a porous structure that facilitates air exchange with the atmosphere, but the presence of trace amounts of oxygen in flooded rice field soil and its impact on microbial-mediated formation of methylmercury is still unclear. We compared the microbial communities and their functions in oxygen-depleted and oxygen-limited paddy soil. We discovered that oxygen-limited paddy soil had higher methylmercury concentration, which was strongly correlated with soil properties and methylation potential. Compared with oxygen-depleted soil, oxygen-limited soil altered the microbial composition based on 16 S rRNA sequences, but not based on hgcA sequences. Moreover, oxygen-limited soil enhanced microbial activity significantly, increasing the abundance of more than half of the KEGG pathways, especially the metabolic pathways that might be involved in methylation. Our study unveils how microbial communities influence methylmercury formation in oxygen-limited paddy soil. ENVIRONMENTAL IMPLICATIONS: This study examined how low oxygen input affects microbial-induced MeHg formation in anaerobic paddy soil. We found that oxygen-limited soil produced more MeHg than oxygen-depleted soil. Oxygen input altered the microbial community structure of 16 S rRNA sequencing in anaerobic paddy soil, but had little impact on the hgcA sequencing community structure. Microbial activity and metabolic functions related to MeHg formation were also higher in oxygen-limited paddy soil. Agricultural N losses strongly dominate the N delivery (average 72 % of total N delivery to rivers in the period 1980-2010) in the rivers discharging into the Bohai Sea, a semi-enclosed marginal sea, which has been suffering from eutrophication and deoxygenation since the 1980s. In this paper we investigate the relationship between N loading and deoxygenation in the Bohai Sea, and consequences of future N loading scenarios. Using modeling for the period 1980-2010, the contributions of different oxygen consumption processes were quantified and the main controlling mechanisms of summer bottom dissolved oxygen (DO) evolution in the central Bohai Sea were determined. Model results show that the water column stratification during summer impeded the DO exchange between oxygenated surface water and oxygen-poor bottom water. Water column oxygen consumption (60 % of total oxygen consumption) was strongly correlated with elevated nutrient loading, while nutrient imbalances (increasing N:P ratios) enhanced harmful algal bloom proliferation. Future scenarios show that deoxygenation may be reduced in all scenarios owing to increasing agricultural efficiency, manure recycling and wastewater treatment. Mercury (Hg) poses a significant risk in paddy fields, particularly when it is converted to methylmercury (MeHg) and accumulates in rice. However, the bioavailability and resupply kinetics of Hg in the paddy soil-water environment are not well understood. In this study, the diffusive gradients in thin films (DGT) and the 'DGT-induced fluxes in sediments' model (DIFS) were first adopted to investigate the Hg resupply kinetics, diffusion fluxes and bioavailability in a paddy environment subjected to flood-drain-reflood treatment and straw amendment. Our results show that although the straw amendment limited the bioavailability of Hg (38.2%-47.9% lower than control) in porewater by decreasing its resupply capacity, especially with smaller straw particles, the net production of MeHg in paddy fields was significantly increased after straw amendment (73.5%-77.9% higher than control). The results of microbial sequencing indicate that enhanced methylators (e.g., family Geobacter) and non-Hg methylators (e.g., Methanosarcinaceae) played a crucial role in MeHg production following straw amendment. Moreover, Hg-containing paddy soils generally tend to release Hg into the overlying water, while drain-reflood treatment changes the direction of Hg diffusion fluxes in the paddy soil-water interface. The drainage-reflooded treatment decreases the Hg reactive and resupply capacity of the paddy soil, thereby hindering the release of Hg from soil into overlying water during the early stages of reflooding. Based on historical data from 1976 to 2019, the effects of anthropogenic activities on long-term changes in nutrients and their ecological effects in the South Yellow Sea were investigated. The dissolved inorganic nitrogen (DIN) concentrations increased continuously from 1990 until the mid-2000s, followed by a shift from an upward trend to a downward trend. The phosphate (PO 4 -P) and silicate (SiO 3 -Si) concentrations also showed obvious interannual variations throughout the study period. The concentrations of DIN, PO 4 -P and SiO 3 -Si have decreased significantly in recent decade and more. These changes mainly resulted from the reduction in terrestrial input, while the main reason for the decrease in DIN and PO 4 -P concentrations is the reduction in anthropogenic input. Better documentation and understanding of long-term temporal dynamics of nutrients in watersheds are necessary to support effective water quality management. We examined the hypothesis that the recent management of fertilizer use and pollution control in the Changjiang River Basin could govern the fluxes of nutrients from the river to the sea. Results based on historical data since 1962 and surveys in recent years show that concentrations of dissolved inorganic nitrogen (DIN) and phosphorus (DIP) in the mid- and downstream reaches were higher than those in the upper reaches due to intensive anthropogenic activities, while dissolved silicate (DSi) was distributed evenly from the up- to downstream reaches. Fluxes of DIN and DIP increased rapidly, and DSi declined during the 1962-1980 and 1980-2000. After the 2000s, concentrations and fluxes of DIN and DSi remained almost unchanged; those of DIP remained stable until the 2010s and slightly decreased afterward. The decline in fertilizer use explains 45 % of the variance in the decline of DIP flux, followed by pollution control, groundwater and water discharge. As a result, the molar ratio of DIN:DIP, DSi:DIP and ammonia:nitrate varied largely during 1962-2020, and the excess DIN relative to DIP and DSi lead to increased limitations of silicon and phosphorus. A turning point probably occurred for nutrient fluxes in the Changjiang River in the 2010s, with the pattern of DIN from continuous increase to stability and DIP from increase to decrease. This decline in phosphorus in the Changjiang River has many similarities with the rivers worldwide. Contaminants in high-altitude mountains such as the Tibetan Plateau (TP) have attracted extensive attention due to their potential impact on fragile ecosystems. Rapid development of the economy and society has promoted pollution caused by local traffic emissions in the TP. Among the pollutants emitted by traffic, polycyclic aromatic hydrocarbons (PAHs) and oxygenated PAHs (OPAHs) are of particular concern due to their high toxicity. The TP provides an environment to explore the degree and range of contribution for traffic-induced PAHs and OPAHs. In this study, soils and moss were collected at different altitudes and distances from the G318 highway in the southeast TP. The total concentrations of PAHs (∑ 16 PAHs) and OPAHs (∑ 6 OPAHs) in soils were in the range of 3.29-119 ng/g dry weight (dw) and 0.54-9.65 ng/g dw, respectively. ∑ 16 PAH and ∑ 6 OPAH concentrations decreased logarithmically with increasing distance from traffic. A significantly positive correlation between ∑ 16 PAHs and altitude was found at sampling points closest to traffic. Dominant PAHs constituents in soil and moss included chrysene (CHR), benzo[g,h,i]perylene (BghiP), and benzo[b]fluoranthene (BbF); prevalent OPAH compounds were 9-fluorenone (9-FO) and 9,10-anthraquinone (ATQ). These compounds were related to characteristics of traffic emissions. The multiple diagnosis ratio and correlation analysis showed that exhaust emissions were the main source of the PAHs and OPAHs in the studied environment. PMF modeling quantification of the relative contribution of traffic emissions to PAHs in roadside soils was 45 % on average. N 2 O is among the most potent greenhouse gases. In this study, we investigated one of the important N 2 O production hotspots, the continental margins. We looked at N 2 O spatiotemporal distributions in situ as well as the potential contributions of nitrification and denitrification to N 2 O production in sediment cores from the Bohai and South Yellow Seas. Real-time PCR and shotgun metagenomics sequencing were used to analyze the microbial communities related to N 2 O production. The results showed that N 2 O concentrations roughly decreased with depth-a trend that was consistent throughout the year and showed no significant seasonal variations. When all the research stations along the continental margin were considered, the estuary exhibited the lowest average N 2 O concentration. Moreover, nitrification was identified as the main process responsible for N 2 O production in estuary areas. Silicon (Si) plays an essential role in the biogeochemistry of rivers. This study explored how damming, eutrophication and climate change alters the abundance and flux of DSi in the Changjiang (Yangtze) River based on long-term observations. The results showed that Three Gorges Reservoir (TGR) could enhance DSi transfer only during low-flow time period, and a downstream DSi retention effect by the TGR was found between the Yichang and Jianli stations in the Changjiang River. This resulted in a DSi loss during March and April in the mainstream from Three Gorges Dam (TGD) to Jianli but a DSi addition during July and October along the main channel of the Changjiang River. Long-term data showed a sharp decrease in DSi abundance at the Cuntan, Hankou and Datong stations between the 1960s and 1980s, but a slight increase in DSi between the 1990s and 2010s at these stations. The decrease in DSi during the 1960s -1980s was primarily the result of a decrease trend of silicate weathering, while a slight DSi increase compared to the temperature/DSi relation after the 1990s was largely due to increased DSi retention in the basin by damming and eutrophication. Non-caloric artificial sweeteners are being widely used as safe table sugar substitutes with highly intensive sweetness but low calories. Previous studies have suggested that some of the sweeteners can alter the gut microbiota composition and promote horizontal transfer of antibiotic resistance genes across bacterial genera. However, little is known about whether these sweeteners could show antibiotic-like antimicrobial activity against bacteria, especially gut relevant bacteria. Whether they could affect evolutional trajectory of antibiotic resistance or tolerance in bacteria is also not clear yet. Here we investigated four commonly used artificial sweeteners (saccharin, sucralose, aspartame, and acesulfame potassium) against both Gram-negative (Escherichia coli and Klebsiella pneumoniae) and positive (Bacillus subtilis) strains. Results show that all four sweeteners exhibit antimicrobial effects on these strains. The antimicrobial mechanism is due to increased reactive oxygen species (ROS) and cell envelope damage. Compared to sucrose and glucose, the treatment of artificial sweeteners stimulates bacterial efflux pumps and promotes bacterial evolution of antibiotic tolerance. Increased chloride concentration can cause salinization, which has become a serious and widespread environmental problem nowadays. This review aims at providing comprehensive and state-of-the-art knowledge and insights of technologies for chloride removal. Mechanisms for chloride removal mainly include chemical precipitation, adsorption, oxidation and membrane separation. In chemical precipitation, chloride removal by forming CuCl, AgCl, BiOCl and Friedel's salt. Adsorbents used in chloride removal mainly include ion exchangers, bimetal oxides and carbon-based electrodes. Oxidation for chloride removal contains ozone-based, electrochemical and sulfate radical-based oxidation. Membrane separation for chloride removal consists of diffusion dialysis, nanofiltration, reverse osmosis and electrodialysis. In this review, we specifically proposed the factors that affect chloride removal process and the corresponding strategies for improving removal efficiency. We established long-term surveillance sites in Jinan city to monitor PM 2.5 particles (PM2.5) and PM2.5-bound PAHs (2014-2020). The range of PM2.5 was 15-230 µg/m 3 . The average annual ƩPAH 16 were 433 ± 271 ng/m 3 (industrial area) and 299 ± 171.8 ng/m 3 (downtown). PAHs captured in winter accounted for 61.5% (industrial area) and 59.1% (downtown) of total PAHs. A hazardous seasonal benzo[a]pyrene level was detected in 2015-2016 winter as 14.03 ng/m 3 (14 folds of EU standard). The dominant PM2.5-bound PAHs were benzo[b]fluoranthene (24-26%), chrysene (19-20%), benzo[g,h,i]perylene (15%), Indeno(1,2,3-cd)pyrene (12%) and Benzo[a]pyrene (10%). Toxic equivalent quotients of PAHs were 4.93 ng/m 3 (industrial area) and 3.13 ng/m 3 (downtown). Excess cancer risks (ECRs) were 4.3 × 10 -4 ng/m 3 and 2.7 × 10 -4 ng/m 3 , respectively. The ECRs exceeded EPA regulatory limit of 1 × 10 -6 ng/m 3 largely. Non-negligible excess lifetime cancer risks were found as 36 and 26 related cancer incidences per 1,000,000 people. Consistently, local prevalence of lung cancer raise from 56.97/100,000 to 72.38/100,000; the prevalence of thyroid cancer raise from 10.12/100,000 to 45.26/100,000 from 2014 to 2020. River damming reduces sediment load in rivers, leading to instability and erosion of coastal deltas; this is a global issue in many large-river delta-front estuaries (LDEs). The total organic carbon, stable carbon isotope, total nitrogen, lignin phenols in suspended particulate matter, and three sediment cores collected from the Changjiang LDE were analyzed to study the effects of river damming and delta erosion on the redistribution and burial of organic carbon (OC) in the coastal margin. The OC budgets that include the delta erosion process were established to better constrain the burial fluxes of OC in the Changjiang River system before and after river damming. We used a three-end-member mixing model with Monte Carlo simulation and found that the ratio of deltaic OC to riverine OC significantly increased in estuarine SPM and sediment cores in the Changjiang LDE after the operation of the Three Gorges Dam (TGD), reflective of reduced sediment load and enhanced delta erosion. Our budget showed that, compared with the pre-dam period (1953-1985), the burial of terrestrial OC in the Changjiang LDE sediments had been reduced by ~97.3%, on the basis of 72.7% and 82.7% reductions in sediment load and particulate OC flux, respectively. Although trapping of terrestrial OC in reservoirs is the key driver in these reductions, re-exposure and decomposition of OC from eroding delta sediments also contribute significantly to OC losses in coastal sediments. On the contrary, the ratio of marine OC to riverine OC increased in sediment cores after TGD building, due in part to more inputs of marine OC because of serious eutrophication in recent decades. One of the largest "green tide" (Ulva prolifera) outbreaks in the world has occurred every year from 2007 to present in the Southern Yellow Sea, China. Currently, the coastal area around Jiangsu Province (Subei Shoal region) is thought to be the origination point of these giant green tide blooms. The combination of high nutrient demand but low river discharge and other inputs suggests that there is a significant flux of submarine groundwater discharge (SGD) in this area. By using a radium mass balance model, we estimated the SGD flux in the area to be (0.7-1.4) × 10 9  m 3 d -1 (6.1-12 cm d -1 ), at the high end of SGD fluxes worldwide. Geographically, Subei Shoal is less than 5% of the entire Southern Yellow Sea area, while our calculated SGD flux just for the shoal area is ~3 times larger than previously documented for the whole Southern Yellow Sea. Therefore, Subei Shoal may be considered a SGD hotspot that plays an important role in SGD associated material fluxes. Compared to inputs from local rivers, atmospheric deposition, and anthropogenic activities, SGD-derived nutrients are the main source term that can support the growth of macroalgae. The Changjiang (Yangtze River) Diluted Water (CDW) plume substantially impacts the biogeochemical processes off the estuary and its adjacent area, resulting in considerable environmental and ecological effects. Based on survey data in the northeastern area off the Changjiang Estuary (CE) obtained in the summers of 2008 and 2013, the hypoxia induced by the offshore detached CDW plume and the associated controlling mechanisms were investigated. The results show that the offshore transport of the CDW plume caused a dispersed low-salinity area in the northeastern area off the CE during summer, in sharp contrast with the surrounding high-salinity and high-density waters. There was a hypoxic area with low-pH (i.e., acidification) near the 40-m isobath in bottom waters in the northeastern area off the CE, and its position generally corresponded to the surface offshore CDW plume. In the area affected by the offshore low-salinity water, the surface patch-like phytoplankton bloom and the organic debris produced in situ were the material drivers of the bottom oxygen consumption and led to the corresponding relationship between the bottom hypoxic zone and the high chlorophyll-a (Chl-a) area at the surface. We consider that the local stratification caused by the offshore low-salinity water and the stable environment within the detached CDW plume constituted the external dynamic conditions for maintaining the bottom hypoxia. Our results demonstrate that the offshore detached CDW plume in the northeastern area off the CE may contribute to the formation of a local hypoxic center with low pH. Dissolved oxygen (DO) decline (i.e., deoxygenation) is an ongoing process in parts of the coastal and open oceans as a result of increased greenhouse gas emissions and nutrient discharges. Yet its controlling mechanisms remain unclear and patchy. Based on continuous observational data collected in a temperate margin, the southern Yellow Sea (SYS), we quantitatively evaluate how deoxygenation responds to warming and eutrophication in different seasons by using an evaluation method that allows us to distinguish the effects of temperature, salinity and biological activities. Results show that during winter, when the water column is vertically well-mixed, and in summer surface waters, deoxygenation is dominated by warming-induced decreases of O 2 solubility due to a quick exchange of O 2 between the ocean and atmosphere. Moreover, we find a regionally accelerated deoxygenation with enhanced warming along the pathway of the YSCC (Yellow Sea Coastal Current) in winter. In contrast, for bottom waters in summer when O 2 exchange is inhibited due to high stratification, deoxygenation appears to be dominated by biological respiration associated with eutrophication. Also, we find the summer bottom deoxygenation can be accelerated by warming, indicating that the bottom waters or the hypolimnion may be vulnerable to deoxygenation in the future. Our study further demonstrates that the deoxygenation mechanisms in shallow coastal oceans are associated with water column structures, i.e., well-mixed vs. stratified water column. The biogeography of active microbial communities and the underlying mechanisms in marine sediments are important in microbial ecology but remain unclear. Here, using qPCR and high-throughput sequencing, we investigated bacterial and archaeal community abundances and activities by quantifying the abundance and expression of the 16S rRNA gene respectively, RNA-derived bacterial and archaeal community biogeography, assembly mechanisms and co-occurrence relationships in surface sediment samples from the Bohai Sea (BS), South Yellow Sea (SYS) and the north East China Sea (NECS) of the eastern Chinese marginal seas. The results revealed a higher heterogeneity of bacterial and archaeal community activities than of abundances and heterogeneous ecological functions among areas reflected by community compositions. Furthermore, clear geographic groups (i.e., the BS, SYS and NECS groups) were observed for all, abundant and rare active bacterial and archaeal communities, accompanied by significant distance-decay patterns. However, the abundant and rare taxa showed inconsistent geographic patterns. More importantly, deterministic processes played a greater role than stochastic processes in active bacterial and archaeal community assembly. The rare taxa had weaker abilities to disperse and/or adapt and more complex ecological processes than the abundant taxa. In addition, this study also showed that intertaxa competition was the dominant interaction between active bacterial and archaeal members, which could greatly contribute to dispersal limitation. Based on field survey in the southwestern Yellow Sea (SWYS) during April-September 2017, the spatiotemporal variations in the hydrological characteristics and nutrient conditions were coupled and analyzed; the intra-seasonal variations in the upwelling in the front of the Yellow Sea Cold Water Mass (YSCWM) and impacts on nutrient transport were explored. The coastal area was controlled by the low-salinity high-nutrient Lubei Coastal Current, Subei Coastal Current, and Yangtze River Diluted Water from north to south; at bottom, the northeastern SWYS was controlled by the low-temperature high-salinity high-nutrient YSCWM. Temperature, salinity and nutrient fronts formed around YSCWM. The upwelling velocity in the front increased during April to late June and decreased in early September; the upwelled fluxes of dissolved inorganic nitrogen (0.29×10 3 -7.77×10 3 μmol·m -2 d -1 ), phosphate (0.02×10 3 -0.27×10 3 μmol·m -2 d -1 ) and silicate (0.98×10 3 -8.75×10 3 μmol·m -2 d -1 ) showed similar variations during April-September. The Changjiang (Yangtze) River Estuary (CJE) is one of the largest and most intense seasonal hypoxic zones in the world. Here we examine the possibility that submarine groundwater discharge (SGD) may contribute to the summer hypoxia. Spatial distributions of bottom water 222 Rn suggest a hotspot discharge area in the northern section of the CJE. SGD fluxes were estimated based on a 222 Rn mass balance model and were found to range from 0.002 ± 0.004 to 0.022 ± 0.011 m 3 /m 2 /day. Higher SGD fluxes were observed during summer hypoxia period. The well-developed overlap of the distribution patterns for SGD flux and dissolved oxygen (DO) implies that SGD could be an important contributor to summer hypoxia in the region off the CJE. Anthropogenic activities have greatly accelerated phosphorus (P) inputs from land to coastal seas. The increased P inputs from major rivers can cause adjacent coastal areas to experience seasonal hypoxia with the enhancing coastal eutrophication, which can subsequently increase P cycling and alter long term preservation. Analysis of sediment core measurements including SEDEX P speciation coupled with diagenetic kinetic models were performed on two cores in the coastal area under the Changjiang river plume, that experiences seasonal hypoxia. It was found that the benthic flux of dissolved reactive phosphate (DRP) in the Changjiang Estuary (CJE) was higher than that of adjacent areas of the Chinese coastal shelf. Sedimentary phosphorus transformations of Fe-bound P and organic P resulted in the in-situ formation of authigenic P (probably apatite), which was the major form of reactive P buried in the sediment. P burial efficiency (PBE) was lower than that of the oxic Chinese shelf but higher than that of other seasonally hypoxic areas in the world away from major river inputs. An exponential relationship between PBE and bottom water dissolved oxygen was developed, which suggested a positive feedback mechanism of increased hypoxia increasing P recycling, and hence intensifying eutrophication. Two typical kanamycin-A (KAN-A) electrochemical aptamer-based sensors employing different signal transduction mechanisms were deliberately designed and constructed with a similar structure. One sensor (sensor-1) was based on the classical probe conformation changing mode (PCCM) with a methylene blue (MB) label used as an electrochemical tag; the other sensor (sensor-2) used the target-induced signal probe shifting (TISPS) method with a free MB label in the assay solution. The difference in signal transduction mechanisms resulted in big differences in basic electrochemical behavior and comprehensive sensing performance. The results show that both sensor types exhibit different electrochemical behavior in square wave voltammetry, cyclic voltammetry, and in sensitivity, with detection limits of 3.0 nM for sensor-1 and 60.0 pM for sensor-2 in buffer. When validated for practical and quantitative detection of tap water and milk samples, both sensing methods performed well with detection limits of <260 nM and measurement times of <40 min. In addition, accuracy was good with mean recoveries of 72.3-92.6% and precision was acceptable with a relative standard deviation (RSD) of ≤14.2%. Global food and water safety issues have prompted the development of highly sensitive, specific, and fast analytical techniques for food and water analysis. The electrochemical aptamer-based detection platform (E-aptasensor) is one of the more promising detection techniques because of its unique combination of advantages that renders these sensors ideal for detection of a wide range of target analytes. Recent research results have further demonstrated that this technique has potential for real world analysis of food and water contaminants. This review summaries the recently developed E-aptasensors for detection of analytes related to food and water safety, including bacteria, mycotoxins, algal toxins, viruses, drugs, pesticides, and metal ions. Ten different electroanalytical techniques and one opto-electroanalytical technique commonly employed with these sensors are also described. Based on field observations in the summer of 2006 and long-term series data, this paper investigates the spatiotemporal variations of hypoxia and associated physical-biogeochemical driving mechanisms in the Bohai Sea (BS), China. Results show that the benthic hypoxic zone is mainly distributed in the "V"-shaped trough region in the western BS, and it tends to form two hypoxic centers which generally correspond to the bottom cold-water core. The regional difference in the intensity of stratification has a significant impact on the spatial distribution of hypoxia. The relatively weak stratification and the mesoscale anticyclonic eddy in the central shoal of the BS weaken the connectivity between the southern and northern hypoxic zones. Organic matter decomposition contributes to hypoxia and results in corresponding nutrient pool with a "dual (southern and northern)-core" structure. The hydro-biogeochemical processes in the world's largest area of green tide occurrence, off the Jiangsu coast in the Yellow Sea, are investigated in the summer, and their implications for Ulva prolifera blooms are discussed. The results show that the offshore transport of coastal water and the inshore upwelling of offshore bottom water both occur off the Jiangsu coast, and the upwelling position is consistent with the 20- to 30-m isobath off the Subei Shoal. The upwelling results in nutrient supplementation off the shoal, where a rapid decrease in the suspended particulate matter content contributes to good light conditions. As a result, a high-value area of phytoplankton is formed within the 20- to 30-m isobath. Eutrophication in the shoal has provided nutrients for the frequent occurrence of Ulva prolifera in recent years, whereas the upwelling area off the shoal has served as a "service station" or "courier station" for floating Ulva prolifera and promoted the species' propagation. The propagation of Ulva prolifera in the upwelling area and its blooms within the Subei Shoal can have a spatially synergistic effect, leading to its large-scale development. Glioma is one of the most lethal brain malignancies with unknown etiologies. Many metabolomics analysis aiming at diverse kinds of samples had been performed. Due to the varied adopted analytical platforms, the reported disease-related metabolites were not consistent across different studies. Comparable metabolomics results are more likely to be acquired by analyzing the same sample types with identical analytical platform. For tumor researches, tissue samples metabolomics analysis own the unique advantage that it can gain more direct insight into disease-specific pathological molecules. In this light, a previous reported capillary electrophoresis - mass spectrometry human tissues metabolomics analysis method was employed to profile the metabolome of rat C6 cell implantation gliomas and the corresponding precancerous tissues. It was found that 9 metabolites increased in the glioma tissues. Of them, hypotaurine was the only metabolite that enriched in the malignant tissues as what had been reported in the relevant human tissues metabolomics analysis. Furthermore, hypotaurine was also proved to inhibit α-ketoglutarate-dependent dioxygenases (2-KDDs) through immunocytochemistry staining and in vitro enzymatic activity assays by using C6 cell cultures. The growing potential of quantum dots (QDs) in biomedical applications has raised considerable concerns regarding their toxicological impact. Consequently, there has been a need to understand the underlying toxicity mechanism of QDs. In this work, we comprehensively investigated the bioaccumulation and toxicity of three CdSe/ZnS QDs (COOH CdSe/ZnS 525, NH 2 CdSe/ZnS 525, and NH 2 CdSe/ZnS 625) in Phanerochaete chrysosporium (P. chrysosporium) using confocal laser scanning microscopy, reactive oxygen species (ROS) measurements, and cell viability assays. Confocal laser scanning microscopy analytical results indicated that all the CdSe/ZnS QDs, with the concentration ranging from 10 to 80nM, could accumulate largely in the hyphae and induce the generation of ROS, showing a direct toxicity to P. chrysosporium. And the bioaccumulation and toxicity of CdSe/ZnS QDs presented dose-dependent and time-dependent effects on P. chrysosporium. Furthermore, the CdSe/ZnS QDs-induced cytotoxicity was also related to their physicochemical properties, including particle size and surface charges: NH 2 CdSe/ZnS 525 with small size was more cytotoxic as compared to NH 2 CdSe/ZnS 625 with large size, and the smaller negative charged NH 2 CdSe/ZnS 525 resulted in greater cytotoxicity than the larger negative charged COOH CdSe/ZnS 525. Anthropogenic activities alter the natural states of large rivers and their surrounding environment. The Yellow River is a well-studied case of a large river with heavy human control. An artificial managed water and sediment release system, known as the Water-Sediment Regulation Scheme (WSRS), has been carried out annually in the Yellow River since 2002. Nutrient concentrations and composition display significant time and space variations during the WSRS period. To figure out the anthropogenic impact of nutrient changes and transport in the Yellow River, biogeochemical observations were carried out in both middle reaches and lower reaches of the Yellow River during 2014 WSRS period. WSRS has a direct impact on water oxidation-reduction environment in the middle reaches; concentrations of nitrite (NO 2 - ) and ammonium (NH 4 + ) increased, while nitrate (NO 3 - ) concentration decreased by enhanced denitrification. WSRS changed transport of water and sediment; dissolved silicate (DSi) in the middle reaches was directly controlled by sediments release during the WSRS while in the lower reaches, DSi changed with both sediments and water released from middle reaches. During the WSRS, the differences of nutrient fluxes and concentrations between lower reaches and middle reaches were significant; dissolved inorganic phosphorous (DIP) and dissolved inorganic nitrogen (DIN) were higher in low reaches because of anthropogenic inputs. Phosphorus (P) speciation, burial, and transformation are poorly constrained under low-oxygen conditions. Sequential chemical extraction techniques, in-situ incubation, and laboratory incubation were employed to explore P cycling in the low-oxygen area of coastal Rushan. The study determined that the total P concentrations in the coastal area of Rushan Bay were higher than those of other China shelf seas, and largely affected by anthropogenic activities. The phosphate (DRP) fluxes in the study area calculated using an incubation method (0-1960μmolm(-2)day(-)(1)) and measured based on pore water gradients (1.5-50.4μmolm(-2)day(-)(1)) were both highly correlated with oxygen conditions. Sediment incubations showed that DRP diffusion from the sediment mainly originates from Fe-P and Auth-P dissolution and that Org-P recycling contributed only a small portion of the total released P pool. The benthic phosphate flux can be 60 times higher under low bottom-water oxygen levels of 63-150μmolL(-1) than under oxygen levels exceeding 150μmolL(-1) in the study area. The P accumulation rates and burial efficiencies in this study area ranged from 16.5-33.3μmolcm(-2)year(-1) and 81.1-83.4%, respectively, and were regulated by the oxygen level and diffusive DRP flux. Suspended particulate matter (SPM) samples were collected along a salinity gradient in the Changjiang Estuary in June 2011. A custom-built water elutriation apparatus was used to separate the suspended sediments into five size fractions. The results indicated that Cr and Pb originated from natural weathering processes, whereas Cu, Zn, and Cd originated from other sources. The distribution of most trace metals in different particle sizes increased with decreasing particle size. The contents of Fe/Mn and organic matter were confirmed to play an important role in increasing the level of heavy metal contents. The Cu, Pb, Zn, and Cd contents varied significantly with increasing salinity in the medium-low salinity region, thus indicating the release of Cu, Pb, Zn, and Cd particles. Thus, the transfer of polluted fine particles into the open sea is probably accompanied by release of pollutants into the dissolved compartment, thereby amplifying the potential harmful effects to marine organisms., (Copyright © 2015 Elsevier Ltd. This study investigated the performance of immobilized Phanerochaete chrysosporium loaded with nitrogen-doped TiO2 nanoparticles in the treatment of raw landfill leachate with a very low biodegradability ratio (BOD5/COD) of 0.09. The effects of various operating parameters, such as initial chemical oxygen demand (COD) concentration, pH, temperature, and biosorbent dosage, were evaluated with respect to the removal efficiency of total organic carbon (TOC) and ammonia nitrogen (NH3-N). For the immobilized biosorbents, an optimum pH of 6.0 for TOC and 7.0 for NH3-N were found suitable for TOC and NH3-N removal at temperature of 37°C, respectively. The most superior removal efficiencies of TOC and NH3-N of landfill leachate were over 75% and 74% in 72 h at an initial COD concentration of 200 mg L(-1), respectively. In addition, heavy metals were partly removed by the immobilized biosorbents during the process of landfill leachate treatment. The species and mass percentage of organic compounds in landfill leachate after the treatment were found to have considerably declined according to the gas chromatography coupled with mass spectrometry (GC-MS) system. Introduction: Phosphodiesterase 10A (PDE10A) is an intracellular enzyme responsible for the breakdown of cyclic nucleotides which are important second messengers for neurotransmission. Inhibition of PDE10A has been identified as a potential target for treatment of various neuropsychiatric disorders. To assist drug development, we have identified a selective PDE10A positron emission tomography (PET) tracer, AMG 580. We describe here the radiosynthesis of [(18)F]AMG 580 and in vitro and in vivo characterization results., Methods: The potency and selectivity were determined by in vitro assay using [(3)H]AMG 580 and baboon brain tissues. [(18)F]AMG 580 was prepared by a 1-step [(18)F]fluorination procedure. Dynamic brain PET scans were performed in non-human primates. Regions-of-interest were defined on individuals' MRIs and transferred to the co-registered PET images. Data were analyzed using two tissue compartment analysis (2TC), Logan graphical (Logan) analysis with metabolite-corrected input function and the simplified reference tissue model (SRTM) method. A PDE10A inhibitor and unlabeled AMG 580 were used to demonstrate the PDE10A specificity. KD was estimated by Scatchard analysis of high and low affinity PET scans., Results: AMG 580 has an in vitro KD of 71.9 pM. Autoradiography showed specific uptake in striatum. Mean activity of 121 ± 18 MBq was used in PET studies. In Rhesus, the baseline BPND for putamen and caudate was 3.38 and 2.34, respectively, via 2TC, and 3.16, 2.34 via Logan, and 2.92, and 2.01 via SRTM. A dose dependent decrease of BPND was observed by the pre-treatment with a PDE10A inhibitor. In baboons, 0.24 mg/kg dose of AMG 580 resulted in about 70% decrease of BPND. The in vivo KD of [(18)F]AMG 580 was estimated to be around 0.44 nM in baboons., Conclusion: [(18)F]AMG 580 is a selective and potent PDE10A PET tracer with excellent specific striatal binding in non-human primates. With the help of the interaction mode of capture probe-target-signal probe (CP-T-SP), an electrochemical sensing method based on the synergism strategy of dual-hybridized signaling probes modified with 6 MB (methylene blue), background suppression and large surface area Au electrode is developed for the detection of Streptococcus pneumoniae (S. pneumoniae) Lyt-1 gene sequence. The proposed sensor features a very low detection limit (LOD) of ∼0.5 fM for the target. In this study, sediment grain size (MGS), specific surface area (SSA), total organic carbon (TOC) contents, C/N molar ratios, stable carbon isotope, and P species in a sediment core, collected from the East China Sea (ECS) inner-shelf were measured to explore the applicability of detrital phosphorus (De-P) as a potential indicator of past flooding events in the Changjiang River Basin (CRB). In particular, we examined the linkages between the evolution of floods with regional climate changes and anthropogenic activities in the CRB. Peaks of De-P concentrations in sediments corresponded well with the worst flooding events of the CRB over the past two centuries (e.g., 1850s, 1860s, 1900s, 1920s, 1950s, 1980s, and 2000s). Moreover, De-P also corresponded well with the extreme hypoxic events in 1981 and 1998 in the Changjiang Estuary as indicated by Mo/Al ratios, indicating potential linkages between De-P as a flooding proxy to flood-induced hypoxia events in this region. In addition, a robust relationship was found among De-P, the floods in 1950s, 1980s, and 2000s of the CRB, the intensive El Niño-Southern Oscillation (ENSO), the abnormally weak East Asian Summer Monsoon (EASM) and the warm phase of Pacific Decadal Oscillation (PDO), suggesting that De-P also provided insights to linkages between regional climate change and flooding events in this region., (Copyright © 2015 Elsevier B.V. ARRY-403 is a glucokinase activator developed for the treatment of diabetes. Less than dose-proportional exposure was observed during single ascending dose studies with ARRY-403. A physiologically based pharmacokinetic (PBPK) model for ARRY-403 was developed through integration of in vitro physicochemical data with precipitation time estimations based on results from the single ascending dose studies; PBPK modeling indicated that the primary cause of the less than dose-proportional exposure was dose-limited absorption because of pH-dependent solubility. The impact of dose, particle size, and fasted or fed state on ARRY-403 exposure was examined through sensitivity analyses and used to refine the PBPK model. On the basis of the marked pH-dependent solubility of ARRY-403, the refined PBPK model was used to simulate the effects of acid-reducing agents (ARAs) on ARRY-403 exposure, as these agents are widely available and could be coadministered with ARRY-403. The simulations indicated that a clinical study with an ARA was warranted; in a clinical study, famotidine had a marked effect on ARRY-403 exposure. After damming of the Yangtze River, in order to explore the impacts of the Three-Gorge Dam (TGD) on the aquatic ecosystem, phytoplankton composition, abundance and biomass spatial distribution were studied in the Three-Gorge Reservoir (TGR), and the closest upstream anabranch Xiangxi River, which is 38 km away from the Three-Gorge Dam (TGD) during August (rainy season) 2004 and April (dry season) 2005. In surveys, 6 transects (2 downstream and 4 cross-stream) and 25 stations have been investigated and 314 samples were collected from the surface to the river bed with water samplers. In TGR, 63 taxa and 60 taxa were identified in the rainy and dry seasons, respectively. In the Xiangxi River, 39 taxa were observed in the rainy and dry seasons. Algal blooms occurred in the Xiangxi River and at the influx region of the Yangtze and Xiangxi in both seasons, but had not occurred prior to damming. In the rainy season, the dominant species was Chroomonas acuta with 1.84x10(7) cells l-1, and in the dry season the dominant species were Asterionella formosa and Cryptomonas ovata with 1.34x10(7) cells l-1 and 1.79x10(6) cells.l-1, respectively. In the main channel of TGR, there were no significant correlations between phytoplankton abundance and the concentrations of the main soluble nutrients. In the Xiangxi River, significant negative correlations were observed between phytoplankton abundance and nitrate (Spearman, p<0.01, n=21), phosphate (Spearman, p<0.05, n=21) and silicate (Spearman, p<0.01, n=21) in the rainy season, and similar correlations were also observed with nitrate (Spearman, p<0.05, n=28) and silicate (Spearman, p<0.01, n=28), but not with phosphate in the dry season. Since the damming of the Yangtze River, eutrophication in the anabranch within the backwater has occurred and become severe, and the frequency of algal bloom within TGR and anabranches is expected to increase.

An unresolved question in the study of the polyglutamine neurodegenerative disorders is the extent to which partial loss of normal function of the mutant protein contributes to the disease phenotype. To address this, we studied Kennedy disease, a degenerative disorder of lower motor neurons caused by a CAG/glutamine expansion in the androgen receptor (Ar) gene. Signs of partial androgen insensitivity, including testicular atrophy and decreased fertility, are common in affected males, although the underlying mechanisms are not well understood. Here, we describe a knock-in mouse model that reproduces the testicular atrophy, diminished fertility, and systemic signs of partial androgen insensitivity that occur in Kennedy disease patients. Using this model, we demonstrate that the testicular pathology in this disorder is distinct from that mediated by loss of AR function. Testes pathology in 113 CAG knock-in mice was characterized by morphological abnormalities of germ cell maturation, decreased solubility of the mutant AR protein, and alterations of the Sertoli cell cytoskeleton, changes that are distinct from those produced by AR loss-of-function mutation in testicular feminization mutant mice. Our data demonstrate that toxic effects of the mutant protein mediate aspects of the Kennedy disease phenotype previously attributed to a loss of AR function.
