Background: Cardiovascular diseases (CVDs) remain the leading cause of global mortality, with dyslipidemia recognized as a significant risk factor. Dyslipidemia, characterized by elevated levels of total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and triglycerides (TG), is associated with an increased lifetime risk of CVD. Although alfalfa’s cholesterol-lowering effects have been demonstrated in animal models, clinical trial data supporting its efficacy in human dyslipidemia treatment are limited. Methods: A randomized controlled trial was conducted from September 20, 2021, to January 12, 2023, involving patients diagnosed with dyslipidemia. Approved by the China Ethics Committee of Registering Clinical Trials, the study included multiple sites in China. Eligible participants were adults aged 18 years or older, meeting criteria from the Chinese Guidelines for the Prevention and Treatment of Adult Blood Lipid Disorders. Participants were randomly assigned to either a health education-only group or a combined alfalfa powder supplementation and health education group for a three-month intervention period. Results: A total of 160 eligible patients were enrolled and randomized into two groups: the Alfalfa group (n = 80) and the Education group (n = 80). The intervention resulted in significant improvements in key lipid parameters in the Alfalfa group, including reductions in TC, LDL-C, and Apo A1 levels. Although changes in HDL-C levels were not significantly different between the groups, the Alfalfa group exhibited a larger increase. Serum lipid metabolomics analysis using liquid chromatography-tandem mass spectrometry (LC-MS/MS) indicated that the combined intervention did not significantly alter the overall blood lipid metabolome but identified fifteen regulated differential metabolites, with the glycerophospholipid metabolism pathway significantly affecting lipid metabolism levels. Conclusion: Supplementation with alfalfa powder combined with health education significantly reduced TC, LDL-C, and Apo A1 levels in patients with dyslipidemia. KEGG pathway analysis suggests an association between the glycerophospholipid metabolism pathway and dyslipidemia. Further research is warranted to validate these findings and explore the underlying mechanisms.

Power flow is an extensively used tool in various operations and planning of power systems. In this paper, we propose an efficient hybrid method for solving the power flow problems in ill-conditioned power systems. This method is implemented in a framework of two-part alternating iteration. In Part I of this method, PV bus voltages are regarded as constant and PQ bus voltages are updated by the Z-bus Gauss method. In Part II of this method, PQ bus loads are equivalent to dynamic admittances, and PV bus voltage angles are updated by Newton’s method. The values of PQ bus voltage magnitudes and PV bus voltage angles are passed between Part I and Part II. The proposed method is validated in well- and ill-conditioned systems and compared with several well-known power flow methods. Results show that the proposed method is robust and efficient to address the issues related with large-scale ill-conditioned power systems and it is not significantly affected by the considered initial guess.

A growing number of clinical and animal studies suggest that the nucleus accumbens (NAc), especially the shell, is involved in the pathogenesis of temporal lobe epilepsy (TLE). However, the role of parvalbumin (PV) GABAergic neurons in the NAc shell involved in TLE is still unclear. In this study, we induced a spontaneous TLE model by intrahippocampal administration of kainic acid (KA), which generally induce acute seizures in first 2 h (acute phase) and then lead to spontaneous recurrent seizures after two months (chronic phase). We found that chemogenetic activation of NAc shell PV neurons could alleviate TLE seizures by reducing the number and period of focal seizures (FSs) and secondary generalized seizures (sGSs), while selective inhibition of PV exacerbated seizure activity. Ruby-virus mapping results identified that the hippocampus (ventral and dorsal) is one of the projection targets of NAc shell PV neurons. Chemogenetic activation of the NAc-Hip PV projection fibers can mitigate seizures while inhibition has no effect on seizure ictogenesis. In summary, our findings reveal that PV neurons in the NAc shell could modulate the seizures in TLE via a long-range NAc-Hip circuit. All of these results enriched the investigation between NAc and epilepsy, offering new targets for future epileptogenesis research and precision therapy.

Water Resourcing in China has historically been a complex issue requiring the ability to deal with regular floods, droughts and diverse water needs. Climate change represents another challenge to this sector, albeit one that is not traditionally considered by water managers. In this sector in China water management is predominantly based on historic, seasonal and annual forecast data while multi-annual and (multi-)decadal data are seldom used. In this paper, we present the co-development of a climate service prototype designed to provide water managers with insights into the impacts of climate change on the Upper Yellow River region for the next century. The paper is an outcome from our project that encouraged water resource planners and water resource managers to utilise long-term climate information to understand the uncertainties and the challenges our changing climate is likely to have in the region. Using an interdisciplinary team and adopting a user-centred, co-production approach, a prototype web-based data visualisation tool was developed. The development of the prototype was based on a design specification constructed from the findings of detailed interviews that allowed it to be developed and tested under SARS-CoV-2 pandemic restrictions that prevented the typical development process to be undertaken. The developed prototype presents climate information and communicates uncertainties regarding climate change in the remainder of the century through data sets that are typically used by the water sector in China in a simple, easy to understand style. Models that estimate river levels under different extraction scenarios and results about estimated river level and flow, and flood risk are also presented. The prototype was shown to be successful, as key messages relating to the impact of climate change and the challenges for water resource management could be effectively communicated through the tool interface. Practical implications: Understanding the impacts of climate change on water resourcing is complicated and multifaceted. There is a need for better data about what water there is and how it is moving around between and within catchments. Estimates of past, present and future climate variables along with historical measurements of river flow can be used to help visualise some of the uncertainties and changes that may happen in the next 50 years. In addition, there is a need to understand changing water demands and water resource management practices. Current water resource management practices are based on historical conditions and assumptions that are less likely to hold true in a more variable and warmer climate. Communicating how future changes will impact future water resourcing is critical to water resources in a changing climate (Belcher et al. 2018).This research outlines the construction of a tool to visualise the impacts of climate change on water availability in part of China that is typically water scarce, using models developed using the Soil Water Assessment Toolkit (SWAT). A model of the Upper Yellow River (UYR) was developed to demonstrate the impact of climate change on river levels in the catchment based on climate variables. The rainfall-runoff model was based on climate predictions from the CMIP5 assessment HadGEM3-GC3.05 climate model and incorporated information about water resource allocations for different administrative regions of the catchment The general climate trend for the region is that it is expected to become significantly warmer. The total amount of precipitation is likely to be about the same, and yet it is expected that overall, the catchment will become significantly drier over time as winter shortens and summer lengthens. The outputs from the model reflect the changes in climate variables. The uncertainties were communicated via a Web based tool. Water resource managers in China helped to coproduce the tool by participating in workshops and providing feedback on prototypes. The workshops helped scientists and water resource managers to communicate about climate change impacts on water resources and water resource management.

Microbes are critical for flavor formation in fermented foods; however, their mechanisms of action are not fully understood. The microbial composition of 51 dairy and 47 vegetable products was functionally annotated and the carbohydrate-active enzyme (CAZyme) profiles of Lactiplantibacillus plantarum 84-3 (Lp84-3), isolated from dairy samples, can promote resistant starch (RS) degradation, were analyzed. Lactobacillus, Streptococcus, and Lactococcus were the predominant genera in dairy products, whereas the major genera in vegetables were Lactobacillus, Weissella, and Carnimonas. Phages from Siphoviridae, Myoviridae, and Herelleviridae were also present in dairy products. Additionally, the glycosyl hydrolase (GHs) family members GH1 and GH13 and the glycosyltransferase (GTs) family members GT2 and GT4 were abundant in Lp84-3. Moreover, Lp84-3 was enriched in butanoate metabolism enzymes and butanoate metabolite compounds. Therefore, fermented food microbes, especially Lp84-3, have an abundant repertoire of enzymes that promote flavor production, as starter improving the flavor of fermented dairy and vegetable products.

Summary: Mosaic symptoms are commonly observed in virus-infected plants. However, the underlying mechanism by which viruses cause mosaic symptoms as well as the key regulator(s) involved in this process remain unclear. Here, we investigate maize dwarf mosaic disease caused by sugarcane mosaic virus (SCMV). We find that the manifestation of mosaic symptoms in SCMV-infected maize plants requires light illumination and is correlated with mitochondrial reactive oxidative species (mROS) accumulation. The transcriptomic and metabolomic analyses results together with the genetic and cytopathological evidence indicate that malate and malate circulation pathways play essential roles in promoting mosaic symptom development. Specifically, at the pre-symptomatic infection stage or infection front, SCMV infection elevates the enzymatic activity of pyruvate orthophosphate dikinase by decreasing the phosphorylation of threonine527 under light, resulting in malate overproduction and subsequent mROS accumulation. Our findings indicate that activated malate circulation contributes to the manifestation of light-dependent mosaic symptoms via mROS.

Objective: functional gastroduodenal disease is the main type of functional gastrointestinal disease in the clinical department of Gastroenterology and psychosomatic medicine at present, which accounts for a large proportion of outpatients in gastroenterology. The main manifestations are epigastric pain, dyspepsia, belching, chronic nausea, and vomiting. The purpose of this study is to explore the changes in brain function in patients with functional gastroduodenal diseases through experiments to reveal the possible central etiology and development process. Methods: the functional changes of the prefrontal lobe in patients with functional gastroduodenal diseases and normal controls were detected and analyzed by near-infrared brain imaging. At the same time, SCL-90 was used to evaluate the mental health status of patients with functional gastroduodenal diseases and normal controls. The changes in the autonomic nerve system in patients and normal controls were detected and compared by heart rate variability trend chart. Results: the activity of left prefrontal lobe areas s8-d8, s10-d4, s10-d10 and s10-d15 in patients with functional gastroduodenal disease was significantly lower than normal controls (p 0.05). Conclusion: the function of the left frontal lobe is decreased in patients with functional gastroduodenal disease. The autonomic nervous system may be related to the connection system between the brain center and internal organs.

Although the complete nucleotide sequence of strawberry vein banding virus (SVBV) has been determined and bioinformatic analysis has revealed that the SVBV genome could encode seven proteins, the precise function of each protein is unclear. This study provided evidence that the P1 protein of SVBV (SVBV-P1) possesses the following features. Bioinformatic and subcellular localization analyses showed that SVBV-P1 is localized in the cytoplasm and cell walls of epidermal cells in Nicotiana benthamiana, and it forms inclusion bodies associated with microtubules and the endoplasmic reticulum. Dilution experiments demonstrated that SVBV-P1 could move from the original agro-infiltrated cells to adjacent cells in N. benthamiana leaves. Further trans-complementation experiments demonstrated that SVBV-P1 could facilitate the intercellular movement of a movement-deficient potato virus X mutant in N. benthamiana leaves. Finally, yeast two-hybrid and bimolecular fluorescence complementation assays revealed that SVBV-P1 could interact with the SVBV coat protein, which is a major component of Caulimovirus virions. Results of the electrophoretic mobility shift assay indicated that SVBV-P1 lacks DNA-binding capability. In summary, the results suggest that SVBV-P1 is probably a movement protein of SVBV, providing new insights into the function of movement proteins of the Caulimovirus genus.

Bread was prepared using wheat flour with grape seed proanthocyanidin (GSP) (0.4%). GSP improved the textural properties of bread including hardness, cohesiviness, gumminess and chewiness. At the last stage of fermentation, GSP reinforced the gluten microstructure with increased the disulfide bonds and hydrophobic interaction and α-helix in the secondary structures. Moreover, GSP addition could increase the total phenolics and antioxidative acitivity of the bread significantly. In addition, the degree of fermentation had a strong influence on the dough forces, and the reasonable control of bread fermentation time was beneficial to improve the bread quality, which provided a reference for the bread processing industry.

Background: Ginsenoside Rh2 (GRh2) is a characterized component in red ginseng widely used in Korea and China. GRh2 exhibits a wide range of pharmacological activities, such as anti-inflammatory, antioxidant, and anticancer properties. However, its effects on Toxoplasma gondii (T. gondii) infection have not been clarified yet. Methods: The effect of GRh2 against T. gondii was assessed under in vitro and in vivo experiments. The BV2 cells were infected with tachyzoites of T. gondii RH strain, and the effects of GRh2 were evaluated by MTT assay, morphological observations, immunofluorescence staining, a trypan blue exclusion assay, reverse transcription PCR, and Western blot analyses. The in vivo experiment was conducted with BALB/c mice inoculated with lethal amounts of tachyzoites with or without GRh2 treatment. Results and conclusion: The GRh2 treatment significantly inhibited the proliferation of T. gondii under in vitro and in vivo studies. Furthermore, GRh2 blocked the activation of microglia and specifically decreased the release of inflammatory mediators in response to T. gondii infection through TLR4/NF-κB signaling pathway. In mice, GRh2 conferred modest protection from a lethal dose of T. gondii. After the treatment, the proliferation of tachyzoites in the peritoneal cavity of infected mice markedly decreased. Moreover, GRh2 also significantly decreased the T. gondii burden in mouse brain tissues. These findings indicate that GRh2 exhibits an anti–T. gondii effect and inhibits the microglial activation through TLR4/NF-κB signaling pathway, providing the basic pharmacological basis for the development of new drugs to treat toxoplasmic encephalitis.

China is one of the countries where landslides caused the most fatalities in the last decades. The threat that landslide disasters pose to people might even be greater in the future, due to climate change and the increasing urbanization of mountainous areas. A reliable national-scale rainfall induced landslide susceptibility model is therefore of great relevance in order to identify regions more and less prone to landsliding as well as to develop suitable risk mitigating strategies. However, relying on imperfect landslide data is inevitable when modelling landslide susceptibility for such a large research area. The purpose of this study is to investigate the influence of incomplete landslide data on national scale statistical landslide susceptibility modeling for China. In this context, it is aimed to explore the benefit of mixed effects modelling to counterbalance associated bias propagations. Six influencing factors including lithology, slope, soil moisture index, mean annual precipitation, land use and geological environment regions were selected based on an initial exploratory data analysis. Three sets of influencing variables were designed to represent different solutions to deal with spatially incomplete landslide information: Set 1 (disregards the presence of incomplete landslide information), Set 2 (excludes factors related to the incompleteness of landslide data), Set 3 (accounts for factors related to the incompleteness via random effects). The variable sets were then introduced in a generalized additive model (GAM: Set 1 and Set 2) and a generalized additive mixed effect model (GAMM: Set 3) to establish three national-scale statistical landslide susceptibility models: models 1, 2 and 3. The models were evaluated using the area under the receiver operating characteristics curve (AUROC) given by spatially explicit and non-spatial cross-validation. The spatial prediction pattern produced by the models were also investigated. The results show that the landslide inventory incompleteness had a substantial impact on the outcomes of the statistical landslide susceptibility models. The cross-validation results provided evidence that the three established models performed well to predict model-independent landslide information with median AUROCs ranging from 0.8 to 0.9. However, although Model 1 reached the highest AUROCs within non-spatial cross-validation (median of 0.9), it was not associated with the most plausible representation of landslide susceptibility. The Model 1 modelling results were inconsistent with geomorphological process knowledge and reflected a large extent the underlying data bias. The Model 2 susceptibility maps provided a less biased picture of landslide susceptibility. However, a lower predicted likelihood of landslide occurrence still existed in areas known to be underrepresented in terms of landslide data (e.g., the Kuenlun Mountains in the northern Tibetan Plateau). The non-linear mixed-effects model (Model 3) reduced the impact of these biases best by introducing bias-describing variables as random effects. Among the three models, Model 3 was selected as the best national-scale susceptibility model for China as it produced the most plausible portray of rainfall induced landslide susceptibility and the highest spatially explicit predictive performance (median AUROC of spatial cross validation 0.84) compared to the other two models (median AUROCs of 0.81 and 0.79, respectively). We conclude that ignoring landslide inventory-based incompleteness can entail misleading modelling results and that the application of non-linear mixed-effect models can reduce the propagation of such biases into the final results for very large areas.

Strawberry vein banding virus (SVBV)-infected strawberry cells contain cytoplasmic inclusions with isometric particles. To identify the components of the inclusions, green fluorescent protein (GFP) was fused to the carboxy-terminus (C-terminus) of SVBV open reading frames, these constructs were separately transformed into Agrobacterium tumefaciens and infiltrated into Nicotiana benthamiana leaves. Results showed that the SVBV P6 protein assembled into prominent and amorphous inclusion bodies (IBs). To investigate P6 subcellular localization, P6-GFP was ectopically expressed in N. benthamiana leaves by agroinfiltration and then stained with 4’,6-diamidino-2-phenylindole (DAPI). We found the P6 protein accumulated in the nuclei and also formed cytoplasmic IBs with different sizes. To further determine the location of P6 IBs in the cytoplasm, and explore whether the P6 IBs move freely or depend on cytoskeleton and endoplasmic reticulum (ER), the microfilament marker protein (GFP-ABD2-GFP), microtubules marker protein (mCherry-MAP65-1) and ER marker protein (mCherry-HDEL) were separately coexpressed with P6-GFP and into N. benthamiana leaves by agroinfiltration, exhibiting that P6 IBs aligned with cytoskeleton and endoplasmic reticulum. Meanwhile, coinfiltration of P1 and P6 indicated the P6 colocalized with the P1 protein at periphery of cells. The P6 protein contains one C-terminal nuclear localization signal (NLS) region, a P6 protein mutant with a deleted NLS did not localize in the nucleus, did not form IBs, and was unable to facilitate exogenous GFP expression. These results demonstrate that the deleted NLS region is an important P6 domain required for biological functions. In summary, the mobile P6 IBs are associated with ER, microfilaments and microtubules and move along microfilaments to the SVBV P1 protein in the PD.

Long-term high sugar accelerates skin ageing via glycosylation. In this study, microalgae-derived active compounds were investigated for their potentially beneficial effects on skin ageing. Two carotenoids, namely astaxanthin (in the form of either monoester or diester) and lutein, were separated from the green alga Haematococcus pluvialis. These carotenoid fractions significantly attenuated the high glucose (HG)-induced cell injury and oxidative stress. They also inhibited the formation of Nε-Carboxymethyllysine (CML), a typical product of glycosylation. The degradation of collagen I and III caused by HG was substantially recovered, in which astaxanthin monoester exhibited the most prominent effect. This was attributed to the down-regulation of matrix metalloproteinase (MMP)-2 mRNA expression. Through proteomics analyses, six hub proteins were identified, which work as highly connected nodes in the protective effects of astaxanthin monoester against HG stress. These findings strongly suggest the benefits of microalgae-derived carotenoids in alleviating HG-induced skin ageing.

Objectives: To explore the effectiveness of the mobile app-based multidisciplinary exercise management on patients who receive percutaneous coronary intervention (PCI). Methods: From January to October 2020, 54 patients after PCI were randomly assigned to the intervention group (n = 27) and the control group (n = 27). The intervention group received the mobile app-based multidisciplinary exercise management, whereas the control group received routine care. The patients after PCI began to take intervention one month after the operation, and the intervention lasted for two months. Before and after the intervention, 6-Minute Walking Distance was used to evaluate the patient’s exercise tolerance, and the patient’s exercise compliance was evaluated according to the patient’s exercise status recorded by the mobile app. The cognitive questionnaire on knowledge about PCI treatment for Coronary Heart Disease, the Self-efficacy for Chronic Disease Scale and the Perceived Social Support Scale were used to evaluate patients’ disease-related cognition, self-efficacy and perception of social support. This study was registered on Clinical Trials.gov with registration number ChiCTR2000028930. Results: Totally 51 patients after PCI who completed this study (25 patients in the intervention group and 26 patients in the control group) were included in the analysis. After 2 months of intervention, the exercise compliance of patients in the intervention group was better than that in the control group. And 6-Minute Walking Distance (469.36 ± 57.48 vs. 432.81 ± 67.09), and the scores of knowledge of PCI treatment for coronary heart disease (52.64 ± 9.82 vs. 42.42 ± 8.54), Self-efficacy for Chronic Disease Scale (42.40 ± 8.04 vs. 36.88 ± 7.73) and Perceived Social Support Scale (74.04 ± 5.73 vs. 66.69 ± 6.86) in the intervention group were higher than those in the control group with statistical significance (P

Coal fly ash, a solid waste from powder plant, was fully used to produce sintered glass-ceramics by direct sintering in air after dry pressing at 10 MPa. The influence of particle size on the sinterability and properties of sintered glass-ceramics had been systematically discussed. Compared with CFA (d50 = 12.6 μm), sintered glass-ceramics issued from 20CFA (d50 = 7.5 μm) and 40CFA (d50 = 4.9 μm) had higher bulk density, higher linear shrinkage and lower porosity. The sintering activation energy of the sintered glass-ceramics obtained from the three sizes of coal fly ash (CFA, 20CFA and 40CFA) were estimated by bulk density and linear shrinkage, which were 127, 102, 81 kJ/mol and 124, 106, 83 kJ/mol respectively, meaning the smaller the particle size, the easier the sintering. According to the XRD analysis, anorthite was formed by the reaction of portion dissolved quartz with the amorphous metastable glassy phase in coal fly ash. The smaller the particle size of coal fly ash is, the easier the formation of anorthite. Moreover, the properties (water absorption, flexural strength and chemical resistance) of the sintered glass-ceramics were enhanced with the decrease of the particle size and reached the standards of architectural decoration materials. This study examines the bioleaching of municipal solid waste incineration fly ash by Aspergillus niger, and its effect on the fungal morphology, the fate of the ash particles, and the precipitation of metallic salt crystals during bioleaching. The fungal morphology was significantly affected during one-step and two-step bioleaching; scanning electron microscopy revealed that bioleaching caused distortion of the fungal hyphae (with up to 10 μm hyphae diameter) and a swollen pellet structure. In the absence of the fly ash, the fungi showed a linear structure (with 2–4 μm hyphae diameter). Energy-dispersive X-ray spectroscopy and X-ray diffraction confirmed the precipitation of calcium oxalate hydrate crystals at the surface of hyphae in both one-step and two-step bioleaching. Calcium oxalate precipitation affects bioleaching via the weakening of the fly ash, thus facilitating the release of other tightly bound metals in the matrix.

UDP-glucuronosyltransferase (UGT) 1A1, one of the most important isoforms in UGTs superfamily, has attracted increasing concerns for its special role in the clearance and detoxification of endogenous and exogenous substances. To avoid the clinical drug-drug interactions, it is of great importance to have the knowledge of the metabolic profile of UGT1A1 substrates early. Herein, we purposed to establish machine learning models to predict the metabolic propeties of UGT1A1 substrates. On the basis of the literature-derived substrates database of UGT1A1, automatic metabolism prediction models for the aromatic hydroxyl (ArOH) and carboxyl (COOH) groups were developed with eight machine learning methods, among which, three methods, i.e. Random Forest, Random Subspace and J48, illustrated the best performance either for the aromatic hydroxyl and the carboxyl model. The models illustrated good robustness when they were evaluated with functions like "Precision", "Recall", "F-Measure", "AUC", "MCC", etc. Nice accuracy was observed for the aromatic hydroxyl and carboxyl model of these methods, whose AUCs ranged from 0.901 to 0.997. Additionally, the ArOH model was applied to predict the UGT1A1-mediated metabolism of an external set. Two new unknown substrates, cytochrome P450 (CYPs)-mediated metabolites of gefitinib, were predicted and identified, which were validated by in vitro assays. In summary, this study provides a reliable and robust strategy to predict UGT1A1 metabolites, which will be helpful either in rational-optimization of drug metabolism or in avoiding drug-drug interactions in clinic., Competing Interests: Declaration of competing interest None Declared., (Copyright © 2022 Elsevier Ltd.
