Endometriosis is a hormone-responsive gynecologic disease, signifying its connotations across a woman’s life span. Previous studies suggested that endocrine disrupting chemicals were risk factors for endometriosis. Nevertheless, little is known on exposure to organophosphate, pyrethroid and phenoxy acid pesticides on endometriosis diagnosis. In this study, we determined the concentrations of 11 pesticides, metabolites of organophosphate and pyrethroid insecticides, and phenoxy herbicides, in urine collected from 619 reproductive-age women in Utah and California, using liquid chromatography-tandem mass spectrometry. The association of urinary concentrations of pesticides with an increase in the odds of endometriosis diagnosis was examined in 594 women who underwent laparoscopy/laparotomy (operative cohort: n = 471) or pelvic magnetic resonance imaging (population cohort: n = 123), during 2007–2009. 2-Isopropyl-4-methyl-6-hydroxypyrimidine (IMPY), malathion dicarboxylic acid (MDA), para-nitrophenol (PNP), 3,5,6-trichloro-2-pyridinol (TCPY), 3-phenoxybenzoic acid (3-PBA), and 2,4-dichlorophenoxyacetic acid (2,4-D) were detected in ≥95% of the urine samples analyzed. Urinary concentrations of IMPY, MDA, PNP, 3-PBA and 2,4-D tended to be higher in younger, non-Hispanic black, nulliparous and less affluent women. IMPY was the most dominant compound in urine followed by PNP and TCPY. When women in the 4th quartile of IMPY and the 2nd quartile of TCPY concentrations (μg/g creatinine) were compared with women in the 1st quartile, the odds ratios (ORs) for diagnosis of endometriosis increased significantly in unadjusted models (IMPY OR = 1.89, 95% confidence interval (Cl) = 1.12–3.20; TCPY OR = 1.65, 95% Cl = 1.02–2.69) for the operative (n = 471) and entire data set (n = 594), respectively. Our results suggest that exposure to elevated concentrations of diazinon (the parent compound of IMPY) and chlorpyrifos and chlorpyrifos-methyl (parent compounds of TCPY) may be associated with endometriosis. Organophosphorus (OP) and pyrethroid (PYR) compounds are the most widely used insecticides. OPs and PYRs are developmental neurotoxicants. Understanding the extent of exposure in the general population and especially in young children is important for the development of public health policy on regulation and use of these chemicals. Presented here are the results of the first investigation into the extent of environmental exposure to neurotoxic insecticides in preschool children in South Australia (SA).Children were enrolled from different areas of SA and assigned into urban, periurban and rural groups according to their residential address. Residential proximity to agricultural activity, parental occupational contact to insecticides and use of insecticides within the household were investigated as potential indirect measures of exposure. We used liquid chromatography/tandem mass spectrometry to measure the following metabolites of OPs and PYRs in urine samples as direct indicators of exposure: dialkylphosphates, p-nitrophenol, 3-methyl-4-nitrophenol, 3,5,6-trichloro-2-pyridinol, cis- and trans-3-(2,2-dichlorovinyl)-2,2-dimethyl-cyclopropane-1-carboxylic acid, cis-3-(2,2-dibromovinyl)-2,2-dimethyl-cyclopropane-1-carboxylic acid, 2-methyl-3phenylbenzoic acid and 3-phenoxybenzoic acid. Results were analysed to assess factors affecting the risk and level of exposure. Results were also compared to the published data in similar age groups from US and German studies.The results of this study demonstrate that there was widespread chronic exposure to OPs and and PYRs in SA children. OP metabolites were detected more commonly than PYR. Exposure to more than one chemical and contemporaneous exposure to chemicals from both OP and PYR groups was common in the study population. There were some differences in risks and levels of exposure between the study groups. Exposure to some restricted use of chemicals, for example, fenitrothion, was higher in periurban and rural children. There was no difference among the study groups in exposure to chlorpyrifos, used commonly in agriculture and in domestic settings and most frequently found OP pesticide in food in Australia. South Australian children appear to have higher levels of exposure compared their peers in US and Germany. A method has been developed for the analysis of a wide polarity range of the currently used organophosphorus pesticides (OPs) and their metabolites in sewage sludge samples. Extraction was carried out using ultrasonic assisted extraction (UAE) with different solvents. The levels of OPs in sludge were determined by liquid chromatography-tandem mass spectrometry (LC-MS/MS) using electrospray ionization (ESI) in positive or negative ion mode. Extraction with acetonitrile containing 1% acetic acid gave the best results with recoveries between 83.2% and 106.4% and RSD ≤ 8.7%. Evaluation of matrix effect showed high ion suppression for OPs of intermediate polarity, which decreased to approximately 50% by matrix dilution to an equivalent of 0.5 g of sludge per millilitre; however, for polar OPs, the matrix effect was negligible at the same concentration. Therefore, matrix-matched standards were used for OPs quantification. The limits of quantification were in the range of 1-14 ng g-1. This method was successfully applied to real sludge samples collected from Madrid Province and chlorpyrifos along with its metabolite TCPY were the OPs found in sludge at the highest levels, 113-344 ng g-1 and 33-307 ng g-1, respectively. © 2009 Elsevier B.V.
