Parabens are esters of p-hydroxybenzoic acid, which have been used as preservatives and considered safe for nearly a century, until the last two decades when concerns began to be raised about their association with cancers. Knowledge of the mode of action of parabens on the metastatic properties of different cancer cells is still very limited. In the present study, we investigated the effects of methylparaben (MP) and propylparaben (PP) on cell invasion and/or migration in multiple human cancerous and noncancerous cells, including hepatocellular carcinoma cells (HepG2), cervical carcinoma cells (HeLa), breast carcinoma cells (MCF-7), and human placental trophoblasts (HTR-8/SVneo). MP and PP at concentrations in a range of 5-500 μg/L significantly promoted the invasion of four cell lines, with a minimum effective concentration of 5 μg/L. MP and PP up-regulated the expression levels and enzymatic activities of matrix metalloproteinase 2 and 9 (MMP2 and MMP9), as well as altered the expression of the tissue inhibitors of metalloproteinase 1 and 2 (TIMP1 and TIMP2) in four cell lines, suggesting MMPs/TIMPs as potential key events (KEs) for paraben-induced cell invasion. Activation of the p38 mitogen-activated protein kinase (p38 MAPK) and c-Jun N-terminal protein kinases 1/2 (JNK1/2) signaling pathways was required for MP- and PP-promoted invasion of four cell lines, suggesting MAPK signaling pathways as candidates for KEs in cancer or noncancerous cells response to paraben exposure. This study showed for the first time that the two widely used parabens, MP and PP, promoted invasive capacity of multiple human cells through a common mode of action. Pharmaceuticals and personal care products (PPCPs) are frequently detected in marine environments, posing a threat to aquatic organisms. Our previous research demonstrated the occurrence of neuroactive compounds in effluent and sediments from a wastewater treatment plant (WWTP) in a fjord North of Stavanger, the fourth-largest city in Norway. To better understand the influence of PPCP mixtures on fish, Atlantic cod (Gadus morhua) were caged for one month in 3 locations: site 1 (reference), site 2 (WWTP discharge), and site 3 (6.7 km west of discharge). Transcriptomic profiling was conducted in the brains of exposed fish and detection of PPCPs in WWTP effluent and muscle fillets were determined. Caffeine (47.8 ng/L), benzotriazole (10.9 ng/L), N,N-diethyl-meta-toluamide (DEET) (5.6 ng/L), methyl-1H-benzotriazole (5.5 ng/L), trimethoprim (3.4 ng/L), carbamazepine (2.1 ng/L), and nortriptyline (0.4 ng/L) were detected in the WWTP effluent. Octocrylene concentrations were observed in muscle tissue at all sites and ranged from 53 to 193 ng/g. Nervous system function and endocrine system disorders were the top enriched disease and function pathways predicted in male and female fish at site 2, with the top shared canonical pathways involved with estrogen receptor and Sirtuin signaling. At the discharge site, predicted disease and functional responses in female brains were involved in cellular assembly, organization, and function, tissue development, and nervous system development, whereas male brains were involved in connective tissue development, function, and disorders, nervous system development and function, and neurological disease. The top shared canonical pathways in females and males were involved in fatty acid activation and tight junction signaling. Polycyclic aromatic hydrocarbons found in crude oil can impair fish health following sublethal exposure. However, the dysbiosis of microbial communities within the fish host and influence it has on the toxic response of fish following exposure has been less characterized, particularly in marine species. To better understand the effect of dispersed crude oil (DCO) on juvenile Atlantic cod (Gadus morhua) microbiota composition and potential targets of exposure within the gut, fish were exposed to 0.05 ppm DCO for 1, 3, 7, or 28 days and 16 S metagenomic and metatranscriptomic sequencing on the gut and RNA sequencing on intestinal content were conducted. In addition to assessing species composition, richness, and diversity from microbial gut community analysis and transcriptomic profiling, the functional capacity of the microbiome was determined. Mycoplasma and Aliivibrio were the two most abundant genera after DCO exposure and Photobacterium the most abundant genus in controls, after 28 days. Metagenomic profiles were only significantly different between treatments after a 28-day exposure. The top identified pathways were involved in energy and the biosynthesis of carbohydrates, fatty acids, amino acids, and cellular structure. Biological processes following fish transcriptomic profiling shared common pathways with microbial functional annotations such as energy, translation, amide biosynthetic process, and proteolysis. There were 58 differently expressed genes determined from metatranscriptomic profiling after 7 days of exposure. Predicted pathways that were altered included those involved in translation, signal transduction, and Wnt signaling. EIF2 signaling was consistently dysregulated following exposure to DCO, regardless of exposure duration, with impairments in IL-22 signaling and spermine and spermidine biosynthesis in fish after 28 days. Data were consistent with predictions of a potentially reduced immune response related to gastrointestinal disease. The continual discharge of pharmaceuticals from wastewater treatment plants (WWTPs) into the marine environment, even at concentrations as low as ng/L, can exceed levels that induce sublethal effects to aquatic organisms. Amitriptyline, a tricyclic antidepressant, is the most prescribed antidepressant in Norway, though the presence, potential for transport, and uptake by aquatic biota have not been assessed. To better understand the release and bioaccumulative capacity of amitriptyline, laboratory exposure studies were carried out with field-collected sediments. Influent and effluent composite samples from the WWTP of Stavanger (the 4th largest city in Norway) were taken, and sediment samples were collected in three sites in the proximity of this WWTP discharge at sea (WWTP discharge (IVAR), Boknafjord, and Kvitsøy (reference)). Polychaetes (Nereis virens) were exposed to field-collected sediments, as well as to Kvitsøy sediment spiked with 3 and 30 μg/g amitriptyline for 28 days. The WWTP influent and effluent samples had concentrations of amitriptyline of 4.93 ± 1.40 and 6.24 ± 1.39 ng/L, respectively. Sediment samples collected from IVAR, Boknafjord, and Kvitsøy had concentrations of 6.5 ± 3.9, 15.6 ± 12.7, and 12.7 ± 8.0 ng/g, respectively. Concentrations of amitriptyline were below the limit of detection in polychaetes exposed to sediment collected from Kvitsøy and IVAR, and 5.2 ± 2.8 ng/g in those exposed to Boknafjord sediment. Sediment spiked with 3 and 30 μg/g amitriptyline had measured values of 423.83 ± 33.1 and 763.2 ± 180.5 ng/g, respectively. Concentrations in worms exposed to the amended sediments were 9.5 ± 0.2 and 56.6 ± 2.2 ng/g, respectively. Antibiotic residues in the aquatic environment have been shown to induce significant adverse effects on the early-life stage development of aquatic organisms, though the underlying molecular mechanisms of these effects have not been well characterized. In this study, we performed global mRNA-miRNA sequencing, canonical pathway analyses, morphological, physiological, immunohistochemical, and behavioral analyses to comprehensively assess the cross-generational cardiotoxicity and mechanisms of antibiotic mixtures in zebrafish. Following parental treatment to 1 and 100 μg/L antibiotic mixtures (15 of the most commonly detected antibiotics) for 150 days, all 15 assessed antibiotics were detected in the F1 eggs, indicating the cross-generational transfer of antibiotics. Global mRNA-miRNA sequencing functional analysis predicted cardiotoxicity in the F1 generation by using the F1 whole fish. Consistent with canonical pathway analyses, significant cardiotoxicity was observed in F1 larvae, as well as the apoptosis of cardiac cells. Furthermore, let-7a-5p regulated the cardiac hypertrophy signaling pathway, suggesting mechanisms of miRNA of let-7 family mediating cross-generational cardiotoxicity of antibiotics in zebrafish. Ultraviolet (UV) radiation can significantly increase the toxicity of polycyclic aromatic hydrocarbons (PAHs) in crude oil to early life stage (ELS) fishes through photo-induced /photo-enhanced toxicity. However, little is known about the sub-lethal effects and mechanisms of photo-induced PAH toxicity in ELS fishes. The present study investigated apoptosis and global transcriptomic effects in larval red drum (Sciaenops ocellatus) (24-72 h post-fertilization) following co-exposure to oil (0.29-0.30 μg/L ∑PAH 50 ) and UV. Apoptosis was quantified using the TUNEL assay, and transcriptomic effects were assessed using RNA sequencing analysis. Apoptotic fluorescence was greatest in the eyes and skin following 24 and 48 h co-exposure to oil and UV, indicating photo-induced toxicity. Consistent with these phenotypic responses, pathways associated with phototransduction, eye development, and dermatological disease were among the top predicted pathways impacted. Oil and gas extraction activities occur across the globe, yet species-specific toxicological information on the biological and ecological impacts of exposure to petrochemicals is lacking for the vast majority of marine species. To help prioritize species for recovery, mitigation, and conservation in light of significant toxicological data gaps, a trait-based petrochemical vulnerability index was developed and applied to the more than 1700 marine fishes present across the entire Gulf of Mexico, including all known bony fishes, sharks, rays and chimaeras. Using life history and other traits related to likelihood of exposure, physiological sensitivity to exposure, and population resiliency, final calculated petrochemical vulnerability scores can be used to provide information on the relative sensitivity, or resilience, of marine fish populations across the Gulf of Mexico to oil and gas activities. Based on current knowledge of traits, marine fishes with the highest vulnerability scores primarily occur in areas of high petrochemical activity, are found at or near the surface, and have low reproductive turnover rates and/or highly specialized diet and habitat requirements. Crude oil is known to induce developmental defects in teleost fish exposed during early life stages (ELSs). While most studies in recent years have focused on cardiac endpoints, evidence from whole-animal transcriptomic analyses and studies with individual polycyclic aromatic hydrocarbons (PAHs) indicate that the developing kidney (i.e., pronephros) is also at risk. Considering the role of the pronephros in osmoregulation, and the common observance of edema in oil-exposed ELS fish, surprisingly little is known regarding the effects of oil exposure on pronephros development and function. Using zebrafish (Danio rerio) ELSs, we assessed the transcriptional and morphological responses to two dilutions of high-energy water accommodated fractions (HEWAF) of oil from the Deepwater Horizon oil spill using a combination of qPCR and whole-mount in situ hybridization (WM-ISH) of candidate genes involved in pronephros development and function, and immunohistochemistry (WM-IHC). To assess potential functional impacts on the pronephros, three 24 h osmotic challenges (2 hypo-osmotic, 1 near iso-osmotic) were implemented at two developmental time points (48 and 96 h post fertilization; hpf) following exposure to HEWAF. Changes in transcript expression level and location specific to different regions of the pronephros were observed by qPCR and WM-ISH. Further, pronephros morphology was altered in crude oil exposed larvae, characterized by failed glomerulus and neck segment formation, and straightening of the pronephric tubules. The osmotic challenges at 96 hpf greatly exacerbated edema in both HEWAF-exposed groups regardless of osmolarity. By contrast, larvae at 48 hpf exhibited no edema prior to the osmotic challenge, but previous HEWAF exposure elicited a concentration-response increase in edema at hypo-osmotic conditions that appeared to have been largely alleviated under near iso-osmotic conditions. Effluent produced during the electroplating process can contain high concentrations of heavy metals that can enter the environment and induce toxicity to aquatic organisms. Relatively high concentrations of zinc (Zn) and mercury (Hg) have been detected in treated electroplating industrial effluent (TEPIE), though the cytotoxic potential of these compounds has not been well assessed in fish gills. A novel cell line, Danio rerio gill (DrG), were exposed to TEPIE and concentrations of Zn, Hg, and Zn + Hg previously measured in treated effluent to evaluate the use of the DrG cell line following exposure to environmental pollutants. Several cytotoxic assays were employed to assess the effect of TEPIE, Zn, and Hg on this cell line. The percent cell viability was significantly reduced in a concentration-dependent manner following exposure to TEPIE, Zn, Hg, and Zn + Hg (p < 0.05) for 24 h, with additional morphological changes observed in exposure treatments relative to controls. Additionally, there was a significant induction of DNA damage detected in all exposure treatments determined through comet assay tail length. An increase in intracellular ROS generation was also observed in cells exposed to TEPIE, Zn, Hg, and Zn + Hg, corresponding to dose-dependent increases in apoptosis. The choriogenin H - EGFP transgenic medaka (Oryzias melastigma) has been used to test estrogenic substances and quantify estrogenic activity into 17β-estradiol (E2) equivalency (EEQ). The method uses 8 eleutheroembryos in 2 ml solution per well and 3 wells per treatment in 24-well plates at 26 ± 1 °C for 24 ± 2 h, with subsequent measurements of induced GFP signal intensity. EEQ measurements are calculated using a E2 probit regression model with a coefficient of determination (R 2 ) > 0.90. The selectivity was confirmed evaluating 27 known estrogenic and 5 known non-estrogenic compounds. Limit of quantitation (LOQ), recovery rate and bias were calculated to be 1 ng/ml EEQ, 104% and 4% respectively. Robustness analysis revealed exposure temperature is a sensitive parameter that should be kept at 26 ± 1 °C. The repeatability of intra- and inter-laboratories achieved CV < 30% for most tested food and cosmetics samples. The lot-lot stability was confirmed by the stable EEQ qualitative control (QC, 1 ng/mL E2) and calibration curve results. The stability of standard reagents, samples and sample extracts was also investigated. A fundamental understanding of the impact of petrochemicals and other stressors on marine biodiversity is critical for effective management, restoration, recovery, and mitigation initiatives. As species-specific information on levels of petrochemical exposure and toxicological response are lacking for the majority of marine species, a trait-based assessment to rank species vulnerabilities to petrochemical activities in the Gulf of Mexico can provide a more comprehensive and effective means to prioritize species, habitats, and ecosystems for improved management, restoration and recovery. To initiate and standardize this process, we developed a trait-based framework, applicable to a wide range of vertebrate and invertebrate species, that can be used to rank relative population vulnerabilities of species to petrochemical activities in the Gulf of Mexico. Through expert consultation, 18 traits related to likelihood of exposure, individual sensitivity, and population resilience were identified and defined. The resulting multi-taxonomic petrochemical vulnerability framework can be adapted and applied to a wide variety of species groups and geographic regions. Estrogenically active compounds (EACs) in surface waters can disrupt the endocrine system of biota, raising concern for aquatic species. Concentrations of EACs are generally higher in effluent-dominated aquatic systems, such as California's Santa Ana River (SAR). Addressing estrogenicity of effluent-dominated waters is increasingly important due to both increasing urbanization and climate change. To this end, water samples were collected from multiple sites downstream of wastewater treatment plants (WWTPs) and intermittent points along the SAR during 2018-2019 and cell-based bioassays were used to determine estrogen receptor activity. During baseflow conditions, the highest estradiol equivalencies (EEQs) from all SAR water between summer (August and September) and fall (November) sampling events in 2018 were from Yorba Linda (EEQ = 1.36 ± 0.38 ng/L) and Prado (1.14 ± 0.13 ng/L), respectively. Water extracts in January 2019 following a major rainfall generally had higher EEQs with the highest EEQ of 10.0 ± 0.69 ng/L observed at Yorba Linda. During low flow conditions in November 2018, male Japanese medaka (Oryzias latipes) fish were exposed to SAR water to compare to cell bioassay responses and targeted analytical chemistry for 5 steroidal estrogens. Chemical-based EEQ correlations with in vitro EEQs were statistically significant. However, vitellogenin (vtg) mRNA expression in the livers of medaka exposed to SAR water was not significantly different compared to controls. Perfluoroalkyl substances (PFAS) are contaminants that are applied in a wide range of consumer products, including ski products. The present study investigated the neuro-dopamine (DA) and cellular steroid hormone homeostasis of wild Bank voles (Myodes glareolus) from a skiing area in Norway (Trondheim), in relation to tissue concentrations of PFAS. We found a positive association between brain DA concentrations and the concentration of several PFAS, while there was a negative association between PFAS and dopamine receptor 1 (dr1) mRNA. The ratio between DA and its metabolites (3,4-dihydroxyphenylacetic acid: DOPAC and homovanillic acid: HVA) showed a negative association between DOPAC/DA and several PFAS, suggesting that PFAS altered the metabolism of DA via monoamine oxidase (Mao). This assumption is supported by an observed negative association between mao mRNA and PFAS. Previous studies have shown that DA homeostasis can indirectly regulate cellular estrogen (E2) and testosterone (T) biosynthesis. We found no association between DA and steroid hormone levels, while there was a negative association between some PFAS and T concentrations, suggesting that PFAS might affect T through other mechanisms. Boscalid is a persistent fungicide that is frequently detected in surface waters and may be neurotoxic to aquatic organisms. Herein, we evaluated the effects of environmentally relevant boscalid concentrations to zebrafish to explore its potentially neurotoxic mechanisms of effect. Behavioral responses (swimming, phototaxis, and predation), histopathology, transcriptomics, biochemical parameter analysis and gene expression of larval and adult zebrafish following boscalid treatment were assessed. We found that boscalid significantly inhibited the locomotor ability and phototactic response of larvae after an 8-d exposure, and altered the locomotor activity, predation trajectories and ability in adults after a 21-d exposure. It was noted that predation rates of zebrafish were significantly decreased by 30% and 100% after exposure to 0.1 and 1.0 mg/L boscalid, respectively. Adverse alterations in the cell differentiation of eyes and brain injury were also observed in both larvae and adults following boscalid exposure. The expression of genes related to neurodevelopment, neurotransmission, eye development, and visual function, in conjunction with RNA-Seq results, indicated that boscalid may impair visual phototransduction and nervous system processes in larval zebrafish. The Orange County Sanitation District (OCSD) is a public agency that provides treatment of wastewater for residents in Orange County, California. The final effluent characteristics at OCSD have been altered in the last 18 years due to the ~20-fold increase of sodium hypochlorite usage for disinfecting waste streams from 2002 to 2010, implementation of the Ground Water Replenishment System (GWRS) in 2008, and conversion to full secondary treatment, together with a major reduction of sodium hypochlorite usage in 2011. This study analyzed infauna data gathered from 1994 to 2016 at the zone of initial dilution (ZID) and reference stations located on the San Pedro Shelf to assess the effects of these treatment process changes on biota in the receiving water. Impacts to the infauna community within the ZID were observed during the period of high chlorine usage, including the increased proportion of the pollution tolerant polychaete Capitella capitata complex from 6.2% in 2003 to nearly 60% in 2010, the lowest Infaunal Trophic Index score in 2009, and the highest Benthic Response Index score in 2006. The degradation of the infauna community in the ZID was correlated with chlorination and was coincidental with the initiation of the GWRS. The infauna community at the ZID recovered rapidly after the implementation of full secondary treatment as indicated by the significant reduction of the relative abundance of C. capitata complex from 59.8% in 2010 to less than 0.1% after 2012, and by the markedly improved community health index scores. This study demonstrated the composition and biointegrity of the infauna community at the ZID varied in response to changes in the wastewater treatment process. In this study, fragments of polyethylene plastic bags were treated with simulated gastric juice of fish for 16 h. Following solid-phase extraction, methanol eluents caused acute toxicity to embryos and larvae of Japanese medaka. Chromatographic fractions (polar to more non-polar with numbers increasing) of the extract were evaluated for toxicity and estrogenic activity using medaka and an estrogen receptor (ER) cell-line. Fractions 6 and 9 had the highest estrogenic effects with relative hydrophobic chemicals. The vtg expression in fraction 6 was 22-fold higher than control, and the ER cellular response in fraction 9 was 8.5-fold higher than controls. Following non-target screening (NTS), several novel phthalates and phenols were identified in the above two fractions. Fractions 1 and 2 appeared to be primarily responsible for the acute toxicity observed with the whole extract. The hatching rate decreased to 36 % in fraction 2, and was not observed following exposure to fraction 1. NTS of these fractions indicated 635 and 808 entities, respectively, most without toxicity information. These results indicate plastic leachates from gastric juices of fish are complex mixtures of many compounds that can have acute reproductive and sublethal endocrine impacts in fish., (Copyright © 2020 Elsevier B.V. Due to overuse, misuse, and poor absorption during treatment, antibiotics are consistently released into the environment, raising concerns about their impacts on ecological sustainability and health. In this study we performed transcriptome profiling to assess potential reproductive effects of an antibiotic mixture in gravid female zebrafish. Gravid fish (150 dpf) were exposed to a mixture of 15 commonly detected antibiotics at 0, 1, and 100 μg/L for 4 weeks. Concentrations of all the 15 antibiotics, especially chlortetracycline, were detected in the F0 ovary and F1 eggs after treatment, indicating maternal transfer of antibiotics. Impaired F0 growth (average 2.2 % and 24.3 % inductions in body length and ovary weight, respectively), and reduced F1 offspring survival (average 4.2 % reductions in survival at 120 hpf) was observed after maternal exposure to the 100 μg/L treatment. Pathway analyses of whole-transcriptome expression profiles from F0 ovaries predicted colorectal disorders. Similarly, pathways of F1 larval transcriptomes from treated females also predicted colorectal disorders along with intestinal apoptosis and oxidative stress, which may be related to growth impairment. These results show that maternal transfer of antibiotics occurs in zebrafish, resulting in transgenerational changes in F1 offspring survival and transcription that predict adverse gastrointestinal effects in offspring., (Copyright © 2020 The Author(s). Bifenthrin (BF) is a pyrethroid insecticide used in urban and agricultural applications. Previous studies in early life stages of fish have indicated anti-estrogenic activity; however, estrogenic activity has been observed in adults. To test the hypothesis that BF impairs sex differentiation, larval Japanese Medaka were exposed to BF during a critical developmental window for phenotypic sexual differentiation. Fish were exposed to environmentally relevant concentrations of BF (0.15 μg/L and 1.5 μg/L), a single concentration (0.3 mg/L) of an estrogen receptor (ER) antagonist (ICI 182,780), and an ER agonist (0.2 ug/L) (17β-estradiol). Fish were exposed at 8 days post hatch (dph) larvae for 30 days. Phenotypic sex, secondary sexual characteristics (SSC) and genotypic sex were investigated at sexual maturity (8 weeks). A trend towards masculinization (p = 0.06) based on the presence of papillary processes in anal fin rays of Japanese Medaka was observed in fish exposed to the lowest concentration of BF. Bifenthrin (BF) is a pyrethroid insecticide widely used in urban and agricultural applications. Previous studies in embryos of zebrafish have shown that BF can affect estradiol biosynthesis and the dopaminergic system. To examine the role of the estrogen receptor (ER) in the endocrine effects of BF, embryos were exposed for 96 h to a mixture of 0.15 and 1.5 μg/L BF and an ER agonist (17α-ethynylestradiol - EE2) at 0.09 μg/L. Transcripts related to estrogenic (vitellogenin VTG) and dopaminergic (tyrosine hydroxylase (TH), dopamine receptor 1 (DR1), monoamine oxidase (MAO), and catechol-O-methyltransferase b (COMTb)) signaling pathways were investigated by qRT-PCR. Dopamine (DA) and its metabolites (homovanillic acid (HVA) and 3,4-dihydroxyphenylacetic acid (DOPAC)) were also measured. There was a significant increase in VTG, DR1, MAO and COMTb mRNA levels and HVA-DA ratios within all zebrafish embryos exposed to EE2, including EE2 alone, 0.15 μg/L BF + EE2 and 1.5 μg/L BF + EE2. A significant decrease in homogenate concentrations of DA was observed within all zebrafish embryos exposed to EE2, which included EE2 alone, 0.15 μg/L BF + EE2 and 1.5 μg/L BF + EE2. Co-exposure of BF with EE2 failed to diminish estrogenic or dopaminergic signaling in embryos. Additionally, embryos with diminished ERα expression by morpholino injection were exposed to 0.15 μg/L BF, 1.5 μg/L BF and 0.09 μg/L EE2, with subsequent gene expression measurements. ERα knockdown did not prevent the effects of BF, indicating ERα may have a limited role in the estrogenic and dopaminergic effects caused by BF in zebrafish embryos., (Copyright © 2018 Elsevier B.V. Wastewater treatment plant effluent introduces a mixture of pollutants into marine environments; however, the impacts of chronic sublethal exposures on populations are often unclear. Presence of estrogenic agents in sediments and uptake of these compounds by demersal flatfishes has been reported at the Orange County Sanitation District (OCSD) wastewater outfall. Furthermore, estrogenic activity has been identified in male flatfish in the area, potentially contributing to observed population declines in the OCSD region. Rising ocean temperatures may further contribute to flatfish declines as relationships between temperature and abundance have been reported in the Southern California Bight. To investigate declines, sex ratios, condition factor, organ health indices, hormones, and vitellogenin were quantified in flatfish collected at OCSD outfall and reference sites. Additionally, historical temperature data was examined for trends with population abundances. Fluoxetine is a selective serotonin reuptake inhibitor used as an antidepressant and has been frequently detected in aquatic environments. However, its effects in fish from Asia remain relatively less studied. In this study, the topmouth gudgeon Pseudorasbora parva was exposed to 0, 50, and 200 µg/L of fluoxetine for 4 h and 42 d. The effects of fluoxetine on biometrics were compared to biochemical endpoints indicative of stress in different fish tissues (brain, liver, gills and intestine) following exposures. In fish exposed for 42 d, lipid peroxidation endpoints were enhanced 80% in the liver and gills. Acetylcholinesterase (AChE) activity was increased 40% after exposure to 50 µg/L and 55% at 200 µg/L following 4 h exposure. In contrast AChE was increased 26% (at 50 µg/L) after 42 d of exposures. Enhanced ethoxyresorufin-O-deethylase activity (EROD) was detected only in fish exposed to 50 µg/L of fluoxetine for 4 h. The activity of α-glucosidase (α-Glu) was also induced (at 200 µg/L) after 4 h of exposure. After 4 h of exposure, the activities of proteases in the intestine were generally inhibited at 200 µg/L. Both 4 h and 42 d exposures resulted in an increased hepatosomatic index (HSI) but did not affect the condition factor (CF). Our results demonstrate that fluoxetine significantly altered biochemical endpoints in P. parva after acute exposure and the morphological changes in liver size were not observed until 42 d of exposure., (Copyright © 2018. Selenite(IV) and selenate(VI) are the major forms of Se in aquatic ecosystem. In this study, Pseudorasbora parva were exposed to 10, 200 and 1000 μg L -1 selenite and selenate for 28 days. Selenium accumulation, antioxidant enzyme levels, glutathione concentrations, lipid peroxidation and histology were evaluated in livers following exposure. Our results showed that Se(IV) and Se(VI) caused different accumulation patterns in the liver, with a more rapid accumulation of Se with Se(IV) treatment. Both Se species increased hepatic lipid peroxidation after 14 and 28 d (~ 30%). Among the antioxidants examined, the activity of SOD (except day 28) and the cellular levels of GSH were induced by 72-137% at lower concentrations, while the activity of GST was at least 24% lower than that of the control at 200 and 1000 μg L -1 for both Se species at all sampling points. Both forms of Se reduced the hepatosomatic index at 1000 μg L -1 after 28 d. In addition, marked histopathological alterations (10-31%) were observed in the liver of P. parva after exposure to both Se species, with higher frequency in the Se(IV) exposed fish. Liver local necrosis was observed only in the liver of fish exposed to 1000 μg L -1 of Se(IV) (~ 20%). Diuron is one of the most used herbicide in the world, and its field application has been particularly increased in Brazil due to the expansion of sugarcane crops. Diuron has often been detected in freshwater ecosystems and it can be biodegraded into three main metabolites in the environment, the 3,4-dichloroaniline (DCA), 3,4-dichlorophenylurea (DCPU) and 3,4-dichlorophenyl-N-methylurea (DCPMU). Negative effects under aquatic biota are still not well established for diuron, especially when considering its presence in mixture with its different metabolites. In this study, we evaluated the effects of diuron alone or in combination with its metabolites, DCPMU, DCPU and 3,4-DCA on biochemical stress responses and biotransformation activity of the fish Oreochromis niloticus. Results showed that diuron and its metabolites caused significant but dispersed alterations in oxidative stress markers and biotransformation enzymes, except for ethoxyresorufin-O-deethylase (EROD) activity, that presented a dose-dependent increase after exposure to either diuron or its metabolites. Glutathione S-transferase (GST) activity was significant lower in gills after exposure to diuron metabolites, but not diuron. Diuron, DCPMU and DCA also decreased the multixenobiotic resistance (MXR) activity. Lipid peroxidation levels were increased in gill after exposure to all compounds, indicating that the original compound and diuron metabolites can induce oxidative stress in fish. The integration of all biochemical responses by the Integrated Biomarker Response (IBR) model indicated that all compounds caused significant alterations in O. niloticus, but DCPMU caused the higher alterations in both liver and gill. Our findings imply that diuron and its metabolites may impair the physiological response related to biotransformation and antioxidant activity in fish at field concentrations. Calculating risk from seafood exposure to persistent organic pollutants continues to be problematic as estimates of exposure from diet require extensive monitoring of fish species and limited assessments of bioavailability from sediments where the contaminants tend to reside. Previous studies in our laboratory utilized a laboratory-based isotope dilution method (IDM) to estimate the bioavailability of DDT [1,1,1-trichloro-2, 2-bis(p-chloro-phenyl)ethane] and its metabolites from sediment to biota from a superfund site on the shelf of the Palos Verdes (PVS) Peninsula in California (USA). Using a biota-sediment accumulation factor (BSAF) derived from IDM and biomagnification factors (BMF) calculated from previous studies as well as seafood-consumption data specific to anglers in the PVS area, we estimated cancer and non-cancer risks for anglers and nursing infants representing sensitive groups. Predicted cancer risks from consumption of White croaker (Genyonemus lineatus) to the 50th and 95th percentile to all shore mode anglers were, respectively, 2×10 -7 and 7×10 -7 , which were similar to field studies using fish concentrations of all DDT isomers and their environmental degradates (ΣDDT) from collected animals. The calculated non-cancer hazard quotient values for the 50th and 95th percentile shore mode anglers consuming White croaker from this study (0.008 and 0.023, respectively) were also of similar magnitude as those obtained from studies based on samples obtained solely from fish. For nursing infants, similar results were also observed. Marine sediments at many locations in the world are contaminated with a wide range of persistent organic pollutants. The Palos Verdes Shelf (PVS) is located in the ocean off the coast of Los Angeles, California and has been listed as a Superfund site by the US EPA since 1997, because of heavy contamination of DDTs and PCBs. However, little is known about the historical trend in the deposition of DDTs as a result of decades-long discharge of wastewater effluents. In this study, sediment cores were taken from the PVS site and determined for DDT and its metabolites including DDE and DDD (denoted as DDTs). Individual DDTs were found in the majority (95%) of the samples analyzed. The highest ∑DDT concentrations were found in three cores along the 60-meter isobath with geometric means of 31300, 7490, and 5010ng/gdw and medians of 82400, 17300, and 5200ng/g dw, respectively. Among DDT congeners, p,p'-DDE, o,p'-DDE and p,p'-DDD were predominant, contributing to approximately 54%, 27%, and 14% of the ΣDDTs in sediment. The vertical profiles of concentrations of contaminants in the sediment cores were examined. For most of the cores, a steady increase in the concentrations of DDTs during 1940s to 1980s was observed, while the concentrations declined gradually toward the surficial layers. On the basis of the mass flux of DDTs calculated and the area of the PVS Superfund site, we estimated the total deposition amount of DDTs in sediment and the deposition amount of ΣDDTs in this region during 1947-1971 was 132 tons, which was fairly close to what was reported in earlier studies for industrial wastewater discharge in the PVS site (870-1450tons). The Salton Sea, the largest inland surface water body in California, has been designated as a sensitive ecological area by federal and state governments. Its two main tributaries, the New River and Alamo River are impacted by urban and agriculture land use wastes. The purpose of this study was to temporally and spatially evaluate the ecological risks of contaminants of concern in water, sediments and fish tissues. A total of 229 semivolatile organic compounds and 12 trace metals were examined. Among them Selenium, DDTs, PAHs, PCBs, chlorpyrifos and some current-use pesticides such as pyrethroids exceeded risk thresholds. From 2002 to 2012, measurements of chlorpyrifos in sediments generally declined and were not observed after 2009 at the river outlets. In contrast, pyrethroid concentrations in sediments rose consistently after 2009. In water samples, the outlets of the two rivers showed relatively higher levels of contamination than the main water body of the Salton Sea. However, sediments of the main water body of the Salton Sea showed relatively higher sediment concentrations of contaminants than the two rivers. This was particularly true for selenium which showed reductions in concentrations from 2002 to 2007, but then gradual increases to 2012. Consistent with water evaluations, contaminant concentrations in fish tissues tended to be higher at the New River boundary and at the drainage sites for the Alamo River compared to sites along each river. The persistent contaminants DDTs, PAHs, chlorpyrifos and several pyrethroid insecticides were associated with the toxicity of sediments and water collected from the rivers. Despite being considered friendlier to the environment, biodiesel fuel can be harmful to aquatic organisms, especially when combined with petroleum diesel fuel. In this work we evaluated the effects of mineral diesel fuel containing increasing concentrations of biodiesel (5% and 20%, namely B5 and B20) and pure biodiesel (B100), at concentrations of 0.001 and 0.01mLL(-1), after 15 days of exposure, in armored catfish (Pterygoplichtys anisitsi). Toxicity tests were also performed to estimate LC50 values (96h) for each compound. Biotransformation enzymes [ethoxyresorufin-O-deethylase (EROD), and glutathione S-transferase (GST)] as well as oxidative stress markers (superoxide dismutase, SOD, catalase, CAT, glutathione peroxidase, GPx, and the level of lipid peroxidation) were measured in liver and gills after treatment. Genotoxic effects were also accessed in erythrocytes using the comet assay and by evaluating the frequency of micronuclei formation. Further, the mRNA of cytochrome P450 1A (CYP1A) was also measured in liver. Mortality was not observed even exposure to concentrations as high as 6.0mLL(-1). EROD and GST activities were increased after B5 and B20 treatments; however, CYP1A mRNA induction was not observed. SOD and CAT activities were decreased, but GPx was significantly higher for all treatments in gills. There were no significant changes in lipid peroxidation, but genotoxicity markers revealed that all treatments increased comet scores. Fuels B5 and B20 increased micronuclei frequency. The Cd exposure for 14 days significantly increased both the molecular (DNA single-strand breaks) and biochemical (metallothionein concentrations) biomarkers in the freshwater pale chub, Zacco platypus, whereas changes in the histological and physiological biomarker responses were negligible. The BkF exposure for 14 days led to significant increases in the mRNA expression of catalase and superoxide dismutase, 7-ethoxyresorufin-O-deethylase enzymatic activity and DNA single-strand breakage at the molecular and biochemical levels. In addition, exposure to 50μg/L of BkF induced histological alteration in the liver, with significant changes to the liver somatic index and condition factor at the physiological level. Permethrin (PM) is a pyrethroid insecticide that exists as 4 enantiomers. Biotransformation of PM to estrogen receptor agonists (3-phenoxybenzyl alcohol (PBOH) and 3-(4'-hydroxyphenoxy)-benzyl alcohol (3,4 PBOH)) has been shown to be stereoselective in other vertebrate species. This study evaluated the biotransformation of PM enantiomers in human liver microsomes and with recombinant CYP3A4 and CYP2C19. PBOH and 3,4 PBOH were the only metabolites detected from in vitro incubations including each of the 4 enantiomers of PM with 1R-trans PM having the most efficient NADPH-catalyzed biotransformation to both metabolites. Coincubation with the CYP inhibitor ketoconazole and time course experiments with liver microsomes and recombinant CYP2C19 and CYP3A4 indicated CYP-catalyzed stereoselective cleavage of the ester followed by 4-hydoxylation to 3,4' PBOH. These data indicate potential dispositional differences may occur with PM enantiomers and a shift in putative molecular targets. While cleavage of pyrethroid esters lead to detoxification of the acute neurological effects, formation of the benzyl alcohol and hydroxylated metabolite may lead to estrogenic responses, since each of these metabolites are estrogen receptor ligands., (Copyright © 2014 Elsevier Ireland Ltd. Oryzias melastigma, also called O. dancena, is becoming a very useful model for estuarine and marine ecotoxicity studies. With O. melastigma being adopted by ILSI Health and Environmental Science Institute (HESI) for embryo toxicity testing, improved knowledge of biomarker based embryonic development becomes especially important for mechanism-based toxicity evaluations. Using whole mount in situ hybridization and immunostaining techniques together with widely used molecular markers, this study describes the molecular development of marine medaka embryos, focusing on the brain, eye, heart, pectoral fin, pancreas, liver, muscle and neuron system. Although the importance of chirality in organophosphorus compounds (OPs) is well recognized in relation to their biological effects, as with most chiral pesticides, OPs are generally marketed, used and released to the environment as racemates (i.e., equimolar mixtures of enantiomers). In addition, research on enantioselective environmental fate and effects of chiral OPs is still limited, particularly in the evaluation of enantioselectivity in their environmental degradation. A large number of OPs are chiral compounds, and yet enantioselectivity in their environmental fate and effects is rarely addressed. This paper highlights the current state of knowledge on the environmental occurrence and behavior of chiral OP pesticides. Developments in enantioselective analytical techniques, specifically gas chromatography (GC), high-performance liquid chromatography (HPLC) and capillary electrophoresis (CE), as applied in the evaluation of enantiomer-specific fate and effects of chiral OPs, are also discussed., (Copyright (c) 2009 Elsevier B.V. In this study, toxicity of effluents in a wastewater treatment plant and of receiving water in an adjacent stream was periodically monitored from November 2007 to June 2008, in order to trace and reduce sources of toxicants. The results showed that toxicity of final effluent (FE) changed greatly over different sampling events, and appeared to have impacts on toxicity of downstream water with a significant correlation (r(2)=0.87, p<0.05). In particular, FE toxicity was always higher than that of secondary effluent (SE). Toxicity identification evaluation (TIE) for the FE sample collected in March 2008 showed that FE toxicity was attributed to low quality of Fenton reagent with Zn contamination used for SE treatment. Furthermore, Zn concentrations in FE samples significantly correlated with FE toxicity during the sampling period (r(2)=0.95, p<0.05). After changing the Fenton reagent to one containing low Zn, Zn concentration and toxicity of FE greatly decreased in the following months.

Studies throughout the southern California bight have indicated persistent estrogenic activity in male hornyhead turbot (Pleuronichthys verticalis). Plasma 17beta-estradiol (E2) concentrations correlated with gonadal DNA damage in fish collected near a wastewater treatment plant outfall, but not from fish collected at the reference location. When the same species was collected from the same reference location and treated with E2, no relationship between uptake and gonadal DNA damage was observed. To evaluate the site-specific effects of E2 in fish from a wastewater outfall and fish from a reference location, male hornyhead turbot from each location were exposed to 15 microg/L aqueous E2 in a time-course experiment, with fish sampled every 12 h for 48 h. Concentrations of E2 were measured in the aqueous exposure and in plasma from the fish. Vitellogenin (vtg) was also measured in the plasma, and 8-oxo-7,8-dihydro-2'-deoxyguanosine levels in male gonads were measured as an indicator of DNA damage. Untreated fish from the outfall had significantly lower E2 in the plasma relative to the untreated reference fish, and this trend was consistent at each time point in the E2-treated fish. Vtg was significantly induced after 36 h of exposure in fish from both sites and no significant differences were observed between the sites. A significant increase of oxidative DNA damage was observed in E2-treated fish from the outfall population and the damage was significantly correlated with plasma E2 concentrations only in fish from the outfall after 48 h. These results indicated that there were significant differences in E2 disposition and gonadal genotoxicity between the hornyhead turbot populations following exposure to E2, suggesting that fish at wastewater outfalls may be more sensitive to DNA damage, which may be temporally related to concentrations of E2 in plasma.

Flavin-containing monooxigenases (FMOs) are a polymorphic family of drug and pesticide metabolizing enzymes, found in the smooth endoplasmatic reticulum that catalyze the oxidation of soft nucleophilic heteroatom substances to their respective oxides. Previous studies in euryhaline fishes have indicated induction of FMO expression and activity in vivo under hyperosmotic conditions. In this study we evaluated the effect of hypersaline conditions in rat kidney. Male Sprague-Dawley rats were injected intraperitoneal with 3.5M NaCl at a doses ranging from 0.3cm(3)/100g to 0.6cm(3)/100g in two separate treatments. Three hours after injection, FMO activities and FMO1 protein was examined in the first experiment, and the expression of FMO1 mRNA was measured in the second experiment from kidneys after treatment with NaCl. A positive significant correlation was found between FMO1 protein expression and plasma osmolarity (p<0.05, r=0.6193). Methyl-p-tolyl sulfide oxidase showed a statistically significant increase in FMO activity, and a positive correlation was observed between plasma osmolarity and production of FMO1-derived (R)-methyl-p-tolyl sulfoxide (p<0.05, r=0.6736). Expression of FMO1 mRNA was also positively correlated with plasma osmolality (p<0.05, r=0.8428). Similar to studies in fish, these results suggest that expression and activities of FMOs may be influenced by hyperosmotic conditions in the kidney of rats.

Oxidative compounds have been demonstrated to decrease the fertilization capability and viability of offspring of treated spermatozoa. As estrogen and its hydroxylated metabolites readily undergo redox cycling, this study was undertaken to determine if estrogens and other oxidants could damage DNA and impair sperm function. Sperm was preexposed to either 17beta-estradiol (E2), 4-hydroxyestradiol (4OHE2) or the oxidant t-butyl hydroperoxide (t-BOOH), and allowed to fertilize untreated eggs. The fertilization rates and development of the larvae were assessed, as well as the amount of 8-oxodeoxyguanosine (8-oxodG) as an indication of oxidative DNA damage. All compounds caused significant decreases in fertilization and increases in pathological abnormalities in offspring, with 4OHE2 being the most toxic. Treatment with 4OHE2 caused a significant increase of 8-oxodG, but E2 failed to show any effect. Pathological abnormalities were significantly correlated (r(2)=0.44, p< or =0.05) with 8-oxodG levels in sperm treated with t-BOOH and 4OHE2, but not E2. 8-OxodG levels also were somewhat weakly correlated with impaired fertilization in 4OHE2-treated sperm (r(2)=0.33, p< or =0.05). The results indicate that biotransformation of E2 to 4OHE2 enhances oxidative damage of DNA in sperm, which can reduce fertilization and impair embryonic development, but other mechanisms of action may also contribute to these effects.

Male and female hornyhead turbot (Pleuronichthys verticalis) were exposed to four concentrations (0, 0.75, 14.7 and 46.5 ng/g dry weight) of E2-amended sediment for 7 days. Sediment-derived E2 was bioavailable to the flatfish, though the route of uptake was unclear. A concentration of 46.5 ng/g E2 in sediment led to a significant increase in vitellogenin concentrations in the plasma in both sexes after 7 days of exposure. Though plasma E2 concentrations increased significantly in males at sediment E2 concentrations of 0.75 ng/g dry weight and above, a dose-dependent increase was not observed. There was also no correlation between sediment E2 concentrations, plasma E2 concentrations, and oxidative DNA damage in male gonads. The results suggest that the DNA damage previously seen in the gonads of feral hornyhead turbot at a sewage outfall is likely not caused by acute exposure to exogenous E2 from sediments.

Two cholinesterases (ChE) present in vertebrates, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) are distributed in tissues in species-specific patterns. English sole (Pleuronectes vetulus, ES) and Hornyhead turbot (Pleuronichthys verticalis, HT) muscle ChE activities were measured in fish from a location near a municipal wastewater outfall and a far-field reference site off the coast of Southern California. ES muscle ChE correlated with total fish length (r=-0.3271) in fish from two sites. No significant differences were observed between gender or sampling sites. Substrate preference assays showed that both ChEs were found in muscle with BChE representing about 30% of the total ChE activity. Data resulting from the use of selective inhibitors suggested the presence of an atypical BChE. Higher sensitivity to pesticide inhibition was observed in ChEs from smaller fish. In HT muscle, ChE activities presented significant differences between males and females, but no correlation was found with total length. No significant differences were observed between sampling sites. Both ChEs were also present with BChE activity constituting 90% of the total ChE activity. Selective inhibitor results indicated the presence of an atypical ChE. Male ChE was more sensitive to pesticide inhibition. These data indicate that species, size, and gender dependent differences in ChE may contribute to differences in susceptibility to ChE-inhibiting toxicants encountered in the marine environment.

Feminization of fish has been reported throughout the world in freshwater and marine systems. While the population impacts are conflictive, enough negative effects warrant additional research into causation. In order to ascertain the identities of specific feminizing agents, variants of toxicity identification evaluations (TIEs) have been employed. The majority of these evaluations have utilized in vitro estrogen receptor-based cell-lines to identify chromatographic fractions that possess biological activity from predominately wastewater derived from municipal treatment facilities and have concluded that synthetic and natural estrogens are the primary cause for feminization of fish. This paper will focus on three aquatic systems impacted by wastewater originating from purely domestic, and industrial/domestic secondary treatment systems. Wastewater and sediment extracts were evaluated by in vitro and in vivo biological responses in a TIE fractionation design. While in vitro responses tended to mirror in vivo responses in purely domestic wastewater systems, in vitro responses tended to severely underestimate in vivo estrogenic activity when normalized to estradiol equivalents in more complex systems. TIE fractionation schemes using in vivo biological responses failed to indicate any relationship to steroids in either wastewater or sediment extractions. These data consistently support the view that mechanisms other than direct ER binding and activation by toxicants may be important in the feminization of fish particularly residing in habitats that receive complex wastewater or agricultural effluents.

Pentachlorophenol (PCP) is widely used to control termites and protect wood from fungal-rot and wood-boring insects, and is often detected in the aquatic environment. Few studies have evaluated PCP as an environmental endocrine disruptor. In the present work, Japanese medaka (Oryzias latipes) was exposed to PCP for 28 days (F0 generation) with subsequent measurements of vitellogenin (VTG), hepatic 7-ethoxyresorufin-O-deethylase (EROD), and reproductive endpoints. Plasma VTG significantly increased in male fish treated with PCP concentrations lower than 200 microg/l and decreased in male and female animals exposed to 200 microg/l. Hepatic EROD from female fish increased when PCP exposure concentrations exceeded 20 microg/l, but decreased in the 200 microg/l PCP treatment group. Fecundity and mean fertility of female medaka decreased significantly in the second and third week following exposure concentrations greater than 100 microg/l, and testis-ova of male medaka was observed at PCP concentrations greater than 50 microg/l. Histological lesions of liver and kidney occurred when exposure concentrations exceeded 50 microg/l. In F1 generations, the hatching rates and time to hatch of offspring were significantly affected in fish exposed to 200 microg/l. These results indicated that PCP exposure caused responses consistent with estrogen and aryl hydrocarbon receptor activation as well as reproductive impairment at environmentally relevant concentrations.

Coal Oil Point (COP) is a natural oil seep off the coast of Santa Barbara, California. Although most studies examining the fate and effects of petroleum have focused upon urbanized or anthropogenic sources of inputs, few have examined the effects of polyaromatic hydrocarbons (PAHs) derived from natural seeps. In order to evaluate the effects of PAHs derived from COP on marine fish populations, hatchery-reared California Halibut (Platichthys californicus) were exposed for 30 days to seven dilutions of sediments collected from COP. Hepatic cytochrome P450 1A (CYP1A), biliary fluorescent aromatic compounds (FACs), gonadal somatic indices, and plasma steroid concentrations. Sixteen USEPA priority PAHs were targeted for analysis in each sediment dilution. In general, biochemical responses were somewhat recalcitrant to dose-response relationships and were less sensitive than the literature values established for the same indicators following exposure to urbanized PAHs. Trends toward reductions in plasma 17beta-estradiol concentrations were observed, but reductions in gonadal somatic indices were not observed. FAC values for naphthalene, benzo(a)pyrene, phenanthrene-related compounds reached maximums at 33-100% COP sediment. The resulting insensitivity may be unique for exposure to "natural" petroleum due to a higher concentration of lower molecular weight PAHs or uncharacterized inhibitors.
