We investigated the biogenic volatile organic compound (BVOC) emission rates and composition of Cupressaceae species and how the emissions change in response to moderate warming and more severe heat stress. A total of 8 species from 7 distinct Cupressaceae genera were targeted in this study and exposed to laboratory-simulated heatwaves. Each plant was enclosed in a temperature-controlled glass chamber and allowed to equilibrate at 30 °C for 24 h. The temperature was then increased stepwise from 33 °C to 43 °C in 2 °C increments, with each step lasting 2 h, and was finally kept at 45 °C for 12 h. The BVOC emissions were measured periodically using an automated air sampler coupled to a gas chromatograph. Most of the sampled Cupressaceae species (6 out of 8) were low BVOC emitters (<0.3 μgC g -1  h -1 ) at 30 °C. However, the BVOC emissions of all 8 species increased strongly with temperature, and in most species (5 out of 8), the emissions continued to increase with longer exposure times to heat stress. The largest increase was observed in Thuja occidentalis and Chamaecyparis thyoides, which reached maximum emissions of 350 and 190 μgC g -1  h -1 , respectively. Of the different BVOCs, monoterpenes responded most strongly to heat stress, with Q 10 temperature coefficients typically ranging between 7.6 and 22, which were significantly greater than the model-predicted value of 2.7. Other BVOCs including sesquiterpenes, C 9 aromatics (only detected in Calocedrus decurrens), methylbutenols, and other C 5 oxygenates were also induced by heat stress, but generally at a lower magnitude than monoterpenes. Our results indicate that Cupressaceae are a large but typically dormant source of reactive volatile hydrocarbons (mostly monoterpenes) whose emissions can be activated by heat stress. Latest studies confirmed that abnormal function of histone deacetylase (HDAC) plays a pivotal role in formation of tumors and is a potential therapeutic target for treating breast cancer. In this research, in-silico drug discovery approaches via quantitative structure activity relationship (QSAR) and molecular docking simulations were adapted to 43 compounds of indazole derivatives with HDAC inhibition for anticancer activity against breast cancer. The QSAR models were built from multiple linear regression (MLR), and models predictability was cross-validated by leave-one-out (LOO) method. Based on these results, compounds C32, C26 and C31 from model 3 showed superior inhibitory activity with pIC 50 of 9.30103, 9.1549 and 9.1549. We designed 10 novel compounds with molecular docking scores ranging from -7.9 to -9.3 kcal/mol. The molecular docking simulation results reveal that amino acid residues ILE1122 and PRO1123 play a significant role in bonding with 6CE6 protein. Furthermore, newly designed compounds P5, P2 and P7 with high docking scores of -9.3 kcal/mol, -8.9 kcal/mol and -8.8 kcal/mol than FDA-approved drug Raloxifene (-8.5 kcal/mol) and aid in establishment of potential drug candidate for HDAC inhibitors. The in-silico ADME functionality is used in the final phase to evaluate newly designed inhibitors as potential drug candidates. Through the revolutionization of artificial intelligence (AI) technologies in clinical research, significant improvement is observed in diagnosis of cancer. Utilization of these AI technologies, such as machine and deep learning, is imperative for the discovery of novel anticancer drugs and improves existing/ongoing cancer therapeutics. However, building a model for complicated cancers and their types remains a challenge due to lack of effective therapeutics that hinder the establishment of effective computational tools. In this review, we exploit recent approaches and state-of-the-art in implementing AI methods for anticancer drug discovery, and discussed how advances in these applications need to be considered in the current cancer therapeutics. Considering the immense potential of AI, we explore molecular docking and their interactions to recognize metabolic activities that support drug design. In this study, the physiology of symbiotic 'peacock-tail' shrimp Ancylocaris brevicarpalis and its host 'Haddon's carpet' sea anemone Stichodactyla haddoni were tested under lowered pH (7.7) and control (8.1) conditions. The biochemical responses such as digestive enzyme (AP), organic acids (lactate and succinate), oxidative damages (MDA), antioxidants metabolites/enzymes (ASC, GSH, SOD, CAT, APX, GPX, GR, POX, and PHOX), and detoxification enzyme (GST) were measured. The AP showed insignificantly reduced values in both the organisms in lowered pH conditions compared to control indicating the effect of abiotic stress. The hierarchical clustering analysis indicated low MDA in sea anemone can be explained by higher POX, APX, GR, ASC, and GSH levels compared to shrimps. However, the detoxification enzyme GST showed less activity in sea anemones compared to shrimps. The results suggest that A. brevicarpalis and sea anemone S. haddoni may have deleterious effects when exposed to short-term acidification stress., (Copyright © 2022 Elsevier Ltd. Background: Biliary anastomotic stricture (BAS) is an uncommon complication of pancreaticoduodenectomy (PD). As PDs are performed more frequently, BAS may become a more common pathologic entity requiring clinical engagement. The aim of this study was to report the incidence of BAS in the modern era of pancreatic surgery and identify risk factors associated with it., Methods: Patients undergoing PD at the Johns Hopkins Hospital between 2007 and 2016 were identified using an institutional registry and clinicopathological features were analyzed to identify risk factors associated with BAS., Results: Of 2125 patients identified, 103 (4.9%) developed BAS. Factors independently associated with BAS included laparoscopic approach (HR:2.83,95%CI:1.35-5.92, p = 0.006), postoperative pancreatic fistula (HR:2.45,95%CI:1.56-4.16,p < 0.001), postoperative bile leak (BL) (HR:5.26,95%CI:2.45-11.28,p < 0.001), and administration of adjuvant radiation therapy (HR:6.01,95%CI:3.19-11.34,p < 0.001). Malignant pathology was associated with lower rates of BAS (HR:0.52,95%CI:0.30-0.92, p = 0.025). BL was associated with higher rates of early-BAS (HR:16.49,95%CI:3.28-82.94, p = 0.001) while use of Vicryl suture for biliary enteric anastomosis was associated with lower rates of early-BAS (HR:0.20,95%CI:0.05-0.93, p = 0.041)., Conclusion: Approximately 5% of patients undergoing PD experience BAS. Simultaneous atomic force microscope (AFM) and sample scanning confocal fluorescence microscope measurements are widely used to obtain mechanistic and structural insights into protein dynamics in live cells. However, the absence of a robust technique to synchronously scan both AFM and confocal microscope piezo stages makes it difficult to visualize force-induced changes in fluorescent protein distribution in cells.  To address this challenge, we have built an integrated AFM-confocal fluorescence microscope platform that implements a synchronous scanning method which eliminates image artifacts from piezo motion ramping, produces accurate pixel binning and enables the collection of a scanned image of a sample while applying force to a single point on the sample. As proof of principle, we use this instrument to monitor the redistribution of fluorescent E-cadherin, an essential transmembrane protein, in live cells, upon application of mechanical force., (Copyright © 2020 Elsevier B.V. The clinical success of coated implants in executing biological functions inclusive of sustainable drug release and long term antibacterial activity without antibiotics is critical. To this aim, a nanohybrid of silver nanoparticles (AgNPs) cored in polyvinyl alcohol nanocapsules (Ag-PVA NCs) embedded in chitosan (CS) matrix loaded with anti-inflammatory drug naproxen was prepared. The synthesized nanohybrids that were subjected to coatings on (3-aminopropyl)triethoxysilane (APTES) treated titanium (Ti) metal exhibited dual role of excellent inhibition on biofilm formation and sustained drug release. These dual characteristics are achieved mainly based on intrinsic antibacterial property of AgNPs and differential entrapment of drug in PVA polymeric shell of AgNPs and CS matrix. The coatings also demonstrated enhanced mechanical properties with increasing inorganic filler and stress shielding on Ti metal. The biocompatibility tests involving adhesion, proliferation and differentiation of osteoblast cells demonstrated the efficacy of Ag-PVA NCs embedded in CS matrix as a suitable coating material for orthopedic applications., (Copyright © 2016 Elsevier Inc. The incidence of esophageal cancer has been steadily increasing in the United States over the past 25 years. Even with standardized surgical techniques, esophagectomy is a complex, multi-phase operation with a wide range of possible complications. The Ivor-Lewis esophagectomy is a commonly used technique where the right gastroepiploic artery (RGEA) becomes the sole source of blood to the stomach. We describe a case of accidental transection of the RGEA which was then re-anastomosed followed by successful use of the gastric conduit. Cadherins are Ca(2+)-dependent cell-cell adhesion proteins that maintain the structural integrity of the epidermis; their principle function is to resist mechanical force. This review summarizes the biophysical mechanisms by which classical cadherins tune adhesion and withstand mechanical stress. We first relate the structure of classical cadherins to their equilibrium binding properties. We then review the role of mechanical perturbations in tuning the kinetics of cadherin adhesion. In particular, we highlight recent studies that show that cadherins form three types of adhesive bonds: catch bonds, which become longer lived and lock in the presence of tensile force; slip bonds, which become shorter lived when pulled; and ideal bonds, which are insensitive to tugging.

Classical cadherins are the principle adhesive proteins at cohesive intercellular junctions, and are essential proteins for morphogenesis and tissue homeostasis. Because subtype-dependent differences in cadherin adhesion are at the heart of cadherin functions, several structural and biophysical approaches have been used to elucidate relationships between cadherin structures, biophysical properties of cadherin bonds, and cadherin-dependent cell functions. Some experimental approaches appeared to provide conflicting views of the cadherin binding mechanism. However, recent structural and biophysical data, as well as computer simulations generated new insights into classical cadherin binding that increasingly reconcile diverse experimental findings. This review summarizes these recent findings, and highlights both the consistencies and remaining challenges needed to generate a comprehensive model of cadherin interactions that is consistent with all available experimental data., (Copyright © 2012 Elsevier Ltd. A CaO-P(2)O(5)-SiO(2)-ZnO bioglass was formed by the sol-gel technique and characterized by Raman spectroscopy, X-ray diffraction, energy dispersive X-ray analysis (EDXA) and scanning electron microscopy (SEM). The surface reactivity of the resultant glass-ceramic specimens was analyzed by immersion studies in simulated body fluid (SBF). SEM-EDXS and inductively coupled plasma atomic emission spectrometry techniques were used to monitor changes in the glass surface and SBF composition. Osteoblast cell culture experiments were performed to assess the biocompatibility and the alkaline phosphatase activity. Cell counts of the osteoblasts cultured on the bioglass samples were studied and compared with the polystyrene plates. The cells cultured on the bioglass disks consistently showed a higher alkaline phosphatase activity and cell counts compared to cells cultured on either polystyrene plates or the base CaO-P(2)O(5)-SiO(2) bioglass. This was due to cell proliferation and differentiation promoted by the zinc-substituted bioglass.
