1. Listening to patients: Incidence and distribution of sleep disorders in Prader-Willi syndrome

Abstract: While the medical community continues to recognize the contributions of disordered sleep to the burden of Prader-Willi syndrome (PWS), the medical literature lacks characterization of the patient burden of sleep disorders by people living with PWS and their caregivers. We developed and fielded a 72-question survey to the online PWS community, to query caregivers about their experiences with sleep symptoms. Respondents for all age groups reported sleep-disordered breathing (40 %), cataplexy-like symptoms (28.4 %), and insomnia (43.5 %). The presentation of cataplexy-like symptoms tended to change as children aged, presenting initially (ages 0–4 years) as head bobbing while eating, then transitioning to knee buckling between the ages of 5–12 years. Finally, loss of generalized tone associated with extreme emotions became more common in the teenage years. Frequent screening for potentially treatable sleep disorders should be considered the standard of care for individuals with PWS.

2. Mitochondria embedded in degalactosylated xyloglucan hydrogels to improve mitochondrial transplantation

Abstract: Mitochondria are the major source of intracellular adenosine triphosphate (ATP) and play an essential role in a plethora of physiological functions, including regulating metabolism and maintaining cellular homeostasis. Mitochondrial dysfunction is associated with the onset of several cardiovascular diseases and, although several approaches currently exist to counteract it, no treatment using mitochondria as a therapeutic target exists to date. Recently, mitochondrial transplantation (MT) has been identified as a potential therapy that leads to increased ATP production, reduced oxidative stress, and improved bioenergetics. MT involves the replacement of damaged mitochondria, following injury or diseases.With MT, mitochondria must survive an inhospitable extracellular environment often characterized by oxidizing agents due to pathological and/or inflammatory conditions. Furthermore, only a small percentage of the injected mitochondria reaches the target site due to dispersion throughout the body.In this work, an MT strategy involving degalactosylated xyloglucan hydrogel encapsulating mitochondria, to overcome MT problems and improve its efficiency, is illustrated for the treatment of cardiovascular damage. The presence of the hydrogel presents the following advantages: improves the health of mitochondria; plays a protective role towards mitochondria from the extracellular environment and oxidative stress; allows for sustained release of viable mitochondria and local transfer into host cells.

3. The value of waiting spaces: Tourists’ willingness to pay for high-quality bus terminals

Abstract: Transport quality has long been recognized as an important factor in influencing travelers’ behavior, and transport terminal quality undoubtedly plays no small part. Indeed, transit promotion policies explicitly based on qualitative factors and high-standard architecture are increasingly being adopted in designing new bus terminals and stops. High architectural standards can be found in several bus terminals worldwide. Nevertheless, the literature in transport sector has yet to explore the impact of bus terminal “hedonic quality” on users’ behavior or use their willingness to pay (WTP) in cost–benefit analyses or other transport policy applications. Hedonic value is here intended as the aggregate of all elements related to travelers’ pleasure in spending time in a terminal where architectural beauty and the passenger services offered are arguably the most visible and representative attributes. Within this context, we propose a quantitative analysis of the perceived hedonic value of bus nodes in terms of users’ WTP (pure preference) for a high-quality bus terminal. To this end a discrete choice experiment based on a visual immersive experience was performed, and data were collected from 324 residents of Milan (Italy) traveling for tourism. Different binomial Mixed Logit models with panel data and random coefficients were estimated for the purpose. Monetary valuation of pure preference for using a high-quality bus terminal was estimated with a mean of about 25 % of the average trip cost observed for the case study considered. This pure preference means that the Italian tourist is willing to spend up to €4.35/trip more for a high-quality bus waiting space or travel up to 28.2 min/trip more, instead of using a traditional bus terminal for the same trip. Category-specific analyses show that females > 30 years old have a greater pure preference for a high-quality bus terminal (up to + 220 %) against males and young users. The employed also have a higher (+42 %) pure preference for the beauty of a bus terminal against others. The results of this paper should be compared with those from other case studies as they have a potential impact for transportation planning applications, underling the importance of also incorporating “hedonic quality” as explicit design variables for new/revamped transportation hub. At the same time, new challenges are posed for modeling user behavior and determining quality-related indicators and measures.

Abstract: Summary: Human genetic studies of critical COVID-19 pneumonia have revealed the essential role of type I interferon-dependent innate immunity to SARS-CoV-2 infection. Conversely, an association between the HLA-B∗15:01 allele and asymptomatic SARS-CoV-2 infection in unvaccinated individuals was recently reported, suggesting a contribution of pre-existing T cell-dependent adaptive immunity. We report a lack of association of classical HLA alleles, including HLA-B∗15:01, with pre-omicron asymptomatic SARS-CoV-2 infection in unvaccinated participants in a prospective population-based study in the United States (191 asymptomatic vs. 945 symptomatic COVID-19 cases). Moreover, we found no such association in the international COVID Human Genetic Effort cohort (206 asymptomatic vs. 574 mild or moderate COVID-19 cases and 1,625 severe or critical COVID-19 cases). Finally, in the Human Challenge Characterisation study, the three HLA-B∗15:01 individuals infected with SARS-CoV-2 developed symptoms. As with other acute primary infections studied, no classical HLA alleles favoring an asymptomatic course of SARS-CoV-2 infection were identified.

Abstract: Background: COVID-19 is primarily known as a respiratory illness; however, many patients present to hospital without respiratory symptoms. The association between non-respiratory presentations of COVID-19 and outcomes remains unclear. We investigated risk factors and clinical outcomes in patients with no respiratory symptoms (NRS) and respiratory symptoms (RS) at hospital admission. Methods: This study describes clinical features, physiological parameters, and outcomes of hospitalised COVID-19 patients, stratified by the presence or absence of respiratory symptoms at hospital admission. RS patients had one or more of: cough, shortness of breath, sore throat, runny nose or wheezing; while NRS patients did not. Results: Of 178,640 patients in the study, 86.4 % presented with RS, while 13.6 % had NRS. NRS patients were older (median age: NRS: 74 vs RS: 65) and less likely to be admitted to the ICU (NRS: 36.7 % vs RS: 37.5 %). NRS patients had a higher crude in-hospital case-fatality ratio (NRS 41.1 % vs. RS 32.0 %), but a lower risk of death after adjusting for confounders (HR 0.88 [0.83–0.93]). Conclusion: Approximately one in seven COVID-19 patients presented at hospital admission without respiratory symptoms. These patients were older, had lower ICU admission rates, and had a lower risk of in-hospital mortality after adjusting for confounders.

6. Biomimetic proteolipid vesicles for reverting GPI deficiency in paroxysmal nocturnal hemoglobinuria

Abstract: Summary: Nano-vesicular carriers are promising tissue-specific drug delivery platforms. Here, biomimetic proteolipid vesicles (BPLVs) were used for delivery of glycosylphosphatidylinositol (GPI)-anchored proteins to GPI deficient paroxysmal nocturnal hemoglobinuria (PNH) cells. BPLVs were assembled as single unilamellar monodispersed (polydispersity index, 0.1) negatively charged (ζ-potential, −28.6 ± 5.6 mV) system using microfluidic technique equipped with Y-shaped chip. GPI-anchored and not-GPI proteins on BPLV surface were detected by flow cytometry. Peripheral blood mononuclear cells (PBMCs) from healthy and PNH subjects were treated with BPLVs (final concentration, 0.5 mg/mL), and cells displayed an excellent protein uptake, documented by flow cytometry immunophenotyping and confocal microscopy. BPLV-treated cells stressed with complement components showed an increased resistance to complement-mediated lysis, both healthy and PNH PBMCs. In conclusion, BPLVs could be effective nanocarriers for protein transfer to targeted cells to revert protein deficiency, like in PNH disease. However, further in vivo studies are required to validate our preclinical in vitro results.

7. Validity of Home Blood Pressure Devices Sold in Canada

Abstract: Background: Measurement of home blood pressure is an important tool for the management of hypertension. However, the validity of home devices is of concern. The Recommended Blood Pressure Devices Program of Hypertension Canada reviews and recommends blood pressure devices using international validation standards. We sought to determine the proportion of Hypertension Canada-recommended devices available for purchase in pharmacies and online. Methods: We visited 16 community pharmacies in the Edmonton area to record the blood pressure devices they sold. We also reviewed the 50 most popular devices from online retailers (Amazon, Walmart, Best Buy, and Canadian Tire). All devices were referenced against the Recommended Blood Pressure Device Program of Hypertension Canada (www.hypertension.ca/bpdevices) to determine if the models were recommended. Results: We reviewed 170 devices. Of those sold in pharmacies, 61 of 68 (89.7%) were Hypertension Canada-recommended devices, whereas online retailers had only 46 of 102 (45.1%) recommended devices; P < 0.001. Conclusions: Most blood pressure devices sold in pharmacies are Hypertension Canada recommended, in contrast to less than one-half from online retailers. The lack of validation of many home blood pressure devices could have important clinical implications, leading to over- or undertreatment of hypertension. Clinicians should advise patients on the importance of home blood pressure device validation and direct them to resources such as Hypertension Canada (https://hypertension.ca/public/recommended-devices) for guidance. Résumé: Contexte: La mesure de la pression artérielle au domicile est un outil important dans la prise en charge de l’hypertension. Or, il semble que les appareils utilisés à la maison à cette fin ne soient pas toujours des dispositifs validés. Dans le cadre de son Programme de recommandation d’appareils de mesure de la pression artérielle, Hypertension Canada analyse et recommande des tensiomètres selon des normes de validation internationales. Nous avons donc cherché à déterminer la proportion des tensiomètres recommandés par Hypertension Canada qu’il est possible d’acheter dans les pharmacies et en ligne. Méthodologie: Nous avons rendu visite à 16 pharmacies communautaires dans la région d’Edmonton pour recenser les tensiomètres qui y étaient vendus. Abstract: Background: Prior research has established a link between thalamic pathology and cognitive impairment (CI) in people with multiple sclerosis (pwMS). However, the translation of these findings to pwMS in everyday clinical settings has been insufficient. Objective: To assess which global and/or thalamic imaging biomarkers can be used to identify pwMS at risk for CI and cognitive worsening (CW) in a real-world setting. Methods: This was an international, multi-center (11 centers), longitudinal, retrospective, real-word study of people with relapsing-remitting MS (pwRRMS). Brain MRI exams acquired at baseline and follow-up were collected. Cognitive status was evaluated using the Symbol Digit Modalities Test (SDMT). Thalamic volume (TV) measurement was performed on T2-FLAIR, as well as on T1-WI, when available. Abstract: Natural products are a rich source of bioactive molecules that have potential pharmacotherapeutic applications. In this study, we focused on Artemisia annua (A. annua) and its enriched extracts which were biologically evaluated in vitro as virucidal, antiviral, and antioxidant agents, with a potential application against the COVID-19 infection. The crude extract showed virucidal, antiviral and antioxidant effects in concentrations that did not affect cell viability. Scopoletin, arteannuin B and artemisinic acid (single fractions isolated from A. annua) exerted a considerable virucidal and antiviral effect in vitro starting from a concentration of 50 µg/mL. Data from Surface Plasmon Resonance (SPR) showed that the inhibition of the viral infection was due to the interaction of these compounds with the 3CLpro and Spike proteins of SARS-CoV-2, suggesting that the main interaction of compounds may interfere with the viral pathways during the insertion and the replication process. The present study suggests that natural extract of A. annua and its components could have a key role as antioxidants and antiviral agents and support the fight against SARS-CoV-2 variants and other possible emerging Coronaviruses.

Abstract: A flexible Surface-Enhanced Raman Spectroscopy (SERS) substrate based on silver nanoparticles encapsulated in an agar gel has been probed to detect the fungicide thiabendazole (TBZ) reaching a limit of detection (LOD) of 30 ng/cm2. In addition, a simple and sensitive strategy was employed for in-situ detection of TBZ on fruit and vegetable peels. For that purpose, peels of different fruits and vegetables were intentionally contaminated with different amount of TBZ, and the analyte was subsequently extracted within few seconds by gently rubbing the surface with the SERS substrate. The lowest value of TBZ detected on eggplant and green pepper peels was 50 ng/cm2. The values achieved for apple and pear peels were 0.20 μg/cm2 and 40 ng/cm2, respectively. On the other hand, for tomato and strawberry peels the lowest value achieved was 0.50 μg/cm2. The variation in sensitivity can be attributed to differences in the surface properties of the different peels. The above results show that this flexible SERS substrate can be further employed for the detection of contaminants in practical applications for food safety inspection.

Abstract: Concern about atmospheric microplastic (MP) contamination has increased in recent years. This study assessed the abundance of airborne anthropogenic particles, including MPs, deposited in rainfall in Bahia Blanca, southwest Buenos Aires, Argentina. Rainwater samples were collected monthly from March to December 2021 using an active wet-only collector consisting of a glass funnel and a PVC pipe that is only open during rain events. Results obtained show that all rain samples contained anthropogenic debris. The term “anthropogenic debris” is used to refer to the total number of particles as not all the particles found could be determined as plastic. Among all the samples, an average deposition of 77 ± 29 items (anthropogenic debris) m−2d−1 was found. The highest deposition was observed in November (148 items m−2d−1) while the lowest was found in March (46 items m−2d−1). Anthropogenic debris ranged in size from 0.1 mm to 3.87 mm with the most abundant particles being smaller than 1 mm (77.8%). 15. Incidence, causes, and risk factors of stillbirth in an Amazonian context: Saint Laurent du Maroni maternity ward 2016–2021

Abstract: Objective: We aimed to describe the epidemiology of intrauterine fetal deaths in multiethnic western French Guiana and to assess its main causes and risk factors. Study design: A retrospective descriptive study was conducted based on data from January 2016 to December 2021. All information on stillbirth with a gestational age ≥20 weeks in the Western French Guiana Hospital Center was extracted. Terminations of pregnancy were excluded. We focused on medical history, clinical investigation, biological findings, placental histology, and autopsy examination to elucidate the cause of death. We used the Initial Cause of Fetal Death (INCODE) classification system for assessment. Univariable and multivariable logistic regression analyses were performed. Results: Overall, 331 fetuses in 318 stillbirth deliveries were reviewed and compared to live births that occurred during the same period. The rate of fetal death varied between 1.3 % and 2.1 %, with an average of 1.8 % over the 6-year period. Poor antenatal care (104/318, 32.7 %), obesity ≥30 kg/m2 (88/318, 31.7 %), and preeclampsia (59/318, 18.5 %) were the main risk factors associated with fetal death in this group. Four hypertensive crises were reported. According to the INCODE classification, the main causes of fetal death were obstetric complications (112/331, 33.8 %), particularly intrapartum fetal death with labor-associated asphyxia under 26 weeks (64/112, 57.1 %), and placental abruption (29/112, 25.9 %). Maternal-fetal infections were common, particularly mosquito-borne diseases (e.g., Zika virus, dengue, and malaria), re-emerging infectious agents such as syphilis, and severe maternal infections (8/331, 2.4 %). 19.3 % of fetal deaths (64/331) remained unexplained. Conclusion: Change in lifestyle as well as social deprivation and isolation adversely affect pregnancy in western French Guiana, in the context of a poor health care system that is similar to what is found in the Amazonian basin. Particular attention must be paid to emerging infectious agents in pregnant women and travelers returning from the Amazon region.

Abstract: Lactoferrin (L) and gellan gum (G) nanoparticles were produced in different biopolymer proportions through electrostatic complexation to enhance the antimicrobial properties of lactoferrin. The nanoparticles were characterized according to size, charge density, morphology and antimicrobial activity against S. aureus and E. coli, in two different broths to show the effect of the broth composition on the nanoparticle activity. The 9L:1G particles showed the highest positive zeta potential (+21.20 mV) and reduced diameter (92.03 nm) which resulted in a minimum inhibitory concentration six times smaller (0.3 mg/ml) than pure lactoferrin (2 mg/ml). However, the bacteriostatic action of nanoparticles was inhibited in the presence of divalent cations. When applied to strawberries as a coating, lactoferrin nanoparticles extended fruit shelf-life up to 6 days in the presence of carboxymethylcellulose (CMC). Therefore, lactoferrin-gellan gum complexation was proved to be a promising tool to enhance lactoferrin antimicrobial action and broaden its application as a food preserver.

Abstract: Surfactant-polyelectrolyte complexes (SPECs) based on lecithin and sodium caseinate were produced and the effects of such binding on the physical, chemical and emulsifying properties were evaluated and compared with the two ingredients in isolation. Negative, neutral, and positive charged SPECs were obtained. Zeta potential values and size distributions of the SPECs were dependent on the mass ratio between compounds. Electrostatic association decreased the polydispersity index in comparison with pure compounds solutions. Analysis of interfacial properties showed that solutions containing SPECs promoted a greater reduction of surface tension and interfacial tension with sunflower oil when compared with pure compounds solutions. Emulsions produced with SPECs in 10:1 lecithin:sodium caseinate ratio proved to be more stable than emulsions prepared with pure compounds. Thus, the complexation improved the emulsifying properties of lecithin and sodium caseinate establishing SPECs as potential natural emulsifiers.

Abstract: COronaVIrus Disease 2019 (COVID-19) is a newly emerging infectious disease that spread across the world, caused by the novel coronavirus Severe Acute Respiratory Syndrome CoronaVirus 2 (SARS-CoV-2). Despite the advancements in science that led to the creation of the vaccine, there is still an urgent need for new antiviral drugs effective against SARS-CoV-2. This study aimed to investigate the antiviral effect of Paulownia tomentosa Steud extract against SARS-CoV-2 and to evaluate its antioxidant properties, including respiratory smooth muscle relaxant effects. Our results showed that P. tomentosa extract can inhibit viral replication by directly interacting with both the 3-chymotrypsin-like protease and spike protein. In addition, the phyto complex does not reduce lung epithelial cell viability and exerts a protective action in those cells damaged by tert-butyl hydroperoxide , a toxic agent able to alter cells’ functions via increased oxidative stress. These data suggest the potential role of P. tomentosa extract in COVID-19 treatment, since this extract is able to act both as an antiviral and a cytoprotective agent in vitro.

20. The neglected status of the vermetid reefs in the Mediterranean Sea: A systematic map

Abstract: Studied since late 1800, vermetid reefs are marine bioconstructions of well-acknowledged importance in the Mediterranean Sea. Despite their persistence being jeopardized in the whole basin, recent studies have referred to this bioconstruction as a neglected habitat. In this study, we assessed the neglected status of the Mediterranean vermetid reefs in the scientific literature producing a systematic map through a multi-method bibliometric protocol. Scopus and Web of Science databases were jointly used for data collection. Vermetid reefs publication rate (i.e., number of publications per year) was investigated compared to the other Mediterranean bioconstructions using ANOVA analysis and Zero-Inflated Poisson regression. Later, VOSviewer software was used to perform a bibliometric network analysis and for mapping visualization. The analysis aimed at investigating gaps, patterns, and trends of the vermetid reefs together with the other main Mediterranean bioconstructions (i.e., Astroides calycularis, Cladocora caespitosa and coralligenous formations, and sabellariid and Lithophyllum reefs). The ANOVA analysis of the number of publications from 1966 to 2020 found statistically significant differences between coralligenous and vermetid reefs publication rates in the 2006–2010, 2011–2015, and 2016–2020 timeframes and pointed out a clear before/after-2010 pattern in coralligenous publication rate, which was also confirmed by the Zero-Inflated Poisson regression model. The bibliometric network analysis of the bioconstructions literature revealed the same temporal pattern, with the vermetid reefs poorly investigated and weakly connected to newer research lines and conservation topics. Instead, coralligenous showed strong connections with biodiversity conservation and was indicated as a recent research hotspot. Overall, the results of this study confirm previous references of the vermetid reefs as a neglected habitat and, among others, show an increasing research interest in the coralligenous topic.

Abstract: ABSTRACT: To describe a rare case of symptomatic pseudoaneurysm of the superficial termporal artery. ear old man after cranial trauma. A 79-year-old man presented to our hospital with an appearance of a pulsating swelling in the right front-temporal region, with signs of ischemic suffering of the overlying skin. The patient reported having accidentally fallen about 7 days earlier and reported a non-commotional blunt head injury apparently without any subcutaneous hematoma. The patient underwent an echocolordoppler of the pulsating swelling, reporting the presence of a pseudo-aneurysm of the temporal artery and pre-occlusive carotid stenosis. The patient underwent urgent surgery of the pseudoaneurysm and after 30 days, the patient was once again hospitalized in order to perform the carotid endoarterectomy. The course was regular and the patient was discharged on the third postoperative day. At the level of the right frontotemporal region, complete healing of the skin and the absence of pulsating fear were observed.

Abstract: Introdução: Pessoas imunocomprometidas apresentam alto risco de infecção persistente pelo Papiloma Vírus Humano (HPV) e de cânceres associados. A melhor estratégia de prevenção é a vacinação de suscetíveis. Objetivo: Avaliar imunogenicidade e segurança da vacina HPV quadrivalente (HPV4V) em transplantadas de órgãos sólidos, em comparação a mulheres imunocompetentes da mesma faixa etária. Método: Ensaio clínico aberto, em que foram incluídas mulheres de 18 a 45 anos transplantadas de órgãos sólidos e imunocompetentes. Todas as participantes receberam três doses da vacina quadrivalente (HPV4V, aos 0, 2 meses e 6 meses). Foram coletadas amostra de sangue antes e um mês após a terceira dose da vacina para avaliação da resposta imune à vacinação, avaliada pela frequência de soroconversão e títulos médios geométricos [GMT] de anticorpos anti-HPV. Foi realizado ensaio de anticorpos neutralizantes baseado em pseudovírions (multiplexed pseudovirion-based serological assay [PsV-Luminex]). Eventos adversos (EA) solicitados, locais e sistêmicos, foram avaliados desde o momento da vacinação até sete dias após. EA graves não solicitados foram avaliados durante todo o período do estudo. Resultados: Foram incluídas 125 transplantadas de órgãos sólidos (69 rim, 4 rim e pâncreas, 28 fígado, 17 pulmão e 8 coração) e 132 mulheres imunocompetentes, das quais 105 transplantadas e 119 imunocompetentes completaram o estudo. Houve diferença estatisticamente significativa na soroconversão entre os dois grupos: entre as transplantadas soronegativas na inclusão, a soroconversão variou conforme o tipo de HPV, 57% (HPV18), 69% (HPV6 e HPV16) e 72% (HPV11), enquanto 100% das imunocompetentes apresentaram soroconversão para os quatro tipos de HPV vacinais. Os GMT de anticorpos tipo-específicos também foram significativamente maiores nas imunocompetentes, para todos os tipos de HPV incluídos na vacina HPV4V. A frequência de eventos adversos após a vacinação foi semelhante nos dois grupos, com exceção de dor no local, mais frequente nas imunocompetentes, e náuseas após a 2ª dose, mais frequente nas transplantadas. Houve um óbito não relacioando à vacinação e uma transplantada apresentou rejeição e não retornou para avaliação. Conclusão: A vacina HPV4V é segura nas mulheres transplantadas de órgãos sólidos, porém a resposta imune foi significativamente inferior quando comparada a mulheres imunocompetentes da mesma faixa etária.Ag. Abstract: Introdução: Alguns estudos sugerem que medicamentos antirretrovirais (ARV) possuem efeito inibitório sobre o SARS-CoV-2 em modelos in vitro, porém não foram observados benefícios do uso de ARV no tratamento de pessoas com COVID-19. O efeito potencial do uso contínuo de medicamentos ARV sobre o risco de infecção por SARS-CoV-2 não é conhecido. Objetivo: Comparar a porcentagem de infecções por SARS-CoV-2 em PVHIV e controles durante o período anterior à implementação de vacinas no Brasil. Método: Estudo de coorte incluindo PVHIV sob tratamento ARV e contactantes próximos sem diagnóstico de infecção por HIV acompanhados por ?120 dias com avaliação clínica semanal e avaliação sorológica (IgM/IgG) ao início (TS1) e final (TS2) do seguimento. A infecção foi definida pela soroconversão de IgG (TS1 negativo e TS2 positiva) e/ou positividade em exame laboratorial (PCR ou antígeno) durante o período do estudo. Resultados: Entre abril/2020 e janeiro/2021, foram incluídos 267 PVHIV e 175 controles com mediana de idade de 52 e 44 anos, respectivamente; desses, 25 PVHIV e 56 controles não realizaram o TS2. Sintomas gripais foram relatados por 56 PVHIV e 35 controles ao longo do estudo, e infecções confirmadas por PCR foram registradas para 6 PVHIV e 3 controles. Um total de 74 amostras tiveram IgG positivo; entre PVHIV, 14 tiveram resultado reagente no TS1 e TS2, e 18 tiveram resultado reagente somente no TS2. No grupo controle, 5 indivíduos tiveram resultado reagente no TS1 e TS2, e 2 tiveram resultado reagente somente no TS2. A incidência de infecção estimada foi de 10% entre PVHIV (23/242; IC95% 6-14) e 3% entre controles (4/119; IC95% 1-8), sugerindo ausência de efeito protetor estatisticamente significante do uso contínuo de ARV sobre o risco de infecção por SARS-CoV-2. Conclusão: Apesar do efeito antiviral in vitro demonstrado em alguns estudos, o uso de ARV como profilaxia pré-exposição ao SARS-CoV-2 não parece ser relevante.

Abstract: Introdução: Testes de neutralização têm se tornado a principal referência para avaliar proteção após exposição ao SARS-CoV-2 ou após a vacinação. Alguns estudos sugerem que pessoas que vivem com HIV (PVHIV) têm menor probabilidade de soroconversão após a vacinação para COVID-19, porém a resposta humoral após infecção natural é pouco conhecida. Objetivo: Avaliar a positividade e títulos de anticorpos neutralizantes em PVHIV e controles com IgG positivo identificados no Estudo Prevent, realizado antes da implementação das vacinas para COVID-19 no Brasil. Método: O Estudo Prevent incluiu PVHIV sob tratamento ARV e contactantes próximos sem diagnóstico de infecção por HIV acompanhados por 120 dias com avaliação clínica semanal e avaliação sorológica (IgM/IgG) ao início (TS1) e final (TS2) do seguimento, entre abril/2020 e janeiro/2021. Todas as amostras com IgG reagente (+) foram submetidas a um teste correlato de anticorpos neutralizantes (TCAN). Resultados: Um total de 74 amostras tiveram IgG reagente; entre PVHIV, 9 tiveram TS1+ e TS2 não reagente (NR); 14 tiveram TS1+ e TS2+; e 18 tiveram TS1 NR e TS2+. No grupo controle, 6 tiveram TS1+/TS2 NR; 5 tiveram TS1+ e TS2+ e apenas 2 tiveram TS1 NR e TS2+. Quanto à avaliação do TCAN, houve positividade em 39/56 (69%; IC95% 56-81) amostras de PVHIV, e em 14/18 (78%; IC95% 52-94) amostras de controles. 21 amostras foram positivas no TS e negativas no TCAN (17 PVHIV e 4 controles) além de 1 amostra TNeutrAc indeterminada após TS positivo (PVHIV). Embora as medianas de porcentagens de neutralização tenham sido mais altas entre controles em relação a PVHIV tanto nas amostras iniciais quanto ao término do estudo, essa diferença não atingiu significância estatística. Conclusão: Testes de neutralização para SARS-CoV-2 ainda possuem aplicabilidade e interpretação controversos. Entretanto, até o momento consistem na metodologia mais aceita para avaliar níveis de proteção contra o vírus. Nossos resultados sugerem tendência a resposta neutralizante inferior entre PVHIV comparadas com controles.

Abstract: Summary: Background: Early death after cancer surgery is higher in low-income and middle-income countries (LMICs) compared with in high-income countries, yet the impact of facility characteristics on early postoperative outcomes is unknown. The aim of this study was to examine the association between hospital infrastructure, resource availability, and processes on early outcomes after cancer surgery worldwide. Methods: A multimethods analysis was performed as part of the GlobalSurg 3 study—a multicentre, international, prospective cohort study of patients who had surgery for breast, colorectal, or gastric cancer. The primary outcomes were 30-day mortality and 30-day major complication rates. Potentially beneficial hospital facilities were identified by variable selection to select those associated with 30-day mortality. Adjusted outcomes were determined using generalised estimating equations to account for patient characteristics and country-income group, with population stratification by hospital. Findings: Between April 1, 2018, and April 23, 2019, facility-level data were collected for 9685 patients across 238 hospitals in 66 countries (91 hospitals in 20 high-income countries; 57 hospitals in 19 upper-middle-income countries; and 90 hospitals in 27 low-income to lower-middle-income countries). The availability of five hospital facilities was inversely associated with mortality: ultrasound, CT scanner, critical care unit, opioid analgesia, and oncologist. Abstract: Introdução/Objetivo: Entre pessoas que vivem com HIV/Aids (PVHA), a realização anual de teste tuberculínico cutâneo (PPD) ou interferon-γ release assay (IGRA) é recomendada pelo Ministério da Saúde do Brasil para investigação da tuberculose (TB) latente para pacientes com contagem de linfócitos T CD4+ >350 cel/mm3 que não tenham tratamento prévio, indicação atual de tratamento de TB ou TB latente. Ainda há incertezas sobre a eficácia e concordância dos dois testes no diagnóstico da TB latente em PVHA no cenário epidemiológico brasileiro. A concordância entre os testes pode variar de acordo com diversos fatores, incluindo histórico clínico, tratamento do HIV e exposição prévia à BCG. O objetivo deste estudo foi avaliar a concordância entre os testes PPD e IGRA entre PVHA. Métodos: Após consentimento, PVHA maiores de 18 anos em seguimento em São Paulo, Fortaleza, Natal e Campo Grande, sem histórico de TB ou PPD positivo prévios, foram submetidos aos exames IGRA (Quantiferon Gold-Plus - Qiagen) e PPD, preferencialmente no mesmo dia. A concordância entre os testes foi avaliada pela concordância global e estatística kappa. Resultados: 523 voluntários foram incluídos para a análise de concordância. A maioria era parda (46%), do sexo masculino (64%), com mediana de idade de 48 anos (IIQ 36-58). A mediana de tempo desde o diagnóstico do HIV foi de 10 anos (IIQ 3-20); a mediana dos linfócitos T CD4+ foi de 573 cel/mm3 (IQR 411-792); e 88% possuíam carga viral indetectável. A concordância global entre os testes foi de 89%; 23 (4,4%) apresentaram PPD+/IGRA- e 41 (7,8%) apresentaram IGRA+/PPD-, com estatística kappa de 0,38 (concordância razoável), sem alteração significativa na performance do teste kappa ao estratificar a análise por categorias de valor de linfócitos T CD4+. Conclusão: Nessa coorte de PVHA com alta cobertura de vacinação por BCG na primeira infância, PPD e IGRA apresentaram concordância razoável, com a maioria dos testes discordantes resultando de IGRA positivo e PPD negativo. Estudos prospectivos avaliando risco de desenvolvimento de tuberculose ativa na população com resultados divergentes são necessários para esclarecer qual a melhor estratégia diagnóstica nessa população. Abstract: Introdução: A tuberculose continua a ser a doença oportunista mais frequente na população vivendo com HIV/aids (PVHA), sendo fundamental o diagnóstico precoce e tratamento da infecção latente por Mycobacterium tuberculosis (ILMTB) para evitar a progressão para doença ou óbito. Nesse estudo, descrevemos e comparamos a aceitabilidade dos testes tuberculínico (TT) e teste de resposta linfocitária (IGRA) para diagnóstico da ILMTB em PVHA. Métodos: Este é um estudo de corte transversal aninhado ao protocolo “Custo-efetividade do rastreamento da tuberculose latente em população vivendo com HIV/aids - Estudo CERTH”. A avaliação baseou-se em um questionário estruturado com 12 perguntas, utilizando respostas em escala Likert. Resultados: Um total de 664 participantes responderam ao questionário. Destes, 65% eram homens cis, 37% brancos e 50% pardos, com idade mediana de 46 anos (intervalo interquartil [IIQ] 34-56 anos) e mediana de 11 anos de estudo (IIQ 8-14); 70% já tinham realizado o TT, 6% já haviam falhado em retornar para leitura e 2% não compareceram para a leitura do TT. Porcentagens semelhantes de participantes relataram ter tido medo do exame (6% para TT, 5% para IGRA), enquanto o relato de dor ao realizar o exame foi mais frequente para o TT (13%, IC95% 10-15%) em relação ao IGRA (8%, IC 95% 6-10%). Quanto à probabilidade de recomendar os testes diagnósticos (net promoter score) não observamos diferença significativa entre os testes, com 92 e 93% dos participantes classificados como promotores do TT e IGRA, respectivamente. Conclusão: Nãoobservamos diferenças clinicamente relevantes na aceitabilidade do IGRA em comparação ao TT. Outros benefícios conhecidos do IGRA (menor demanda de visitas ao serviço, ausência de falha de leitura, ausência de eventos adversos tardios) devem ser considerados na implementação, assim como aspectos ainda em estudo (custo, custo-efetividade, concordância e acurácia).

Abstract: Demand for products containing natural compounds has grown in recent decades and synthetic emulsifiers probably will be replaced. Lecithin is a natural emulsifier, but it has low emulsifying capacity for stabilizing oil-in water emulsions and does not show antimicrobial property. Thus, we proposed the development of surfactant-polyelectrolyte complexes (SPECs) based on lecithin (Lip) and chitosan (Chi) in order to provide antimicrobial properties and to enhance the emulsifying properties of lecithin. The physicochemical properties of SPECs were depending on the mass ratios between compounds. SPECs showed sizes ranging from 1.5 to 5.5 microns and Zeta potential values ranging from −49.5 mV to +38.5 mV. All SPECs showed surfactant properties reducing superficial tension and interfacial tension with sunflower oil. SPECs with positive Zeta potentials values and at concentration of 2.5 mg/mL were able to inhibit the Escherichia coli growth. Through droplet size measurements, may be observed that emulsions prepared with SPECs showed coalescence decreasing when compared with those prepared only with lecithin or chitosan. These results showed that SPECs can be modulated in their physicochemical properties to be potential emulsifiers with antimicrobial properties.

Abstract: Introduction: Double occurrence of TTF1 and ΔNp63/p40 (henceforth, p40) within the same individual cells is exceedingly rare in lung cancer. Little is known on their biological and clinical implications. Methods: Two index cases immunoreactive for both p40 and TTF1 and nine tumors selected from The Cancer Genome Atlas (TCGA) according to the mRNA levels of the two relevant genes entered the study. Results: The two index cases were peripherally located, poorly differentiated, and behaviorally unfavorable carcinomas, which shared widespread p40 and TTF1 decoration within the same individual tumor cells. They also retained SMARCA2 and SMARCA4 expression, while variably stained for p53, cytokeratin 5, and programmed death-ligand 1. A subset of basal cells p40+/TTF1+ could be found in normal distal airways. Biphenotypic glandular and squamous differentiation was unveiled by electron microscopy, along with EGFR, RAD51B, CCND3, or NF1 mutations and IGF1R, MYC, CCND1, or CDK2 copy number variations on next-generation sequencing analysis. The nine tumors from TCGA (0.88% of 1018 tumors) shared the same poor prognosis, clinical presentation, and challenging histology and had activated pathways of enhanced angiogenesis and epithelial-mesenchymal transition. Mutation and copy number variation profiles did not differ from the other TCGA tumors. Conclusions: Double p40+/TTF1+ lung carcinomas are aggressive and likely underrecognized non–small cell carcinomas, whose origin could reside in double-positive distal airway stem-like basal cells through either de novo-basal-like or differentiating cell mechanisms according to a model of epithelial renewal.

Abstract: Neurodegenerative diseases affect millions of people worldwide and the presence of various physiological barriers limits the accessibility to the brain and reduces the efficacy of various therapies. Moreover, new carriers having targeting properties to specific brain regions and cells are needed in order to improve therapies for the brain disorder treatment. In this study, for the first time, Myelin nanoVesicles (hereafter defined MyVes) from brain-extracted myelin were produced. The MyVes have an average diameter of 100–150 nm, negative zeta potential, spheroidal morphology, and contain lipids and the key proteins of the myelin sheath. Furthermore, they exhibit good cytocompatibility. The MyVes were able to target the white matter and interact mainly with the microglia cells. The preliminary results here presented allow us to suppose the employment of MyVes as potential carrier to target the white matter and microglia in order to counteract white matter microglia-related diseases.

Abstract: Photolysis products of dichloroacetyl chloride (DCAC) isolated in Ar matrix and in gas phase, in the absence and presence of molecular oxygen, were studied by means of FTIR spectroscopy. The samples were exposed to light of different energy ranges (200–800, 280–320, 350–450 and 400–800 nm). DCAC is photostable when irradiated with light of wavelengths above 400 nm, and gives dichloroketene after photolysis with light between 280–320 and 350–450 nm. Exposure of DCAC to broad-band radiation (200–800 nm) produces dichloroketene as an intermediate photoproduct, and different 1:1 CHCl3:CO molecular complexes after further irradiation. HCl, CO and CHCl3 were detected in gas-phase DCAC photolysis. ClC(O)CCl2CCl2H molecule was also proposed, in a mechanism that involve the insertion of:CCl2 biradical into the C−C bond of DCAC. The photochemical reaction of DCAC with O2 in Ar matrix gives Cl2CO and CO2. The same photoproducts, together with HCl, were observed in the gas-phase photochemical reaction. Additionally, the FTIR spectra of DCAC isolated in solid Ar were fully interpreted in terms of an equilibrium between syn (the H−C bond syn with respect to the C=O bond) and gauche conformers. Some absorptions, which grow as the DCAC:Ar ratio increases, were attributed to dimeric forms. The most stable dimer was predicted by DFT calculations as composed by two DCAC molecules with syn conformations, interacting through two H-bonds in a structure with Ci symmetry.

Abstract: Fragrance materials (FMs) are used in a variety of detergents and cosmetics, including household and personal care products. Despite their widespread use and the growing evidence of their occurrence in surface waters worldwide, very little is known about their toxicity towards marine species, including a key component of the marine food webs such as copepods. Thus, we investigated the toxicity of six of the more long-lasting and stable commercial fragrances, including Amyl Salicylate (AMY), Oranger Crystals (ORA), Hexyl Salicylate (HEX), Ambrofix (AMB), Peonile (PEO), and Benzyl Salicylate (BZS), to assess their ability to impair the larval development of the calanoid copepod Acartia tonsa. FMs inhibited the development of A. tonsa significantly at concentrations by far lower than the effect-concentrations reported in the literature for aquatic species. The more toxic FMs were HEX (EC50 = 57 ng L−1), AMY (EC50 = 131 ng L−1) and ORA (EC50 = 766 ng L−1), while the other three compounds exerted toxic effects at concentrations higher than 1000 ng L−1 (LOEC at 1000 ng L−1 for PEO and BZS, and at 10,000 ng L−1 for AMB). Early life-stage mortality was unaffected by FMs at all the tested concentrations. A comparison with water concentrations of FMs reported in the literature confirmed that FMs, especially HEX and AMY, may act as contaminants of potential concern in many aquatic habitats, including urban areas and remote and polar environments.

Abstract: Background: Antiretroviral therapy (ART) has decreased AIDS incidence and mortality, rendering comorbidities, such as hepatitis B more relevant for people living with human immunodeficiency virus (HIV). Since antiretroviral drugs may also inhibit hepatitis B virus (HBV) replication, analyzing the impact of ART on management of hepatitis B in this population is important. Objective: To assess HBV viremia among HIV/HBV coinfected individuals on ART and its associated factors. Method: For this cross-sectional study, HIV/HBV-coinfected individuals, aged over 18 years, who were on ART for over six months and receiving care at an outpatient clinic in São Paulo were recruited. Sociodemographic characteristics, information about viral exposure, clinical and laboratory data, including evaluation of liver fibrosis were obtained. Plasma HBV DNA was measured by polymerase chain reaction. Viral genome sequencing was conducted for genotyping and identification of drug resistance-conferring mutations if viral load exceeded 900 IU/mL. Results: Out of 2,946 patients who attended the clinic in 2015, 83 were eligible and 56 evaluated. Plasma HBV DNA was detected in 16 (28.6%) (95% CI: 18.0–41.3%), all on lamivudine and tenofovir treatment. HBV DNA detection was associated with lower education (p = 0.015), higher international normalized ratios (p = 0.045), history of an AIDS-defining illness [OR: 3.43 (95% CI: 1.10–11.50)], and HBeAg detection [OR: 6.60 (95% CI: 1.84–23.6)]. In contrast, a last CD4+ count above 500 cells/mm3 in the year prior to inclusion [OR: 0.18 (95% CI: 0.04–0.71)] and detection of anti-HBe [OR: 0.21 (95% CI: 0.04–0.99)] were negatively associated. Patients with HBV DNA above 900 IU/mL were infected with subgenotypes A1 (n = 3) and D2 (n = 1), and exhibited viral mutations associated with total resistance to lamivudine and partial resistance to entecavir. Conclusions: Despite being on ART, a significant proportion of HIV/HBV-coinfected individuals present HBV viremia. Characterization of factors that are associated with this finding may help professionals provide better management to these patients. 34. Glucagon-like peptide-2 reduces the obesity-associated inflammation in the brain

Abstract: Growing evidence suggests a link between obesity and neurodegeneration. The purpose of the present study was to explore the neuroprotective potential of glucagon-like peptide-2 (GLP-2) in the brain of high fat diet (HFD)-fed mice. Markers of inflammation and oxidative stress were analysed in the brains of obese mice chronically treated with [Gly2]-GLP-2 (teduglutide), the stable analogue of the GLP-2, and they were compared to age-matched untreated obese and lean animals. Neurodegeneration was examined by TUNEL assay. HFD feeding increased the expression of pro-inflammatory mediators (NF-kB, IL-8, TNF-α, IL-1β and IL-6), glial fibrillary acidic protein (GFAP), index of gliosis and neurodegeneration, stress marker proteins (p-ERK, Hsp60 and i-NOS), amyloid-β precursor protein (APP). [Gly2]-GLP-2 treatment significantly attenuated the HFD-induced increased expression of the various markers, as well as the higher levels of reactive oxygen species found in brains of untreated-HFD mice. Immunofluorescence confirmed that the increase of GFAP or APP in the brain cortex of HFD mice were less prominent in the [Gly2]-GLP-2 treated group. TUNEL-positive cell number in brain sections of [Gly2]-GLP-2-treated HFD-fed mice was significantly lesser in comparison with untreated-HFD animals and similar to STD fed mice. In conclusion, the results of the present study suggest that GLP-2 stable analogue improves the obesity-associated neuroinflammation and the central stress conditions, it reduces the neuronal apoptotic death, providing evidence for a neuroprotective role of the peptide.

36. Evaluation of artificial intelligence systems for assisting neurologists with fast and accurate annotations of scalp electroencephalography data

Abstract: Background: Assistive automatic seizure detection can empower human annotators to shorten patient monitoring data review times. We present a proof-of-concept for a seizure detection system that is sensitive, automated, patient-specific, and tunable to maximise sensitivity while minimizing human annotation times. The system uses custom data preparation methods, deep learning analytics and electroencephalography (EEG) data. Methods: Scalp EEG data of 365 patients containing 171,745 s ictal and 2,185,864 s interictal samples obtained from clinical monitoring systems were analysed as part of a crowdsourced artificial intelligence (AI) challenge. Participants were tasked to develop an ictal/interictal classifier with high sensitivity and low false alarm rates. We built a challenge platform that prevented participants from downloading or directly accessing the data while allowing crowdsourced model development. Findings: The automatic detection system achieved tunable sensitivities between 75.00% and 91.60% allowing a reduction in the amount of raw EEG data to be reviewed by a human annotator by factors between 142x, and 22x respectively. The algorithm enables instantaneous reviewer-managed optimization of the balance between sensitivity and the amount of raw EEG data to be reviewed. Interpretation: This study demonstrates the utility of deep learning for patient-specific seizure detection in EEG data. Furthermore, deep learning in combination with a human reviewer can provide the basis for an assistive data labelling system lowering the time of manual review while maintaining human expert annotation performance. Abstract: Digital twins are virtual software copies of physical objects and systems, and represent a strategic technology enabler to support Internet of Things devices and systems. Existing software frameworks for digital twins mainly operate in the cloud and are based on platform-specific solutions, harming interoperability and adaptability. However, it is getting recognized that Internet of Things and digital twins architectures can take advantage of microservices and platform-independent distributed architectures (also on the edge), promoting higher scalability, adaptability, and interoperability. In this context, we introduce WLDT (White Label Digital Twins), a general-purpose library that allows developers to create digital twins in terms of modular, adaptable, and inter-operable software agents. Among different features, WLDT library supports multiple standard protocols, caching, software processing pipeline and metrics monitoring.

Abstract: The Java Programming Language revolutionized the world of software development in the last decades. Thanks to its portability, Java makes it possible to develop software that can run everywhere, in a truly cross-platform computing environment. Although running the same Java code anywhere works smoothly on major desktop and server platforms, this becomes much more complicated when different devices and platforms, such as smartphones or embedded systems, are taken into account. Furthermore, even if we consider devices that natively support the Java programming language, the same application may not run without re-writing part or the entire source code. This is mainly due to the existence of platform specific libraries for accessing input/output peripherals or system-specific features. In particular, the main limitation is usually associated to the different APIs that must be used for programming the Graphical User Interface (GUIs). In this paper, we present a novel framework that can be used by developers to write Java applications with portable GUIs that are truly platform-independent and thus can run on different systems such as PCs, Workstations, Android devices or mobile phones and embedded systems with Java MicroEdition (Java ME).

Abstract: Proteolytic resistance is a relevant aspect to be tested in the formulation of new nanoscale biomaterials. The action of proteolytic enzymes is a very fast process occurring in the range of few minutes. Here, we report data concerning the proteolytic resistance of a heat-set BSA hydrogel obtained after 20-hour incubation at 60 °C prepared at the pH value of 3.9, pH at which the hydrogel presents the highest elastic character with respect to gel formed at pH 5.9 and 7.4 “Heat-and pH-induced BSA conformational changes, hydrogel formation and application as 3D cell scaffold” (G. Navarra, C. Peres, M. Contardi, P. Picone, P.L. San Biagio, M. Di Carlo, D. Giacomazza, V. Militello, 2016) [1]. We show that the BSA hydrogel produced by heating treatment is protected by the action of proteinase K enzyme. Abstract: Next generation sequencing platforms have recently been used to rapidly characterize transcriptome sequences from a number of non-model organisms. The present study compares two of the most frequently used platforms, the Roche 454-pyrosequencing and the Illumina sequencing-by-synthesis (SBS), on the same RNA sample obtained from an intertidal gastropod mollusc species, Haliotis midae. All the sequencing reads were deposited in the Short Read Archive (SRA) database are retrievable under the accession number [SRR071314 (Illumina Genome Analyzer II)] and [SRR1737738, SRR1737737, SRR1737735, SRR1737734 (454 GS FLX)] in the SRA database of NCBI. Three transcriptomes, composed of either pure 454 or Illumina reads or a mixture of read types (Hybrid), were assembled using CLC Genomics Workbench software. Illumina assemblies performed the best de novo transcriptome characterization in terms of contig length, whereas the 454 assemblies tended to improve the complete assembly of gene transcripts. Both the Hybrid and Illumina assemblies produced longer contigs covering more of the transcriptome than 454 assemblies. However, the addition of 454 significantly increased the number of genes annotated.

Abstract: Multiple sclerosis (MS) is a complex autoimmune disease with multiple players. In particular, peripheral (myelin-reactive CD4+ T lymphocytes) and central immune cells (microglia) are involved in the neuroinflammatory process and are found in MS brain lesions. New nanotechnological approaches that can cross the blood-brain barrier and specifically target the key players in the disease using biocompatible nanomaterials with low immunoreactivity represent an important challenge. To this end, nanoparticles and nanovesicles have been studied to induce immune tolerance to a wide range of myelin-derived antigens as potential approaches against MS. To this aim, we extracted myelin from bovine brain and produced myelin-based nanovesicles (MyVes) by nanoprecipitation. MyVes have a diameter of about 100 nm, negative zeta potential and contain the typical proteins of the myelin sheath. The results showed that MyVes are not cytotoxic, are hemocompatibile and do not induce an inflammatory response. In vitro experiments showed that MyVes are specifically taken up by microglial cells and are able to induce the expression of the anti-inflammatory cytokine IL-4. In addition, we have used biodistribution experiments to show that MyVes are able to reach the brain after intranasal administration. Finally, MyVes induced the production of the anti-inflammatory cytokines IL-10 and IL-4 in peripheral blood mononuclear cells isolated from MS patients. Taken together, these data provide proof of concept that MyVes may represent a safe nanosystem capable of promoting anti-inflammatory effects by modulating both central and peripheral immune cells to treat neuroinflammation in MS. STATEMENT OF SIGNIFICANCE: Recently, nanoparticles and nanovesicles have been investigated as potential approaches for the treatment of neurodegenerative diseases. We propose the use of myelin nanovesicles (MyVes) as a potential application to counteract neuroinflammation in multiple sclerosis (MS). Approximately 2.8 million people worldwide are estimated to live with MS. It is an autoimmune disease directed toward various myelin-derived antigens. Both peripheral immune cells (lymphocytes) and central immune cells (microglia) actively contribute to MS brain lesions. Abstract: Coronaviruses (CoVs) are large positive stranded enveloped RNA viruses that generally cause enteric and respiratory diseases in humans and in animals. Most human CoVs have recently attracted global attention to their lethal potential and great infectious capacity. A highly pathogenic CoV, called COVID-19 or SARS-CoV-2, dramatically emerged in December 2019 in Wuhan, China. This new CoV has caused severe pneumonia in China and rapidly spreads around the world, the COVID-19 pandemic. Growing evidence pieces show that viruses, such as CoVs, can enter the central nervous system from different pathways and inducing neurotoxicity. Therefore, it is urgent to make clear whether SARS-CoV-2 has access to the central nervous system and can cause direct neuronal effects. Moreover, a brain-lung-brain axis is been proposed from the scientific community where severe neurological dysfunction and injury are associated with lung injury, and vice versa. In this axis, virus-induced inflammation and oxidative stress could be the common mechanisms responsible for CoV neurological symptoms. Abstract: Crosslinked xyloglucan-poly(vinyl alcohol) based hydrogel films are interesting materials for wound healing applications. This work focuses on the hydrolytic degradation and consequent morphological modification of a XG-PVA film and on its interaction with cells, blood, bacteria. Biocompatibility of the film was assessed in vitro by investigating different aspects, such as cell viability, oxidative stress level, mitochondrial dysfunction and specific stress biomarkers. Partial adhesiveness was demonstrated by performing different attaching assays and phalloidin staining. Hemocompatibility of XG-PVA film after interaction with blood was evaluated by using a multi-parametric approach, including human Red Blood Cells (RBC) count, hemolytic response and platelets activation. Thrombin and fibrinogen concentrations were examined as marker of the coagulation cascade. After direct contact with human blood and peripheral blood mononuclear cells (PBMC), no evidence of cell defense response was observed. Antimicrobial activity of XG-PVA film was tested against Escherichia coli (E.coli). XG-PVA film promotes bacterial retentivity and provides mechanical protection against bacterial infiltration. After loading the film with ampicillin, an inhibitory E. coli growth zone was observed. All together these results indicate that the XG-PVA system is a promising material to be tested in vivo for wound healing applications., (Copyright © 2018. Published by Elsevier B.V.)

Abstract: Aggregation and gelation of globular proteins can be an advantage to generate new forms of nanoscale biomaterials based on the fibrillar architecture. Here, we report results obtained by exploiting the proteins' natural tendency to self-organize in 3D network, for the production of new material based on BSA for medical application. In particular, at five different pH values the conformational and structural changes of the BSA during all the steps of the thermal aggregation and gelation have been analyzed by FTIR spectroscopy. The macroscopic mechanical properties of these hydrogels have been obtained by rheological measurements. The microscopic structure of the gels have been studied by AFM and SEM images to have a picture of their different spatial arrangement. Abstract: Alzheimer's disease (AD) is the most common form of dementia. The cause of AD is closely related to the accumulation of amyloid beta peptide in the neuritic plaques. The use of animal model systems represents a good strategy to elucidate the molecular mechanism behind the development of this pathology. Here we use the Paracentrotus lividus embryo to identify molecules and pathways that can be involved in the degenerative process. As a first step, we identified the presence of an antigen related to the human APP, called PlAPP. This antigen, after gastrula stage, is processed producing a polypeptide of about 10kDa. By immunohistochemistry we localized the PlAPP antigen in some serotonin expressing cells. Similarly, after 48 or 96h incubation, a recombinant beta-amyloid peptide, rAbeta42, accumulates around the intestinal tube and oesophagus. In addition, incubation of sea urchin embryos with two different solutions rich in oligomers and fibrillar aggregates of rAbeta42 induce activation of apoptosis as detected by TUNEL assay. Moreover, we demonstrate that aggregates induce apoptosis by extrinsic pathway activation, whereas oligomers induce apoptosis both by extrinsic and intrinsic pathway activation. Utilizing an apoptotic inhibitor, caspases activation was offset and morphological damage rescued. Taken together all these observations suggest that the sea urchin may be a simple and suitable model to characterize the mechanism underlining the cytotoxicity of Abeta42.
