Urban areas are often hotspots for the dissemination of non-native (invasive) plant species, some of which release (potentially) allergenic pollen. Given the high population density in cities, a considerable number of people can be regularly and potentially intensively exposed to the pollen from these plants. This study delves into the Tree-of-Heaven (Ailanthus altissima, [Mill.] Swingle), native to East Asia, which is known for its high invasiveness in temperate regions worldwide, particularly favoring urban colonization. This study explores the botanical and aerobiological dimensions of this species using the central European metropolitan region of Berlin, Germany, as a case study, and provides a comprehensive global overview of allergological insights. The number of Ailanthus trees decreased markedly from the center to the periphery of Berlin City, following a temperature gradient. The same spatial trend was mirrored by airborne Ailanthus pollen concentrations measured with volumetric spore traps (Hirst-type) at five sites using seven traps. Ailanthus pollen was most abundant around midday and in the afternoon, with concentrations tenfold higher at street level than at roof level. The Ailanthus flowering period in June and July coincided well with the pollen season. To the best of our knowledge this is the first study to investigate Ailanthus altissima pollen production. On average, 5539 pollen grains were found per anther. A literature review on the allergy relevance of Ailanthus altissima pollen indicates the high allergenic potential of pollen from this species. Considering the anticipated expansion of suitable habitats for Ailanthus owing to global warming and the allergological significance of its pollen, it is recommended to include Ailanthus pollen in routine pollen monitoring, particularly in areas colonized by this species. Bisphenol A (BPA) is one of key raw materials used in the production of epoxy resins and plastics, which has toxicological effects on humans by disrupting cell functions through a variety of cell signaling pathways. Therefore, it is of great significance to develop a simple, rapid, and accurate BPA detection method in real water samples. In this study, a ratiometric fluorescence method based on yellow-emitting surface-functionalized polymer dots (PFBT@L Pdots) and blue-emitting carbon dots (Cdots) was described for the detection of BPA. Pdots as the detecting part were synthesized by using highly fluorescent hydrophobic Poly[(9,9-dioctylfluorenyl-2,7-diyl)-alt-co-(1,4-benzo-(2,1',3)-thiadiazole)] (PFBT) polymer and (R)-5,11,17,23-Tetra-tert-butyl-25,27-bis[(diphenylphosphinoyl)methoxy]-26-(3-oxabutyloxy)-28-[(1-phenylethyl)- carbamoylmethoxy]calix [4]arene (L) functionalizing ligand, and Cdots as internal reference were prepared by hydrothermal treatment of citric acid and urea. In the presence of BPA, chemical binding of the phosphorus atoms of nearby PFBT@L Pdots with BPA hydroxyl functional groups led to the aggregation of the PFBT@L Pdots aggregation and quenching their yellow emission, but the blue emission of Cdots, on the other hand, remained stable. The proposed PFBT@L Pdots probe was successfully applied for the detection of BPA in real water samples, and the results were in good agreement with those obtained by HPLC-FLD. Stressful situations may result in serum chromium (Cr) depletion with increased urinary excretion of the mineral and increased Cr requirements. The objective of this study was to investigate the effects of Cr supplementation on growth performance, feeding behavior, blood metabolites and hormones, indicators of oxidative stress and glucose-insulin kinetics of summer-exposed weaned dairy calves. In total, 48 Holstein female calves (63 days of age; 77.0±1.45 kg of BW) were assigned randomly to one of two treatments: (1) a control group with no supplemental Cr (Cr-), and (2) a supplemental Cr group (Cr+) to supply 0.05 mg Cr as Cr-methionine/kg of BW0.75. Chromium was provided in the starter feed and adjusted weekly based on BW over the experimental period. All calves were on experiment for 4 weeks after weaning. The average maximum temperature-humidity index was 76.1 units during the study period, indicating a mild degree of environmental heat load. Results indicated that in summer-exposed dairy calves, increased dietary Cr provision tended to decrease fecal score, tended to change rumination pattern, increased antioxidant capacity by increasing serum concentration of catalase, but had no effects on growth performance, metabolic status or peripheral glucose and insulin metabolism.

Background: The identification of human remains is an essential part of forensic science. Studying paranasal sinuses is very useful in identification of mutilated or burnt bodies from accidents such as plane crashes. We aimed to assess the diagnostic value of anthropometric indices of maxillary sinuses for sex determination using CT-scan images in Iranian adults., Methods: CT-scan slices of 228 maxillary sinuses (from 144 men and 144 women) were studied. The maximum height, maximum anterior-posterior diameter, maximum width, and maximum distance between the sinuses were measured in both sexes. Our participants were divided into three age groups of 20-34, 35-49, and over 50 to compare the recorded measurements in different age groups., Results: We found a significant difference between men and women regarding the maximum height, maximum width, maximum A-P diameter of sinuses and the maximum distance between the right and left maxillary sinuses in all of our participants. We also found that the highest accuracy for sex determination was related to the maximum distance between the sinuses (65.6%) and the lowest to the maximum width of the right sinus (56.2%). The most accurate identification resulted from assessing the maximum distance between the sinuses in the 20-34 age group (74.3%), the maximum anterior-posterior diameter of the sinuses in the 35-49 age group (62.8%) and the maximum height of the left sinus in the over 50 age group (65.7%)., Conclusion: Regardless of age, according to our study the parameters of height, width, anterior-posterior diameter of sinuses and the maximum distance between the right and left maxillary sinuses are partially valuable for sex determination. The most accurate sex identification was examined in the 20-34 age group from studying the maximum distance between the sinuses. Ailanthus altissima pollen (AAP) is considered as an emerging cause of respiratory allergy in United States, Italy and Iran. However, the allergenic composition of AAP is still unknown and has yet to be characterized. The present study aimed to identify AAP allergens using a proteomics-based approach. For this purpose, optimized AAP protein extracts were analyzed using 1D- and 2D- gel electrophoresis and confronted to twenty sera from individuals with respiratory allergy during the AAP season. Candidate allergens were detected using the serum from an allergic patient with clinical history of AAP pollinosis. IgE-binding spots were identified using MALDI-TOF/TOF mass spectrometry and database searching. According to our results, AAP extracts were rich in proteins (up to 16.25mg/ml) with a molecular-weight distribution ranging from 10 to 175kDa. Two-D electrophoresis of AAP extracts revealed 125 protein spots from which 13 were IgE reactive. These IgE-binding proteins were identified as enolase, calreticulin, probable pectate lyase 6, conserved hypothetical protein and ras-related protein RHN1-like. By our knowledge, this study is the first report identifying AAP allergens. These findings will open up further avenues for the diagnosis and immunotherapy of the AAP allergy as well as for the cloning and molecular characterization of relevant allergens., Biological Significance: Ailanthus altissima colonizes new areas every year in Iran and is spreading aggressively worldwide. According to USDA, the tree of heaven is now present as an invasive plant in 30 states in US (www.invasivespeciesinfo.gov/plants/treeheaven.shtml) and come to dominate large areas in many regions. Up to now, several cases of allergy to A. altissima pollen have been reported in United States, Italy and Iran [1-4]. However, there is still no information on the sensitizing allergens and the molecular origin of these clinical symptoms, which constitutes a serious threat to patients suffering from respiratory allergies in these regions. To our knowledge, the current study describes, therefore, the first panel of proteins responsible for IgE-mediated A. altissima pollinosis by using a gel-based proteomic approach. In metabolomics, the workflow for quantitative and comprehensive metabolic mapping of cellular metabolites can be a very challenging undertaking. Sampling and sample preparation play a significant role in untargeted analysis, as they may affect the composition of the analyzed metabolome. In the current work, different solid phase microextraction (SPME) coating chemistries were developed and applied to provide simultaneous extraction of a wide range of both hydrophobic and hydrophilic cellular metabolites produced by a model organism, Escherichia coli. Three different LC-MS methods were also evaluated for analysis of extracted metabolites. Finally, over 200 cellular metabolites were separated and detected with widely varying hydrophobicities ranging within -7 < log P < 15, including amino acids, peptides, nucleotides, carbohydrates, polycarboxylic acids, vitamins, phosphorylated compounds, and lipids such as hydrophobic phospholipids, prenol lipids, and fatty acids at the stationary phase of the E. coli life cycle using the developed 96-blade SPME-LC-MS method., (Copyright © 2015 Elsevier B.V. 1-Vinyl-3-octadecylimidazolium bromide ionic liquid [C18VIm]Br was prepared and used for the modification of mercaptopropyl-functionalized silica (Si-MPS) through surface radical chain-transfer addition. The synthesized octadecylimidazolium-modified silica (SiImC18) was characterized by thermogravimetric analysis (TGA), infrared spectroscopy (IR), (13)C NMR and (29)Si NMR spectroscopy and used as an extraction phase for the automated 96-blade solid phase microextraction (SPME) system with thin-film geometry using polyacrylonitrile (PAN) glue. The new proposed extraction phase was applied for extraction of aminoacids from grape pulp, and LC-MS-MS method was developed for separation of model compounds. Extraction efficiency, reusability, linearity, limit of detection, limit of quantitation and matrix effect were evaluated. The whole process of sample preparation for the proposed method requires 270min for 96 samples simultaneously (60min preconditioning, 90min extraction, 60min desorption and 60min for carryover step) using 96-blade SPME system. Inter-blade and intra-blade reproducibility were in the respective ranges of 5-13 and 3-10% relative standard deviation (RSD) for all model compounds. Limits of detection and quantitation of the proposed SPME-LC-MS/MS system for analysis of analytes were found to range from 0.1 to 1.0 and 0.5 to 3.0μgL(-1), respectively. An automated 96-thin-film solid phase microextraction system was optimized for high throughput analysis of phenolic compounds in wine, berry, and grape samples. Liquid chromatography and tandem mass spectrometry methods were optimized and applied for separation and detection of compounds. Evaluation of five different stationary phases showed that polystyrene-divinylbenzene-polyacrylonitrile (PS-DVB-PAN) is the optimum extraction phase for the extraction of phenolic compounds under study. The thin-film PS-DVB-PAN SPME coating provided almost exhaustive extraction recovery for all phenolics compounds under study, except for naringenin with 80% recovery. Extraction efficiency, inter- and intra-day reproducibility, sensitivity, linearity, limit of detection and quantitation, and matrix effect were evaluated. Intra-day and inter-day reproducibility were in the respective range of 4-8 and 7-13% relative standard deviation (RSD) for all eight phenolic compounds. Limits of detection and quantitation of the proposed SPME-LC-MS/MS system for the analysis of analytes under study were found in the range of 0.2-3 and 0.5-10 ng/mL, respectively. Standard addition calibration was applied for the quantitative determination of unknown phenolic compounds from wine, berry, and grape samples.
