Aims: Cell adhesion molecule 1 (CADM1) mediates interepithelial adhesion and is upregulated in crowded epithelial monolayers. This study aimed to examine CADM1 expression in the human endometrium of proliferative and secretory phases, and its transcriptional regulation in terms of estrogen stimuli and higher cellularity., Main Methods: CADM1 immunohistochemistry was conducted on endometrial tissues from women in their 40s and adult mice subcutaneously injected with estradiol following ovariectomy. Dual-luciferase reporter assays were conducted using human endometrial HEC-50B and HEC-1B cells and reporter plasmids harboring the human CADM1 3.4-kb promoter and its deleted and mutated forms. Cells were transfected with estrogen receptor α cDNA and reporter plasmids, and treated with estradiol before luciferase activity measurement., Key Findings: Immunohistochemistry revealed that CADM1 was clearly expressed on the lateral membranes of the simple columnar glandular cells in the proliferative phase, but not in the secretory phase, from both women and the mouse model. The glandular cell density increased two-fold in the proliferative phase. Reporter assays identified three Sp1-binding sites as estradiol-responsive elements in the proximal region (from -223 to -84) of the transcription start site (+1) in HEC-50B cells. When the cell culture was started at eight-fold higher cell density, the CADM1 3.4-kb promoter was transactivated at a two-fold higher level in HEC-50B cells. This cell density effect was not detected for the CADM1 2.3-kb or 1.6-kb promoter., Significance: Two (proximal and distal) promoter regions are suggested to function additively to transactivate CADM1 in endometrial glandular cells that crowd in the proliferative phase., (Copyright © 2021 Elsevier Inc. Aims: Stroke-prone spontaneously hypertensive rats (SHRSP) show significantly lower body weight than normotensive Wistar-Kyoto rats (WKY). Our hypotheses are as follows: weight loss of the skeletal muscle is related to hypertension-related diseases, and muscle hypotrophy is useful as a therapeutic target for hypertension and hypertension-related diseases. In this study, we aimed to investigate the pathophysiological characteristics of muscle hypotrophy in SHRSP to determine the therapeutic target molecule(s)., Main Methods: The difference in skeletal muscles in the lower leg between WKY and SHRSP was evaluated mainly through weight/tibial length, histological, gene expression, and protein expression analyses., Key Findings: SHRSP had a significantly lower weight/tibial length in soleus and gastrocnemius, but not in plantaris and tibialis anterior, indicating that muscles consisting of a relatively high amount of slow muscle fiber were affected. This result was confirmed by the histological analysis of soleus, showing that type I fiber mainly decreased the fiber size. Microarray and protein expression analyses showed that the muscle-specific ubiquitin ligase, muscle RING finger 1 (MuRF1), but not atrogin-1, was highly expressed in soleus, but not in plantaris, in SHRSP. TNF-like weak inducer of apoptosis receptor (TWEAKR) was predicted as a MuRF1 up-regulator by Ingenuity Pathway Analysis and immunostained only in type II fiber in WKY but in both type I and II fibers in SHRSP., Significance: TWEAKR is a type II-specific receptor in the skeletal muscle. Motor imagery is the mental execution of an action without any actual movement. Although numerous studies have utilized questionnaires to evaluate the vividness of motor imagery, it remains unclear whether it is related to the accuracy of motor imagery. To examine the relationship between vividness and accuracy, we investigated brain activity during kinesthetic and visual motor imagery, by using a novel sequential finger-tapping task. We estimated accuracy by measuring the fidelity of the actual performance and evaluated vividness by using a visual analog scale. We found that accuracy of visual motor imagery was correlated with the activity in the left visual cortex, as well as with bilateral sensorimotor regions. In contrast, vividness of visual motor imagery was associated with the activity in the right orbitofrontal cortex. However, there was no correlation in the brain activity between the right orbitofrontal cortex and visuomotor regions or between vividness and accuracy of motor imagery. In addition, we did not find any correlation in the kinesthetic imagery condition. We conclude that vividness of visual motor imagery is associated with the right orbitofrontal cortex and is independent of processes occurring in sensorimotor regions, which would be responsible for the accuracy of visual motor imagery., (Copyright © 2018 Elsevier B.V. and Japan Neuroscience Society. Functional plasticity of the sensorimotor cortex occurs following motor practice, as well as after limb amputation. However, the joint effect of limb amputation and intensive, long-term motor practice on cortical plasticity remains unclear. Here, we recorded brain activity during unilateral contraction of the hip, knee, and ankle joint muscles from a long jump Paralympic gold medalist with a unilateral below-knee amputation (Amputee Long Jumper, ALJ). He used the amputated leg with a prosthesis for take-off. Under similar conditions to the ALJ, we also recorded brain activity from healthy long jumpers (HLJ) and non-athletes with a below-knee amputation. During a rhythmic isometric contraction of knee extensor muscles with the take-off/prosthetic leg, the ALJ activated not only the contralateral primary sensorimotor cortex (M1/S1), but also the ipsilateral M1/S1. In addition, this ipsilateral M1/S1 activation was significantly greater than that seen in the HLJ. However, we did not find any significant differences between the ALJ and HLJ in M1/S1 activation during knee muscle contraction in the non-take-off/intact leg, nor during hip muscle contraction on either side. Region of interest analysis revealed that the ALJ exhibited a greater difference in M1/S1 activity and activated areas ipsilateral to the movement side between the take-off/prosthetic and non-take-off/intact legs during knee muscle contraction compared with the other two groups. However, difference in activity in M1/S1 contralateral to the movement side did not differ across groups. These results suggest that a combination of below-knee amputation and intensive, prolonged long jump training using a prosthesis (i.e. fine knee joint control) induced an expansion of the functional representation of the take-off/prosthetic leg in the ipsilateral M1/S1 in a muscle-specific manner. Many studies have demonstrated that training utilizing action observation and/or motor imagery improves motor performance. These two techniques are widely used in sports and in the rehabilitation of movement-related disorders. Motor imagery has also been used for brain-machine/computer interfaces (BMI/BCI). During both action observation and motor imagery, motor-related regions such as the premotor cortex and inferior parietal lobule are activated. This is common to actual execution and are involved with the underlying mechanisms of motor learning without execution. Since it is easier to record brain activity during action observation and motor imagery than that during actual sport movements, action observation, and motor imagery of sports skills or complex whole body movements have been utilized to investigate how neural mechanisms differ across the performance spectrum ranging from beginner to expert. However, brain activity during action observation and motor imagery is influenced by task complexity (i.e., simple vs complex movements). Furthermore, temporal changes in brain activity during actual execution along the long time course of motor learning are likely nonlinear and would be different from that during action observation or motor imagery. We utilized functional magnetic resonance imaging to investigate the brain regions activated during motor imagery of an action with an object both with and without passively holding the object. Participants performed the following tasks: (1) 'Imagery with Ball' condition: subjects imagined squeezing a foam ball (7cm diameter) while holding the ball, (2) 'Imagery' condition: subjects imagined squeezing a ball without holding the ball, and (3) 'Ball' condition: subjects held the ball without motor imagery. Regions activated by the 'Imagery with Ball' condition were located in the left dorsolateral prefrontal cortex (DLPFC), supplemental motor areas (SMA), inferior parietal lobule (IPL), superior parietal lobule (SPL), insula, cerebellum and basal ganglia. A direct comparison revealed that the right DLPFC and the right IPL showed a higher level of activation during the 'Imagery with Ball' than during the 'Imagery'+'Ball' conditions. The present study investigated whether combining observation and imagery of an action increased corticospinal excitability over the effects of either manipulation performed alone. Corticospinal excitability was assessed by motor-evoked potentials in the biceps brachii muscle following transcranial magnetic stimulation over the motor cortex during observation, imagery or both. The action utilized was repetitive elbow flexion/extension. Simultaneous observation and imagery of the elbow action facilitated corticospinal excitability as compared to that recorded during observation or imagery alone. However, facilitation due to the combination of observation and imagery was not obtained when the participants imagined the action pattern while they observed the same action presented out of phase. These findings suggest that a combination of observation and imagery can enhance corticospinal excitability. This enhancement depends on phase consistency between the observed and imagined actions.
