1. Effects of resistance exercise and whey protein supplementation on cognitive function in older men: secondary analysis of a randomised, double-blind, placebo-controlled trial

Abstract: Purpose: Ageing is associated with cognitive decline. This study investigated the individual and combined effects of resistance exercise (RE) and whey protein supplementation (PRO) on cognitive function in older men. Methods: In a pooled-groups analysis, 36 older men (age: 67 ± 4 years) were randomised to either RE (2 x/week; n = 18) or no exercise (NE; n = 18), and either PRO (2 × 25 g/d whey protein isolate; n = 18) or control (CON, 2 × 23.75 g maltodextrin/d; n = 18). A sub-analysis was also conducted between RE + CON (n = 9) and RE + PRO (n = 9). At baseline and 12 weeks, participants completed a battery of neuropsychological tests (CANTAB; Cambridge Cognition, UK) and neurobiological, inflammatory, salivary cortisol and insulin sensitivity biomarkers were quantified. Results: PRO improved executive function z-score (+0.31 ± 0.08) greater than CON (+0.06 ± 0.08, P = 0.03) and there was a trend towards improved global cognitive function (P = 0.053). RE and RE + PRO did not improve any cognitive function domains (p ≥ 0.07). RE decreased tumor necrosis factor-alpha (P = 0.02) and interleukin-6 (P = 0.048) concentrations compared to NE, but changes in biomarkers did not correlate with changes in cognitive domains. 2. A MYC-controlled redox switch protects B lymphoma cells from EGR1-dependent apoptosis

Abstract: Summary: Refractory and relapsed B cell lymphomas are often driven by the difficult-to-target oncogene MYC. Here, we report that high MYC expression stimulates proliferation and protects B lymphoma cells from apoptosis under normal oxidative stress levels and that compounds including N-acetylcysteine (NAC) and vitamin C (VitC) induce apoptosis by reducing oxidative stress. NAC and VitC injections effectively reduce tumor growth in lymphoma cells with high MYC expression but not in those with low MYC expression. MYC knockdown confers tumor resistance to NAC and VitC, while MYC activation renders B cells sensitive to these compounds. Mechanistically, NAC and VitC stimulate MYC binding to EGR1 through Cys117 of MYC, shifting its transcriptional output from cell cycle to apoptosis gene expression. These results identify a redox-controlled mechanism for MYC’s role in maintaining proliferation and preventing apoptosis, offering a potential therapeutic rationale for evaluating NAC or VitC in patients with MYC-driven B cell lymphoma.

3. ROS-lowering doses of vitamins C and A accelerate malignant melanoma metastasis

Abstract: Oxidative stress is a barrier of migration and metastasis for malignant melanoma cells. Consequently, reducing oxidative stress with the antioxidant N-acetylcysteine (NAC) stimulates melanoma cell migration in vitro and metastasis in vivo. However, it is not yet known whether the NAC effect is shared with other antioxidants. Here, we screened 104 redox-active compounds and identify 27 that increase migration of human malignant melanoma cells in two doses. Validation experiments in four cell lines and four drug doses resulted in a list of 18 compounds which were ranked based on their ability to increase migration and reduce ROS levels; vitamin C (VitC) ranked as number one, followed by the vitamin E analogue Trolox and several carotenoids and Vitamin A–related compounds. Four diet-relevant compounds from this list—VitC, β-carotene, retinyl palmitate, and canthaxanthin—were selected and found to accelerate metastasis in mice with BRAFV600E-driven malignant melanoma. Genomics analyses revealed that the transcription factor BACH1 is activated following antioxidant administration and knockout of Bach1 in mouse melanoma cells reduced lymph node and liver metastasis in xenograft mouse models. We conclude that a broad range of antioxidants accelerate melanoma migration and metastasis and that BACH1 is functionally linked to melanoma metastasis in vivo.

6. Using CO2-Plume geothermal (CPG) energy technologies to support wind and solar power in renewable-heavy electricity systems

Abstract: CO2-Plume Geothermal (CPG) technologies are geothermal power systems that use geologically stored CO2 as the subsurface heat extraction fluid to generate renewable energy. CPG technologies can support variable wind and solar energy technologies by providing dispatchable power, while Flexible CPG (CPG-F) facilities can provide dispatchable power, energy storage, or both simultaneously. We present the first study investigating how CPG power plants and CPG-F facilities may operate as part of a renewable-heavy electricity system by integrating plant-level power plant models with systems-level optimization models. We use North Dakota, USA as a case study to demonstrate the potential of CPG to expand the geothermal resource base to locations not typically considered for geothermal power. We find that optimal system capacity for a solar-wind-CPG model can be up to 20 times greater than peak-demand. CPG-F facilities can reduce this modeled system capacity to just over 2 times peak demand by providing energy storage over both seasonal and short-term timescales. The operational flexibility of CPG-F facilities is further leveraged to bypass the ambient air temperature constraint of CPG power plants by storing energy at critical temperatures. Across all scenarios, a tax on CO2 emissions, on the order of hundreds of dollars per tonne, is required to financially justify using renewable energy over natural-gas power plants. Our findings suggest that CPG and CPG-F technologies may play a valuable role in future renewable-heavy electricity systems, and we propose a few recommendations to further study its integration potential.

7. Early Response to Chemotherapy in Malignant Pleural Mesothelioma Evaluated Using Diffusion-Weighted Magnetic Resonance Imaging: Initial Observations

Abstract: Introduction: We compared the magnetic resonance imaging total tumor volume (TTV) and median apparent diffusion coefficient (ADC) of malignant pleural mesothelioma (MPM) before and at 4 weeks after chemotherapy, to evaluate whether these are potential early markers of treatment response. Methods: Diffusion-weighted magnetic resonance imaging was performed in 23 patients with MPM before and after 4 weeks of chemotherapy. The TTV was measured by semiautomatic segmentation (GrowCut) and transferred onto ADC maps to record the median ADC. Test-retest repeatability of TTV and ADC was evaluated in eight patients. TTV and median ADC changes were compared between responders and nonresponders, defined using modified Response Evaluation Criteria In Solid Tumors on computed tomography (CT) at 12 weeks after treatment. TTV and median ADC were also correlated with CT size measurement and disease survival. Results: The test-retest 95% limits of agreement for TTV were −13.9% to 16.2% and for median ADC −1.2% to 3.3%. A significant increase in median ADC in responders was observed at 4 weeks after treatment (p = 0.02). Correlation was found between CT tumor size change at 12 weeks and median ADC changes at 4 weeks post-treatment (r = −0.560, p = 0.006). An increase in median ADC greater than 5.1% at 4 weeks has 100% sensitivity and 90% specificity for responders (area under the curve = 0.933, p < 0.001). There was also moderate correlation between median tumor ADC at baseline and overall survival (r = 0.45, p = 0.03). Conclusions: Diffusion-weighted magnetic resonance imaging measurements of TTV and median ADC in MPM have good measurement repeatability. Increase in ADC at 4 weeks post-treatment has the potential to be an early response biomarker.

Abstract: Summary: Background: The clonal diversity underpinning trends in multidrug resistant Escherichia coli causing bloodstream infections remains uncertain. We aimed to determine the contribution of individual clones to resistance over time, using large-scale genomics-based molecular epidemiology. Methods: This was a longitudinal, E coli population, genomic, cohort study that sampled isolates from 22 512 E coli bloodstream infections included in the Norwegian surveillance programme on resistant microbes (NORM) from 2002 to 2017. 15 of 22 laboratories were able to share their isolates, and the first 22·5% of isolates from each year were requested. We used whole genome sequencing to infer the population structure (PopPUNK), and we investigated the clade composition of the dominant multidrug resistant clonal complex (CC)131 using genetic markers previously reported for sequence type (ST)131, effective population size (BEAST), and presence of determinants of antimicrobial resistance (ARIBA, PointFinder, and ResFinder databases) over time. We compared these features between the 2002–10 and 2011–17 time periods. We also compared our results with those of a longitudinal study from the UK done between 2001 and 2011. Findings: Of the 3500 isolates requested from the participating laboratories, 3397 (97·1%) were received, of which 3254 (95·8%) were successfully sequenced and included in the analysis. Abstract: CO2-Plume Geothermal (CPG) power plants can produce heat and/or electric power. One of the most important parameters for the design of a CPG system is the CO2 mass flowrate. Firstly, the flowrate determines the power generated. Secondly, the flowrate has a significant effect on the fluid pressure drawdown in the geologic reservoir at the production well inlet. This pressure drawdown is important because it can lead to water flow in the reservoir towards and into the borehole. Thirdly, the CO2 flowrate directly affects the two-phase (CO2 and water) flow regime within the production well. An annular flow regime, dominated by the flow of the CO2 phase in the well, is favorable to increase CPG efficiency. Thus, flowrate optimizations of CPG systems need to honor all of the above processes. We investigate the effects of various operational parameters (maximum flowrate, admissible reservoir-pressure drawdown, borehole diameter) and reservoir parameters (permeability anisotropy and relative permeability curves) on the CO2 and water flow regime in the production well and on the power generation of a CPG system. We use a numerical modeling approach that couples the reservoir processes with the well and power plant systems. Our results show that water accumulation in the CPG vertical production well can occur. However, with proper CPG system design, it is possible to prevent such water accumulation in the production well and to maximize CPG electric power output. ISSN:0375-6505

Abstract: Electropulse stimulation provides an energy-efficient means of excavating hard rocks through repeated application of high voltage pulses to the rock surface. As such, it has the potential to confer significant advantages to mining and drilling operations for mineral and energy resources. Nevertheless, before these benefits can be realized, a better understanding of these processes is required to improve their deployment in the field. In this paper, we employ a recently developed model of the grain-scale processes involved in electropulse stimulation to examine excavation of hard rock under realistic operating conditions. To that end, we investigate the maximum applied voltage within ranges of 120–600 kV, to observe the onset of rock fragmentation. We further study the effect of grain size on rock breakage, by comparing fine (granodiorite) and coarse grained (granite) rocks. Lastly, the pore fluid salinity is investigated, since the electric conductivity of the pore fluid is shown to be a governing factor for the electrical conductivity of the modeled system. Abstract: Protein farnesyltransferase (FTase) and protein geranylgeranyltransferase-I (GGTase-I) add 15- or 20-carbon lipids, respectively, to proteins that terminate with a CaaX motif. These posttranslational modifications of proteins with lipids promote protein interactions with membrane surfaces in cells, but the in vivo importance of the CaaX prenyltransferases and the protein lipidation reactions they catalyze remain incompletely defined. One study concluded that a deficiency of FTase was inconsequential in adult mice and led to little or no tissue pathology. To assess the physiologic importance of the CaaX prenyltransferases, we used conditional knockout alleles and an albumin–Cre transgene to produce mice lacking FTase, GGTase-I, or both enzymes in hepatocytes. The hepatocyte-specific FTase knockout mice survived but exhibited hepatocellular disease and elevated transaminases. Mice lacking GGTase-I not only had elevated transaminases but also had dilated bile cannaliculi, hyperbilirubinemia, hepatosplenomegaly, and reduced survival. Of note, GGTase-I–deficient hepatocytes had a rounded shape and markedly reduced numbers of actin stress fibers. Hepatocyte-specific FTase/GGTase-I double-knockout mice closely resembled mice lacking GGTase-I alone, but the disease was slightly more severe. Our studies refute the notion that FTase is dispensable and demonstrate that GGTase-I is crucial for the vitality of hepatocytes.

Abstract: Introduction: We compared the magnetic resonance imaging total tumor volume (TTV) and median apparent diffusion coefficient (ADC) of malignant pleural mesothelioma (MPM) before and at 4 weeks after chemotherapy, to evaluate whether these are potential early markers of treatment response. Methods: Diffusion-weighted magnetic resonance imaging was performed in 23 patients with MPM before and after 4 weeks of chemotherapy. The TTV was measured by semiautomatic segmentation (GrowCut) and transferred onto ADC maps to record the median ADC. Test-retest repeatability of TTV and ADC was evaluated in eight patients. TTV and median ADC changes were compared between responders and nonresponders, defined using modified Response Evaluation Criteria In Solid Tumors on computed tomography (CT) at 12 weeks after treatment. TTV and median ADC were also correlated with CT size measurement and disease survival. Results: The test-retest 95% limits of agreement for TTV were −13.9% to 16.2% and for median ADC −1.2% to 3.3%. A significant increase in median ADC in responders was observed at 4 weeks after treatment (p = 0.02). Correlation was found between CT tumor size change at 12 weeks and median ADC changes at 4 weeks post-treatment (r = −0.560, p = 0.006). An increase in median ADC greater than 5.1% at 4 weeks has 100% sensitivity and 90% specificity for responders (area under the curve = 0.933, p < 0.001). There was also moderate correlation between median tumor ADC at baseline and overall survival (r = 0.45, p = 0.03). Conclusions: Diffusion-weighted magnetic resonance imaging measurements of TTV and median ADC in MPM have good measurement repeatability. Increase in ADC at 4 weeks post-treatment has the potential to be an early response biomarker.

21. Flow-through Drying during CO Injection into Brine-filled Natural Fractures: A Tale of Effective Normal Stress

Abstract: Injecting supercritical CO2 (scCO2) into brine-filled fracture-dominated reservoirs causes brine displacement and possibly evaporite precipitations that alter the fracture space. Here, we report on isothermal laboratory experiments on scCO2-induced flow-through drying in a naturally fractured granodiorite specimen under effective normal stresses of 5-10 MPa, where two drying regimes are identified. A novel approach is developed to delineate the evolution of brine saturation and relative permeability from fluid production and differential pressure measurements. Under higher compressive stresses, the derived relative permeability curves indicate lower mobility of brine and higher mobility of the scCO2 phase. The derived fractional flow curves also suggest an increase in channelling and a decrease in brine sweep efficiencies under higher compressive stresses. Finally, lowering compressive stresses seems to promote less water evaporation. Our experimental results assist in understanding the injectivity of single fractures and eventually of fracture networks during subsurface applications that involve scCO2 injection into saline formations. 22. Quantification of mineral accessible surface area and flow-dependent fluid-mineral reactivity at the pore scale

Abstract: Accessible surface areas (ASAs) of individual rock-forming minerals exert a fundamental control on the maximum mineral reactivity with formation fluids. Notably, ASA efficiency during fluid-rock reactions can vary by orders of magnitude, depending on the inflow fluid chemistry and the velocity field. Due to the lack of adequate quantification methods, determining the mineral-specific ASAs and their reaction efficiency still remain extremely difficult. Here, we first present a novel joint method that appropriately calculates ASAs of individual minerals in a multi-mineral sandstone. This joint method combines SEM-image processing results and Brunauer-Emmett-Teller (BET) surface area measurements by a Monte-Carlo algorithm to derive scaling factors and ASAs for individual minerals at the resolution of BET measurements. Using these atomic-scale ASAs, we then investigate the impact of flow rate on the ASA efficiency in mineral dissolution reactions during the injection of CO2-enriched brine. This is done by conducting a series of pore-scale reactive transport simulations, using a two-dimensional (2D) scanning electron microscopy (SEM) image of this sandstone. The ASA efficiency is determined employing a domain-averaged dissolution rate and the effective surface area of the most reactive phase in the sandstone (dolomite). As expected, the dolomite reactivity is found to increase with the flow rate, due to the on average high fluid reactivity. The surface efficiency increases slightly with the fluid flow rate, and reaches a relatively stable value of about 1%. The domain averaged method is then compared with the in-out averaged method (i.e the “Black-box” approach), which is often used to analyzed the experimental observations. The in-out averaged method yields a considerable overestimation of the fluid reactivity, a small underestimation of the dolomite reactivity, and a considerable underestimation of the ASA efficiency. The discrepancy between the two methods is becoming smaller when the injection rate increases. Our comparison suggests that the result interpretation of the in-out averaged method should be contemplated, in particular, when the flow rate is small. Nonetheless, our proposed ASA determination method should facilitate accurate calculations of fluid-mineral reactivity in large-scale reactive transport simulations, and we advise that an upscaling of the ASA efficiency needs to be carefully considered, due to the low surface efficiency. We then discuss the rock mechanics applications and limitations at two extreme scales, the Earth's crust (tens of kilometers) and intact rocks (meters). We finally offer thoughts on bridging these scales. The context of the article is essential not only to the rock mechanics/geomechanics community but also to a broader powder technology community. Abstract: Globally, hepatitis E virus (HEV) is a major cause of acute viral hepatitis. Epidemiology and clinical presentation of hepatitis E vary greatly by location and are affected by the HEV genotype. Nucleic acid amplification technique (NAT)-based assays are important for the detection of acute HEV infection as well for monitoring chronic cases of hepatitis E. The aim of the study was to evaluate a panel of samples containing different genotypes of HEV for use in nucleic NAT-based assays. The panel of samples comprises eleven different members including HEV genotype 1a (2 strains), 1e, 2a, 3b, 3c, 3e, 3f, 4c, 4g as well as a human isolate related to rabbit HEV. Each laboratory assayed the panel members directly against the 1 World Health Organization (WHO) International Standard (IS) for HEV RNA (6329/10) which is based upon a genotype 3 a strain. The samples for evaluation were distributed to 24 laboratories from 14 different countries and assayed on three separate days. Of these, 23 participating laboratories returned a total of 32 sets of data; 17 from quantitative assays and 15 from qualitative assays. The assays used consisted of a mixture of in-house developed and commercially available assays. The results showed that all samples were detected consistently by the majority of participants, although in some cases, some samples were detected less efficiently. Based on the results of the collaborative study the panel (code number 8578/13) was established as the "1st International Reference Panel (IRP) for all HEV genotypes for NAT-based assays" by the WHO Expert Committee on Biological Standardization. This IRP will be important for assay validation and ensuring adequate detection of different genotypes and clinically important sub-genotypes of HEV.

Abstract: Endogenous pediatric Cushing's syndrome (CS) is very rare. Cushing's disease (CD) secondary to a corticotroph adenoma represents the commonest cause of CS in the pediatric age range. It is important to recognize the features of CD early to allow timely diagnosis and management and to minimize the development of comorbidities. Once diagnosed, the primary aim should be the normalization of serum cortisol to minimize the adverse effects of prolonged hypercortisolaemia. Once remission is achieved, posttreatment management should focus on the optimization of growth, pubertal development, bone mineral density and body composition.

Abstract: Accessing the intrinsic hydrogen content within ammonia, NH3, has the potential to play a very significant role in the future of a CO2-free sustainable energy supply. Inexpensive light metal imides and amides are effective at decomposing ammonia to hydrogen and nitrogen (2NH3 → 3H2 + N2), at modest temperatures, and thus represent a low-cost approach to on-demand hydrogen production. Building upon this discovery, this paper describes the integration of an ammonia cracking unit with a post-reactor gas purification system and a small-scale PEM fuel cell to create a first bench-top demonstrator for the production of hydrogen using light metal imides.

Abstract: The development of a safe technique for the supply of hydrogen to small portable fuel cells has emerged as a significant barrier to their deployment in recent years, with solutions centering on the use of hydrogen absorption materials, or the generation of hydrogen through chemical reaction. In the present work we demonstrate that the ball-milling of Si under inert conditions in the presence of KOH and sucrose results in the formation of a fine Si-based powder which reacts spontaneously with water at ambient starting temperature to evolve hydrogen rapidly at high yield. Embedded KOH is capable of accelerating the hydrolysis reaction of silicon by the self-heating effect attributed to dissolution heat of KOH, obviating the need for external heating to initiate the reaction; it also reduces the sensitivity of the reaction to oxide contamination of the Si surface by enabling its dissolution in the form of soluble silicates. Moreover, the silicon–water reaction can be switched on and off by adjusting the ambient temperature. It is shown that ball-milled, KOH-embedded Si powder is able to react with different water sources, such as tap water, river water, and salt water, to produce H2 under aerobic conditions. The method represents a cheap scalable approach for the safe provision of hydrogen fuel to small fuel cells.

Abstract: Background Conserved pneumococcal proteins are potential candidates for inclusion in vaccines against pneumococcal diseases. In the first part of a two-part study, an investigational vaccine (PHiD-CV/dPly/PhtD-30) containing 10 pneumococcal serotype-specific polysaccharide conjugates (10VT) combined with pneumolysin toxoid and pneumococcal histidine triad protein D (30 μg each) was well tolerated by Gambian children. Part two, presented here, assessed the efficacy of two PHiD-CV/dPly/PhtD formulations against pneumococcal nasopharyngeal carriage (NPC) prevalence in infants. Methods In this phase 2, randomized, controlled, observer-blind trial, healthy infants aged 8–10 weeks, recruited from a peri-urban health center, were randomized (1:1:1:1:1:1) into six groups. Four groups received PHiD-CV/dPly/PhtD (10 or 30 μg of each protein), PHiD-CV, or 13-valent pneumococcal conjugate vaccine at ages 2–3–4 months (3 + 0 infant schedule) and two groups PHiD-CV/dPly/PhtD-30 or PHiD-CV at 2–4–9 months (2 + 1 infant schedule). The primary objective was impact on non-10VT NPC at ages 5–9–12 months. Secondary objectives included confirmatory analysis of protein dose superiority and safety/reactogenicity. Impact on pneumococcal NPC acquisition, bacterial load, and ply and phtD gene sequencing were explored. Results 1200 infants were enrolled between June 2011 and May 2012. Prevalences of pneumococcal (60–67%) and non-10VT (55–61%) NPC were high at baseline. Across all post-vaccination time points, efficacy of PHiD-CV/dPly/PhtD-10 and PHiD-CV/dPly/PhtD-30 against non-10VT NPC (3 + 0 schedule) was 1.1% (95% CI −21.5, 19.5) and 2.1% (−20.3, 20.3), respectively; efficacy of PHiD-CV/dPly/PhtD-30 (2 + 1 schedule) was 0.5% (−22.1, 18.9) versus PHiD-CV. No differences were observed in pneumococcal NPC acquisition, clearance, or bacterial load. Both protein-based vaccines elicited immune responses to pneumococcal proteins. Conclusions In this high carriage prevalence setting, inclusion of pneumococcal proteins in the PHiD-CV/dPly/PhtD investigational vaccine had no impact on pneumococcal NPC in infants, regardless of protein dose or schedule. 40. Growth of 18O isotopically-enriched ZnO nanorods by two novel VPT methods

Abstract: We have developed two novel vapour phase transport methods to grow ZnO nanorod arrays isotopically enriched with 18O. Firstly, a three-step process used to grow natural and Zn-enriched ZnO nanorods has been further modified, by replacing the atmospheric O2 with enriched 18O2, in order to grow 18O-enriched ZnO nanorods using this vapour-solid method on chemical bath deposited buffer layers. In addition, 18O-enriched ZnO nanorods were successfully grown using 18O isotopically enriched ZnO source powders in a vapour-liquid-solid growth method. Scanning electron microscopy studies confirmed the success of both growth methods in terms of nanorod morphology, although in the case of the vapour-liquid-solid samples, the nanorods’ c-axes were not vertically aligned due to the use of a non-epitaxial substrate. Raman and PL studies indicated clearly that O-enrichment was successful in both cases, although the results indicate that the enrichment is at a lower level in our samples compared to previous reports with the same nominal enrichment levels. The results of our studies also allow us to comment on both levels of enrichment achieved and on novel effects of the high temperature growth environment on the nanorod growth, as well as suggesting possible mechanisms for such effects. Very narrow photoluminescence line widths, far narrower than those reported previously in the literature for isotopically enriched bulk ZnO, are seen in both the vapour-solid and vapour-liquid-solid nanorod samples demonstrating their excellent optical quality and their potential for use in detailed optical studies of defects and impurities using low temperature photoluminescence.

Abstract: The interaction of electromagnetic fields with the human body is quantified by the dielectric properties of biological tissues. These properties are incorporated into complex numerical simulations using parametric models such as Debye and Cole-Cole, for the computational investigation of electromagnetic wave propagation within the body. These parameters can be acquired through a variety of optimisation algorithms to achieve an accurate fit to measured data sets. A number of different optimisation techniques have been proposed, but these are often limited by the requirement for initial value estimations or by the large overall error (often up to several percentage points). In this work, a novel two-stage genetic algorithm proposed by the authors is applied to optimise the multi-pole Debye parameters for 54 types of human tissues. The performance of the two-stage genetic algorithm has been examined through a comparison with five other existing algorithms. The experimental results demonstrate that the two-stage genetic algorithm produces an accurate fit to a range of experimental data and efficiently out-performs all other optimisation algorithms under consideration. Accurate values of the three-pole Debye models for 54 types of human tissues, over 500 MHz to 20 GHz, are also presented for reference. Abstract: The new compounds (5-methyl-2-thiophene-2-pyridyl(R))imine [R = methyl (L1); R = ethyl (L2)] and (5-bromo-2-thiophene-2-pyridyl(R)imine [R = methyl (L3); R = ethyl (L4)] were successfully synthesized via Schiff base condensation reaction and obtained in good yields. These potential ligands were reacted with [PdCl2(COD)] and [PdClMe(COD)] to give the corresponding complexes [PdCl2(L)] (L = L1-L4; 1–4) and [PdClMe(L)] (L = L1-L4; 5–8). All compounds were characterized by IR, 1H and 13C NMR spectroscopy, elemental analysis and mass spectrometry. The molecular structures of 1, 2, 6 and 8 were confirmed by X-ray crystallography. The complexes were evaluated as catalyst precursors for standard Heck coupling reactions and showed significant catalytic activities that could be correlated with steric and electronic influences. peerReviewed

Abstract: Cancer cells undergoing starvation- and treatment-induced autophagy were found to exhibit reduced intracellular lactate, reduced rates of steady-state lactate excretion and reduced real-time pyruvate-lactate exchange rates, indicating that glycolytic metabolism was altered in autophagic cells. In this chapter, we describe the technical details of the use of 1H-magnetic resonance spectroscopy (MRS) to measure endogenous cellular concentrations of lactate and glucose in autophagic cells and tissues, how to measure the rate of steady-state lactate excretion and glucose uptake by 1H-MRS in autophagic cells, and details of the real-time measurement of [1-13C] pyruvate to lactate exchange in autophagic cells by 13C-MRS-DNP (dynamic nuclear polarization).

Abstract: Drug addiction is a brain disease that involves the disruption of important brain neurocircuitry. One important brain system implicated in drug addiction is the mesolimbic system, which includes dopaminergic projections from the ventral tegmental area to the nucleus accumbens (NAc). Within the NAc are neurons containing cocaine- and amphetamine-regulated transcript (CART) peptide, which interacts with the mesolimbic dopaminergic system. This interaction may indicate that CART peptide is involved in drug abuse and addiction. In support of this idea, several studies have shown that endogenous and exogenous changes in the concentrations of CART peptide in the NAc modulate dopamine- and cocaine-mediated behavioral effects. CART peptide also modulates some of the behavioral effects of other drugs of abuse such as amphetamine and ethanol. While the CART peptide receptor has not yet been cloned and characterized, understanding CART peptide pharmacology may enable us to better understand the mechanisms involved in drug addiction.

Abstract: Machine learning has been key to the success and advance of artificial intelligence in recent years. However, it usually requires significant quantities of labeled data with a supervised training regime, and obtaining exact labeling with expert knowledge can be very expensive and challenging, especially for medical images. This is especially so for datasets with abnormalities, as tissue types and the shapes of the organs in these datasets can be widely different. Deep learning is an approach to learn representative features of data from the given dataset in an unsupervised manner using deep neural networks. This can be promising in medical image analysis, because it can be applied to automatically analyze a diverse, large set of medical images, despite the difficulty of obtaining libraries of accurately labeled example datasets and the intrinsic abnormalities present in patient datasets. This chapter investigates the concept of deep learning for medical image analysis, demonstrating the application of multiple organ detection in multiparametric magnetic resonance images of tumor patients using stacked autoencoder models.

Abstract: Protein farnesyltransferase (FTase) and protein geranylgeranyltransferase-I (GGTase-I) add 15- or 20-carbon lipids, respectively, to proteins that terminate with a CaaX motif. These posttranslational modifications of proteins with lipids promote protein interactions with membrane surfaces in cells, but the in vivo importance of the CaaX prenyltransferases and the protein lipidation reactions they catalyze remain incompletely defined. One study concluded that a deficiency of FTase was inconsequential in adult mice and led to little or no tissue pathology. To assess the physiologic importance of the CaaX prenyltransferases, we used conditional knockout alleles and an albumin–Cre transgene to produce mice lacking FTase, GGTase-I, or both enzymes in hepatocytes. The hepatocyte-specific FTase knockout mice survived but exhibited hepatocellular disease and elevated transaminases. Mice lacking GGTase-I not only had elevated transaminases but also had dilated bile cannaliculi, hyperbilirubinemia, hepatosplenomegaly, and reduced survival. Of note, GGTase-I–deficient hepatocytes had a rounded shape and markedly reduced numbers of actin stress fibers. Hepatocyte-specific FTase/GGTase-I double-knockout mice closely resembled mice lacking GGTase-I alone, but the disease was slightly more severe. Our studies refute the notion that FTase is dispensable and demonstrate that GGTase-I is crucial for the vitality of hepatocytes.

Abstract: CO2-Plume Geothermal (CPG) technology involves injecting CO2 into natural, highly permeable geologic units to extract energy. The subsurface CO2 absorbs heat from the reservoir, buoyantly rises to the surface, and drives a power generation system. The CO2 is then cooled and reinjected underground. Here, we analyze the effects of multi-layered geologic reservoirs on CPG system performance by examining the CO2 mass fraction in the produced fluid, pore-fluid pressure buildup during operation, and heat energy extraction rates. The produced CO2 mass fraction depends on the stratigraphic positions of highly permeable layers which also affect the pore-fluid pressure drop across the reservoir. ISSN:1876-6102

Abstract: Carbon dioxide (CO2) sequestration in deep saline aquifers and exhausted oil and natural gas fields has been widely considered as a means for reducing CO2 emissions to the atmosphere as a counter-measure to global warming. However, rather than treating CO2 merely as a waste fluid in need of permanent disposal, we propose that it could also be used as a working fluid in geothermal energy capture, as its thermodynamic and fluid mechanical properties suggest it transfers geothermal heat more efficiently than water. Energy production and sales in conjunction with sequestration would improve the economic viability of CO2 sequestration, a critical challenge for large-scale implementation of the technology. In addition, using CO2 as the working fluid in geothermal power systems may permit utilization of lower temperature geologic formations than those that are currently deemed economically viable, leading to more widespread utilization of geothermal energy.
