Ericifolin, an eugenol 5-O-beta-(6'-O-galloylglucopyranoside) possessing the naturally unknown phenolic moiety, 5-hydroxyeugenol, together with the two new phenolics, 2-O-p-hydroxybenzoyl-6-O-galloyl-(alpha/beta)-4C1-glucopyranose and 3-methoxyellagic acid 4-O-rhamnopyranoside have been isolated from the antibacterial leaves extract of Melaleuca ericifolia. In addition, 19 known phenolics were also separated and characterized. All structures were elucidated on the basis of analysis of 1H, 13C NMR, HMQC, HMBC and FTMS spectral data.

The Type III restriction endonuclease EcoP15I forms a hetero-oligomeric enzyme complex that consists of two modification (Mod) subunits and two restriction (Res) subunits. Structural data on Type III restriction enzymes in general are lacking because of their remarkable size of more than 400 kDa and the laborious and low-yield protein purification procedures. We took advantage of the EcoP15I-overexpressing vector pQEP15 and affinity chromatography to generate a quantity of EcoP15I high enough for comprehensive proteolytic digestion studies and analyses of the proteolytic fragments by mass spectrometry. We show here that in the presence of specific DNA the entire Mod subunit is protected from trypsin digestion, whereas in the absence of DNA stable protein domains of the Mod subunit were not detected. In contrast, the Res subunit is comprised of two trypsin-resistant domains of approximately 77-79 kDa and 27-29 kDa, respectively. The cofactor ATP and the presence of DNA, either specific or unspecific, are important stabilizers of the Res subunit. The large N-terminal domain of Res contains numerous functional motifs that are predicted to be involved in ATP-binding and hydrolysis and/or DNA translocation. The C-terminal small domain harbours the catalytic center. Based on our data, we conclude that both structural Res domains are connected by a flexible linker region that spans 23 amino acid residues. To confirm this conclusion, we have investigated several EcoP15I enzyme mutants obtained by insertion mutagenesis in and around the predicted linker region within the Res subunit. All mutants tolerated the genetic manipulation and did not display loss of function or alteration of the DNA cleavage position.

The mass spectrometric decompositions of beta-cyclodextrin (beta-CD) complexed with a number of common divalent metal cations (Mg, Ca, Cd, Cu, Co and Pb), obtained under electrospray ionization conditions, are reported. The main fragmentation pathways of [beta-CD+Cat](2+) ions studied (Cat stands for divalent cation) consist of consecutive losses of sugar units. The rupture of C-C bond in sugar units, which occurs via hydrogen atom transfer from the fragment ion formed to the eliminated species, was also observed. Isotope labelling consisting of the exchange of all hydroxyl hydrogens for deuteriums, has been applied in order to understand better the formation of fragment ions. It was found that C-H hydrogen transfer proceeds only during fragmentation across C-C bonds.

The reaction of styrene oxide with DNA components was studied using separation by capillary zone electrophoresis (CZE) and detection by negative-ion electrospray mass spectrometry (MS). The CZE-MS interface was built for a sector field mass spectrometer. The reaction of styrene oxide with mononucleotides (dGMP, dAMP) was used to optimize the relevant separation parameters and to gather the first information about the behaviour of the possible products. With these mixtures, sample stacking procedures were developed and the scope of collision-induced dissociations were studied. From the fragments recorded, information about the reaction sites in the nucleotides was obtained. Further, the reaction with intact calf thymus DNA was investigated. The DNA was digested into oligonucleotides using the previously described approach with Benzonase, an unspecific nuclease, and alkaline phosphatase. Styrene oxide mono-adducts in dinucleotides, trinucleotides and tetranucleotides were detected, whereas pentanucleotides exhibit mono- and discernible amounts of di-adducts. The hexanucleotides were generally modified twice. The alkylated species moved faster than the unmodified oligomers.

For the determination of the cytotoxic drug 5-fluorouracil in tissue, a sensitive and selective assay has been developed based on microbore high-performance liquid chromatography and fluorescence detection after pre-column derivatization with 4-bromomethyl-7-methoxycoumarin. 5-Chlorouracil was found to be an appropriate internal standard. A column switching protocol has been devised to separate the analyte from late eluting matrix components and the natural uracil. The drug can be determined at concentrations in the low ng/g wet tissue range with a detection limit of 3 ng/g; furthermore, the same protocol can be applied to analyze serum and plasma samples. The procedure was employed to determine the drug in samples taken from human liver tumors and metastases after an intra-arterial bolus administration and from epithelial carcinomas after systemic continuous application.

Incorporated tetraalkyllead compounds are metabolized in the liver and the highly toxic trialkyllead species are excreted via the urine. The procedure for the determination of these metabolites in urine consists of solid-phase enrichment, reversed-phase pre-column high-performance liquid chromatography (HPLC) and chemical reaction detection. As urine is a very complex matrix, it must be questioned whether the retention time alone is a sufficient criterion for the identification of the analytes. For the trimethyllead ion the validity of the results was examined by selectivity checks of the chemical reaction detector, by the application of different stationary and mobile phases in single and dual pre-column HPLC systems and by the use of thermospray LC-mass spectrometry as an independent method. The results demonstrated that the recommended method is accurate for the determination of trimethyllead in urine samples.

Periodic acid oxidation in methanol followed by incubation with 1, 1-dimethylhydrazine results in release of diacylglycerols from 1,2-diacyl-3-glycosyl-sn-glycerols. During hydrazinolysis of oxidized monogalactosyl diacylglycerol, an intermediate hydrazone derivative was observed which was isolated and identified. The diacylglycerols recovered are 1,2-diacyl isomers containing the same fatty acid mixtures as the intact glycolipids. The yields of diacylglycerols released from plant monogalactosyl-, digalactosyl-, and sulfoquinovosyl diacylglycerols were in the range of 30-50%. The method may be used for analysis of molecular species and for preparative purposes.
