The performance of rats on the conditioned blocking test (CB) of learned inattention was measured in a two-way shuttle avoidance task after sham and dopamine (DA) - depleting lesions of the frontal cortex, the limbic septum, and the ventral tegmental area (VTA - A10). Methods: Animals were trained on two sessions with tone and / or light as conditioned stimuli. One group was trained with both stimuli on both sessions. A second group was trained on the first session with one stimulus and on the second with both stimuli. The blocking of conditioning to the added stimulus (b) was tested by presenting the stimuli (a and b) separately and measuring the blocking ration (avoidance to b/a + b) and response latencies. Results: 1/ No deficits were recorded on tests of sensory and motor ability; 2/ The VTA group alone showed a hyperlocomotor response to apomorphine treatment, - and did not acquire the avoidance response (i.e. did not learn the active avoidance task); 3/ The appearance of blocking in the septally lesioned group was delayed until the end of the 20-trial test session - then it was exaggerated; 4/ Blocking was mildly attenuated in the frontally lesioned group. 5/ Dopamine (DA) levels were depleted by about 80% and noradrenaline (NA) levels by, respectively, 20% and 50% in the frontal and septal regions. Figure 2 illustrates a) the CB impairment in the frontal group relative to sham controls, and b) the late development of "supr-blocking" in septally-damage animals. Figure 3 illustrates the results of the HPLC analysis for NA, DA and DOPAC in frontal cortex, septum, N. accumbens and striatum after 6-OHDA lesions (& vehicle treatment) in the frontal, septal, and VTA areas. Conclusions: The results show that the levels of DA activity, or rather the balance between the activity of DA and NA in frontal and limbic regions can contribute to efficient associative conditioning and / or the normal ability of rats not to attend to a redundant stimulus.

Eutrophication is one of the most common causes of water quality impairment of inland and marine waters. Its best-known manifestations are toxic cyanobacteria blooms in lakes and waterways and proliferations of green macro algae in coastal areas. The term eutrophication is used by both the scientific community and public policy-makers, and therefore has a myriad of definitions. The introduction by the public authorities of regulations to limit eutrophication is a source of tension and debate on the activities identified as contributing or having contributed decisively to these phenomena. Debates on the identification of the driving factors and risk levels of eutrophication, seeking to guide public policies, have led the ministries in charge of the environment and agriculture to ask for a joint scientific appraisal to be conducted on the subject. Four French research institutes were mandated to produce a critical scientific analysis on the latest knowledge of the causes, mechanisms, consequences and predictability of eutrophication phenomena. Background: Sleep and cognitive impairments are two of the most prevalent neuropsychiatric disorders in the aged population. Age-related memory dysfunctions can result from alterations in sleep/wake circadian rhythm. However, the underlying mechanism of these alterations is unknown. Here, we demonstrate the role of alterations in brain steroid levels in age-related sleep-dependent memory impairment in rats., Methods: Sleep/wake circadian activity and spatial memory performance were evaluated in adult, middle-aged, and aged rats, and steroid levels were measured in brain structures involved in mediating sleep-dependent memory processes using gas chromatography/mass spectrometry. The causal relationship between circadian activity and allopregnanolone levels was assessed using an inhibitor of allopregnanolone synthesis (indomethacin)., Results: Similar to observations in humans, a subpopulation of middle-aged and aged rats show flattened amplitude of circadian activity associated with impaired spatial long-term memory performance. Sleep-dependent memory dysfunction was associated with a low level of allopregnanolone in the hypothalamus, pedunculopontine nucleus, and ventral striatum. Inhibition of allopregnanolone synthesis in young rats decreased allopregnanolone in the hypothalamus and produced flattened amplitude of circadian activity similar to aged rats., Conclusions: These findings identify brainstem and basal forebrain allopregnanolone as an essential endogenous substrate involved in mediating sleep-dependent memory function in young and aged rats. Allopregnanolone may play a critical role in preserving individuals from age-induced alterations in sleep and memory processes and may represent a novel target for attenuating age-related declines in sleep and memory., (Copyright © 2010 Society of Biological Psychiatry. Drug addiction is a medical condition, a chronic relapsing disease. As in other domains of experimental medicine, appropriate experimental investigations are needed in order to better understand the disease. However, to understand the diverse facets of drug effects and of the underlying pathophysiology it is necessary to keep in mind the complexity of the psychopathological processes. The main symptoms that characterize addiction correspond to expressions of dysfunctions within specific circuits and regions. Pathways to addiction are numerous and comorbidity and in the real world poly-drug use are common. Some of these aspects will be examined as well as the role of life events and stress. Theoretical considerations will be proposed [see also: Koob, G.F., & Le Moal, M.. 2005a. Neurobiology of Addiction. Elsevier. 570 pp] to account for the stages of the disease from impulse control disorder to compulsive disorders, for affective dynamics and for the relations between the symptoms and pathophysiology.

In humans, memory impairments are highly prevalent in the aged population, but their functional and structural origins are still unknown. We hypothesized that circadian rhythm alterations may predict spatial memory impairment in aged rats. We demonstrate an association between sleep/wake circadian rhythm disturbances (non-REM sleep fragmentation) and spatial memory impairments in aged rats. We show by light and electron microscopy that these age-related disruptions in circadian rhythm and spatial memory are also associated with degeneration of cholinergic neurons of the pedunculopontine nucleus (PPT), a structure known to be involved in sleep and cognitive functions and which is altered during aging. Finally, we demonstrate that a trophic deregulation of the PPT occur in aged impaired rats, involving an over activation of the TGFbeta-Smad cascade, a signalling pathway involved in neurodegeneration. In conclusion these results provide a new pathophysiological mechanism for age-related sleep-dependent memory impairments opening the ground for the development of new therapeutic approaches of these pathologies.

Background: Prenatal stress constitutes a developmental risk factor for later psychopathology. The behavioral disorders are sustained by neurobiological alterations including long-term reduction of hippocampal neurogenesis; its deregulation has been involved in cognitive impairments, mood disorders and addiction. A major goal is to define periods in development and strategies for intervening to prevent the effects of early stressful events. We investigated the ability of a postnatal infantile stimulation to prevent prenatal stress-induced alteration in hippocampal neurogenesis., Methods: The influence of postnatal handling on prenatal stress-induced changes in hippocampal neurogenesis was examined in 4 and 26 month-old male rats. Three distinct phases of the neurogenesis were studied: proliferation, survival and neuronal differentiation., Results: Prenatal stress reduced hippocampal cell proliferation all throughout life. Furthermore, the survival rate of newborn cells, the number of immature neurons and the number of differentiated new neurons were reduced in young and old prenatally-stressed rats. All those deleterious effects were counteracted by neonatal handling., Conclusions: These data show that finer aspects of brain shaping can be rewired by environmental influences occurring at sensitive phase of development. They also suggest that infantile stimulation may reverse the appearance of behavioral disorders induced by early life stress.

Ageing is accompanied by an alteration of spatial memory, a decline in hippocampal neurogenesis and a dysregulation of the hypothalamic-pituitary axis (HPA) leading to elevated levels of circulating corticosterone. However, the role of the HPA axis in age-related decline in cognitive functions and in neurogenesis decline remains unclear. We found that suppression of glucocorticoids secretion from midlife to the rest of the animals' life increases neurogenesis in old animals and prevents the emergence of age-related memory disorders. Reciprocally, aged rats with a chronic upregulation of the HPA axis exhibit not only spatial memory impairments but also very low levels of hippocampal cell proliferation and survival. Altogether, these results indicate that the extent of lifetime exposure to glucocorticoids determines the extent of age-related decline in hippocampal neurogenesis and consequently age-related cognitive dysfunctions.

Age-dependent cognitive impairments have been correlated with functional and structural modifications in the hippocampal formation. In particular, the brain endogenous steroid pregnenolone-sulfate (Preg-S) is a cognitive enhancer whose hippocampal levels have been linked physiologically to cognitive performance in senescent animals. However, the mechanism of its actions remains unknown. Because neurogenesis is sensitive to hormonal influences, we examined the effect of Preg-S on neurogenesis, a novel form of plasticity, in young and old rats. We demonstrate that in vivo infusion of Preg-S stimulates neurogenesis and the expression of the polysialylated forms of NCAM, PSA-NCAM, in the dentate gyrus of 3- and 20-month-old rats. These influences on hippocampal plasticity are mediated by the modulation of the gamma-aminobutyric acid receptor complex A (GABA(A)) receptors present on hippocampal neuroblasts. In vitro, Preg-S stimulates the division of adult-derived spheres suggesting a direct influence on progenitors. These data provide evidence that neurosteroids represent one of the local secreted signals controlling hippocampal neurogenesis. Thus, therapies which stimulate neurosteroidogenesis could preserve hippocampal plasticity and prevent the appearance of age-related cognitive disturbances.

In the present study the effect of aging on the basal expression of three different immediate early genes (IEGs) was investigated. The protein products of c-fos, c-jun, and egr-1 genes were visualized immunohistochemically in the rat hippocampus of young adult (4-month-old) and old rats (20-month-old). Astrocytes were quantified by GFAp immunostaining to determine whether changes in the expression of IEGs were correlated with modifications in this marker of degenerative changes. In the young adult rat brain, basal levels of c-Jun and Egr-1 but not c-Fos were detected within the hippocampal formation. Whereas very high basal levels of c-Jun were found in the dentate granule cells and in the pyramidal cells of the ventral hippocampus, Egr-1 was highly expressed in the CA1 pyramidal cells of the dorsal hippocampus. Aging was accompanied by a decrease in Egr-1 expression, by a decrease in total cell density, as well as by a loss of astrocytes in CA1 subfields. In contrast, basal expression of c-Fos and c-Jun as well as astrocyte density within the dentate gyrus were not affected by aging. No difference in these markers was observed in aged rats with or without impairment in spatial learning in a water maze. It was concluded that although these changes may reflect senescence-induced decline of brain function, they do not constitute the defect underlying the age-associated reduction in mnesic capability.

Urtica dioica agglutinin (UDA) is a mouse T-lymphocyte-specific mitogen endowed with proliferative characteristics different from ConA, the prototypic T-lymphocyte mitogen. In particular, UDA induces 2-3-fold-reduced thymidine incorporation as compared to ConA. In an attempt to define the basis of this reduced proliferation, we analysed whether UDA binds to a unique subset of T lymphocytes or whether it activates only a T-cell subset. Cytofluorimetric analysis showed that this lectin binds uniformly to all T lymphocytes and does not, on this criterion, distinguish a particular T-cell subset. We next analysed whether UDA provokes the activation of all T lymphocytes. This was carried out by measuring the increase in cell size and the induction of the p55 chain of the IL2 receptor. The analysis showed that, throughout the kinetics of cell activation, only one subset of T lymphocytes increased in size and expressed the p55 chain of the IL2 receptor, suggesting that UDA activates only a subpopulation of T cells. This conclusion was strengthened by the analysis of 5-bromo-2-deoxyuridine (BrdU) incorporation into the DNA of UDA-activated cells. Two populations were easily identifiable: a BrdU-negative subset consisting of all the small p55-negative lymphocytes, and a BrdU-labelled subset including all the large p55-positive cells. BrdU was incorporated in both CD4+ and CD8+ cells, indicating that UDA did not distinguish helper from cytotoxic T lymphocytes. In addition to the p55 chain of the IL2R, all cycling cells expressed the Pgp-1 activation marker. The T lymphocytes, which bound UDA but did not proliferate, remained fully susceptible to subsequent stimulation by ConA. In conclusion, the capacity to proliferate upon UDA binding differentiates a UDA-sensitive from a UDA-refractory subset among splenic mouse T lymphocytes.

Urtica dioica agglutinin (UDA) is a T-lymphocyte-specific polyclonal activator that differs from ConA, the classical mouse T-cell mitogen, by inducing a late and limited proliferation of a distinct T-cell subset recruited among both CD4+ and CD8+ lymphocytes. We investigated the possibility that the particular kinetics may originate from UDA-specific activation processes in which the known early mandatory signals were completed only after an extended delay. We report that the time of contact required between lectin and the cell membrane to acquire the capacity to proceed into cell cycle was much longer (36-40 h) for UDA than for ConA (8-10 h). Addition of phorbol ester, which artificially induces PKC translocation, or ionomycin, which provokes Ca2+ mobilization, did not accelerate the proliferative kinetics, suggesting that these early mandatory signals are not the limiting factors in the delayed proliferation. The induction of c-myc was retarded in the UDA group, and there was a good correlation between the kinetics of c-myc induction and the kinetics of cell proliferation. The comparison of the level of transcription of the genes encoding different cytokines revealed additional differences between the two mitogens: the whole wave of cytokine gene expression was delayed with UDA. In particular, IL2, IL3 and IFN gamma gene expression was retarded compared to the ConA-induced single wave. An even later transcriptional wave took place at around 72 h for IL4 and IL5. Finally, this particular kinetics corresponded to an unusually high level of IL3 and IFN gamma and a low level of IL4 and IL5 gene transcripts.(ABSTRACT TRUNCATED AT 250 WORDS)

The influence of a chronic social stress upon immunity was investigated in Wistar rats, submitted for four weeks to two different behavioral situations, balanced in a factorial design: housing with three females and membership rotation. The combination of these two factor led to adrenal enlargement (43.3%), thymus involution (39.5%) and increased basal corticosterone levels, all indices of activation of the hypothalamic-hypophysis-adrenal axis. However, neither natural killer cell activity, splenocyte reactivity to mitogen nor the rate of spontaneous development of antibodies against Mycoplasma pulmonis, a common pathogen of the respiratory tract, were changed in the endocrine activated animals. Analysis of the data on kinetics of stress at 1, 7 and 28 days after the initial mixing of the animals gave the same results. These data question the immunosuppressant activity usually conferred to corticosteroids, at least when adrenal hyperactivity is induced by chronic environmental stressors.

The brain is known to modulate the immune system in an asymmetrical way. In mice, there is an association between handedness and immune response and it has also been shown that hemicortical ablation has opposite effects on some immune parameters. An association between autoantibody production and paw preference was previously observed in female mice, but not in males, suggesting that the association between immune reactivity and functional brain asymmetry is a sex-dependent phenomenon. In three independent experiments, natural killer (NK) cell activity, lymphocyte subset distribution, and mitogen-induced lymphoproliferation were assessed in male C3H/OuJIco mice selected for handedness and after unilateral cortical ablation. Handedness was shown to be associated with NK cell activity but not with lymphocyte subset distribution or lymphoproliferation. Left-handers exhibited lower NK cell activity compared to right-handed or ambidextrous animals. In contrast to previous results in female mice, mitogen-induced lymphoproliferation was not associated with handedness in males. Left cortical ablations depressed NK cell activity, while right lesions had no effect. Neither left or right lesions affected lymphocyte subsets. No interaction between paw preference and side of the lesion was found in the modulation of NK cell activity. These and previous data show that the association between paw preference and immune reactivity varies according to the sex of the animal and the immunological parameters studied. This indicates that the brain may modulate different components of the immune system in different ways, through mechanisms apparently involving sex hormones.

Transcutaneous electrostimulation is a somewhat controversial technique used in the management of the opiate withdrawal syndrome. We report an animal study of a particular transcutaneous electrostimulation called transcutaneous cranial electrostimulation, based on a technique used for many years on heroin addicts for the rapid severance of their addiction, which has been validated in a clinical setting by a double-blind trial. This technique involves the application of an intermittent high-frequency current (Limoge's current). Our experimental data show that this transcutaneous cranial electrostimulation increases morphine analgesia by threefold on the tail flick latency measure and produces a 48% attenuation of the abstinence syndrome observed after abrupt cessation of morphine administration. These results were obtained using a double-blind paradigm.

The effects of the new tricyclic antidepressant tianeptine were investigated on dopaminergic (DAergic) metabolism in the anteromedian prefrontal cortex and the nucleus accumbens of the rat. DAergic metabolism was assessed by the measurement of DOPAC, the main presynaptic metabolite of dopamine, using in vivo voltammetry in rats ventilated with halothane (0.5-0.75% in air). Acute treatment with tianeptine (10 mg/kg, 20 mg/kg) only increased significantly DOPAC levels in the anteromedian prefrontal cortex. After chronic treatment with tianeptine (15 days, 2 times/day) the increases in DOPAC levels in this structure were altered and less pronounced with the 20 mg/kg dose. Previous studies led to suggest that both acute and chronic effects on DAergic terminals in the anteromedian prefrontal cortex may be involved in the therapeutic action of this new antidepressant.

Behavioral sensitization can be observed with repeated administration of amphetamine where the intensity of motor stimulation increases over time. The process of sensitization has been well characterized, however, the neurochemical mechanisms that are critical for the development of sensitization are not known. In the present study, the role of the hypothalamic pituitary adrenal axis (HPA) in the development of behavioral sensitization to amphetamine was explored by pretreating rats with an intravenous administration of an antiserum to corticotropin-releasing factor in a volume that has been shown to block significantly stress- and cocaine-induced activation of the HPA. Four groups of eight rats were pretreated intravenously with either heparinized saline or CRF antiserum and subcutaneously with saline or d-amphetamine in a balanced design. The rats were then returned to their home cages and left undisturbed for seven days after which they were given three consecutive behavioral tests with saline SC, 0.75 mg/kg d-amphetamine SC, and 3.0 mg/kg d-amphetamine SC. The rats pretreated with intravenous CRF antiserum showed a significant attenuation of the development of d-amphetamine-induced sensitization but the antiserum did not alter the magnitude of the behavioral response to the initial, sensitizing dose of d-amphetamine. These results suggest that activation of the hypothalamic pituitary adrenal axis may be of critical importance to the development of behavioral sensitization to amphetamine.

Buspirone is a non-benzodiazepine drug with anxiolytic properties. It has been reported to induce a marked increase in the metabolism of dopamine in the striatum and the nucleus accumbens which is similar to that induced by neuroleptics. It has been suggested that the effect observed in the striatum reflects an action of buspirone on dopaminergic autoreceptors in both terminals and cell bodies. In the present study, presynaptic effects of buspirone on dopaminergic metabolism in the nucleus accumbens were investigated, and they were compared to the effects of the classical neuroleptic, haloperidol. Dopaminergic terminals were isolated by infusion of tetrodotoxin into the median forebrain bundle in order to evaluate the effects of buspirone and haloperidol on presynaptic receptors. Changes in dopamine metabolism were determined by in vivo voltammetry. Buspirone administered after interruption of the impulse flow did not affect dopamine metabolism. In contrast haloperidol treatment led to an increase in metabolism of dopamine. It is concluded that buspirone did not act at the presynaptic level and furthermore on dopaminergic autoreceptors.

Urtica dioica agglutinin, a small-molecular-weight lectin purified from stinging nettle rhizomes, induces murine cell proliferation. U. dioica agglutinin is a specific T-cell mitogen for both thymocytes and spleen T lymphocytes; its mitogenic properties are strictly dependent on the presence of accessory cells. The kinetics of proliferation are markedly different from those of the classical T-cell mitogen concanavalin A, with a 2 to 3-day delay for both splenic and thymic populations and a rate of DNA synthesis twofold lower than that observed with concanavalin A. The late T-lymphocyte proliferation induced by U. dioica agglutinin correlates well with (i) the observed late interleukin-2 production and interleukin-2 receptor expression, and (ii) the long-lasting cyclosporin A-sensitive early activation period. In contrast, the production of interleukin-1 is not different, both in terms of concentration and kinetics, from that observed with concanavalin A.

The thymocyte costimulator (LAF) assay, the standard biological test used for IL-1 titration, has a low sensitivity and lacks specificity since it can be potentiated by the IL-2 which is frequently present in IL-1-containing biological fluids. We describe here a new IL-1 titration method which takes advantage of the capacity of a thymoma line, EL4-6.1, to differentiate and express IL-2 receptors upon stimulation by IL-1 in the presence of a suboptimal dose of phorbol diester. Membrane IL-2R measurement on this indicator cell line permits the detection of 1-2 X 10(-4) ng/ml IL-1, compared to 5 X 10(-2) ng/ml in the LAF assay. In addition, rIL-2 up to 250 U/ml has no effect on IL-1 measurement by this assay, which also exhibits a 100-fold lower sensitivity to inhibitory effects of prostaglandin, compared to the LAF assay. Finally, tumor necrosis factor alpha only exerts a weak costimulation effect at very high doses. A flow cytometry technique and an ELISA are described for IL-2 receptor detection. Due to its high sensitivity and specificity, this novel assay should now permit reliable IL-1 titration in biological fluids such as IL-2-rich lymphocyte culture supernatants.

We improved an ELISA bioassay for murine IFN-gamma (MuIFN-gamma) based on measurement of Ia antigen on P388D1, a mouse macrophagic tumour line. Cells were cultured in microtitre plates in medium containing dilutions of IFN-gamma source. They were then washed and stained with a rat anti-mouse I amAb followed by mouse anti-rat peroxidase-labelled antibody. After incubation with substrate, the OD was read directly from microtitre plates. Standard curves obtained with reference NIH MuIFN-gamma showed that this assay allowed for the definition of unit values (giving 50% of the maximal effect) comparable to NIH international units (IU). It detected as low as 0.2 IU/ml of MuIFN-gamma and, in contrast to antiviral assays, was insensitive to IFN-alpha/beta. We used a concanavalin A-conditioned supernatant, which is a mixed source of lymphokines, to assess the specificity of our assay. Indeed, Ia expression induced by ConA-conditioned supernatant was fully inhibited by preincubation with anti-MuIFN-gamma antibodies. Using a stable indicator cell line, the present cell surface assay is easier to perform than other ELISA using bone-marrow-derived macrophages, and does not require cell fixation; its high sensitivity and specificity are comparable to that of immunoradiometric assays. It is thus valuable for routine MuIFN-gamma quantitations in culture supernatant and biological fluids.

The primary and secondary responses to Tnp-Ficoll, a class 2 thymus-independent antigen, were assessed in various inbred strains of mice. The eventual implication of H-2 or IgH linked genes was searched for. Contrasting with our previous reports using Tnp-LPS, a class 1 thymus-independent antigen, no homologous memory-type response to Tnp-Ficoll and consequently no genetic control was observed. However Tnp-specific B-memory lymphocytes were induced in most strains since a heterologous challenge with Tnp-LPS evoked a typical memory type response characterized by an increased number of antibody-secreting cells and/or significant amount of anti-Tnp antibodies of the IgG isotype. The lack of memory revelation by Tnp-Ficoll is discussed in terms of a possible humoral or cellular regulation and of B-memory cell generation and maturation.

Since the primary PFC response to TNP-Ficoll, a thymus-independent type 2 antigen, displays an important variability in vivo among diverse inbred mouse strains, we used, in the present report, H-2 congenic strains possessing different genetic backgrounds to show that the amplitude of this response is governed by MHC genes, with one regulating locus situated in or near the centromeric part of the I-A subregion. In addition, this H-2 control was largely modulated by gene(s) located outside MHC and IgH haplotypes, as evidenced by the response of recombinant inbred strains (BXH) between the high responder C3H/HeJ and the low responder C57BL/6J. Our results are discussed in terms of humoral regulations and the requirement for self-recognition in cellular interactions which lead to activation of B lymphocytes in the in vivo primary response towards TI-2 antigens.

The lesion of the ventral mesencephalic tegmentum region (VMT) induces a behavioral syndrome characterized mainly by locomotor hyperactivity and reduction of attention processes. Previous data indicated that this syndrome was due at least in part to the destruction of dopaminergic (DA) A10 neurons. In order to test this hypothesis, we examined the effects of two dopaminomimetics drugs: apomorphine (APO) and d-amphetamine (d-AMPH) on the VMT behavioral syndrome. The acute administration of very low doses of APO (30 microgram/KG; Sc) reduces the behavioral deficits; similarly a chronic administration of d-aMPH (two injections daily for 43 days) reduces locomotor hyperactivity. In these two cases, the lesioned rats activity reaches the control level. These results confirm the primary role of DA-A10 neurons in the VMT behavioral syndrome. Acute APO and chronic d-AMPH effects are discussed in terms of (i) reactivation of DA postsynaptic receptors disafferented after DA-A10 group destruction and (ii) strengthening of the hyperfunctioning of the remaining DA-A10 neurons. VMT-A10 syndrome could be a good animal model for pathophysiological studies.

The present experiments investigated changes in dopaminergic mesocorticolimbic neurones originating from the A10 cell group, in animals exposed to electric shocks in pairs or individually, in comparison to animals receiving no shock and tested in pairs or alone. The social setting under which shock occurred had no influence on the increases in DOPAC levels observed in animals exposed acutely or chronically to electric shocks. In contrast, subordinate rats in the paired shock condition had lower tyrosine hydroxylase activity in the accumbens than dominant rats. Pairing of animals in the test cage without shock induced an increase in accumbens DOPAC levels.

The behavioral effects of systemic heroin administration were examined in rats subjected to flupentixol impregnation prior to and during behavioral testing. In the first experiment, the dose of heroin required to produce a place preference was determined in two groups of rats, one of which had received chronic flupentixol decanoate (12 mg/kg, sc, every 10 days for 6 weeks) and the other which had received the palm oil vehicle during the same time period. It was found that whereas 60 micrograms/kg of heroin was required to produce a place preference in control rats, only 7.5 micrograms/kg was sufficient to do so in chronic neuroleptic-treated rats. In a second experiment, the locomotor activating effect of heroin was evaluated in two groups of rats that had been subjected to the same chronic regimen as in Experiment 1. Locomotor activity was enhanced in both groups following 120 micrograms/kg heroin, whereas 30 micrograms/kg was ineffective in either group. Finally, it was found that neuroleptic-treated, but not control, rats rapidly learned to self-administer intravenous infusions of an unusually low dose of heroin (4 micrograms) and to discriminate these infusions from vehicle infusions. Together, these data show that chronic dopamine (DA) receptor blockade produces a marked increase in the sensitivity to the reinforcing and discriminative stimulus properties of systemic heroin administration and that this increase is not attributable to heroin-induced locomotor activation. The results are discussed in terms of the role of DA systems in opiate reinforcement processes.

Relations between a peptide, hormone or neurotransmitter and behavior involve a molecule and a frequently complex process. The psychobiologist suggest the need for caution against extensive, operative and pharmacologic use of the notion of behavior. Multifactorial organization of adaptive behavior lends itself poorly to a reductionism leading to the equation molecule-behavior; however, neuropeptides do modulate behavior, sometimes potently. These subjects are discussed from a methodologic and critical point of view. Neurobiologists and endocrinologists have too often neglected the organizational and controlling roles of behavior on global neurobiologic and neuroendocrine reactions. Several examples are given to illustrate these various propositions.

Bovine serum albumin haloperidol (BSA-Hal) is a macromolecular complex with 14 molecules of haloperidol immobilized on the BSA protein backbone. This compound produces a selective, long lasting and reversible blockade of dopamine receptor activity. Its action was demonstrated by the ability to trigger a high rate of ipsilateral amphetamine-induced rotation up to 6 days after a single unilateral injection of the conjugate into the striatum. In the present study, the effect of the blockade of dopamine transmission in the nucleus accumbens (n.Acc) on spontaneous and learned behaviors was tested. The results indicate that the specific and long lasting blockade of dopamine receptors by bilateral injection of BSA-Hal in the n.Acc (1.6 micrograms/2 microliters) induced deficits in spontaneous alternation in a Y-maze on the 2nd and 5th days after the injection but not on the 11th day of the experiment, impaired acquisition but not retention in a radial 8-arm maze, increased latency to escape during learning in the place navigation task. These findings confirm the involvement of the n.Acc system in processes that have been generally attributed to the limbic system.

1. Slow rhythmic theata-type waves were recorded in the sub-interpeduncular ventral mesencephalic tegmentum (VMT) in the rat. This region is one of the parts of the "limbic midbrain area". These waves are recorded during certain attentive waking states, during the execution of voluntary movements and during paradoxical sleep. 2. These rhythmic VMT waves seem to be of the same nature as the theta rhythm recorded in the limbic structures. A lesion of the septum suppresses the slow rhythmic waves at the mesencephalic level as well as at the telencephalic level. 3. Although we have now shown that the mesencephalic and telencephalic slow rhythmic waves have the same pacemaker, the pathways which transmit the theta rhythm to the VMT are not well known. However, it is reasonable to suppose that the fibres of the post-commissural fornix are involved in this transmission.

The effect of electric footshocks and of exposure to environmental stimuli paired with electrical shocks upon the dopaminergic activity in various cortical and limbic areas of the rat were evaluated by measuring dihydroxyphenylacetic acid (DOPAC) levels in these areas. In animals exposed to a 20 min electric footshock session DOPAC concentrations were significantly increased in the antero-medial and sulcal frontal cortices, olfactory tubercle, nucleus accumbens and amygdaloid complex (by 66, 37, 28, 55 and 90% respectively). Re-exposure of rats to an environment where they had been shocked 24 h earlier induced an elevation of DOPAC content only in the anteromedial frontal cortex (by 47%). Plasma corticosterone levels were elevated in both situations. No change in serotonin or 5-hydroxyindolacetic acid content of these areas could be detected in either situation. The results show that electric footshocks and environmental stimuli associated to previous shocks both activate central dopaminergic systems, although the patterns of activation are different.

After training in two different passive avoidance tasks, the platform box of Ader and De Wied (1972) and the Jarvik box of Jarvik and Kopp (1967), rats injected with vasopressin immediately following the training trial showed a significant enhancement of retention 24 hours later. This vasopressin effect was reversed by high doses of the vasopressor antagonist, dPtyr(Me) AVP. These results support the hypothesis that the visceral afferent signals may be involved in the apparent memory-enhancing effects of AVP, but the high doses of antagonist required suggest that factors other than a simple reversal of the pressor effects of AVP may be important.

The influence of the cerebral neocortex on the immunological status was studied in groups of C3H/He mice after lesioning the right or left fronto-parietal cortex. In left lesioned mice, mitogenesis induced either by phytohemagglutinin (PHA) or concanavalin A (Con A) was depressed by about 50% compared to controls. On the contrary, in animals with right lesions, T cell mitogenesis was enhanced by about 140% as compared to controls and by 220-300% as compared to that observed in left lesioned animals. Mitogenesis of B cells induced by lipopolysaccharide was modified by cortical lesions in exactly the same way as that of T cell proliferation although not reaching statistical significance. These results confirm the lateralization in the cortical modulation of the immune system.

Rats were tested in a simple one-trial water-finding task for the effects of arginine vasopressin (AVP) on performance of an appetitive task. On the training day, each animal was exposed for 5 min to a novel open-field environment that contained a water-tube located in an alcove set into one of the walls of the enclosure. Immediately upon removal from the enclosure, the animals received a subcutaneous injection of either AVP (1 microgram/rat) or vehicle solution. When water-deprived and tested 48 hr later, vasopressin-treated rats found the water tube reliably faster than controls. In other groups of animals, this potentiation in learned performance was prevented by concurrently treating the rats with a vasopressin analog having potent pressor antagonist properties. These results are consistent with the notion that vasopressin may play a role in memory consolidation, but peripheral visceral factors may mediate this action.

1. Multi-unit activity (MUA) was recorded together with gross activity in different structures during various states of sleep in 43 rats. The records were made by means of telemetry from lateral hypothalamus, nucleus ruber, formatio reticularis, dorsal hippocampus and the ventral mesencephalic tegmentum (VMT) surrounding the nucleus interpeduncularis. 2. The records during paradoxical sleep showed slow rhythmic theta waves located in the VMT and the dorsal hippocampus. In these two structures the MUA was mainly organized in periodic patterns of discharges. In the other structures a continuous increment of the basic level of discharge was superimposed on the periodic patterns. 3. The similarity of the hippocampal and VMT records strengthens the hypothesis that the VMT region has anatomical and functional connections with the limbic system.

The effects of buspirone, a buspirone analogue (BMY 13805) and a buspirone metabolite (1-PP) on dopaminergic metabolism in the nucleus accumbens were investigated using in vivo voltammetry. Differential pulse voltammetry coupled with electrochemically pretreated carbon fiber electrodes was used to provide a continuous and selective measure of the 3,4-dihydroxyphenylacetic acid (DOPAC). An implanted micromanipulator enabled the use of freely moving animals. Buspirone injections induced a marked and rapid increase of the DOPAC peak in the nucleus accumbens. Buspirone was 10 times more potent when injected subcutaneously than intraperitoneally. BMY 13805 and 1-PP were without effect on dopaminergic metabolism in the nucleus accumbens. In conclusion, the anxiolytic properties of these drugs and their effects on dopaminergic metabolism do not appear related.

Trinitrophenyl (Tnp)-Ficoll, a class 2 thymus-independent (TI) antigen, generates in most mouse strains Tnp-specific B-memory cells which can be detected in situ 1 week after priming by a heterologous stimulation with Tnp-lipopolysaccharide (LPS) but not by a homologous Tnp-Ficoll challenge. We have investigated the secondary responses raised in CB.20 congenic mice by a homologous challenge in situ occurring at various time intervals after priming. We report that a memory-type response is obtained, culminating when the challenge is performed at 4 weeks; this finding assesses definitely the ability of TI-2 antigens to produce immunological memory under standard conditions. However, the same immunization procedure elicits no memory-type response in the majority of other mouse strains, suggesting a possible genetic control of the expression of memory to class 2 TI antigens. The utilization of F1 hybrids between C57BL/6 and BALB/c and of appropriate congenic strains shows indeed that this memory expression is under multigenic control: Igh-V or closely linked genes are clearly involved but a complementation with other gene(s), located outside the H-2 complex, is required for a memory-type response to Tnp-Ficoll. We have also analyzed the secondary heterologous response to Tnp-LPS in CB.20 mice at different times after Tnp-Ficoll priming. The difference in the kinetic profile of the heterologous (TI-2----TI-1) versus homologous (TI-2----TI-2) secondary responses is discussed in terms of B-memory-cell ontogeny and humoral regulation.
