Background and Aims: Peripheral arterial disease (PAD) is a leading cause of morbimortality worldwide. Lipocalin-2 (LCN2) has been associated with higher risk of amputation or mortality in PAD and might be involved in muscle regeneration. Our aim is to unravel the role of LCN2 in skeletal muscle repair and PAD., Methods and Results: WT and Lcn2 -/- mice underwent hindlimb ischemia. Blood and crural muscles were analyzed at the inflammatory and regenerative phases. At day 2, Lcn2 -/- male mice, but not females, showed increased blood and soleus muscle neutrophils, and elevated circulating pro-inflammatory monocytes (p < 0.05), while locally, total infiltrating macrophages were reduced (p < 0.05). Moreover, Lcn2 -/- soleus displayed an elevation of Cxcl1 (p < 0.001), and Cxcr2 (p < 0.01 in males), and a decrease in Ccl5 (p < 0.05). At day 15, Lcn2 deficiency delayed muscle recovery, with higher density of regenerating myocytes (p < 0.04) and arterioles (αSMA + , p < 0.025). Reverse target prediction analysis identified miR-138-5p as a potential regulator of LCN2, showing an inverse correlation with Lcn2 mRNA in skeletal muscles (rho = -0.58, p < 0.01). In vitro, miR-138-5p mimic reduced Lcn2 expression and luciferase activity in murine macrophages (p < 0.05). Finally, in human serum miR-138-5p was inversely correlated with LCN2 (p ≤ 0.001 adjusted, n = 318), and associated with PAD (Odds ratio 0.634, p = 0.02, adjusted, PAD n = 264, control n = 54)., Conclusions: This study suggests a possible dual role of LCN2 in acute and chronic conditions, with a probable role in restraining inflammation early after skeletal muscle ischemia, while being associated with vascular damage in PAD, and identifies miR-138-5p as one potential post-transcriptional regulator of LCN2., Competing Interests: Declaration of competing interest The authors declare that they have no known competing financial interests or personal relationships that could have appeared to influence the work reported in this paper., (Copyright © 2023 The Authors. Objective: Tobii glasses can record corneal infrared light reflection to track pupil position and to map gaze focusing in the video recording. Eye tracking has been proposed for use in training and coaching as a visually guided control interface. The aim of our study was to test the potential use of these glasses in various situations: explanations of anatomical structures on tablet-type electronic devices, explanations of anatomical models and dissected cadavers, and during the prosection thereof. An additional aim of the study was to test the use of the glasses during laparoscopies performed on Thiel-embalmed cadavers (that allows pneumoinsufflation and exact reproduction of the laparoscopic surgical technique). The device was also tried out in actual surgery (both laparoscopy and open surgery)., Design: We performed a pilot study using the Tobii glasses., Setting: Dissection room at our School of Medicine and in the operating room at our Hospital., Participants: To evaluate usefulness, a survey was designed for use among students, instructors, and practicing physicians., Results: The results were satisfactory, with the usefulness of this tool supported by more than 80% positive responses to most questions. There was no inconvenience for surgeons and that patient safety was ensured in the real laparoscopy., Conclusion: To our knowledge, this is the first publication to demonstrate the usefulness of eye tracking in practical instruction of human anatomy, as well as in teaching clinical anatomy and surgical techniques in the dissection and operating rooms., (Copyright © 2017 Association of Program Directors in Surgery. Unlabelled: There are limited data on the prognostic role of biomarkers in anticoagulated patients with atrial fibrillation (AF). We evaluated the prognostic value of high sensitivity TnT (hsTnT) and high-sensitivity interleukin-6 (hsIL6) in a large cohort of AF patients taking oral anticoagulant therapy (OAC) as both biomarkers have been associated with adverse cardiovascular events., Methods: We studied 930 patients (51% male; median age 76) with permanent/ paroxysmal AF who were stabilized (for at least 6 months) on OAC (INRs 2.0-3.0). Plasma hsTnT and hsIL6 levels were quantified by electrochemiluminescense immunoassay at baseline. Patients were followed-up for up to 2 years, and adverse events (thrombotic and vascular events, mortality and major bleeding) were recorded., Results: At follow-up, 96 patients (3.97%/year) died whilst 107 had an adverse cardiovascular event (3.14%/year). On multivariate analysis, high hsTnT and high hsIL6 remained significantly associated with prognosis even after adjusting for CHADS2 score: HR 2.21 (1.46-3.35, P<0.001) for high hsTnT and 1.97 (1.29-3.02, P=0.002) for high hsIL6, for adverse cardiovascular events. For all-cause mortality, the HRs were 1.79 (1.13-2.83, P=0.013) and 2.48 (1.60-3.85, P<0.001), respectively. The integrated discrimination index (IDI) values of clinical scores (CHADS2 and CHA2 DS2-VASc) were improved by the addition of hsTnT and/or hsIL6 (all P<0.05)., Conclusion: In a large 'real world' cohort of anticoagulated AF patients, both hsTnT and hsIL6 levels provided prognostic information that was complementary to clinical risk scores for prediction of long-term cardiovascular events and death, suggesting that these biomarkers may potentially be used to refine clinical risk stratification in AF., (© 2012 International Society on Thrombosis and Haemostasis.)

Botulinum neurotoxins (BoNTs) are among the most toxic substances known. Surveillance and diagnostics require methods for rapid detection of BoNTs in complex media such as foodstuffs and human serum. We have developed in vitro assays to specifically detect the protease activity of botulinum neurotoxin B (BoNT/B) on a time scale of minutes. Cleavage of the BoNT/B substrate VAMP2, a membrane SNARE protein associated with synaptic vesicles, was monitored using real-time surface plasmon resonance to measure vesicle capture by specific antibodies coupled to microchips. The assay is functional in low-ionic-strength buffers and stable over a wide range of pH values (5.5-9.0). Endoproteolytic cleavage of VAMP2 was detected in 10 min with 2 pM native BoNT/B holotoxin. Contamination of liquid food products such as carrot juice, apple juice, and milk with low picomolar amounts of BoNT/B was revealed within 3h. BoNT/B activity was detected in sera from patients with type B botulism but not in healthy controls or patients with other neurological diseases. This robust, sensitive, and rapid protein chip assay is appropriate for monitoring BoNT/B in food products and diagnostic tests for type B botulism and could replace the current in vivo mouse bioassay., (Copyright © 2010 Elsevier Inc. Photo-solid-phase microextraction (photo-SPME) has been employed to study the photolysis of three common indoor air pollutants: coumarin, butylated hydroxytoluene, 2,6-diisopropylnaphthalene. Analytes were first extracted by SPME, and the fibre was subsequently exposed to an irradiation source (i.e. xenon arc or low-pressure mercury lamp) for the selected time (from 2 to 120 min). Analyses of the irradiated fibres were carried out by gas chromatography-mass spectrometry (GC-MS) detection. Photodecay kinetics exhibited a first-order behaviour and their rate constants and half-life times were estimated. Twenty-five photoproducts have been tentatively identified by means of their mass spectra. On the basis of the identified transformation compounds, some photodegradation pathways were proposed. The photoformation-photodecay kinetics of the identified by-products were also monitored by photo-SPME. To the best of our knowledge, photolytic routes for coumarin, butylated hydroxytoluene and 2,6-diisopropylnaphthalene have not been previously investigated.

A method for the extraction of triethyl lead (TEL(+)), trimethyl lead (TML(+)), and Pb(2+) from sand was developed using supercritical modified CO(2)-CH(3)OH extraction and in situ complexation with sodium diethyldithiocarbamate (NaDDTC) using a 2(5) factorial exploratory design is described. The screened variables were (i) pressure (69-193 bar), (ii) temperature (40-150 degrees C), (iii) ligand amount (0-100 mg), (iv) methanol volume (0.0-0.5 mL) and (v) static time (0-45 min). The optimum extraction conditions found were as follow: pressure, 193 bar; temperature, 40 degrees C; amount of NaDDTC, 100 mg; methanol volume, 0.5 mL; static time 45 min; and CO(2) flow rate, 1 mL min(-1). Under these conditions the following recoveries were obtained (TML(+) 97+/-2%, TEL(+) 70+/-5%, and Pb(2+) 100+/-4%). The presence of NaDDTC is not necessary for the extraction of TML(+) and TEL(+), but it is a very significative parameter for Pb(2+). A second experimental design 2(2)+star for temperature and pressure was realized, but the results were not better than those of the first model. SFE extract derivatization was achieved with pentylmagnesium bromide, and target analyte determination was carried out by gas chromatography-mass spectrometry. Detection limits in the full-scan mode were 4, 10, and 39 pg as lead for TMPeL, TEPeL and PbPe(4), respectively. The method was validated with urban dust containing TML(+) (CRM 605. Pb 7.9 +/-1.2 microg kg(-1)) and river sediment containing inorganic lead (GBW08301. Pb 79.0+/-12.0 mg kg(-1)) as reference materials. The proposed method was applied to lead analysis in sand collected from an oil-polluted beach in Chile.

Benzothiazoles, benzotriazoles and benzosulfonamides are high-production-volume chemicals found in various environmental aqueous samples that should be considered as emerging pollutants. This study examines the suitability of comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC x GC-TOF-MS) for the characterization of benzothiazoles, benzotriazoles and benzosulfonamides in aqueous matrices. Solid-phase extraction was optimized in order to ensure the proper preconcentration of these contaminants prior to their analysis. Column selection, in both the first and second dimensions, was optimized to ensure a good chromatographic separation of the target analytes and of the potential interfering compounds extracted from the matrix. Several column phases were tested. The combined power of two-dimensional separation was fully illustrated by identifying minor compounds and avoiding the overestimations usually made by one-dimensional systems. Finally, the suitability of GC x GC-TOF-MS to quantify the aforementioned emerging pollutants was proven by determining those pollutants in several real samples such as river water, effluent from a wastewater treatment plant, and raw sewage.

Unlabelled: The actual gold standard of Botulin A toxin (BoTx A) batches qualification is the mouse lethality assay. With this assay it is nevertheless impossible to set a therapeutic value unit., Aims: The goal of this research was to study the effects of BoTx A increasing concentrations on glutamatergic rat neurons., Experimental Procedure: We studied the glutamate release with increasing concentrations of BoTx A. We also studied the BoTx A target cleavage with a western blot technique., Results: Our results proved that it is possible to establish a dose-response - like curve of BoTx A effects on glutamate release. Moreover the cleavage of the target protein was visible for the same toxin concentrations that inhibited the glutamate release., Conclusion: This technique could be the first step toward a new way of setting a better pharmaceutical profile for toxin batches.

Simultaneous determination of cyanide and volatile alkylnitriles such as acetonitrile, cis- and trans-crotononitrile, allylnitrile and butyronitrile at low ppb concentration on whole blood (rat and mice) by headspace solid-phase microextraction (HS-SPME) followed by gas chromatography (GC) with nitrogen phosphorus detection has been achieved for the first time. SPME extraction time and temperature were optimized using a star experimental design. Optimum conditions for cyanide extraction were chosen to analyze unspiked blood samples containing alkylnitriles as that analyte occurs at the lowest concentrations. For all analytes, the developed methodology yielded good quality parameters. In all cases, good reproducibility (relative standard deviation < or =12%), detection limits (<3ng mL(-1)) and quantification limits (<4 ng mL(-1)) were recorded.

The adsorption of dissolved organic contaminants on glass fibre filters throughout water dissolved/particulate phase decoupling studies was examined. A total of 49 different compounds were considered at low concentration levels (ng L(-1)), including PAHs, PCBs, organochlorine and organophosphorus pesticides, triazines, thiocarbamates, pyrethroids, phosphate esters and caffeine. Their adsorption on the filters was positively correlated with their log Kow and solubilities, indicating that filter adsorption increased with hydrophobicity. The influence of water properties (i.e. salinity and dissolved organic carbon (DOC) content) was also studied by means of a star experimental design (n=11). Salinity was the main factor in increasing the adsorption, due to the salting out effect. The influence of DOC suggested that part of the contaminant losses during water filtration may have been caused by the retention on the organic matter adsorbed on the filter surface. Nevertheless, a decrease in filter retention was observed for water with the highest DOC contents, which was probably due to an enhancement of the contaminant solubility in these conditions. Although several factors may control the adsorption process in naturally occurring waters, the extent of the retention of dissolved target analytes on the glass fibre filters should not be underestimated in the analysis of hydrophobic contaminants in marine and estuarine waters at very low concentrations (ppt level).

Exposure to mercury species was assessed in the hair of 130 Spanish children (age 4) from the general population in two areas. Both areas are exposed to different sources of mercury: a point source in Ribera d'Ebre (northeastern Spain) and a diffuse source on the island of Menorca (northwestern Mediterranean). The median MeHg values in the hair of children from Ribera d'Ebre (RE) were nearly twice (0.631 microg/g vs. 0.370 microg/g) those of children from Menorca (MC) (p < 0.05). Total Hg showed a similar trend (REmedian: 0.720 microg/g vs. MCmedian: 0.476 microg/g). Nevertheless, inorganic mercury levels were similar in the two groups of children (REmedian: 0.186 microg/g vs. MCmedian: 0.210 microg/g). Two subgroups of the Ribera d'Ebre group were defined: children living in Flix (a village near a chlor-alkali plant) (RE1) and children living on the outskirts of Flix with no clear, direct influence of the plant (RE2). The mercury concentrations in RE1 were also significantly higher than those in Menorca, but no significant differences were found between Menorca and the RE2 subgroup. We evaluated the fish consumption of RE1, RE2 and MC and found that the Menorcan children consumed significantly less fish (p < 0.05) than the other two subgroups. Children who consumed fish more than three times a week had higher MeHg concentrations (beta (SE) = 0.991 (0.279) than those who ate it less than once a week. Nevertheless, the differences in MeHg levels between children from Ribera d'Ebre and Menorca remained statistically significant after adjustment for fish intake and other variables (beta (SE) = 0.779 (0.203) for children from RE1). In conclusion, local sources other than seafood contribute significantly to MeHg content in hair in the two Ribera d'Ebre subgroups.

A twin GC x GC system has been designed which enables the analysis of a sample by means of two different and independent column combinations simultaneously. Both combinations are incorporated in the same oven, using the same temperature programme, and are fed using a 50:50 column-entrance-split. It is demonstrated that, employing combinations of a conventional non-polar x polar and a reversed-type polar x non-polar column set, the information content is as high, and the analytical performance is as good as when using two separate GC x GC systems. That is, there is an appreciable gain of time and a reduction of costs without any loss of quality. The general usefulness of performing, and comparing two mutually different GC x GC runs is further illustrated with FAMEs in olive oil, and pollutants in a sediment sample.

Most lipids are a complex mixture of classes of compounds such as fatty acids, fatty alcohols, diols, sterols and hydroxy acids. In this study, the suitability of comprehensive two-dimensional gas chromatography coupled to a time-of-light mass spectrometer is studied for lipid characterization in complex samples. With lanolin, a refined wool wax, as test sample, it is demonstrated that combined methylation plus silylation is the preferred derivatization procedure to achieve (i) high-quality GC x GC separation and (ii) easily recognizable ordered structures in lipid analysis. Optimization of the GC x GC column combination, the influence of the temperature programme on the quality of the separation, and the potential and limitations of automated TOF-MS-based identification are discussed. The combined power of a 2D separation, ordered structures and MS detection is illustrated by the identification of several minor sample constituents.

A procedure for the determination of perfluorocarboxylic acids (i.e. PFC7-10A) in sediment by pressurized fluid extraction (PFE), derivatization, headspace solid-phase microextraction and GC-MS determination in the negative ion chemical ionisation mode was developed. The PFE extraction variables such as solvent composition, number and time per extraction cycle, and extraction temperature were optimised. In the optimum extraction conditions, recoveries exceeding 95% with a limit of detection and RSDs of 0.5-0.8 ng g(-1) and 15.5-16.8%, respectively, were obtained. The developed analytical procedure was applied to harbour sediments where PFC8A and PFC10A were detected for the first time at low ppb concentrations (i.e. 8-11 ng g(-1).

Improvements in the methylmercury extraction from human hair by solid-phase microextraction followed by gas chromatography coupled to cold-vapour atomic fluorescence spectrometry (GC-CVAFS) have been carried out. They consisted in the optimisation of the digestion step prior to the aqueous-phase ethylation and in the GC-CVAFS interface set-up. The main digestion parameters such as acid type, concentration, temperature and time have been optimised for hair sample analysis, thereby avoiding methylmercury degradation. Moreover, the stability of the digested samples was evaluated to improve the sample throughput.

Steryl esters occurring in lanolin have been characterised by sub-ambient pressure gas chromatography coupled to mass spectrometry. Electron impact and chemical ionisation modes with different reagent gases have been evaluated in order to carry out unambiguous peak identification. Steryl esters with different sterol (i.e. cholesterol, lanosterol and dihydrolanosterol) and acid moieties either according to carbon number (i.e C10-C23) or isomeric forms (i.e. normal, iso and anteiso) have been identified. Identification of the sterol and acid moieties has been carried out by means of the mass spectral information obtained in the electron impact, chemical ionisation mode either in the positive or negative modes using methane, isobutane and ammonia as reagent gases. Isomeric identification has been achieved by chromatographic retention parameters (i.e. entire-chain length and fractional-chain length) and by the free fatty acid profile also present in lanolin.

A detailed structural characterisation of the aliphatic high-molecular-mass esters extracted from raw wool based on high-temperature gas chromatography-electron impact ionisation mass spectrometry is described. The raw wool esters extracted are in the range of C37 to C54 (i.e., molecular mass 550-788). The selected ion chromatogram exhibited four isomers for the esters with an odd number of carbon atoms (i:a, i:n, a:n and n:n) and five for those with an even number of carbon atoms (i:i, a:a, i:n, a:n and n:n). Isomeric structural elucidation is discussed with respect to the long-chain fatty acid and long-chain fatty alcohol structures, on the basis of chromatographic retention behaviour and mass spectral information.

A methodology for multi-class pesticide determination at trace level in lanolin is presented. Gel permeation chromatography on a Bio-Beads SX-3 column followed by a dual GC chromatographic determination has been developed. The effluent of the analytical column (50% diphenyl-methyl- or 14% cyanopropyl-phenylpolysiloxane) was split into an electron-capture and a nitrogen-phosphorus detection system. The chromatographic system was optimised for 28 pesticides commonly used to control sheep pests and corresponding to organochlorine, organophosphorus and pyretroid classes. Identification has been carried out by gas chromatography coupled to negative chemical ionization mass spectrometry. Recoveries ranged from 72 to 94% and the detection limits from 20 to 97 ng/g depending on the pesticide class, the RSDs were below 10%. Finally, the developed analytical methodology has been successfully applied to the determination of pesticides in several lanolin samples.

Azido nitrophenylaminoacetyl [125I]iodo derivative of toxin II from Centruroides suffusus suffusus, a beta-toxin, and azido nitrophenylaminoacetyl [125I]iodo derivative of toxin V from Leiurus quinquestriatus quinquestriatus, an alpha-toxin, have been covalently linked after binding to their receptor sites that are related to the voltage sensitive sodium channel present in rat brain synaptosomes. Both derivatives labeled two polypeptides of 253000 +/- 20000 and 35000 +/- 2000 mol. wt. Labeling was blocked for each derivative by a large excess of the corresponding native toxin but no cross inhibition was obtained. These results suggest that both alpha - and beta - scorpion toxin receptors are located on or near the same two membrane polypeptides which may be part of the voltage dependent sodium channel.

We have compared the concentrations of serum lipoprotein apoproteins in 55 ischaemic heart disease (IHD) patients and 116 apparently healthy control subjects by a simple, precise and reasonably sensitive "rocket" electroimmunoassay technique. Apolipoprotein B and apolipoprotein C levels in the vaery low density lipoprotein class and apolipoprotein B in the low density lipoprotein class were significantly lower IHD patients than in control subjects (p < 0.001). Apolipoprotein A levels within high density lipoproteins were markedly lower in a subpopulation of IHD patients compared with age-and sex-matched controls (p < 0.05). Using levels in excess of the 95th percentile of the levels in the healthy control population to delineate "abnormalities", we showed that there were more patients with "abnormalities" when lipoprotein apoprotein measurements were considered than when lipoprotein lipids were considered. The positive correlations between lipoprotein lipids and their apoproteins were in most instances greater in IHD patients than in controls.
