3. A model system for antiviral siRNA therapeutics using exosome-based delivery

Emerging viral diseases, such as Ebola, SARS, MERS, and the pathogen causing COVID-19, SARS-CoV-2, present a challenge for the development of therapeutics because of strict biosafety handling procedures and rapid mutation of their genomes. To facilitate the development of an adaptable and testable therapeutic model system, a colostrum exosome-based nanoparticle delivery system, EPM (exosome-PEI matrix), that overcomes stringent biosafety containment, was used to mimic the expression of viral proteins. This system would greatly expand the number of laboratories actively participating in the screening of potential therapeutics. EPM technology can deliver both plasmid DNA and siRNA to both simulate viral gene expression and screen potential antiviral siRNA therapeutics. Using this nanoplatform, three key SARS-CoV-2 proteins (the spike glycoprotein, nucleocapsid, and replicase) were expressed in vitro and in vivo. In vitro, several viral gene-targeting siRNAs were screened to determine knockdown efficiency, with some siRNA duplexes resulting in 80%–95% knockdown of corresponding protein expression. Moreover, in vivo experiments introducing the spike protein and nucleocapsid by EPM resulted in the production of antibodies against the viral antigen, measured up to 45 d after target delivery. Together, these findings support the efficacy of the EPM delivery system to establish a model for screening antiviral therapeutics-reduced biosafety level.

To alleviate the problem, a new and unique property for the complex is desirable. Novel hexagonal stratified structures have been observed. This category of hybrid material is prepared with surfactant, polyoxometalate and s-triazine complex through ionic interaction. To know the tenacity of the present study for an inventive property was further studied. A novel technique of shock wave impulsed material was developed in this work; employed to fabricate the FO TFN membrane. The experimental results recommended that notable attention should be paid to the 0.1 wt % POM-AG incorporated TFN membrane which exhibits 9.2 LMH higher pure water flux compared to the composite membranes without shock imposed material incorporation that were synthesized. Those water enhancements of FO membranes could be assigned the presence of the POM-AG, which is allocated evenly throughout the PA active layer. Thus, the formation of porous selective layer is minimized water transport resistance. The dielectric and conduction mechanisms were monitored through dielectric anomalies and intramolecular charge transfer of the covalence of Mo-O and NH+, manifested by dipolar relaxation. An unassociated in intensity originated from the differential of the selective orientation during the analyses of the experimental data for the powder samples. Furthermore, the structural, morphological and thermal properties of hock wave impulsed hybrid material have been investigated using XRD, SEM, FT-IR, TGA, and NMR techniques. However, the FO membrane biological activity was reported above 70%.

Macular corneal dystrophy (MCD) an autosomal recessive disorder leading to severe visual impairment is caused by mutations in the carbohydrate sulfotransferase 6 (CHST6) gene. Here, we describe generation of a iPSC line from peripheral blood mononuclear cells (PBMCs) from a 38 year old MCD patient harboring a homozygous mutation, p.V172L in the CHST6 gene. The iPSC line GLNNFi001-A was generated using an integration-free Sendai virus containing (KLF4, c-MYC, OCT4 and SOX2).

Membranous nephropathy (MN) is the most common cause of primary nephrotic syndrome among adults. The identification of phospholipase A2 receptor (PLA2R) as target antigen in most patients changed the management of MN dramatically, and provided a rationale for B-cell depleting agents such as rituximab. The efficacy of rituximab in inducing remission has been investigated in several studies, including 3 randomized controlled trials, in which complete and partial remission of proteinuria was achieved in approximately two-thirds of treated patients. Due to its favorable safety profile, rituximab is now considered a first-line treatment option for MN, especially in patients at moderate and high risk of deterioration in kidney function. However, questions remain about how to best use rituximab, including the optimal dosing regimen, a potential need for maintenance therapy, and assessment of long-term safety and efficacy outcomes. In this review, we provide an overview of the current literature and discuss both strengths and limitations of “the new standard.”

Summary: The etiology of peripartum cardiomyopathy remains unknown. One hypothesis is that an increase in the 16-kDa form of prolactin is pathogenic and suggests that breastfeeding may worsen peripartum cardiomyopathy by increasing prolactin, while bromocriptine, which blocks prolactin release, may be therapeutic. An autoimmune etiology has also been proposed. The authors investigated the impact of breastfeeding on cellular immunity and myocardial recovery for women with peripartum cardiomyopathy in the IPAC (Investigations in Pregnancy Associated Cardiomyopathy) study. Women who breastfed had elevated prolactin, and prolactin levels correlated with elevations in CD8+ T cells. However, despite elevated prolactin and cytotoxic T cell subsets, myocardial recovery was not impaired in breastfeeding women. Key Words: breastfeeding, immune activation, peripartum cardiomyopathy

24. Sustained expression of CYPs and DNA adduct accumulation with continuous exposure to PCB126 and PCB153 through a new delivery method: Polymeric implants

A new delivery method via polymeric implants was used for continuous exposure to PCBs. Female Sprague-Dawley rats received subcutaneous polymeric implants containing PCB126 (0.15% load), PCB153 (5% load), or both, for up to 45 d and release kinetics and tissue distribution were measured. PCB153 tissue levels on day 15 were readily detected in lung, liver, mammary and serum, with highest levels in the mammary tissue. PCB126 was detected only in liver and mammary tissues. However, a completely different pharmacokinetics was observed on co-exposure of PCB153 and PCB126, with a 1.8-fold higher levels of PCB153 in the liver whereas a 1.7-fold lower levels in the mammary tissue. PCB126 and PCB153 caused an increase in expression of key PCB-inducible enzymes, CYP 1A1/2 and 2B1/2, respectively. Serum and liver activities of the antioxidant enzymes, PON1 and PON3, and AhR transcription were also significantly increased by PCB126. 32P-postlabeling for polar and lipophilic DNA-adducts showed significant quantitative differences: PCB126 increased 8-oxodG, an oxidative DNA lesion, in liver and lung tissues. Adduct levels in the liver remained upregulated up to 45 d, while some lung DNA adducts declined. This is the first demonstration that continuous low-dose exposure to PCBs via implants can produce sustained tissue levels leading to the accumulation of DNA-adducts in target tissue and induction of indicator enzymes. Collectively, these data demonstrate that this exposure model is a promising tool for long-term exposure studies.

A series of 16 N1-(3-chloro-4-fluorophenyl)-N4-substituted semicarbazone derivatives were synthesized and subjected to computational pharmacokinetic studies to predict molecular properties. All the title compounds (4a–p) followed the Lipinski “Rule of Five”. The synthesized compounds were characterized by elemental analyses and spectral data and the compounds (4a–p) were evaluated for antimicrobial activities. Among them the compound 2-(4-hydroxybenzylidene)-N-(3-chloro-4-fluorolphenyl)hydrazinecarboxamide (4f) was found to be the most active compound that showed good antibacterial activity while the compound 2-(4-methoxybenzylidene)-N-(3-chloro-4-fluorolphenyl)hydrazinecarboxamide (4g) was moderately active against fungal strains. We have noticed that the compounds, (4f, 4k and 4d) bearing OH and NO2 groups on the phenyl ring at position 4 exhibited good antibacterial activity while compound (4g) bearing OCH3 on the phenyl ring at position 4 exhibited moderate antifungal activity.

Nanomaterials monitor leading agricultural and environmental controlling activities especially due to its minuscule dimension. Additionally, several potential applications including a reduction in the accumulation of agricultural waste and green synthesis of nanoparticle using biological agents allow nanotechnology tools to substantially revolutionize the world. Agrowastes are ecofriendly to synthesize the typical nanostructures with precise chemical composition, morphology and size. The use of agricultural waste not only minimizes the synthesis cost but also reduces the hazardous chemicals from the environment and thus stimulates the green synthesis. Biosynthesis of various nanomaterials from agrowastes and microbes including fungi, viruses, bacteria and microalgae are being progressively explored to accomplish pollution mitigation and resource recycling objectives. A challenging task for the 21st century is the cleaning up environmental contaminants using ecofriendly, economical, adoptable, and sustainable technologies. Organophosphorus (OP) compounds are mostly disseminated as agricultural pesticides and plasticizers which has led to environmental accumulation, water and soil toxicity, and they are involved in many poisoning cases every year around the globe. Therefore OP residues are in top of the list for environmental bioremediation. Thus an effective and safe detoxicant therapy is needed. In the bioremediation method, several cyanobacteria, fungi, bacteria, and their enzymes with the potential to degrade OP compounds use OP as an energy source; they have been isolated and efficaciously used for environmental bioremediation in different parts of globe. Nanotechnology is a rapidly growing field of study due to its efficient mechanisms, cost-effectiveness, and environment-friendly approach in comparison to other existing treatments for the bioremediation of environmental contaminants. It makes use of several nanomaterials for the removal of pollutants from soil and groundwater through adsorption, photocatalysis, and encapsulation mode of actions.

Environment contamination because of heavy metals is not only restricted to the aquatic ecosystem, even soil is also increasingly getting contaminated. In consideration with toxic metals sources for soil contamination; the most significant anthropogenic sources include effluents from industrial activities and sewage from domestic and industrial sources. In recent days several strategies are being opted to control soil contamination include physical, chemical, microbial, and phytoremediation. Heavy metals that contaminate soil include lead, arsenic, mercury, and chromium. To overcome this, recently researchers have developed advanced and efficient nanobubble technology, which means small bubbles with diameter less than 1 μm, that is, in the nanoscale. Several methods have been optimized for the generation of nanobubbles such as cavitation, hydrodynamic, acoustic, sonication, and agitation methods. This nanobubble technology has made great breakthroughs and its small size makes nanobubbles have different physicochemical and fluid dynamic properties than ordinary macrobubbles. The presence of oxygen content in nanobubbles makes them more efficient for use in several applications related to sterilization, oxidation, and to improve the soil quality. Several properties such as the presence of large area, high concentration, and the higher stability with a hydrophobic nature make nanobubbles efficient and strong in the remediation process. Apart from that these nanobubbles can easily adsorb and aggregate the dispersed particles, making the remediation process stable and effective.

Soil organic compounds are one of the major indicators for soil fertility. Decreased content of soil organic compounds usually implies a decline in soil quality. Therefore detection and measurement of soil organic compounds is very important. In recent years, nanosensors have been getting much attention due to their use in many industrial, medical, agricultural and environmental applications. Nanosensors are nanoscale devices that work on the principle of measuring the physical quantity and converting it into detectable signals which further can be analyzed. Nanosensors applications have transformed the conventional agricultural practices in the smart agriculture system that is a more environment-friendly strategy and energy efficient approach for sustainable agriculture. Nanosensors are being synthesized in laboratory conditions with potential to monitor temperature and seasonal changes in the soil system. In the case of detection of pollutants, soil solution treated with nanosensors gives accurate results for the measurement and detection of organic pollutants. Real-time monitoring of soil through nanosensors has reduced the excessive mounts of fertilizers and pesticides being used, thus helping to avoid the product cost and ecological contamination. Development of such nanosensors continues to play a significant part in agricultural advancements. Different types of nanosensors have been developed by employing several fabrication approaches to monitor the soil condition and measurement of organic compounds.

- Abstract
Primary forms of minimal change disease and focal segmental glomerulosclerosis are rare podocytopathies and clinically characterized by nephrotic syndrome. Glucocorticoids are the cornerstone of the initial immunosuppressive treatment in these two entities. Especially among adults with minimal change disease or focal segmental glomerulosclerosis, relapses, steroid dependence or resistance are common and necessitate re-initiation of steroids and other immunosuppressants. Effective steroid-sparing therapies and introduction of less toxic immunosuppressive agents are urgently needed to reduce undesirable side effects, in particular for patients whose disease course is complex. Rituximab, a B cell depleting monoclonal antibody, is increasingly used off-label in these circumstances, despite a low level of evidence for adult patients. Hence, critical questions concerning drug-safety, long-term efficacy and the optimal regimen for rituximab-treatment remain unanswered. Evidence in the form of large, multicenter studies and randomized controlled trials are urgently needed to overcome these limitations.

Highlights • Low and middle-income (LMIC) countries still suffer with pre-eclampsia. • Risk assessment of pre-eclampsia has evolved in high-income countries. • Biobanks can provide a unique opportunity to understand preeclampsia in LMIC. • Databanks can provide a unique opportunity to understand preeclampsia in LMIC., The Millennium Development Goal 5, a project signed in 2000, intended to improve maternal health and reduce maternal mortality by 75% by 2015. Despite all efforts, little progress has been achieved in low and middle-income countries (LMIC) and 99% of all maternal deaths related to pre-eclampsia (PE) still occur in these settings. It is important to determine whether women in LMIC, where PE carries a greater risk than in high-income countries (HIC), have unique risk factors. Some variances may alter the risk, severity and pertinent pathophysiology of PE. We posit based upon this, that women from LMIC may have biomarkers specific to this population. Discovering such specific biomarkers and testing the relevance of biomarkers developed in high-income populations could increase the clinical usefulness of these analyses without increasing cost-effective approaches for prediction of PE. Here we briefly describe our platform to develop the PREPARE – Biobank in tertiary hospitals or basic units for antenatal care from 6 different cities in Brazil. The PREPARE – Biobank has been developed with two arms. The first arm is a cross-sectional study that will collect clinical information and biosamples from more than 1000 women who developed preterm PE. The second arm is a cohort study of 7000 women. It will collect clinical information and longitudinal biosamples from women at three times during pregnancy

Highlights • Polymeric implants successfully achieved continuous exposure to PCBs in rats. • PCB126 resulted in significant oxidative DNA damage (8-oxodG) in rat liver and lung. • PCB126 or in combination with PCB153 induced PON1 and its activity in the liver. • The induction was even greater for PON3 and AhR gene transcription. • Co-treatment reduced mammary PCB153 and increased liver PCB126 and PCB153 levels., A new delivery method via polymeric implants was used for continuous exposure to PCBs. Female Sprague-Dawley rats received subcutaneous polymeric implants containing PCB126 (0.15% load), PCB153 (5% load), or both, for up to 45 d and release kinetics and tissue distribution were measured. PCB153 tissue levels on day 15 were readily detected in lung, liver, mammary and serum, with highest levels in the mammary tissue. PCB126 was detected only in liver and mammary tissues. However, a completely different pharmacokinetics was observed on co-exposure of PCB153 and PCB126, with a 1.8-fold higher levels of PCB153 in the liver whereas a 1.7-fold lower levels in the mammary tissue. PCB126 and PCB153 caused an increase in expression of key PCB-inducible enzymes, CYP 1A1/2 and 2B1/2, respectively. Serum and liver activities of the antioxidant enzymes, PON1 and PON3, and AhR transcription were also significantly increased by PCB126. 32P-postlabeling for polar and lipophilic DNA-adducts showed significant quantitative differences: PCB126 increased 8-oxodG, an oxidative DNA lesion, in liver and lung tissues. Adduct levels in the liver remained upregulated up to 45 d, while some lung DNA adducts declined. This is the first demonstration that continuous low-dose exposure to PCBs via implants can produce sustained tissue levels leading to the accumulation of DNA-adducts in target tissue and induction of indicator enzymes. Collectively, these data demonstrate that this exposure model is a promising tool for long-term exposure studies.
