Environmental pollution poses a major problem now a day. Several dyes, in the form of industrial waste, pollute water body and may cause adverse effects to human health. In this paper ADME and toxicity of fives Phenothiazinium group of dyes Methylene blue (MB), Azure A (AA), Azure B (AB), Azure C (AC) and Toluidine Blue O (TBO) were predicted using Swiss ADME and Protox II tools. Results showed these dyes may herm for living organism due to their carcinogenic, mutagenic and hepatotoxic properties. Removal efficiency of these dyes using okra plant product were determined using spectroscopic, thermodynamic and molecular modeling study. It was revealed that these dyes adsorb on the surface of okra leaf mostly at pH 7.0 and the adsorption isotherms were found to fit in Langmuir and Freundlich isotherm model, while Temkin model fails to do this. Mucilage present in different parts of okra plant plays a significant role on removal of these dyes and is able to remove near about 71-92 % of dyes from water body by itself. As this process did not fit in any of above said adsorption isotherm model, it may be suggested that some other mechanism may happen. Further studies explore that these dyes bound to the hydrophobic pocket of mucilage with binding affinity in the order of 10 5 M -1 and the bindings were exothermic in nature with enthalpy change in the range of - 2.94 to - 4.28 kcal/mole. Molecular docking study validate all the experimental results obtained from spectroscopic and thermodynamic study and enlighten the role of structure of dyes on their binding affinity to mucilage. DNA double-strand breaks (DSBs) are considered one of the most harmful forms of DNA damage. These DSBs are repaired through non-homologous end joining (NHEJ) and homologous recombination (HR) pathways and defects in these processes can lead to genomic instability and promote tumorigenesis. Phosphatase and Tensin homolog (PTEN) are crucial in HR repair. However, its involvement in the NHEJ repair pathway has remained elusive. In this study, we investigate the function of epigenetic regulation of PTEN in the NHEJ repair pathway. Our findings indicate that both the phosphorylation and phosphatase activity of PTEN are required for efficient NHEJ-mediated DSB repair. During the DNA damage response, we observed a reduced expression and chromatin attachment of the key NHEJ proteins, including Ku70/80, DNA-PKcs, XRCC4, and XLF, in PTEN-null cells. This reduction was attributed to the instability of these NHEJ proteins, as confirmed by our protein half-life assay. We have demonstrated that the DNA-PKcs inhibitor, NU7026, suppresses the DNA damage-induced phosphorylation of the C-terminal of PTEN. Thus, our study indicates that PTEN could be a target of DNA-PKcs. Protein-protein docking analysis also shows that PTEN interacts with the C-terminal region of DNA-PKcs. PTEN null cells exhibit compromised DNA-PKcs foci after DNA damage as it is in a hyper-phosphorylated state. Phospho-PTEN assists in recruiting DNA-PKcs on the DNA damage site by maintaining its hypo-phosphorylated state which also depends on its phosphatase activity. Therefore, after DNA damage, crosstalk between PTEN and DNA-PKcs modulates the NHEJ pathway. Thus, during DNA damage, PTEN gets phosphorylated directly or indirectly by DNA-PKcs and attaches to chromatin, resulting in the dephosphorylation of DNA-PKcs and subsequently recruitment of other NHEJ factors on chromatin occurs for efficient execution of the NHEJ pathway. Saint Martin Island (SMI), the only coral island in Bangladesh, is located in the Bay of Bengal and has been identified as a marine protected area (MPA). Littering cigarette butts (CBs) waste in an ecologically sensitive environment can have numerous adverse effects. The purpose of this research is to investigate the abundance and density of CBs in SMI and to assess the pollution status using the Cigarette Butt Pollution Index (CBPI). This study is conducted based on the visual survey method in the three types of land use zones of SMI. During the peak season, the investigation was carried out from 9 a.m. to 5 p.m. in December 2023. A total of 4481 CBs item were counted, and the density ranged from 0.37 to 1.76 m -2 with an average value of 0.99 m -2 across 12 sampling campaigns. The highest density was observed at service zones, and the fishing zones had the lowest density. The calculated CBPI values revealed that 75 % of the sampling stations were in the "severe pollution" while 25 % were classified as "high pollution" status, underscoring the prevalence of hazardous CBs across most areas of SMI. To tackle these issues requires regulatory measures, public awareness initiatives, and community involvement. The dissolution of cellulose in double salt ionic liquids (DSILs) was studied in detail and compared with the dissolution in individual constituent ionic liquids (ILs). The DSILs, [C 4 mim](CH 3 CO 2 ) x Cl 1-x (x is the mole fraction of the single component ILs), were synthesized using acetate and chloride salts of 1-butyl-3-methylimidazolium. These DSILs were then used for the investigation of the solubility of cellulose in the whole mole fraction range. Commercial cellulose (CC) powder, kraft pulp (KP), and prehydrolysis kraft pulp (PHKP) of jute were chosen as cellulose sources. The solubility of cellulose increased with an increasing temperature for [C 4 mim](CH 3 CO 2 ) 0.6 Cl 0.4 and with increasing amount of [C 4 mim]Cl in DSILs. The maximum solubility of CC powder was 32.8 wt% in [C 4 mim](CH 3 CO 2 ) 0.6 Cl 0.4 at 100 °C, while for KP and PHKP, solubilities were 30.1 and 30.5 wt%, respectively under the identical condition. Cellulose could be regenerated from the DSILs using water as an antisolvent. Structure, morphology, and thermal stability of the regenerated cellulosic materials were analyzed. DSILs could be recycled >99 % without a discernible change in structure. Although Bangladesh is known to be burdened with elevated tropospheric ozone levels, little is known about its effects on food security. We conducted field experiments in four highly polluted rice growing environments of Bangladesh in three cropping seasons (2020-2022), in which we grew 20 different rice varieties with or without application of the ozone protectant ethylene diurea (EDU). The average daytime ozone concentrations at the study sites during the rice growing seasons ranged from 53 ppb to 84 ppb, with the lowest concentrations occurring in the year 2020. EDU increased rice grain yields significantly by an average of 10.4 % across all seasons and locations, indicating that plants were stressed under ambient ozone concentrations. EDU was effective in distinguishing ozone-tolerant from ozone-sensitive varieties, in which yield increased by up to 21 %. Likewise, the EDU treatment positively affected vegetation indices representing chlorophyll (NDVI), the chorophyll:carotenoid ratio (Lic2), and pigments of the xanthophyll cycle (PRI). Stomatal conductance was increased significantly by an average of around 10 % among all varieties when plants were treated with EDU. In all physiological traits, significant genotype by treatment interactions occurred, indicating that different varieties varied in their responses to ozone stress. Background: Copper (Cu), an essential trace mineral regulating multiple actions of inflammation and oxidative stress, has been implicated in risk for preterm birth (PTB)., Objectives: This study aimed to determine the association of maternal Cu concentration during pregnancy with PTB risk and gestational duration in a large multicohort study including diverse populations., Methods: Maternal plasma or serum samples of 10,449 singleton live births were obtained from 18 geographically diverse study cohorts. Maternal Cu concentrations were determined using inductively coupled plasma mass spectrometry. The associations of maternal Cu with PTB and gestational duration were analyzed using logistic and linear regressions for each cohort. The estimates were then combined using meta-analysis. Associations between maternal Cu and acute-phase reactants (APRs) and infection status were analyzed in 1239 samples from the Malawi cohort., Results: The maternal prenatal Cu concentration in our study samples followed normal distribution with mean of 1.92 μg/mL and standard deviation of 0.43 μg/mL, and Cu concentrations increased with gestational age up to 20 wk. The random-effect meta-analysis across 18 cohorts revealed that 1 μg/mL increase in maternal Cu concentration was associated with higher risk of PTB with odds ratio of 1.30 (95% confidence interval [CI]: 1.08, 1.57) and shorter gestational duration of 1.64 d (95% CI: 0.56, 2.73). In the Malawi cohort, higher maternal Cu concentration, concentrations of multiple APRs, and infections (malaria and HIV) were correlated and associated with greater risk of PTB and shorter gestational duration., Conclusions: Our study supports robust negative association between maternal Cu and gestational duration and positive association with risk for PTB. Cu concentration was strongly correlated with APRs and infection status suggesting its potential role in inflammation, a pathway implicated in the mechanisms of PTB. In recent years, the obesity epidemic has escalated into a serious public health catastrophe that is only getting worse. However, research into the pathophysiological pathways behind the obesity development and the illnesses that it is associated with is ongoing. In the last decades, it is now clear that the gut microbiota plays a significant role in the genesis and progression of obesity and obesity-related illnesses, particularly changes in its metabolites and composition as obesity progresses. Here, we provide a summary of the processes by which variations in gut metabolite levels and the composition of gut microbiota affect obesity and associated disorders. The bacteria residing in the gut release several chemicals that influence the appetite control, metabolism, and other systems. Since it can either encourage or restrict the deposition of fat in several different ways, the gut microbiota's role in obesity is debatable. Cigarette butts (CBs) have serious harmful effects on the environment and living organisms despite their small size. This research aims to investigate the abundance, densities and pollution status of CBs in Cox's Bazar Sea beach. The survey was conducted between 11 a.m. and 5 p.m. in February 2023, during the peak season of the year. A total of 13,988 CBs were collected, and the mean density was 0.388 m -2 and ranges from 0.195 m -2 - to 0.689 m -2 . The mean CBPI value was 7.78, which showed the "high pollution" status. According to CBPI analysis, 25 %, 41.67 %, 16.67 %, and 16.67 % were "severe pollution", "considerable pollution", "high pollution", and "pollution" status, respectively. From HII values, 91.67 % beach fell within the "Class III" category, indicating abundant toxic CBs in the study area. A total of 13 brands were identified, of which Derby (19.31 %), B&H (18.54 %), and Star (11.26 %) were more abundant brands. This study measured 22 metal and metalloid concentrations in Hypnea musciformis from the Bakkhali River estuary and Saint Martin's Island, Bangladesh and determined their potential impact on consumption. Student t-tests showed a significant variation in metal concentrations between the two sampling sites (p < 0.05). Mean concentrations of Co (2.49 ± 0.05 mg/kg), Fe (793.29 ± 11.76 mg/kg), Mn (368.72 ± 4.87 mg/kg), Pb (3.82 ± 0.02 mg/kg), V (11.23 ± 0.20 mg/kg) and Zn (16.60 ± 0.28 mg/kg) were higher in samples collected from the Bakkhali River estuary compared to Saint Martin's Island, while mean concentrations of Ca (484.18 ± 4.68 mg/kg), Cd (2.44 ± 0.03 mg/kg), Mg (2112.70 ± 17.80 mg/kg), Mo (1.57 ± 0.06 mg/kg), Sr (2377.57 ± 29.98 mg/kg), and Ti (258.27 ± 4.62 mg/kg) were higher in samples collected from Saint Martin's Island. Eight heavy metals (Pb, Cd, Zn, Cu, Ni, Mn, Cr, Fe) were used to assess potential health risks for adults, but no potential health risk was detected (HQ value>1). Otopetrin 1 (OTOP1) is a proton (H + ) channel which detects acidic stimuli in sour taste receptor cells and plays some sort of role in the formation of otoconia in the inner ear. Although it is known that zinc ion (Zn 2+ ) inhibits OTOP1, Zn 2+ requires high concentrations (mM order) to inhibit OTOP1 sufficiently, and no other inhibitors have been found. Therefore, to identify a novel inhibitor, we screened a chemical library (LOPAC 1280 ) by whole-cell patch clamp recordings, measuring proton currents of heterologously-expressed mouse OTOP1. From the screening, we found that reactive blue 2 inhibited OTOP1 currents. Further evaluations of three analogues of reactive blue 2 revealed that cibacron blue 3G-A potently inhibited OTOP1 currents. Cibacron blue 3G-A inhibited OTOP1 currents in a concentration-dependent manner, and its 50% inhibitory concentration (IC50) and the Hill coefficient were 5.0 μM and 1.1, respectively. The inhibition of OTOP1 currents by cibacron blue 3G-A was less affected by extracellular anion compositions, membrane potentials, and low pH than the inhibition by Zn 2+ . These results suggest that the inhibition of OTOP1 by cibacron blue 3G-A is neither likely to be a pore-blocking inhibition nor a competitive inhibition. Regeneration of damaged cornea can save vision for millions of patients. Most of these patients are waiting for transplantation of a donor cornea or suitable substitute to restore vision. Although donor cornea transplantation is the most clinically accepted treatment, shortage of donor cornea results in almost 69 out of every 70 patients untreated with the waiting list for transplantation drastically increasing every year according to a prepandemic estimation. Therefore, corneal replacements are coming up as a cutting-edge alternative strategy. In view of the peptides, especially collagen-like peptides and peptide amphiphiles with bioactive functional motifs demonstrate promising avenue for the corneal tissue engineering and promoting regeneration, by their hierarchical self-assembling propensity to acquire desired nano- to macroscale 3D architecture. Aim: One of the most anticipated adverse effects of warfarin is over-anticoagulation. There is little to no evidence on the treatment that should be administered in patients with an international normalized ratio (INR)≥10. The primary outcome of this study is to analyze the effects of various treatments on 30-day mortality in patients with INR≥10 and without major bleeding on 30-day all-cause mortality. The secondary outcome is to propose a model that predicts 30-day all-cause mortality in the same patient group., Methods: Patients older than 18 years of age using warfarin and who had an INR≥10 were included in this retrospective cohort study. Patients with major bleeding on admission were excluded. Patients treated with only cessation of warfarin were named as "Group-1", patients who were treated with vitamin-K in addition to cessation of warfarin were named as "Group-2", and patients who were treated with cessation of warfarin and vitamin-K and fresh frozen plasma or prothrombin complex concentrate were named as "Group-3"., Results: 190 patients were included in the analysis. Seven (38.9%) patients in the first group, 3 (8.6%) in the second group, and 21 (15.3%) in the third group died within 30-days (p=0.015) . In the post-hoc analysis, the difference between Group-1 and Group-2 was found to be significant( p=0.036, OR:0.147, 95%CI=0.032 to 0.671). The performance of the model in predicting 30-day all-cause mortality was high (AUC=0.818 (95%CI = 0.716 to 0.920) and found to be compatible with the validation dataset 0.806 (95%CI = 0.631 to 0.981). Carmofur is an acid ceramidase inhibitor with superior efficacy in suppressing and killing fatally aggressive glioblastoma cell lines compared to the FDA-approved drug temozolomide. In addition to brain tumors, carmofur also gained attention as a potential lead inhibitor of the main protease (M PRO ) of SARS-CoV-2. It is also reported efficacious against numerous other cancers and non-cancerous diseases including acute lung injury, dementia, Parkinson's disease, childhood ependymoma, and Krabbe disease etc. Carmofur also possesses antifungal and antimicrobial properties. Therefore, a sensitive bio-analytical method is needed in order to support further in vivo pharmacological investigation, pre-clinical and clinical studies. Herein, we report a sensitive, and reliable LC-MS/MS method for quantitative bioanalysis of carmofur using mouse plasma. The samples were prepared employing liquid-liquid extraction (LLE) technique using ethyl acetate and 2-propanol (85:15). Chromatographic separation was achieved on an XBridge BEH C18 XP column (100 mm × 3 mm, 2.5 µm) with a runtime of eight minutes. Quantification was performed in multiple reaction monitoring (MRM) mode with precursor to product ion transition of m/z 256.25 → m/z 129.01 for carmofur and m/z 145.53 → m/z 42.00 for 5-chlorouracil (IS) in negative electrospray ionization. Carmofur showed good linearity over the range of 5-1,000 ng.mL -1 . The method was validated in terms of specificity, linearity, carry-over, matrix effect, recovery efficiency, accuracy, precision, dilution integrity, and stability. Finally, the method was successfully employed in a pharmacokinetic study in mouse plasma after intraperitoneal administration of the drug solution. Introduction: The use of alcohol and or illicit drugs during pregnancy is a complex public health issue. There are many adverse short- and long-term health implications of substance use during pregnancy that can potentially affect the mother-foetal dyad. Although prevention and treatment options - such as counselling, pharmacotherapy, rehabilitation, support and case management and withdrawal management - are available, a range of barriers impedes women's ability to disclose their substance use, which limits access and engagement with available services., Objective: This research explored barriers women encountered in disclosing substance use and accessing substance use treatment in pregnancy., Methods: Participants were recruited from a longitudinal cohort study of people with a history of injecting drug use from metropolitan Melbourne. One-on-one in-depth interviews with 15 participants were conducted using a semi-structured interview guide. To be included in this study, participants must have reported a history of substance use during one or more of their pregnancies., Results: The fear of losing child custody associated with the involvement of the child protection services was one of the main barriers to disclosing substance use during pregnancy and accessing treatment and rehabilitation services. Other barriers including stigma and perceived limited treatment options impacted women in various ways., Conclusion: Pregnancy is an important time for women and offers opportunities for service providers to support women who are using substances. Human Activity Recognition (HAR) plays a significant role in the everyday life of people because of its ability to learn extensive high-level information about human activity from wearable or stationary devices. A substantial amount of research has been conducted on HAR and numerous approaches based on deep learning have been exploited by the research community to classify human activities. The main goal of this review is to summarize recent works based on a wide range of deep neural networks architecture, namely convolutional neural networks (CNNs) for human activity recognition. The reviewed systems are clustered into four categories depending on the use of input devices like multimodal sensing devices, smartphones, radar, and vision devices. This review describes the performances, strengths, weaknesses, and the used hyperparameters of CNN architectures for each reviewed system with an overview of available public data sources. In addition, a discussion of the current challenges to CNN-based HAR systems is presented. Finally, this review is concluded with some potential future directions that would be of great assistance for the researchers who would like to contribute to this field. We conclude that CNN-based approaches are suitable for effective and accurate human activity recognition system applications despite challenges including availability of data regarding composite or group activities, high computational resource requirements, data privacy concerns, and edge computing limitations. Background: The objective is to investigate whether calculating the PPI (Pulse Pressure Index) and the RRI (Renal Resistive Index) using routinely collected Duplex ultrasound waveforms data obtained from the aorta and renal artery correlates and predicts renal function, and determine whether RRI is affected by the presence of a renal artery stenosis., Methods: The records of 965 patients were evaluated. The RRI or pulsatility index of the aorta, renal artery, hilum, cortex, and medulla were measured with concurrent glomerular filtration rate GFR, Cr, PPI, and HR measurements, among which 75 patients had a 24-hour urine measured for CrCl, and 32 patients had aortic pulse pressure index (API) calculated from the central aortic pressure measured with applanation tonometry. The propagation of the pulsatility was evaluated by Analysis of Variance (ANOVA). The correlation coefficient (r) and the linear regression coefficient of determination R-squared (R 2 ) were determined. The effects of a renal artery stenosis were evaluated with a paired t-Test comparing the RRI in 192 patients where only one side had a renal artery stenosis greater than 60%., Results: The pulsatility indexes and RRIs progressively decreases and are statistically distinct by ANOVA from the aorta to the renal cortex (P = 7.26 × 10 -125 ). CrCl correlates with the PPI, cortex RRI and medulla RRI with r equal to -0.34, -0.23 and -0.42 (P < 0.05). GFR correlates with the PPI, cortex RRI and medulla RRI with r equal to -0.15, -0.12, and -0.20 (P < 0.0001). Cr correlates with the PPI, cortex RRI and medulla RRI with r equal to 0.09, 0.12, and 0.14 (P < 0.005). The CrCl, GFR and Cr were not statistically correlated with the HR. On univariate and multivariate analysis, the R 2 predictive value for PPI, cortex RRI and medulla RRI for CrCl, GFR and Cr were all less than 0.2 (P < 0.05). The cortex and medulla RRI were correlated with the API with r = 0.63 (P < 0.001). The R 2 predictive value of the PPI for the cortex and medulla RRI was 0.41 and 0.28 (P < 0.001), respectively. On paired t-Test analysis renal artery stenosis had no effect on the RRI (P = 0.78)., Conclusions: The RRI is calculated based on velocity waveform propagation where pulsatility slowly decreases in a series of elastic vessels. While CrCl, GFR and Cr do correlate with the PPI, cortex RRI and medulla RRI, the R 2 coefficient of determination for these correlations demonstrate that they are poor predictors of renal function. The present study aimed to assess the levels of heavy metals and metalloids present in six seaweeds and their potential impact on consumption. The highest concentration of 11 metals, i.e., Be (0.47 mg/kg), Co (4.34 mg/kg), Cr (23.46 mg/kg), Cu (11.96 mg/kg), Fe (2290.26 mg/kg), Li (11.55 mg/kg), Ni (13.75 mg/kg), Pb (6.67 mg/kg), Ti (736.62 mg/kg), Tl (0.14 mg/kg), and V (33.09 mg/kg) were observed in Enteromorpha intestinalis (green seaweeds). Besides, the highest concentration of Ca (1071.09 mg/kg), Cd (5.81 mg/kg), Mn (1003.41 mg/kg), Sr (2838.86 mg/kg), and Zn (41.95 mg/kg) were found in Padina tetrastromatica (brown seaweeds). Eight metals (Pb, Cd, Zn, Cu, Ni, Mn, Cr, Fe) have been used to assess the potential health risk for adults, but no potential health risk was detected (HQ value > 1). As the COVID-19 pandemic continues to affect human health across the globe rapid, simple, point-of-care (POC) diagnosis of infectious viruses such as SARS-CoV-2 remains challenging. Polymerase chain reaction (PCR)-based diagnosis has risen to meet these demands and despite its high-throughput and accuracy, it has failed to gain traction in the rapid, low-cost, point-of-test settings. In contrast, different emerging isothermal amplification-based detection methods show promise in the rapid point-of-test market. In this comprehensive study of the literature, several promising isothermal amplification methods for the detection of SARS-CoV-2 are critically reviewed that can also be applied to other infectious viruses detection. Starting with a brief discussion on the SARS-CoV-2 structure, its genomic features, and the epidemiology of the current pandemic, this review focuses on different emerging isothermal methods and their advancement. The potential of isothermal amplification combined with the revolutionary CRISPR/Cas system for a more powerful detection tool is also critically reviewed. Additionally, the commercial success of several isothermal methods in the pandemic are highlighted. Different variants of SARS-CoV-2 and their implication on isothermal amplifications are also discussed. Background: Children in resource-limited settings remain vulnerable to zinc deficiency and its consequences., Objectives: To evaluate the effects of different doses, durations, and frequencies of zinc supplementation on the incidence of diarrhea and change in linear growth among young children., Methods: We conducted a randomized, partially double-blind, controlled, 6-arm, community-based efficacy trial in Dhaka, Bangladesh. Children aged 9-11 mo were randomly assigned to receive 1 of the following interventions for 24 wk: 1) standard micronutrient powder (MNP) containing 4.1 mg zinc and 10 mg iron, daily; 2) high-zinc (10 mg), low-iron (6 mg) (HiZn LoFe) MNP, daily; 3) HiZn (10 mg) LoFe (6 mg)/HiZn (10 mg), no-iron MNPs on alternating days; 4) dispersible zinc tablet (10 mg), daily; 5) dispersible zinc tablet (10 mg), daily for 2 wk at enrollment and 12 wk; 6) placebo powder, daily. Primary outcomes were incidence of diarrhea and change in length-for-age z-score (LAZ) over the 24-wk intervention period. Home visits were conducted twice weekly to assess diarrhea and other morbidity. Incidence and prevalence outcomes were compared among groups with Poisson regression; continuous outcomes were compared using ANCOVA., Results: A total of 2886 children were enrolled between February 2018 and July 2019. The mean incidence and prevalence of diarrhea among all participants was 1.21 episodes per 100 d and 3.76 d per 100 d, respectively. There were no differences in the incidence or prevalence of diarrhea across intervention groups. The decline in LAZ was slightly smaller among children in the daily HiZn LoFe MNP group compared with the placebo powder group (P < 0.05)., Conclusions: The dose of zinc in MNPs as well as the duration and frequency of supplementation evaluated in this trial were not effective in reducing diarrhea; however, the daily HiZn LoFe MNP formulation offered modest improvements in linear growth among young children. As an emerging resource, Gram-negative Burkholderia bacteria were able to produce a wide range of bioactive secondary metabolites with potential therapeutic and biotechnological applications. Genome mining has emerged as an influential platform for screening and pinpointing natural product diversity with the increasing number of Burkholderia genome sequences. Here, for genome mining of potential biosynthetic gene clusters (BGCs) and prioritizing prolific producing Burkholderia strains, we investigated the relationship between species evolution and distribution of main BGC groups using computational analysis of complete genome sequences of 248 Burkholderia species publicly available. We uncovered significantly differential distribution patterns of BGCs in the Burkholderia phyla, even among strains that are genetically very similar. We found various types of BGCs in Burkholderia, including some representative and most common BGCs for biosynthesis of encrypted or known terpenes, non-ribosomal peptides (NRPs) and some hybrid BGCs for cryptic products. We also observed that Burkholderia contain a lot of unspecified BGCs, representing high potentials to produce novel compounds. Analysis of BGCs for RiPPs (Ribosomally synthesized and posttranslationally modified peptides) and a texobactin-like BGC as examples showed wide classification and diversity of RiPP BGCs in Burkholderia at species level and metabolite predication. In conclusion, as the biggest investigation in silico by far on BGCs of the particular genus Burkholderia, our data implied a great diversity of natural products in Burkholderia and BGC distributions closely related to phylogenetic variation, and suggested different or concurrent strategies used to identify new drug molecules from these microorganisms will be important for the selection of potential BGCs and prolific producing strains for drug discovery., (Copyright © 2021 Elsevier Ltd. Background: Variability in the 25-hydroxyvitamin D [25(OH)D] response to prenatal and postpartum vitamin D supplementation is an important consideration for establishing vitamin D deficiency prevention regimens., Objectives: We aimed to examine interindividual variation in maternal and infant 25(OH)D following maternal vitamin D supplementation., Methods: In a randomized trial of maternal vitamin D supplementation (Maternal Vitamin D for Infant Growth Trial), healthy pregnant women (n = 1300) received a prenatal cholecalciferol (vitamin D-3) dose of 0, 4200, 16,800, or 28,000 IU/wk from 17 to 24 wk of gestation followed by placebo to 6 mo postpartum. A fifth group received 28,000 IU cholecalciferol/wk both prenatally and postpartum. In a subset of participants, associations of 25(OH)D with hypothesized explanatory factors were estimated in women at delivery (n = 655) and 6 mo postpartum (n = 566), and in their infants at birth (n = 502) and 6 mo of age (n = 215). Base models included initial 25(OH)D and supplemental vitamin D dose. Multivariable models were extended to include other individual characteristics and specimen-related factors. The model coefficient of determination (R2) was used to express the percentage of total variance explained., Results: Supplemental vitamin D intake and initial 25(OH)D accounted for the majority of variance in maternal 25(OH)D at delivery and postpartum (R2 = 70% and 79%, respectively). Additional characteristics, including BMI, contributed negligibly to remaining variance (<5% increase in R2). Variance in neonatal 25(OH)D was explained mostly by maternal delivery 25(OH)D and prenatal vitamin D intake (R2 = 82%). Variance in 25(OH)D in later infancy could only partly be explained by numerous biological, sociodemographic, and laboratory-related characteristics, including feeding practices (R2 = 43%)., Conclusions: Presupplementation 25(OH)D and vitamin D supplemental dose are the major determinants of the response to maternal prenatal vitamin D intake. Like most sectors, the aquaculture and fisheries sector especially in developing countries like Bangladesh is believed to have been severely affected by this unique coronavirus (COVID-19) pandemic. To overcome the adverse impacts of COVID-19, the aquaculture and fisheries economy needs urgent recovery plans which this study focusses on using a mixed-method including online questionnaire surveys, face-to-face and telephonic interviews and focus group discussions during June - November 2020. This study reveals some positive impacts on ecosystem and fish stock (e.g., increase in fish stock) due to less disturbance of fishing activities, but that are not able to bring societal benefits as all the fisheries sub-sectors are affected differently by the pandemic. There are disrupted transportation systems and increased transportation costs (around 20 - 60% higher than normal), more input and maintenance costs and less demand for/and decreased market price of fish. There are also shortages of labourers and reduced patrolling to implement fishery regulations. Cancellation of orders by foreign buyers has seriously affected the shrimp and crab sub-sectors. The fisheries-dependent people's capital assets and activities have been mostly negatively affected resulting in a worsened livelihood. This study has suggested a set of immediate and long-term changes to policy and action plans to recover this sector and sub-sectors from the pandemic considering economic, social and environmental sustainability. Emotion recognition using Artificial Intelligence (AI) is a fundamental prerequisite to improve Human-Computer Interaction (HCI). Recognizing emotion from Electroencephalogram (EEG) has been globally accepted in many applications such as intelligent thinking, decision-making, social communication, feeling detection, affective computing, etc. Nevertheless, due to having too low amplitude variation related to time on EEG signal, the proper recognition of emotion from this signal has become too challenging. Usually, considerable effort is required to identify the proper feature or feature set for an effective feature-based emotion recognition system. To extenuate the manual human effort of feature extraction, we proposed a deep machine-learning-based model with Convolutional Neural Network (CNN). At first, the one-dimensional EEG data were converted to Pearson's Correlation Coefficient (PCC) featured images of channel correlation of EEG sub-bands. Then the images were fed into the CNN model to recognize emotion. Two protocols were conducted, namely, protocol-1 to identify two levels and protocol-2 to recognize three levels of valence and arousal that demonstrate emotion. We investigated that only the upper triangular portion of the PCC featured images reduced the computational complexity and size of memory without hampering the model accuracy. Background: Postpartum depression (PPD) is among the top mental health issues and affects children's health and cognitive development. This study aimed to identify the prevalence of PPD symptomatology and possible determinants among facility delivered mothers of Dhaka city., Method: A cross-sectional study was conducted among 291 mothers within the 12 months of postpartum during January to May 2019., Results: The prevalence of PPD symptomatology was 29.9 % among postpartum mothers. Multiple adjusted odds ratios of stepwise logistic regression analyses revealed, mothers age more than 30 years (AOR = 2.56:95 %CI = 1.21-5.39), being a mother for the first time (AOR = 2.08:95 %CI = 1.09-3.96), lost job or couldn't able to do paid work due to pregnancy (AOR = 2.60:95 %CI = 1.25-5.43), hypertension history (AOR = 2.48:95 %CI = 1.20-5.10), neonatal complication (AOR = 2.04:95 %CI = 1.05-3.95), and rare or no support from husband (AOR = 4.12:95 %CI = 2.14-7.95) were identified as significant predictors of PPD. However, having a household income of more than 50,000 Bangladeshi Taka per month (AOR = 0.36:95 %CI = 0.17-0.76) and passing less sedentary hours (AOR = 0.41:95 % CI = 0.23-0.75) were protective factors for PPD., Conclusion: Our study has revealed a relatively high prevalence of PPD symptomatology; therefore, mental health counseling and proper management of cases are essential for bettering mothers and the next generation., (Copyright © 2021 Elsevier B.V. AGL, a 15-kDa lectin from Amaranthus gangeticus seeds was isolated using ion-exchange and gel filtration chromatography. AGL contained 8.55% of neutral sugar and became specifically inhibited by N-acetyl-D-galactosamine. Hemagglutination activity of the lectin was maximum over the pH range of 4.0-6.0 and temperatures of 30-60 °C though it lost the activity when treated with urea and EDTA. With an LC 50 value of 250 μg/ml, AGL showed mild toxicity against Artemia nauplii. It inhibited the growth of pathogenic bacteria like Shigella boydii, Shigella dysenteriae and Staphylococcus aureus when treated for 8 and 16 h, respectively, but lost the antibacterial activity during a 24 h treatment. AGL could not inhibit the growth of Escherichia coli and mitogenic growth (7.0-9.0%) was observed instead. AGL inhibited 37.14%, 65.71% and 82.85% of biofilm formation of Escherichia coli at the concentrations of 250, 500 and 1000 μg/ml, respectively. Marked inhibition of the proliferation of Ehrlich ascites carcinoma cells was determined when treated with various doses of AGL. AGL inhibited 65.89% and 81.25% of the in vivo growth of EAC cells in mice at the doses of 2.0 and 4.0 mg/kg/day, respectively. Inadequate supplies of donor corneas have evoked an escalating interest in corneal xenotransplantation. However, innate immune responses contribute significantly to the mechanism of xenograft rejection. We hypothesized that complement component C5 and TLR co-receptor CD14 inhibition would inhibit porcine cornea induced innate immune responses. Therefore, we measured cytokine release in human blood, induced by three forms of corneal xenografts with or without inhibitors. Native porcine cornea (NPC) induced interleukins (IL-1β, IL-2, IL-6, IL-8, IL-1ra), chemokines (MCP-1, MIP-1α, MIP-1β) and other cytokines (TNF, G-CSF, INF-γ, FGF-basic). Decellularized (DPC) and gamma-irradiated cornea (g-DPC) elevated the release of those cytokines. C5-blockade by eculizumab inhibited all the cytokines except G-CSF when induced by NPC. However, C5-blockade failed to reduce DPC and g-DPC induced cytokines. Blockade of CD14 inhibited DPC-induced cytokines except for IL-8, MCP-1, MIP-1α, and G-CSF, while it inhibited all of them when induced by g-DPC. Combined blockade of C5 and CD14 inhibited the maximum number of cytokines regardless of the xenograft type. Finally, by using the TLR4 specific inhibitor Eritoran, we showed that TLR4 activation was the basis for the CD14 effect. Thus, blockade of C5, when combined with TLR4 inhibition, may have therapeutic potential in pig-to-human corneal xenotransplantation. STATEMENT OF SIGNIFICANCE: Bio-engineered corneal xenografts are on the verge of becoming a viable alternative to allogenic human-donor-cornea, but the host's innate immune response is still a critical barrier for graft acceptance. By overruling this barrier, limited graft availability would no longer be an issue for treating corneal diseases. We showed that the xenograft induced inflammation is initiated by the complement system and toll-like receptor activation. Intriguingly, the inflammatory response was efficiently blocked by simultaneously targeting bottleneck molecules in the complement system (C5) and the TLR co-receptor CD14 with pharmaceutical inhibitors. Buruli ulcer (BU), caused by Mycobacterium ulcerans , is currently treated with rifampin-streptomycin or rifampin-clarithromycin daily for 8 weeks recommended by World Health Organization (WHO). These options are lengthy with severe side effects. A new anti-tuberculosis drug, TB47, targeting QcrB in cytochrome bc1:aa3 complex is being developed in China. TB47-containing regimens were evaluated in a well-established murine model using an autoluminescent M. ulcerans strain. High-level TB47-resistant spontaneous M. ulcerans mutants were selected and their qcrB genes were sequenced. The in vivo activities of TB47 against both low-level and high-level TB47-resistant mutants were tested in BU murine model. Here, we show that TB47-containing oral 3-drug regimens can completely cure BU in ≤2 weeks for daily use or in ≤3 weeks given twice per week (6 doses in total). All high-level TB47-resistant mutants could only be selected using the low-level mutants which were still sensitive to TB47 in mice. This is the first report of double mutations in QcrB in mycobacteria. In summary, TB47-containing regimens have promise to cure BU highly effectively and prevent the emergence of drug resistance. We conducted an emission measurement campaign as a part of a multiyear cookstove intervention trial in two rural locations in northern and southern India. 253 uncontrolled cooking tests measured emissions in control and intervention households during three ~3-month-long measurement periods in each location. We measured pollutants including fine particulate matter (PM 2.5 ), organic and elemental carbon (OC, EC), black carbon (BC) and carbon monoxide (CO) from stoves ranging from traditional solid fuel (TSF) to improved biomass stoves (rocket, gasifier) to liquefied petroleum gas (LPG) models. TSF stoves showed substantial variability in pollutant emission factors (EFs; g kg -1 wood) and optical properties across measurement periods. Multilinear regression modeling found that measurement period, fuel properties, relative humidity, and cooking duration are significant predictors of TSF EFs. A rocket stove showed moderate reductions relative to TSF. LPG stoves had the lowest pollutant EFs, with mean PM 2.5 and CO EFs (g MJ delivered -1 ) >90% lower than biomass stoves. However, in-home EFs of LPG were substantially higher than lab EFs, likely influenced by non-ideal combustion performance, emissions from food and possible influence from other combustion sources. Background: The situation of coronavirus disease 2019 (COVID-19) pandemic in the Indian subcontinent is worsening. In Bangladesh, rate of new infection has been on the rise despite limited testing facility. Constraint of resources in the health care sector makes the fight against COVID-19 more challenging for a developing country like Bangladesh. Vascular surgeons find themselves in a precarious situation while delivering professional services during this crisis. With the limited number of dedicated vascular surgeons in Bangladesh, it is important to safeguard these professionals without compromising emergency vascular care services in the long term. To this end, we at the National Institute of Cardiovascular Diseases and Hospital, Dhaka, have developed a working guideline for our vascular surgeons to follow during the COVID-19 pandemic. The guideline takes into account high vascular work volume against limited resources in the country., Methods: A total of 307 emergency vascular patients were dealt with in the first 4 COVID-19 months (March through June 2020) according to the working guideline, and the results were compared with the 4 pre-COVID-19 months. Vascular trauma, dialysis access complications, and chronic limb-threatening ischemia formed the main bulk of the patient population. Vascular health care workers were regularly screened for COVID-19 infection., Results: There was a 38% decrease in the number of patients in the COVID-19 period. Treatment outcome in COVID-19 months were comparable with that in the pre-COVID-19 months except that limb loss in the chronic limb-threatening ischemia patients was higher. COVID-19 infection among the vascular health care professionals was low., Conclusions: Vascular surgery practice guidelines customized for the high work volume and limited resources of the National Institute of Cardiovascular Diseases and Hospital, Dhaka were effective in delivering emergency care during COVID-19 pandemic, ensuring safety of the caregivers. Background: Nipah virus (NiV) infection, often fatal in humans, is primarily transmitted in Bangladesh through the consumption of date palm sap contaminated by Pteropus bats. Person-to-person transmission is also common and increases the concern of large outbreaks. This study aimed to characterize the molecular epidemiology, phylogenetic relationship, and the evolution of the nucleocapsid gene (N gene) of NiV., Methods: We conducted molecular detection, genetic characterization, and Bayesian time-scale evolution analyses of NiV using pooled Pteropid bat roost urine samples from an outbreak area in 2012 and archived RNA samples from NiV case patients identified during 2012-2018 in Bangladesh., Results: NiV-RNA was detected in 19% (38/456) of bat roost urine samples and among them; nine N gene sequences were recovered. We also retrieved sequences from 53% (21 out of 39) of archived RNA samples from patients. Phylogenetic analysis revealed that all Bangladeshi strains belonged to NiV-BD genotype and had an evolutionary rate of 4.64 × 10 -4 substitutions/site/year. The analyses suggested that the strains of NiV-BD genotype diverged during 1995 and formed two sublineages., Conclusion: This analysis provides further evidence that the NiV strains of the Malaysian and Bangladesh genotypes diverged recently and continue to evolve. Direct unimolar one-step valeroylation of methyl α-d-mannopyranoside (MDM) furnished mainly 6-O-valeroate. However, similar reaction catalyzed by DMAP resulted 3,6-di-O-valeroate (21%) and 6-O-valeroate (47%) indicating reactivity sequence as 6-OH>3-OH>2-OH,4-OH. To get potential antimicrobial agents, 6-O-valeroate was converted into four 2,3,4-di-O-acyl esters, and 3,6-di-O-valeroate was converted into 2,4-di-O-acetate. Direct tetra-O-valeroylation of MDM gave a mixture of 2,3,4,6-tetra-O-valeroate and 2,3,6-tri-O-valeroate indicating that the C2-OH is more reactive than the equatorial C4-OH. The activity spectra analysis along with in vitro antimicrobial evaluation clearly indicated that these novel MDM esters had better antifungal activities over antibacterial agents. In this connection, molecular docking indicated that these MDM esters acted as competitive inhibitors of sterol 14α-demethylase (CYP51), an essential enzyme for clinical target to cure several infectious diseases. Furthermore, pharmacokinetic studies revealed that these MDM esters may be worth considering as potent candidates for oral and topical administration. Objective: Although a reasonable volume of research has been conducted around health impacts of age at first drink of alcohol on adverse health outcomes, the effects of age at first drink of alcohol on knowledge of low-risk drinking and drinks counting habits were rarely researched. The objective of this study is to examine the associations between age at first full serve of alcohol, knowledge of low-risk drinking and frequency of drinks counting., Study Design: This is a cross-sectional study., Methods: Data of six rounds of the National Drug Strategy Household Survey, conducted during the period 2001-2016, were analysed. Multivariable log-binomial regression models were used to explore the associations., Results: Most people drank the first full serve of alcohol during adolescence, and the age at first full serve of alcohol was consumed increased over time. The levels of knowledge of low-risk drinking and frequency of drinks counting increased with age at first drink of alcohol more steeply during adolescence than in the later period. Participants' age at drinking the first full serve of alcohol was significantly associated with knowledge of low-risk drinking and drinks counting. There was an increasing trend of significant risk ratio between knowledge score and the frequency of drinks counting., Conclusion: It seems conceivable that those who reported drinking the first full serve of alcohol before 16 years of age were indifferent to drinks counting, and they lacked necessary knowledge of standard drink of alcohol or low-risk drinking. Objective: Aboriginal and Torres Strait Islander Community Controlled Health Organisations (ACCHOs) have been identified as having an important role in improving the health and wellbeing of individuals in prison; however, a lack of information exists on how to strengthen this role. This paper explores the experiences of ACCHO staff in primary health care to individuals inside or leaving prison., Methods: Nineteen staff from four ACCHOs were interviewed. ACCHO selection was informed by proximity to prisons, town size and/or Local Government Area offending rates. Thematic analysis of the interviews was undertaken., Results: While most ACCHOs had delivered post-release programs, primary health care delivery to prisoners was limited. Three themes emerged: i) a lack of access to prisoners; ii) limited funding to provide services to prisoners; and iii) the need for a team approach to primary health care delivery., Conclusion: A holistic model of care underpinned by a reliable funding model (including access to certain Medicare items) and consistent access to prisoners could strengthen ACCHOs' role in primary health care delivery to people inside or leaving prison. Implications for public health: ACCHOs have an important role to play in the delivery of primary health care to prisoners. Background: Maternal alcohol or other drug use during pregnancy is associated with a range of adverse health outcomes for mothers and their unborn child. The antenatal period presents an opportunity for health professionals to offer routine screening for alcohol or other drugs, to then provide intervention and referral for treatment and/or specialised support services. However, literature indicates that limited screening practices currently exist in maternity care settings., Aim: To identify barriers to screening pregnant women for alcohol or other drugs in maternity care settings, from the perspectives of healthcare professionals., Methods: A comprehensive literature search was conducted in October 2017 to identify relevant studies. Seven databases that index health and social sciences literature, and google scholar, were searched. Eligible articles were subjected to critical appraisal. Extracted data from the eligible studies were synthesised using narrative synthesis., Findings: Nine studies were eligible for this review. The review identified seven key barriers to screening for alcohol or other drugs in pregnancy, namely competing priorities and time constraint; lack of adequate screening skills and clear protocol; relationship between healthcare providers and pregnant women; healthcare providers' perceptions; under-reporting or none/false disclosure; inconclusive evidence regarding the risk of alcohol or other drug use in pregnancy; and concerns about guilt and anxiety., Conclusions: The narrative review revealed a range of barriers to screening for alcohol or other drugs in pregnancy. To address the shortcomings associated with corneal transplants, substantial efforts have been focused on developing new modalities such as xenotransplantion. Xenogeneic corneas are anatomically and biomechanically similar to the human cornea, yet their applications require prior decellularization to remove the antigenic components to avoid rejection. In the context of bringing decellularized corneas into clinical use, sterilization is a crucial step that determines the success of the transplantation. Well-standardized sterilization methods, such as gamma irradiation (GI), have been applied to decellularized porcine corneas (DPC) to avoid graft-associated infections in human recipients. However, little is known about the effect of GI on decellularized corneal xenografts. Here, we evaluated the radiation effect on the ultrastructure, optical, mechanical and biological properties of DPC. Transmission electron microscopy revealed that gamma irradiated decellularized porcine cornea (G-DPC) preserved its structural integrity. Moreover, the radiation did not reduce the optical properties of the tissue. Neither DPC nor G-DPC led to further activation of complement system compared to native porcine cornea when exposed to plasma. Although, DPC were mechanically comparable to the native tissue, GI increased the mechanical strength, tissue hydrophobicity and resistance to enzymatic degradation. Despite these changes, human corneal epithelial, stromal, endothelial and hybrid neuroblastoma cells grew and differentiated on DPC and G-DPC. Thus, GI may achieve effective tissue sterilization without affecting critical properties that are essential for corneal transplant survival., (Copyright © 2019 Acta Materialia Inc. The detection and discrimination of serum albumins (SAs) has been transforming as a research work of keen interest to the scientists in recent times. This is in the root of foundation of more and more fluorescent probes to selectively identify and distinguish the SAs in the modern era of research. Fluorescence based sensors are preferably on high demand because of high sensitivity of fluorescence spectroscopy. Herein we have synthesized berberine derivatives with substitutions at two different positions (9 and 13) with the purpose of an analytical study to detect and differentiate the SAs. It was found that only the 9-O substituted derivatives showed a dramatic enhancement in their inherently weak fluorescence intensity after the addition of serum albumins (BSA and HSA) indicating the occurrence of heteroatom induced hydrophobic binding interaction. Lower value of detection limit, 6.8 nM and 6.1 nM for BSA and 17.8 nM and 16.3 nM for HSA respectively for the two compounds N1 and N2 and extended range of linearity for both the probes justify the fruitfulness of the research work. Moreover, the two effective 9-O substituted probes response differently in presence of the two SAs by the nature, intensity of the fluorescence spectra and position of wavelength maxima which enable us in deciphering the two essential proteins. All the results reveal how the presence of a heteroatom influences the hydrophobic sensing of the SAs and divulge the utility of the synthesized berberine derivatives in detection and distinction of two SAs successfully in the coming years., (Copyright © 2019 Elsevier B.V. Objectives: Prothionamide, a structural analogue of isoniazid, is used mainly for treating multidrug-resistant tuberculosis (MDR-TB). Both drugs have a common target InhA, so prothionamide can be ineffective against isoniazid-resistant (INH R ) Mycobacterium tuberculosis. We aimed to investigate the prevalence of mutations in katG, ethA, ndh, ethR, mshA, inhA and/or its promoter associated with independent resistance and cross-resistance to INH R and/or prothionamide-resistant (PTO R ) M. tuberculosis isolates., Methods: We sequenced the above genes in 206 M. tuberculosis isolates with susceptibility testing against ten drugs., Results: Of the 173 INH R PTO R isolates, 170 (98.3%) harboured mutations in katG, 111 (64.2%) in ethA, 58 (33.5%) in inhA or its promoter, 5 (2.9%) in ndh, 3 (1.7 %) in ethR and 2 (1.2%) in mshA. Among the 18 INH R PTO S isolates, mutations in katG were found in all of them; one had a mutation in the inhA promoter and another in ndh. Of the five INH S PTO R isolates, four showed mutations in ethA and two in the inhA promoter. Notably, 55 novel non-synonymous mutations were found in them and 20.2% of the PTO R M. tuberculosis isolates harboured no known mutations., Conclusions: This is the first report to investigate cross-resistance between INH R and/or PTO R isolates. Among INH R (94.4% MDR-TB) M. tuberculosis isolates, the high diversity of mutations for independent resistance and cross-resistance with prothionamide highlight the importance of both phenotypic susceptibility and genotypic diagnosis when using it to treat patients with INH R -TB. Background: Environmental enteric dysfunction (EED), a chronic inflammatory disorder of the small bowel, is suspected to impair absorption of micronutrients, including zinc., Objective: The objective of this study was to compare zinc absorption from micronutrient powder (MNP) over a range of zinc doses in young children screened for EED with use of the lactulose:mannitol ratio (L:M)., Methods: Bangladeshi children aged 18-24 mo, grouped according to high and low L:M (≥0.09 and <0.09, respectively), were randomly assigned to MNP with 0, 5, 10, or 15 mg Zn/sachet (10 subjects per dose per L:M group). Over a day, fractional absorption of zinc was measured from an MNP-fortified meal and from unfortified meals with stable isotope tracers; total daily absorbed zinc (TAZ, milligrams per day) was determined as the primary outcome. Secondary outcomes included investigation of relations of TAZ to intake, to physiologic requirement, and to other variables, including biomarkers of systemic and intestinal inflammation, using nonlinear models. TAZ was also compared with published data on child zinc absorption., Results: In 74 subjects who completed the study, zinc absorption did not differ by L:M grouping. Most biomarkers of intestinal inflammation were elevated in both L:M groups. For combined L:M groups, mean ± SD TAZ for each MNP dose (0, 5, 10, and 15 mg/sachet) was 0.57 ± 0.30, 0.68 ± 0.31, 0.90 ± 0.43, and 1.0 ± 0.39 mg/d, respectively (P = 0.002), and exceeded the estimated physiologic requirement only for the 10- and 15-mg MNP doses. Zinc absorption was notably lower at all intake levels compared with published data (P < 0.0001) and was inversely related to serum α-1 acid glycoprotein and to fecal Entamoeba histolytica (P = 0.02)., Conclusion: Results indicate impaired absorption of zinc, which may predispose to zinc deficiency in young children with evidence of enteropathy. These findings suggest that current doses of zinc in MNP may be insufficient to yield zinc-related preventative benefits in similar settings. Objective: This study examined the inequality in cesarean section (CS) utilization and its socio-economic contributors., Study Design: Retrospective two-stage stratified sample design., Methods: Data were extracted from two rounds of the Bangladesh Demographic and Health Survey conducted in 2004 and 2014. Concentration Index of CS utilization was calculated using the wealth quintile. Regression-based decomposition method was applied to assess the socio-economic contributors of inequality in CS utilization., Results: The rate of CS utilization increased from 4.98% in 2004 to 24.21% in 2014. The utilization of CS was highly concentrated among the women of higher socio-economic status (SES) in both rounds of the survey. Results of the decomposition models revealed wealth quintile, higher education, higher number of antenatal visits, and being overweight or obese as the critical factors contributing to the inequalities of CS utilization., Conclusion: Bangladesh is now observing a rapid rise in CS utilization and women with higher SES are the main client group of this life saving procedure. There may have inadequate access for those who are relatively less advantaged, even when CS is necessary. Careful analysis of sub-visible amorphous aggregates, where proteins associate non-covalently in either native or denatured states without forming a specific quaternary structure, may shed insight into the mechanisms of protein aggregation and solubility. Here we report a biophysical and biochemical analysis of our model protein, a bovine pancreatic trypsin inhibitor variant (BPTI-19A), whose oligomerization were controlled by attaching solubility controlling peptide tags (SCP tags) to its C terminus, which are short peptides composed of a single type of amino acid that modulate protein solubility. The dynamic light scattering and static light scattering at 25°C indicated that 11 out of 15 SCP tags merely affected the hydrodynamic radius and light scattering intensity of our reference variants BPTI-19A and BPTI-C2G. On the other hand, hydrophobic SCP tags composed of 5 Ile (C5I) or 5 Leu (C5L) were associated into sub-visible aggregates. Circular dichroism indicated that all tagged BPTI variants had the same secondary structure contents as the reference BPTI-19A at 25°C, suggesting that BPTI-C5I and C5L kept their native structure upon association. Furthermore, the thermal denaturation of all of the BPTI variants was fully reversible and typical of natively folded small globular proteins, as monitored by CD at 222 nm. However, the thermal stability of BPTI-19A tagged with hydrophobic residues decreased with increasing protein concentration and tag's hydrophobicity, and BPTI-C5I and C5L were partially denatured at 37°C. Biochemical stability assessed by limited proteolysis with pepsin correlated with the extent of the variants' aggregation, and the large sub-visible aggregates formed by BPTI-C5I and C5L significantly increased their resistance to pepsin proteolysis. Altogether, these observations indicated that hydrophobic SCP tags led to the reversible association of native-like proteins into sub-visible soluble amorphous aggregates resistant to pepsin digestion., (Copyright © 2017 Elsevier B.V. The development of new acetylcholinesterase inhibitors (AChEIs) and subsequent assay of their inhibition efficiency is considered to be a key step for AD treatment. The fluorescence intensity of thioflavin-T (ThT) bound in the active site of acetylcholinesterase (AChE) quenches substantially in presence of standard AChEI drugs due to the dynamic replacement of the fluorophore from the AChE active site as confirmed from steady state emission as well as time-resolved fluorescence anisotropy measurement and molecular dynamics simulation in conjunction with docking calculation. The parametrized % quenching data for individual system shows excellent correlation with enzyme inhibition activity measured independently by standard Ellman AChE assay method in a high throughput plate reader system. The results are encouraging towards design of a fluorescence intensity based AChE inhibition assay method and may provide a better toolset to rapidly evaluate as well as develop newer AChE-inhibitors for AD treatment., (Copyright © 2017 Elsevier B.V.
