Aim: This study aimed to identify the optimal toothpaste for removing coffee-induced stains while also evaluating its impact on tooth enamel through roughness and abraded depth parameters, providing a comprehensive understanding of their effects. Materials and methods: Three whitening toothpastes and 2 conventional toothpastes were prepared for a simulated brushing procedure on coffee-stained bovine enamel tooth specimens. Using a toothbrushing machine, up to 10,000 brushstrokes were completed, while spectrophotometric readings were taken at designated intervals. A mixed-effects model for statistical analysis determined the effects of toothpaste and brushing on colour change, roughness, and abraded depth. Results: Whitening toothpastes significantly deviated from the control (P < .001, P < .001, and P < .003, respectively), whereas the conventional toothpaste did not exhibit a significant contrast (P < .081). Regarding colour restoration following coffee staining, whitening toothpastes showed higher restoration than conventional toothpastes. Surface roughness and abraded depth parameters increased with accumulated brushing. Conclusions: Sodium hexametaphosphate-containing toothpaste demonstrated the highest efficacy in removing coffee-induced stains and restoring tooth colour. Nevertheless, this stronger whitening effect was associated with increased abrasion. While conventional toothpastes exhibited some whitening effects, the most substantial improvement in lightness was consistently observed with whitening toothpastes. Clinical Relevance: Understanding how whitening toothpaste affects enamel integrity is crucial for refining formulations and advancing dental care. This knowledge lays the groundwork for more effective oral care products and improved whitening procedures, ultimately enhancing the overall quality of dental treatments.

SUMMARY: Purpose: Just culture refers to a culture that encourages members of an organization to exchange important safety information and compensates them when they perform such information exchanges. The establishment of a just culture in hospital organizations might be an important means of enhancing patient safety incident reporting. This study aimed to investigate the impact of just culture on the attitudes and behaviors toward patient safety incident reporting in perioperative nurses. Methods: A nationwide cross-sectional survey was performed using structured questionnaires. The participants were 208 perioperative nurses in tertiary general hospitals in South Korea. Data were collected by self-reported on-line questionnaires, from August to September 2020. Data were analyzed with descriptive statistics, independent t-test, chi-square test, Fisher's exact test, one-way ANOVA, Scheffé test, Pearson's correlation analysis, Spearman rank correlation analysis, hierarchical multiple regression, and hierarchical logistic regression using the SPSS WIN 23.0 program. Results: Hierarchical multiple regression analysis revealed that just culture explained an additional 34.5%p of the attitudes on patient safety incident reporting. Hierarchical logistic regression analysis showed that just culture was a significant predictor of behaviors regarding patient safety incident reporting (odds ratio = 2.25, p = .017). The final regression model accounted for 16.0% of the behaviors regarding patient safety incident reporting. Conclusion: This study empirically shows that just culture impacted the attitudes and behaviors regarding patient safety incident reporting in perioperative nurses. This study provides an evidence about the importance of the just culture in every day nursing practice setting. Personnel and organizational efforts for improving or implementing just culture are required to ensure greater patient safety by enhancing the patient safety incident reporting of perioperative nurses in hospitals.

Edible insects are a promising source of bioactive compounds with diverse health benefits. This study investigated the immunomodulatory properties of polysaccharides isolated from Protaetia brevitarsis seulensis larvae (PBSL). Polysaccharides were fractionated using a molecular weight cutoff membrane, and among these fractions, the higher molecular weight fraction (H-PBSL, 666.84 kDa) demonstrated significant immunomodulatory activity. Methylation and NMR structural analyses revealed H-PBSL to be an acidic heteroglycan with a complex structure, consisting of a backbone of repeating (1→4)-linked α-glucan units highly branched at O-6 positions with T-α-Glcp, T-α-GlcpA, and T-β-Galp. The immunomodulatory effects of H-PBSL were evaluated using RAW264.7 macrophage cells. H-PBSL treatment enhanced phagocytic activity and increased the expression of immune modulators and macrophage activation surface molecules. In addition, H-PBSL activated the NF-κB signaling pathway, evidenced by increased phosphorylation of IκB and NF-κB, and translocation of NF-κB from cytosol to nucleus. Moreover, inhibition of Toll-like receptor 4 (TLR4) reduced H-PBSL-induced nitric oxide synthesis, indicating TLR4′s pivotal role in H-PBSL-mediated macrophage activation. These findings highlight the potential of H-PBSL as a novel immunomodulatory agent derived from an edible insect, with possible applications in functional foods and nutraceuticals.

Due to their worldwide distribution and persistence, mercury (Hg), and nano- and microplastics (NMPs) pose major threats to global ocean ecosystems. Hg and NMPs co-exist in the ocean and can interact with each other. However, information on the toxicity of this interaction to marine biota remains limited. Thus, we investigated the toxicological interaction between HgCl2 (Hg) and NMPs by studying the influence of different sizes of polystyrene beads (0.05-, 0.5-, and 6-μm) on Hg accumulation in the brackish water flea Diaphanosoma celebensis. The Hg adsorption capacity of NPs (0.05-μm) was higher than that of MPs (0.5- and 6-μm). Only the group co-exposed to both Hg and NPs showed increased Hg content in D. celebensis. Multixenobiotic resistance (MXR) activity and transcriptional modulation of transporter genes (ABCBs and ABCCs) were decreased by NMP exposure, particularly by NPs, suggesting MXR disruption by NPs. However, only the activity of multidrug resistance-associated proteins (MRPs; ABCCs) increased with Hg exposure and decreased upon NP+Hg co-exposure, indicating an important role of ABCC in Hg efflux. Furthermore, in vivo toxicity tests showed a synergistic toxic interaction between Hg and NPs on the reproduction of D. celebensis. Our findings suggest that NPs have the potential to enhance the toxicity of Hg, increasing Hg accumulation not only by serving Hg as a carrier but also by disrupting MXR.

Shape-memory polyurethanes (SMPUs) are promising materials that change shape in response to external heat. These polymers have a dual-segment structure: a hard segment for netpoint and a soft segment for molecular switch. Understanding the molecular behavior of each segment and microphase-separated morphology is crucial for comprehending the shape-memory mechanism. This study aimed to understand the shape-memory behavior by observing the phase separation of SMPU using mesoscale models based on dissipative particle dynamics (DPD) simulations. The SMPU copolymer was modeled using 4,4′-diphenylmethane diisocyanate (MDI, hard segment) and poly(ethylene oxide) (PEO, soft segment). By calculating segment solubility and repulsion parameters, we found that the hard-segment domain changes from isolated form to a lamellar and interconnected structure and eventually to a continuous form as its content increases. Combining these insights with shape-memory performance models can enhance our understanding of better SMPU design and contribute significantly to the optimization of smart stimuli-responsive materials.

Objectives: This research delves into the application of texture analysis in spine computed tomography (CT) scans and its correlation with bone mineral density (BMD), as determined by dual-energy X-ray absorptiometry (DXA). It specifically addresses the discordance between the 2 measurements, suggesting that certain spinal-specific factors may contribute to this discrepancy. Methods: The study involved 405 cases from a single institution collected between May 6, 2012 and June 30, 2021. Each case underwent a spinal CT scan and a DXA scan. BMD values at the lumbar region (T12 to S1) and total hip were recorded. Texture features from axial cuts of T12 to S1 vertebrae were extracted using gray-level co-occurrence matrices, and a regression model was constructed to predict the BMD values. Results: The correlation between CT texture analysis results and BMD from DXA was moderate, with a correlation coefficient ranging between 0.4 and 0.5. This discordance was examined in light of factors unique to the spine region, such as abdominal obesity, aortic calcification, and lumbar degenerative changes, which could potentially affect BMD measurements. Conclusions: Emerging from this study is a novel insight into the discordance between spinal CT texture analysis and DXA-derived BMD measurements, highlighting the unique influence of spinal attributes. This revelation calls into question the exclusive reliance on DXA scans for BMD assessment, particularly in scenarios where DXA scanning may not be feasible or accurate.

Background: Different progressions or prognoses of chronic obstructive pulmonary disease (COPD) have been reported according to structural abnormalities based on chest computed tomography (CT). This study aimed to investigate whether different structural abnormalities independently affect annual lung function changes and clinical prognosis in patients with COPD. Methods: This longitudinal multicenter observational study was conducted using the KOCOSS cohort (NCT02800499) database in Korea from January 2012 to December 2019. For COPD patients with chest CT findings at baseline enrolment and longitudinal spirometric data, annual forced expiratory volume in 1 s (FEV1) decline rate (mL/year) and clinical outcomes were compared according to structural abnormalities, including emphysema, bronchiectasis (BE), and tuberculosis-destroyed lung (TDL). We estimated the adjusted annual FEV1 changes using a mixed-effect linear regression model. Results: Among the enrolled 237 patients, 152 showed structural abnormalities. Emphysema, BE, and TDL were observed in 119 (78.3%), 28 (18.4%), and 27 (17.8%) patients, respectively. The annual decline in FEV1 was faster in COPD patients with structural abnormalities than those without (β = −70.6 mL/year, P-value = 0.039). BE/TDL-dominant or emphysema-dominant structural abnormality contributed to an accelerated annual FEV1 decline compared to no structural abnormality (BE/TDL-dominant, β = −103.7 mL/year, P-value = 0.043; emphysema-dominant, β = −84.1 mL/year, P-value = 0.018). Structural abnormalities made no significant differences in acute exacerbation rate and mortality. Conclusion: The lung function decline rate in COPD differed according to structural abnormalities on CT. These findings may suggest that more focus should be placed on earlier intervention or regular follow-up with spirometry in COPD patients with BE or TDL on chest CT.

Background and objective: The ability to inhibit aggregation has been demonstrated with synthetically derived ginsenoside compounds G-Rp (1, 3, and 4) and ginsenosides naturally found in Panax ginseng 20(S)-Rg3, Rg6, F4, and Ro. Among these compounds, Rk3 (G-Rk3) from Panax ginseng needs to be further explored in order to reveal the mechanisms of action during inhibition. Methodology: Our study focused to investigate the action of G-Rk3 on agonist-stimulated human platelet aggregation, inhibition of platelet signaling molecules such as fibrinogen binding with integrin αIIbβ3 using flow cytometry, intracellular calcium mobilization, dense granule secretion, and thromboxane B2 secretion. In addition, we checked the regulation of phosphorylation on PI3K/MAPK pathway, and thrombin-induced clot retraction was also observed in platelets rich plasma. Key Results: G-Rk3 significantly increased amounts of cyclic adenosine monophosphate (cAMP) and led to significant phosphorylation of cAMP-dependent kinase substrates vasodilator-stimulated phosphoprotein (VASP) and inositol 1,4,5-trisphosphate receptor (IP3R). In the presence of G-Rk3, dense tubular system Ca2+ was inhibited, and platelet activity was lowered by inactivating the integrin αIIb/β3 and reducing the binding of fibrinogen. Furthermore, the effect of G-Rk3 extended to the inhibition of MAPK and PI3K/Akt phosphorylation resulting in the reduced secretion of intracellular granules and reduced production of TXA2. Lastly, G-Rk3 inhibited platelet aggregation and thrombus formation via fibrin clot. Conclusions and implications: These results suggest that when dealing with cardiovascular diseases brought upon by faulty aggregation among platelets or through the formation of a thrombus, the G-Rk3 compound can play a role as an effective prophylactic or therapeutic agent.

Summary: Peptidoglycan recognition protein 1 (PGLYRP1) is a pattern-recognition protein that mediates antibacterial actions and innate immune responses. Its expression and role in neuroinflammatory conditions remain unclear. We observed the upregulation of PGLYRP1 in inflamed human and mouse spinal cord and brain, with microglia being the primary cellular source. Experiments using a recombinant PGLYRP1 protein show that PGLYRP1 potentiates reactive gliosis, neuroinflammation, and consequent behavioral changes in multiple animal models of neuroinflammation. Furthermore, shRNA-mediated knockdown of Pglyrp1 gene expression attenuates this inflammatory response. In addition, we identify triggering receptor expressed on myeloid cell-1 (TREM1) as an interaction partner of PGLYRP1 and demonstrate that PGLYRP1 promotes neuroinflammation through the TREM1-Syk-Erk1/2-Stat3 axis in cultured glial cells. Taken together, our results reveal a role for microglial PGLYRP1 as a neuroinflammation mediator. Finally, we propose that PGLYRP1 is a potential biomarker and therapeutic target in various neuroinflammatory diseases.

Chronic stress is a pervasive and complex issue that contributes significantly to various mental and physical health disorders. Using the previously established chronic unpredictable stress (CUS) model, which simulates human stress situations, it has been shown that chronic stress induces major depressive disorder (MDD) and memory deficiency. However, this established model is associated with several drawbacks, such as limited research reproducibility and the inability to sustain stress response. To resolve these issues, we developed a new CUS model (CUS+C) that included exogenous corticosterone exposure to induce continuous stress response. Thereafter, we evaluated the effect of this new model on brain health. Thus, we observed that the use of the CUS+C model decreased body and brain weight gain and induced an uncontrolled coat state as well as depressive-like behavior in adult mice. It also impaired learning memory function and cognitive abilities, reduced adult hippocampal neurogenesis as well as the number of hippocampal astrocytes, and downregulated glial fibrillary acidic protein expression in the brains of adult mice. These findings can promote the utilization and validity of the animal stress model and provide new information for the treatment of chronic stress-induced depressive and memory disorders.

Ultrasonic surface acoustic wave (SAW)-induced acoustic streaming flow (ASF) has been utilized for microfluidic flow control, patterning, and mixing. Most previous research employed cross-type SAW acousto-microfluidic mixers, in which the SAWs propagated perpendicular to the flow direction. In this configuration, the flow mixing was induced predominantly by the horizontal component of the acoustic force, which was usually much smaller than the vertical component, leading to energy inefficiency and limited controllability. Here, we propose a vertical-type ultrasonic SAW acousto-microfluidic mixer to achieve rapid flow mixing with improved efficiency and controllability. We conducted in-depth numerical and experimental investigations of the vertical-type SAW-induced ASF to elucidate the acousto-hydrodynamic phenomenon under varying conditions of total flow rate, acoustic wave amplitude, and fluid viscosity conditions. We conducted computational fluid dynamics simulations for numerical flow visualization and utilized micro-prism-embedded microchannels for experimental flow visualization for the vertical SAW-induced ASF. We found that the SAW-induced vortices served as a hydrodynamic barrier for the co-flow streams for controlled flow mixing in the proposed device. For proof-of-concept application, we performed chemical additive-free rapid red blood cell lysis and achieved rapid cell lysis with high lysis efficiency based on the physical interactions of the suspended cells with the SAW-induced acoustic vortical flows. We believe that the proposed vertical-type ultrasonic SAW-based mixer can be broadly utilized for various microfluidic applications that require rapid, controlled flow mixing.

This article proposes the variable sampling interval (VSI) exponentially weighted moving average (EWMA) t chart for the process mean using auxiliary information (AI) (denoted as the VSI EWMA t-AI chart). The proposed chart allows different sampling intervals to be adopted based on prior process quality. The average time to signal (ATS) and expected average time to signal (EATS) criteria are used to evaluate the performance of the proposed chart. Numerical analyses are conducted and the results show that the proposed VSI EWMA t-AI chart surpasses the EWMA t-AI chart in the detection of all sizes of shifts in the process mean when there are no changes (or estimation error) in the process standard deviation. Additionally, the proposed VSI EWMA t-AI chart is found to be more robust than the VSI EWMA X¯ -AI chart when there are changes in the process standard deviation.

Natural amino acid salt solutions (NAASs) are paving the way for greener carbon capture. This study developed simple and precise methods for the viscosity modeling of NAASs. Two approaches, namely, empirical correlations and artificial intelligence, were assessed using a large databank (16 NAAs, 3 alkaline compounds, 25 NAASs, and 1582 data points). Two general correlations and a global equation were suggested. Benefitting from the input of single reference-point data, the modified global equation yielded the best results with a 2.28% deviation. The other empirical models represented viscosities with less than a 7.20% error. The second approach, employing artificial neural networks (ANNs) with different algorithms, was also proposed. The best ANNs were a single-layer perceptron network with tansig + trainlm functions, a double-layer perceptron network with logsig + tansig + trainlm functions, and a radial basis function network with the maximum neurons. They managed to calculate the viscosities with errors of 2.82%, 1.82%, and 0.47%, respectively.

Coronavirus disease 2019 (COVID-19) not only targets the respiratory system but also triggers a cytokine storm and a series of complications, such as gastrointestinal problems, acute kidney injury, and myocardial ischemia. The use of natural products has been utilized to ease the symptoms of COVID-19, and in some cases, to strengthen the immune system against COVID-19. Natural products are readily available and have been regularly consumed for various health benefits. COVID-19 has been reported to be associated with the risk of thromboembolism and deep vein thrombosis. These thrombotic complications often affects mortality and morbidity. Panax ginseng, which has been widely consumed for its various health benefits has also been reported for its therapeutic effects against cardiovascular disease, thrombosis and platelet aggregation. In this review, we propose that P. ginseng can be consumed as a supplementation against the various associated complications of COVID-19, especially against thrombosis. We utilized the network pharmacology approach to validate the potential therapeutic properties of P. ginseng against COVID-19 mediated thrombosis, the coagulation pathway and platelet aggregation. Additionally, we aimed to investigate the roles of P. ginseng against COVID-19 with the involvement of platelet-leukocyte aggregates in relation to immunity-related responses in COVID-19.

Background/purpose: Cuspal deflections in composite restoration have been investigated with considering wall compliance of human tooth cavity and light-curing protocol. The purpose of this study was to investigate effects of mold wall compliance and radiant emittance of LED light on the wall deflection of simulated aluminum mold cavities restored with a bulk-fill composite. Materials and methods: Sixty aluminum molds simulating a class II mesio-occluso-distal (MOD) cavity (6 W × 8 L × 4 D mm; W, width; L, length; D, Depth) were prepared and allocated to three groups with varying mold wall thicknesses of 1, 2, and 3 mm. The molds were bulk-filled with a bulk-fill composite and photo-cured. Four light-curing protocols were used: three duty ratios/exposure times: 100%/20 s, 50%/40 s, or an increasing mode (0 → 100%)/40 s with a pulse width modulated (PWM) LED curing light and one 20 s exposure time with a commercial LED light. Results: Mean mold wall deflection at 2000 s decreased with increasing mold wall thickness (1, 2, and 3 mm) (p 0.05). Background: This study aimed to investigate whether working long hours was related to infertility among female Korean workers, while taking age into consideration. Methods: We used data from the 2018 National Survey on Fertility and Family Health and Welfare in Korea that is a cross-sectional, nationally representative, and population-based survey. Infertility was defined as women who were not pregnant after regular unprotected intercourse for a year. Working long hours was classified as ≥52 hours, and subgroups as per age were classified on the basis of being younger or older than 40 years of age. Differences in infertility risk between the long working hour group and none were estimated in crude and fully adjusted logistic regression models with age-group stratification. Results: Of 5,909 Korean female workers, the crude and adjusted odds ratios (95% confidence intervals) of infertility for working long hours were 1.295 (0.948–1.737) and 1.303 (0.921–1.809), respectively. In the subgroup of patients below 40 years of age, the crude and adjusted odds ratios (95% confidence interval) were 1.957 (1.216–3.039) and 1.921 (1.144–3.120), whereas those aged 40 years or older had 0.994 (0.647–1.471) and 0.939 (0.560–1.501), respectively. The weighted prevalence of infertility increased as weekly working hours increased only for the younger than 40-year subgroup. Conclusions: Infertility is associated with working long hours, especially in young-aged workers. Thus, the working schedule must be structured to better suit young female workers.

This study compares high-temperature grain growth behavior in copper matrix composites containing nano-carbons in the form of fullerenes, carbon nanotubes (CNTs), and graphene to investigate the effect of volume fraction and shape of nano-carbon reinforcement on grain growth suppression. The grain growth was significantly suppressed when the nano-carbon content was 5 vol%, but the suppression efficiency was reduced when nano-carbon particle loading increased above 20 vol%. Furthermore, CNTs induced better grain growth suppression than fullerenes and graphene. Grain growth was observed both near the nano-carbon interfaces and in the bulk matrix. The poor wetting ability between copper and the nano-carbons accelerates lattice diffusion at the reinforcement interface. Owing to differences in the thermal expansion coefficients and elastic moduli between copper and the nano-carbons, due to the presence of the reinforcement, stress distribution will be inhomogeneous. In composites containing discontinuous reinforcement, the stress is concentrated at the tip of the reinforcement and at its minimum at the center of the reinforcement. Inhomogeneous stress fields are formed in the far away from the reinforcement, which in turn causes the suppression of overall grain growth in the composites.

Numerical finite element method (FEM) calculation results reveal that the effective strain differs between the ductile Zr-based bulk metallic glass and brittle Hf-based bulk metallic glass (BMG) during rolling at room temperature. The current results demonstrated that the deformation mechanism of ductile Zr-based bulk metallic glass can be explained by perceptibility of multiple shear bands formation, however, the deformation mechanism of brittle Hf-based bulk metallic glass is represented by uniformly distribution of effective strain through overall specimen rather than localized into shear band. The variation of stress and the distribution of effective strain change significantly near the surface region of a Hf-based bulk metallic glass plate with increasing number of rolling passes. Under elasto-plastic deformation by cold rolling, the brittle Hf-based BMG has a thickness strain (εt) of −0.012, and the neutral effective strain (εeff) is 0.011 at the thickness direction, respectively. We present experimental confirmation that when the applied effective strain can be adjusted to below 1.1% during elasto-plastic deformation then even the brittle as-cast Hf-based BMG can be deformed up to 44% thickness reduction and 23% width expansion after multi-pass cold rolling without fracture by localization of shear stress.

Removing hazardous chemicals such as chemical warfare agents is crucial for the environment, defense, and public security. Herein, magnesium oxide-decorated carbon nitride photocatalysts (CN/MgO) were fabricated for detoxification of 2-chloroethyl ethyl sulfide (CEES) in solutions under the irradiation of a solar simulator. CN/MgO samples were prepared via thermal exfoliation followed by precipitation. The surface enhancements of as-prepared photocatalyst, such as high specific surface area, rich-nitrogen defects, and oxygen dopants, improved adsorption capacity, reaction active sites, and localized charge distribution. CN/MgO catalyst showed high performance of detoxification of CEES in solutions (99% of 200 parts per million volume (ppmv) hydroxylethyl ethyl sulfide, a hydrolysis product of CEES, within 120 min). In addition, the detoxification in chloroform solution was conducted to elucidate the hydrolysis effect in the solution phase. Density functional theory calculations revealed that a built-in electric field at the heterostructure interface could facilitate spatial separation of photo-excited electron-hole pairs. Scavenger tests proved that superoxide radicals contributed to good photo-oxidation reactivity. The reaction mechanism was suggested using the results with GC-MS, XPS, FTIR, and XRD. The as-prepared photocatalyst could function under ambient conditions without adding any base or acid. The detoxification exhibited good selectivity to transfer CEES to sulfoxide, avoiding over-oxidation to generate toxic sulfone.

Small interfering RNAs (siRNAs) therapeutically induce RNA interference (RNAi) of disease-causing genes, but they also silence hundreds of seed-matched off-targets as behaving similar to microRNAs (miRNAs). miRNAs control the pathophysiology of tumors, wherein their accessible binding sites can be sequenced by Argonaute crosslinking immunoprecipitation (AGO CLIP). Herein, based on AGO CLIP, we develop potent anticancer siRNAs utilizing miRNA-like activity (mi/siRNAs). The mi/siRNAs contain seed sequences (positions 2–7) of tumor-suppressive miRNAs while maintaining perfect sequence complementarity to the AGO-accessible tumor target sites. Initially, host miRNA interactions with human papillomavirus 18 (HPV18) were identified in cervical cancer by AGO CLIP, revealing tumor-suppressive activity of miR-1/206 and miR-218. Based on the AGO-miRNA binding sites, mi/siRNAs were designed to target E6 and E7 (E6/E7) transcript with seed sequences of miR-1/206 (206/E7) and miR-218 (218/E7). Synergistic anticancer activity of 206/E7 and 218/E7 was functionally validated and confirmed via RNA sequencing and in vivo xenograft models (206/E7). Other mi/siRNA sequences were additionally designed for cervical, ovarian, and breast cancer, and available as an online tool (http://ago.korea.ac.kr/misiRNA); some of the mi/siRNAs were validated for their augmented anticancer activity (206/EphA2 and 206/Her2). mi/siRNAs could coordinate miRNA-like activity with robust siRNA function, demonstrating the potential of AGO CLIP analysis for RNAi therapeutics.

This study aims to optimize the proportion and constituents of the mixture used for fabricating 180 MPa ultra-high-strength concrete (UHSC) including coarse aggregate. Various parameters, such as the supplementary cementitious material (SCM) type and amount, water–binder (W/B) ratio, fine and coarse aggregate type and amount, chemical admixture, and mixing process, were considered and their effect on the compressive strength was examined. Based on several experimental tests, the optimized SCM proportions for cement, silica fume (SF), ground-granulated blast furnace slag (BFS), and gypsum are 55%, 20%, 20%, and 5%, respectively. The most appropriate W/B ratio, unit water content, and sand–total aggregate (S/a) ratio are 12.5%, 150 kg/m3, and 0.35, respectively. The use of very fine SF with a specific surface area of 13.7 m2/g and diorite, which is a type of coarse aggregate, improves the compressive strength of UHSC. The optimum amount of superplasticizer ranges between 1.3% and 1.6% and the most suitable slump flow is 750 mm. Finally, for this type of flowable UHSC, a mixing time and speed of 5 min and 40 rpm, respectively, and natural self-compacting without using a tamping bar are recommended. Based on the optimized mix design, UHSC with a compressive strength of 180 MPa can be certainly fabricated. Tritium is an important nuclide that must be monitored for radiation safety management. In this study, HTO was orally administered to rats at the level of 37 kBq (1 μCi) or 370 kBq (10 μCi) to examine tissue distribution and excretion levels. After sacrifice, wet and dry tissue samples were weighed and analyzed for tissue free-water tritium (TFWT) and organically bound tritium (OBT). The mean tissue concentrations of TFWT (OBT) were 30.9 (17.8) and 4.4 (8.1) Bq/g on days 7 and 13 at the 37 kBq level and 30.8 (64.6) Bq/g on day 17 at the 370 kBq level. To assess the cytogenetic damage due to tritium exposure, a cytokinesis-blocked micronucleus (MN) assay was performed in blood samples from rats exposed to HTO for 14 and 21 days after oral administration. Purpose: Hair cortisol is emerging as a reliable biomarker for measuring retrospective stress hormone levels. Given that social connectedness can buffer psychobiological stress reactivity, increasing attention is being paid to the specific types of social networks associated with the stress response. This study investigated the role played by two components of social life, emotional closeness and network size, to probe which aspects of social networks were related to stress measures. Methods: The scalp hair cortisol level was used to assess the cumulative cortisol production in 179 community-dwelling older adults, in the Korean Social Life, Health, and Aging Project (KSHAP). Multivariate regression approach was used to examine the link between the stress measures (cortisol and perceived stress scale) and social relationships (social network size and emotional closeness). Results: Emotional closeness (the average level of what one feels about one’s relationship) was significantly associated with decreased levels of hair cortisol, whereas no such relationship was found with the network size. Conclusions: The current findings underscore the role of emotional support on reducing cumulative cortisol, thus providing potential resilience mechanisms for the psychobiological stress response.

Advanced LIGO and Advanced Virgo are monitoring the sky and collecting gravitational-wave strain data with sufficient sensitivity to detect signals routinely. In this paper we describe the data recorded by these instruments during their first and second observing runs. The main data products are gravitational-wave strain time series sampled at 16384 Hz. The datasets that include this strain measurement can be freely accessed through the Gravitational Wave Open Science Center at http://gw-openscience.org, together with data-quality information essential for the analysis of LIGO and Virgo data, documentation, tutorials, and supporting software.

Chronic graft-versus-host disease (GVHD) is a significant complication of allogeneic hematopoietic stem cell transplantation, affecting 30%–70% of transplant recipients. One of the most challenging manifestations of chronic pulmonary GVHD is bronchiolitis obliterans syndrome (BOS), a rare and difficult-to-diagnose disease associated with a high mortality rate. BOS results in progressive circumferential fibrosis and, ultimately, cicatrization of the small terminal airways, manifesting as new fixed airflow obstruction. Although BOS patients are typically treated with immunosuppressive agents, there is no strong evidence that any specific therapies are effective in improving long-term outcomes. Thus, the mortality rate remains high. Therefore, there is an increasing need for additional therapies, including pulmonary rehabilitation (PR), in patients with BOS. PR is an evidence-based and comprehensive intervention for patients with chronic obstructive lung disease aimed at alleviating respiratory symptoms and optimizing functional capacity. This present case series demonstrates that comprehensive PR may also improve exercise tolerance and dyspnea in patients with BOS.

Summary: Plants respond to cold stress by inducing the expression of transcription factors that regulate downstream genes to confer tolerance to freezing. We screened an Arabidopsis transfer DNA (T-DNA) insertion library and identified a cold-hypersensitive mutant, which we named stch4 (sensitive to chilling 4). STCH4/REIL2 encodes a ribosomal biogenesis factor that is upregulated upon cold stress. Overexpression of STCH4 confers chilling and freezing tolerance in Arabidopsis. The stch4 mutation reduces CBF protein levels and thus delayed the induction of C-repeat-binding factor (CBF) regulon genes. Ribosomal RNA processing is reduced in stch4 mutants, especially under cold stress. STCH4 associates with multiple ribosomal proteins, and these interactions are modulated by cold stress. These results suggest that the ribosome is a regulatory node for cold stress responses and that STCH4 promotes an altered ribosomal composition and functions in low temperatures to facilitate the translation of proteins important for plant growth and survival under cold stress. : Yu et al. show that the ribosomal biogenesis factor STCH4 confers cold stress tolerance in Arabidopsis by maintaining rRNA processing and promoting CBF protein translation. Bisphenol A (BPA) may be a critical risk factor for metabolic disorders including obesity and diabetes. Sulforaphane is known to inhibit adipocyte differentiation and induce adipocyte lipolysis. This study sought to investigate the possible effects of sulforaphane on BPA-induced adipocyte differentiation in 3T3-L1 cells. During the preadipocytes differentiation process, sulforaphane suppressed the expression of early adipogenic factors, PPARγ and C/EBPα, induced by BPA in 3T3-L1 cells. Based on cell cycle analysis, sulforaphane mediated the inhibition of mitotic clonal expansion via cell cycle arrest at the G0/G1 phase. The compound inhibited cell cycle progression by suppressing cell cycle regulators such as cyclin D1 and pRb and by increasing p21 and p27, cell cycle inhibitors. Moreover, sulforaphane activated phosphorylation of AMPK and ACC and decreased that of ERK1/2 and Akt. Sulforaphane exerts inhibitory effects on BPA-induced adipocyte differentiation in 3T3-L1 cells. Sulforaphane could be adipogenic properties for improving environmental pollutants.

Here, we report animal experimental data associated with the article entitled “AKR1B10-inhibitory Selaginella tamariscina extract and amentoflavone decrease the growth of A549 human lung cancer cells in vitro and in vivo“ (Jung et al., 2017) [1]. We tested the synergistic anti-tumor effects of Selaginella tamariscina extract and amentoflavone combined with doxorubicin hydrochloride in a nude mouse xenograft model of A549 human lung cancer cells. In our experiment, Selaginella tamariscina extract and amentoflavone were administered orally; and doxorubicin hydrochloride was injected intraperitoneally. We expect our preliminary data will be helpful to the development of the anticancer agent using Selaginella tamariscina extract or amentoflavone. Summary: Post-transcriptional RNA processing is a core mechanism of gene expression control in cell stress response. The poly(A) tail influences mRNA translation and stability, but it is unclear whether there are global roles of poly(A)-tail lengths in cell stress. To address this, we developed tail-end displacement sequencing (TED-seq) for an efficient transcriptome-wide profiling of poly(A) lengths and applied it to endoplasmic reticulum (ER) stress in human cells. ER stress induced increases in the poly(A) lengths of certain mRNAs, including known ER stress regulators, XBP1, DDIT3, and HSPA5. Importantly, the mRNAs with increased poly(A) lengths are both translationally de-repressed and stabilized. Furthermore, mRNAs in stress-induced RNA granules have shorter poly(A) tails than in the cytoplasm, supporting the view that RNA processing is compartmentalized. In conclusion, TED-seq reveals that poly(A) length is dynamically regulated upon ER stress, with potential consequences for both translation and mRNA turnover. : Woo et al. develop TED-seq, a method providing robust, sensitive, and cost-efficient transcriptome-wide measurements of poly(A) length in human cells. Using TED-seq, they show that mRNA poly(A) lengths are dynamic and associated with the control of both protein synthesis and mRNA stability under cell stress. Echinochrome A (EchA), a marine-derived natural product, has shown promise in treating cardiovascular and inflammatory diseases due to its antioxidant and anti-inflammatory properties. However, its cardiac safety remains underexplored. In this study, we utilized human induced pluripotent stem cell-derived cardiac organoids (hCOs) to validate their ability to model the cardiac safety profile of EchA in a human-relevant system. While EchA's therapeutic effects have been reported, prior studies have not evaluated its cardiotoxicity or arrhythmogenic potential in a high-fidelity 3D human cardiac model. The hCOs, characterized by expression of key cardiac markers (cTnT) and functional ion channels (Cav1.2, Nav1.5, hERG), exhibited structural and electrophysiological properties reflective of human cardiac physiology. Using multi-electrode array (MEA) analysis, we assessed the effects of EchA at concentrations ranging from 0.1 to 30 µM on electrophysiological parameters, including beat period, field potential amplitude, field potential duration, and spike slope. EchA treatment induced no significant changes in these parameters, confirming its non-toxic electrophysiological profile. Cellular viability and lactate dehydrogenase (LDH) assays revealed no cytotoxic effects of EchA across tested concentrations. Contractility assays further demonstrated that EchA did not affect contraction velocity, relaxation velocity, or time to 50 % maximal contraction and relaxation. Background & Aims: The risk of hepatocellular carcinoma (HCC) and hepatic decompensation persists after hepatitis B surface antigen (HBsAg) seroclearance. This study aimed to develop and validate a machine learning model to predict the risk of liver-related outcomes (LROs) following HBsAg seroclearance., Methods: A total of 4,787 consecutive patients who achieved HBsAg seroclearance between 2000 and 2022 were enrolled from six centers in South Korea and a territory-wide database in Hong Kong, comprising the training (n = 944), internal validation (n = 1,102), and external validation (n = 2,741) cohorts. Three machine learning-based models were developed and compared in each cohort. The primary outcome was the development of any LRO, including HCC, decompensation, and liver-related death., Results: During a median follow-up of 55.2 (IQR 30.1-92.3) months, 123 LROs were confirmed (1.1%/person-year) in the Korean cohort. The model with the best predictive performance in the training cohort was selected as the final model (designated as PLAN-B-CURE), which was constructed using a gradient boosting algorithm and seven variables (age, sex, diabetes, alcohol consumption, cirrhosis, albumin, and platelet count). Compared to previous HCC prediction models, PLAN-B-CURE showed significantly superior accuracy in the training cohort (c-index: 0.82 vs. 0.63-0.70, all p <0.001; area under the receiver-operating characteristic curve: 0.86 vs. 0.62-0.72, all p <0.01; area under the precision-recall curve: 0.53 vs. 0.13-0.29, all p <0.01). PLAN-B-CURE showed a reliable calibration function (Hosmer-Lemeshow test p >0.05) and these results were reproduced in the internal and external validation cohorts., Conclusion: This novel machine learning model consisting of seven variables provides reliable risk prediction of LROs after HBsAg seroclearance that can be used for personalized surveillance., Impact and Implications: Using large-scale multinational data, we developed a machine learning model to predict the risk of liver-related outcomes (i.e., hepatocellular carcinoma, decompensation, and liver-related death) after the functional cure of chronic hepatitis B (CHB). The new model named PLAN-B-CURE was constructed using seven variables (age, sex, alcohol consumption, diabetes, cirrhosis, serum albumin, and platelet count) and a gradient boosting machine algorithm, and it demonstrated significantly better predictive accuracy than previous models in both the training and validation cohorts. The inclusion of diabetes and significant alcohol intake as model inputs suggests the importance of metabolic risk factor management after the functional cure of CHB. Using seven readily available clinical factors, PLAN-B-CURE, the first machine learning-based model for risk prediction after the functional cure of CHB, may serve as a basis for individualized risk stratification., (Copyright © 2024 European Association for the Study of the Liver.
